| 2 | Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual. | Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang | 2020 | Protein & Cell2020,11,4: | 44 |

| 5 | Landslide susceptibility zonation method based on C5.0 decision tree and K-means cluster algorithms to improve the efficiency of risk management显示文摘Machine learning algorithms are an important measure with which to perform landslide susceptibility assessments, but most studies use GIS-based classification methods to conduct susceptibility zonation.This study presents a machine learning approach based on the C5.0 decision tree(DT) model and the K-means cluster algorithm to produce a regional landslide susceptibility map. Yanchang County, a typical landslide-prone area located in northwestern China, was taken as the area of interest to introduce the proposed application procedure. A landslide inventory containing 82 landslides was prepared and subsequently randomly partitioned into two subsets: training data(70% landslide pixels) and validation data(30% landslide pixels). Fourteen landslide influencing factors were considered in the input dataset and were used to calculate the landslide occurrence probability based on the C5.0 decision tree model.Susceptibility zonation was implemented according to the cut-off values calculated by the K-means cluster algorithm. The validation results of the model performance analysis showed that the AUC(area under the receiver operating characteristic(ROC) curve) of the proposed model was the highest, reaching 0.88,compared with traditional models(support vector machine(SVM) = 0.85, Bayesian network(BN) = 0.81,frequency ratio(FR) = 0.75, weight of evidence(WOE) = 0.76). The landslide frequency ratio and frequency density of the high susceptibility zones were 6.76/km^(2) and 0.88/km^(2), respectively, which were much higher than those of the low susceptibility zones. The top 20% interval of landslide occurrence probability contained 89% of the historical landslides but only accounted for 10.3% of the total area.Our results indicate that the distribution of high susceptibility zones was more focused without containing more ' stable' pixels. | 8 | Periodontitis may induce gut microbiota dysbiosis via salivary microbiota显示文摘The aim of this study was to identify whether periodontitis induces gut microbiota dysbiosis via invasion by salivary microbes.First,faecal and salivary samples were collected from periodontally healthy participants(PH group,n=16)and patients with severe periodontitis(SP group,n=21)and analysed by 16S ribosomal RNA sequencing.Significant differences were observed in both the faecal and salivary microbiota between the PH and SP groups.Notably,more saliva-sourced microbes were observed in the faecal samples of the SP group.Then,the remaining salivary microbes were transplanted into C57BL6/J mice(the C-PH group and the C-SP group),and it was found that the composition of the gut microbiota of the C-SP group was significantly different from that of the C-PH group,with Porphyromonadaceae and Fusobacterium being significantly enriched in the C-SP group.In the colon,the C-SP group showed significantly reduced crypt depth and zonula occludens-1 expression.The m RNA expression levels of pro-inflammatory cytokines,chemokines and tight junction proteins were significantly higher in the C-SP group.To further investigate whether salivary bacteria could persist in the intestine,the salivary microbiota was stained with carboxyfluorescein diacetate succinimidyl ester and transplanted into mice.We found that salivary microbes from both the PH group and the SP group could persist in the gut for at least 24 h.Thus,our data demonstrate that periodontitis may induce gut microbiota dysbiosis through the influx of salivary microbes. | Jun Bao Lili Li Yangheng Zhang Min Wang Faming Chen Shaohua Ge Bin Chen Fuhua Yan | 2022 | International Journal of Oral Science2022,14,3: | 9 |
| 9 | Coastal blue carbon: Concept, study method, and the application to ecological restoration显示文摘Coastal blue carbon refers to the carbon taken from atmospheric CO2; fixed by advanced plants(including salt marsh,mangrove, and seagrass), phytoplankton, macroalgae, and marine calcifiers via the interaction of plants and microbes; and stored in nearshore sediments and soils; as well as the carbon transported from the coast to the ocean and ocean floor. The carbon sequestration capacity per unit area of coastal blue carbon is far greater than that of the terrestrial carbon pool. The mechanisms and controls of the carbon sink from salt marshes, mangroves, seagrasses, the aquaculture of shellfish and macroalgae, and the microbial carbon pump need to be further studied. The methods to quantify coastal blue carbon include carbon flux measurements, carbon pool measurements, manipulative experiments, and modeling. Restoring, conserving, and enhancing blue carbon will increase carbon sinks and produce carbon credits, which