Deferoxamine promotes recovery of traumatic spinal cord injury by inhibiting ferroptosis显示文摘Ferroptosis is an iron-dependent novel cell death pathway. Deferoxamine, a ferroptosis inhibitor, has been reported to promote spinal cord injury repair. It has yet to be clarified whether ferroptosis inhibition represents the mechanism of action of Deferoxamine on spinal cord injury recovery. A rat model of Deferoxamine at thoracic 10 segment was established using a modified Allen's method. Ninety 8-week-old female Wistar rats were used. Rats in the Deferoxamine group were intraperitoneally injected with 100 mg/kg Deferoxamine 30 minutes before injury. Simultaneously, the Sham and Deferoxamine groups served as controls. Drug administration was conducted for 7 consecutive days. The results were as follows:(1) Electron microscopy revealed shrunken mitochondria in the spinal cord injury group.(2) The Basso, Beattie and Bresnahan locomotor rating score showed that recovery of the hindlimb was remarkably better in the Deferoxamine group than in the spinal cord injury group.(3) The iron concentration was lower in the Deferoxamine group than in the spinal cord injury group after injury.(4) Western blot assay revealed that, compared with the spinal cord injury group, GPX4, xCT, and glutathione expression was markedly increased in the Deferoxamine group.(5) Real-time polymerase chain reaction revealed that, compared with the Deferoxamine group, mRNA levels of ferroptosis-related genes Acyl-CoA synthetase family member 2(ACSF2) and iron-responsive element-binding protein 2(IREB2) were up-regulated in the Deferoxamine group.(6) Deferoxamine increased survival of neurons and inhibited gliosis. These findings confirm that Deferoxamine can repair spinal cord injury by inhibiting ferroptosis. Targeting ferroptosis is therefore a promising therapeutic approach for spinal cord injury. | Xue Yao Yan Zhang Jian Hao Hui-Quan Duan Chen-Xi Zhao Chao Sun Bo Li Bao-You Fan Xu Wang Wen-Xiang Li Xuan-Hao Fu Yong Hu Chang Liu Xiao-Hong Kong Shi-Qing Feng | 2019 | Neural Regeneration Research2019,14,3: | 35
Achieving over 16% efficiency for single-junction organic solar cells显示文摘To achieve high photovoltaic performance of bulk hetero-junction organic solar cells(OSCs), a range of critical factors including absorption profiles, energy level alignment, charge carrier mobility and miscibility of donor and acceptor materials should be carefully considered. For electron-donating materials, the deep highest occupied molecular orbital(HOMO) energy level that is beneficial for high open-circuit voltage is much appreciated. However, a new issue in charge transfer emerges when matching such a donor with an acceptor that has a shallower HOMO energy level. More to this point, the chemical strategies used to enhance the absorption coefficient of acceptors may lead to increased molecular crystallinity, and thus result in less controllable phase-separation of photoactive layer. Therefore, to realize balanced photovoltaic parameters, the donor-acceptor combinations should simultaneously address the absorption spectra, energy levels, and film morphologies. Here, we selected two non-fullerene acceptors, namely BTPT-4F and BTPTT-4F, to match with a wide-bandgap polymer donor P2F-EHp consisting of an imidefunctionalized benzotriazole moiety, as these materials presented complementary absorption and well-matched energy levels. By delicately optimizing the blend film morphology, we demonstrated an unprecedented power conversion efficiency of over 16% for the device based on P2F-EHp:BTPTT-4F, suggesting the great promise of materials matching toward high-performance OSCs. | Baobing Fan Difei Zhang Meijing Li Wenkai Zhong Zhaomiyi Zeng Lei Ying Fei Huang Yong Cao | 2019 | Science China Chemistry2019,62,6: | 38

Acupuncture's Role in Solving the Opioid Epidemic: Evidence, Cost-Effectiveness, and Care Availability for Acupuncture as a Primary, Non-Pharmacologic Method for Pain Relief and Management-White Paper 2017显示文摘The United States(U.S.) is facing a national opioid epidemic, and medical systems are in need of nonpharmacologic strategies that can be employed to decrease the public's opioid dependence. Acupuncture has emerged as a powerful, evidence-based, safe, cost-effective, and available treatment modality suitable to meeting this need. Acupuncture has been shown to be effective for the management of numerous types of pain conditions, and mechanisms of action for acupuncture have been described and are understandable from biomedical, physiologic perspectives. Further, acupuncture's cost-effectiveness can dramatically decrease health care expenditures, both from the standpoint of treating acute pain and through avoiding addiction to opioids that requires costly care, destroys quality of life, and can lead to fatal overdose. Numerous federal regulatory agencies have advised or mandated that healthcare systems and providers offer non-pharmacologic treatment options for pain. Acupuncture stands out as the most evidence-based, immediately available choice to fulfil these calls. Acupuncture can safely, easily, and cost-effectively be incorporated into hospital settings as diverse as the emergency department, labor and delivery suites, andneonatal intensive care units to treat a variety of commonly seen pain conditions. Acupuncture is already being successful y and meaningfully utilized by the Veterans Administration and various branches of the U.S. Military, in some studies demonstrably decreasing the volume of opioids prescribed when included in care. | arthur yin fan david w.miller bonnie bolash matthew bauer john mcdonald sarah faggert hongjian he yong ming li amy matecki lindy camardella mel hopper koppelman jennifer a.m.stone lindsay meade john pang | 2017 | Journal of Integrative Medicine2017,15,6: | 29
Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further, there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients. Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer. Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks. The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response, time to progression, disease control rate, and safety. Results:Of 179 patients screened,125 were eligible and received RC48 treatment. The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively. The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety, suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29
Orbit determination of Chang'E-3 and positioning of the lander and the rover显示文摘Chang’E-3 landed on the east of Sinus Iridum area on December 14,2013,performing China’s first successful soft landing on the lunar surface. We present the results on precision orbit determination and positioning of the lander and the rover. We describe the data, modeling, and methods used to achieve position knowledge over the period December 2–21,2014.In addition to the radiometric X-band range and Doppler tracking data, delta differential one-way ranging data are also used in the calculation, which show that they strongly improve the accuracy of the orbit reconstruction.Total position overlap differences are about 20 and 30 m for the 100 km 9 100 km and100 km9 15 km lunar orbit, respectively, increased by*50%with respect to CE-2 and at the same level as other lunar spacecrafts of recent era such as SELENE and lunar reconnaissance orbiter(LRO).The position error of the soft landing trajectory is less than 100 m.A kinematic statistical method is applied to determine the position of the lander and relative position of the rover with respect to the lander. The position difference of the lander is better than50 m compared to LRO photograph result.Compared with the delta very long baseline interferometry(VLBI)group delay between the lander and the rover, the delta VLBI phase delay can improve the relative position of the rover from*1,000 to*1 m. | Yong Huang Shengqi Chang Peijia Li Xiaogong Hu Guangli Wang Qinghui Liu Weimin Zheng Min Fan | 2014 | Chinese Science Bulletin2014,59,29: | 28
Analyzing the Flavor Compounds in Chinese Traditional Fermented Shrimp Pastes by HS-SPME-GC/MS and Electronic Nose显示文摘Shrimp paste is a type of condiments with high nutritional value. However, the flavors of shrimp paste, particularly the non-uniformity flavors, have limited its application in food processing. In order to identify the characteristic flavor compounds in Chinese traditional shrimp pastes, five kinds of typical commercial products were evaluated in this study. The differences in the volatile composition of the five products were investigated.Solid phase micro-extraction method was employed to extract the volatile compounds.GC-MS and electronic nose were applied to identify the compounds, and the data were analyzed using principal component analysis(PCA).A total of 62 volatile compounds were identified, including 8 alcohols,7 aldehydes,3 