could be traded on the carbon market. The need to tackle climate change and implement China's commitment to cut carbon emissions requires us to improve studies on coastal blue carbon science and policy. | 12 | A novel CCCH-type zinc finger protein SAW1 activates OsGA20ox3 to regulate gibberellin homeostasis and anther development in rice显示文摘Male sterility is a prerequisite for hybrid seed production.The phytohormone gibberellin(GA)is in-volved in regulating male reproductive development,but the mechanism underlying GA homeostasis in anther development remains less understood.Here,we report the isolation and characterization of a new positive regulator of GA homeostasis,swollen anther wall 1(SAW1),for anther development in rice(Oryza sativa L.).Rice plants carrying the recessive mutant allele saw1 produces abnormal anthers with swollen anther wall and aborted pollen.Clustered regularly interspaced short palindromic repeats(CRISPR)/CRIPSR-associated protein 9-mediated knockout of SAW1 in rice generated similar male sterile plants.SAW1 encodes a novel nucleus-localizing CCCH-tandem zinc finger protein,and this protein could directly bind to the promoter region of the GA synthesis gene OsGA20ox3 to induce its anther-specific expression.In the saw1 anther,the significantly decreased OsGA20ox3 expression resulted in lower bio-active GA content,which in turn caused the lower expression of the GA-inducible anther-regulator gene OsGAMYB.Thus,our results disclose the mechanism of the SAW1-GA20ox3-GAMYB pathway in controlling rice anther development,and provide a new target gene for the rapid generation of male sterile lines by genome editing for hybrid breeding. | Bin Wang Ruiqiu Fang Faming Chen Jingluan Han Yao-Guang Liu Letian Chen Qinlong Zhu | 2020 | Journal of Integrative Plant Biology2020,62,10: | 5 |
| 13 | Synthesis and Characterizations of Nanocrystalline WC-Co Composite Powders by a Unique Ball Milling Process显示文摘In order to explore the high efficiency of fabricating nanocrystalline WC-Co composite powders, this paper presented a unique high energy ball milling process with variable rotation rate and repeatious circulation, by which nanocrystalline WC-10Co0.8VC-0.2Cr3C2 (wt pct) composite powders with mean grain size of 25 nm were prepared in 32 min, and the quantity of the powders for a batch was as much as 800 grams. The as-prepared powders were analyzed and characterized by chemical analysis,X-ray diffraction (XRD), transmission electron microscopy (TEM) and differential thermal analysis (DTA). The results show that high energy ball milling with variable rotation rates and repeatious circulation could be used to produce nanocrystalline WC-Co powder composites with high efficiency. The compositions of the powders meet its specifications with low impurity content. The mean grain size decreases, lattice distortion and system energy increase with increasing the milling time. The morphology of nanocrystalline WC-Co particles displays dominantly sphere shape and their particle sizes are all lower than 80nm. | 14 | The biogeochemical behavior of dissolved aluminum in the southern Yellow Sea:Influence of the spring phytoplankton bloom显示文摘Many recent studies have investigated the nutrient-type profiles of dissolved aluminum(Al) in the ocean.Significant scavenging of dissolved Al can occur during phytoplankton blooms,but the mechanism remains unclear.The distribution of dissolved Al in the southern Yellow Sea(SYS) was investigated in winter and spring 2009.Following measurements at grid stations during the spring sampling cruise,two drifting anchor surveys of more than 100 h were conducted to trace the variation of dissolved Al concentration during the spring phytoplankton bloom(SPB).The concentration of dissolved Al in the SYS decreased from 40 nmol/L in February to 30 nmol/L in March and 10-20 nmol/L in April,while the concentration of Chl a increased from < 2 μg/L in March to > 4 μg/L in April.The concentration of dissolved Al in the SYS decreased significantly with the development of the phytoplankton bloom,which indicated biological scavenging of dissolved Al from water column.The proportion of dissolved Al scavenged from water column by different phytoplankton species differed at the two drifting stations,with greater removal efficiency demonstrated by diatoms than dinoflagellates.Phytoplankton samples collected from the Chl a maximum layer were washed with trace metal clean reagent(oxalate-EDTA-citrate,abbreviate as oxalate solution,Tovar-Sanchez et al.,2003) to enable the surface-scavenged(extracellular) and intracellular Al pools associated with phytoplankton to be differentiated.Thirty-nine to ninetysix percent of the total Al was found to be existed in the interior pools,which indicated that biological absorption was the important way to scavenge dissolved Al during phytoplankton blooms in the SYS. | LI FaMing REN JingLing YAN Li LIU SuMei LIU ChengGang ZHOU Feng ZHANG Jing | 2013 | Chinese Science Bulletin2013,58,2: | 4 |