ketones,7 ethers,7 acids,3 esters,6 hydrocarbons,12 pyrazines,2 phenols, and 7 other compounds. The typical volatile compounds contributing to the flavor of shrimp paste were found as follows:dimethyl disulfide, dimethyl tetrasulfide, dimethyl trisulfide,2,3,5-trimethyl-6-ethyl pyrazine,ethyl-2,5-dimethyl-pyrazine,phenol and indole.Propanoic acid, butanoic acid, furans, and 2-hydroxy-3-pentanone caused unpleasant odors, such as pungent and rancid odors.Principal component analysis showed that the content of volatile compounds varied depending on the processing conditions and shrimp species. These results indicated that the combinations of multiple analysis and identification methods could make up the limitations of a single method, enhance the accuracy of identification, and provide useful information for sensory research and product development. | FAN Yan YIN Li-ang XUE Yong LI Zhaojie HOU Hu XUE Changhu | 2017 | Journal of Ocean University of China2017,16,2: | 25

Plant Prime Editors Enable Precise Gene Editing in Rice Cells显示文摘Genome editing is revolutionizing plant research and crop breeding.Sequence-specific nucleases(SSNs)such as zinc finger nuclease(ZFN)and TAL effector nuclease(TALEN)have been used to create site-specific DNA double-strand breaks and to achieve precise DNA modifications by promoting homology-directed repair(HDR)(Steinert et al.,2016;Voytas,2013).Later,RNA-guided SSNs such as CRISPR-Cas9,Cas12a,Cas12b,and their variants were applied for genome editing in plants(Li et al.,2013;Nekrasov et alM 2013;Tang et al.,2017;Zhong et al.,2019;Ming et al.,2020;Tang et al.,2019).However,HDR relies on simultaneous delivery of SSNs and DNA donors, which has been challenging in plants(Steinert et al.,2016;Zhang et aL,2019).Another challenge for realizing efficient HDR in plants is that DNA repair favors nonhomologous end joining(NHEJ)pathways over HDR in most cell types(Puchta,2005;Qi et al.,2013). | Xu Tang Simon Sretenovic Qiurong Ren Xinyu Jia Mengke Li Tingting Fan Desuo Yin Shuyue Xiang Yachong Guo Li Liu Xuelian Zheng Yiping Qi Yong Zhang | 2020 | Molecular Plant2020,13,5: | 20
Effect of Marine Collagen Peptides on Markers of Metabolic Nuclear Receptors in Type 2 Diabetic Patients with/without Hypertension显示文摘Objective To explore Effects of marine collagen peptides (MCPs) on markers of metablic nuclear receptors, i.e peroxisome proliferator-activated receptor (PPARs), liver X receptor (LXRs) and farnesoid X receptor (FXRs) in type 2 diabetic patients with/without hypertension. Method Study population consisted of 200 type 2 diabetic patients with/without hypertension and 50 healthy subjects, all of whom were randomly assigned to MCPs-treated diabetics (n=50), placebo-treated diabetics (n=50), MCPs-treated diabetics with hypertension (n=50), placebo-treated diabetics with hypertension (n=50), and healthy controls (n=50). MCPs or placebo (water-soluble starch) were given daily before breakfast and bedtime over three months. Levels of free fatty acid, cytochrome P450, leptin, resistin, adiponectin, bradykinin, NO, and Prostacyclin were determined before intervention, and 1.5 months, and 3 months after intervention. Hypoglycemia and the endpoint events during the study were recorded and compared among the study groups. Result At the end of the study period, MCPs-treated patients showed marked improvement compared with patients receiving placebo. The protection exerted by MCPs seemed more profound in diabetics than in diabetics with hypertension. In particular, after MCPs intervention, levels of free fatty acid, hs-CRP, resistin, Prostacyclin decreased significantly in diabetics and tended to decrease in diabetic and hypertensive patients whereas levels of cytochrome P450, leptin, NO tended to decrease in diabetics with/without hypertension. Meanwhile, levels of adiponectin and bradykinin rose markedly in diabetics following MCPs administration. Conclusion MCPs could offer protection against diabetes and hypertension by affecting levels of molecules involved in diabetic and hypertensive pathogenesis. Regulation on metabolic nuclear receptors by MCPs may be the possible underlying mechanism for its observed effects in the study. Further study into its action may shed light on development of new drugs based on bioactive peptides from marine sources. | CuI-FENG ZHU GUAN-ZHI LI HONG-BIN PENG FAN ZHANG YUN CHEN YONG LI | 2010 | Biomedical and Environmental Sciences2010,23,2: | 18