| 15 | Removal of nonimpacted third molars alters the periodontal condition of their neighbors clinically, immunologically, and microbiologically显示文摘Considering the adverse effects of nonimpacted third molars(N-M3s)on the periodontal health of adjacent second molars(M2s),the removal of N-M3s may be beneficial to the periodontal health of their neighbors.This study aimed to investigate the clinical,immunological,and microbiological changes of the periodontal condition around M2s following removal of neighboring N-M3s across a 6-month period.Subjects with at least one quadrant containing an intact first molar(M1),M2,and N-M3 were screened and those who met the inclusion criteria and decided to receive N-M3 extraction were recruited in the following investigation.M2 periodontal condition was interrogated before M3 extraction(baseline)and at 3 and 6 months postoperatively.Improvements in clinical periodontal indexes of M2s in response to their adjacent N-M3 removal,along with changes in inflammatory biomarkers among gingival crevicular fluid(GCF)and the composition of subgingival plaque collected from the distal sites of the M2s of the targeted quadrant were parallelly analyzed.Complete data of 26 tooth extraction patients across the follow-up period were successfully obtained and subsequently applied for statistical analysis.Compared to the baseline,the periodontal condition of M2s was significantly changed 6 months after N-M3 removal;specifically,the probing depth of M2s significantly reduced(P<0.001),the matrix metalloproteinase(MMP)-8 concentration involved in GCF significantly decreased(P=0.025),and the abundance of the pathogenic genera unidentified Prevotellaceae and Streptococcus significantly decreased(P<0.001 and P=0.009,respectively).We concluded that N-M3 removal was associated with superior clinical indexes,decreased GCF inflammatory biomarkers,and reduced pathogenic microbiome distribution within the subgingival plaque.Although the retention or removal of N-M3s continues to be controversial,our findings provide additional evidence that medical decisions should be made as early as possible or at least before the neighboring teeth are irretrievably damaged. | Yi Tian Lijuan Sun Honglei Qu Yang Yang Faming Chen | 2021 | International Journal of Oral Science2021,13,1: | 3 |
| 16 | From contraction to gene expression:nanojunctions of the sarco/endoplasmic reticulum deliver site-and function-specific calcium signals显示文摘Calcium signals determine, for example, smooth muscle contraction and changes in gene expression. How calcium signals select for these processes is enigmatic. We build on the 'panjunctional sarcoplasmic reticulum' hypothesis, describing our view that different calcium pumps and release channels, with different kinetics and affinities for calcium, are strategically positioned within nanojunctions of the SR and help demarcate their respective cytoplasmic nanodomains. SERCA2 b and Ry R1 are preferentially targeted to the sarcoplasmic reticulum(SR) proximal to the plasma membrane(PM), i.e., to the superficial buffer barrier formed by PM-SR nanojunctions, and support vasodilation. In marked contrast, SERCA2 a may be entirely restricted to the deep, perinuclear SR and may supply calcium to this sub-compartment in support of vasoconstriction. Ry R3 is also preferentially targeted to the perinuclear SR, where its clusters associate with lysosome-SR nanojunctions. The distribution of Ry R2 is more widespread and extends from this region to the wider cell. Therefore, perinuclear Ry R3 s most likely support the initiation of global calcium waves at L-SR junctions, which subsequently propagate by calcium-induced calcium release via Ry R2 in order to elicit contraction. Data also suggest that unique SERCA and Ry R are preferentially targeted to invaginations of the nuclear membrane. Due to the combined influence of human activities and natural factors in the past half a century, water quality in the lower reaches of the Shiyang River decreased and ecological environment deteriorated, which influenced sustainable development of this area and surrounding area seriously. This paper analyzed current situation and reasons of the deteriorated ecological environment in the lower reaches of the Shiyang River, and proposed that water environment regulation was the primary task in the eco-environment control of Minqin area, specifically,(a) By using existing policies, accelerating the construction of water-saving agriculture in the middle reaches of the Shiyang River, water quantity in the lower reaches would be guanranted, water shortage for agricultural production and ecological use in Minqin oasis would be relieved;(b) Unreasonable human activities such as overexploitation of underground water should be controlled strictly;(c) Water conservancy project should be developed vigorously to ensure rate of flow.
