| 2 | Chimeric antigen receptor-modified T cells for the immunotherapy of patients with EGFR-expressing advanced relapsed/refractory non-small cell lung cancer显示文摘The successes achieved by chimeric antigen receptor-modified T(CAR-T) cells in hematological malignancies raised the possibility of their use in non-small lung cancer(NSCLC). In this phase I clinical study(NCT01869166), patients with epidermal growth factor receptor(EGFR)-positive(>50% expression), relapsed/refractory NSCLC received escalating doses of EGFR-targeted CAR-T cell infusions. The EGFR-targeted CAR-T cells were generated from peripheral blood after a 10 to 13-day in vitro expansion. Serum cytokines in peripheral blood and copy numbers of CAR-EGFR transgene in peripheral blood and in tissue biopsy were monitored periodically. Clinical responses were evaluated with RECIST1.1 and immune-related response criteria, and adverse events were graded with CTCAE 4.0. The EGFR-targeted CAR-T cell infusions were well-tolerated without severe toxicity. Of 11 evaluable patients, two patients obtained partial response and five had stable disease for two to eight months. The median dose of transfused CAR+ T cells was 0.97×10~7 cells kg^(-1)(interquartile range(IQR), 0.45 to 1.09×10~7 cells kg^(-1)). Pathological eradication of EGFR positive tumor cells after EGFR-targeted CAR-T cell treatment can be observed in tumor biopsies, along with the CAR-EGFR gene detected in tumor-infiltrating T cells in all four biopsied patients. | 4 | Identification and genetic mapping of four novel genes that regulate leaf development in Arabidopsis显示文摘Molecular and genetic characterizations of mutants have led to a better understanding of many developmental processes in the model system Arabidopsis thaliana. However, the leaf development that is specific to plants has been little studied. With the aim of contributing to the genetic dissection of leaf development, we have performed a large-scare screening for mutants with abnormal leaves. Among a great number of leaf mutants we have generated by T-DNA and transposon tagging and ethylmethae sulfonate (EMS) mutagenesis, four independent mutant lines have been identified and studied genetically. Phenotypes of these mutant lines represent the defects of four novel nuclear genes designated LL1 (LOTUS LEAF 1), LL2 (LOTUS LEAF 2), URO (UPRIGHT ROSETTE), and EIL (ENVIRONT CONDITION INDUCED LESION). The phenotypic analysis indicates that these genes play important roles during leaf development. | 5 | Orbit determination and time synchronization for a GEO/IGSO satellite navigation constellation with regional tracking network显示文摘Aiming at regional services,the space segment of COMPASS (Phase I) satellite navigation system is a constellation of Geostationary Earth Orbit (GEO),Inclined Geostationary Earth Orbit (IGSO) and Medium Earth Orbit (MEO) satellites.Precise orbit determination (POD) for the satellites is limited by the geographic distribution of regional tracking stations.Independent time synchronization (TS) system is developed to supplement the regional tracking network,and satellite clock errors and orbit data may be obtained by simultaneously processing both tracking data and TS data.Consequently,inconsistency between tracking system and TS system caused by remaining instrumental errors not calibrated may decrease navigation accuracy.On the other hand,POD for the mixed constellation of GEO/IGSO/MEO with the regional tracking network leads to parameter estimations that are highly correlated.Notorious example of correlation is found between GEO's orbital elements and its clock errors.We estimate orbital elements and clock errors for a 3GEO+2IGSO constellation in this study using a multi-satellite precise orbit determination (MPOD) strategy,with which clock error elimination algorithm is applied to separate orbital and clock estimates to improve numerical efficiency.Satellite Laser Ranging (SLR) data are used to evaluate User Ranging Error (URE),which is the orbital error projected on a receiver's line-of-sight direction.Two-way radio-wave time transfer measurements are used to evaluate clock errors.Experimenting with data from the regional tracking network,we conclude that the fitting of code data is better than 1 m in terms of Root-Mean-Square (RMS),and fitting of carrier phase is better than 1 cm.For orbital evaluation,difference between computed receiver-satellite ranging based on estimated orbits and SLR measurements is better than 1 m (RMS).For clock estimates evaluation,2-hour linear-fitting shows that the satellite clock rates are about 1.E-10 s/s,while receiver clock rates are about 1×10 13-1×10 12 s/s.For the 72-hour POD experiment,the average differences between POD satellite clock rates estimates and clock measurements based on TS system are about 1×10 13 s/s,and for receiver clock rates,the differences are about 1×10 15 s/s. | ZHOU ShanShi HU XiaoGong WU Bin LIU Li QU WeiJing GUO Rui HE Feng CAO YueLing Wu XiaoLi ZHU LingFeng SHI Xin TAN HongLi | 2011 | Science China(Physics,Mechanics & Astronomy)2011,54,6: | 44 |
| 6 | Glycyrrhizic acid inhibits apoptosis and fibrosis in carbontetrachloride-induced rat liver injury显示文摘AIM:To investigate anti-apoptotic effects of glycyrrhizic acid(GA) against fibrosis in carbon tetrachloride(CCl4)-induced liver injury and its contributing factors.METHODS:Liver fibrosis was induced by administration of CCl4 for 8 wk.Pathological changes in the liver of rats were examined by hematoxylin-eosin staining.Collagen fibers were detected by Sirius red staining.Hepatocyte apoptosis was determined by TUNEL assay and the expression levels of cleaved caspase-3,Bax,α-SMA,connective tissue growth factor(CTGF),matrix metalloproteinase(MMP) 2 and MMP9 proteins were evaluated by western blot analysis,and α-SMA m RNA,collagen type Ⅰ and Ⅲ m RNA were estimated by real-time PCR.RESULTS:Treatment with GA significantly improved the pathological changes in the liver and markedly decreased the positive area of Sirius red compared with rats in the CCl4-treated group.TUNEL assay showed that GA significantly reduced the number of TUNEL-positive cells compared with the CCl4-treated group.The expression levels of cleaved caspase-3,Bax,α-SMA,CTGF,MMP2 and MMP9 proteins,and α-SMA m RNA,collagen type Ⅰ and Ⅲ m RNA were also significantly reduced by GA compared with the CCl4-treated group(P < 0.05).CONCLUSION:GA treatment can ameliorate CCl4-induced liver fibrosis by inhibiting hepatocyte apoptosis and hepatic stellate cell activation. | Bo Liang Xiao-Ling Guo Jing Jin Yong-Chun Ma Zheng-Quan Feng | 2015 | World Journal of Gastroenterology2015,21,17: | 44 |
| 7 | Early Paleozoic subduction of the Paleo-Asian Ocean: Geochronological and geochemical evidence from the Dashizhai basalts, Inner Mongolia显示文摘Zircon U-Pb results of basalt from the Dashizhai Town in Inner Mongolia, NE China, shows that the basaltic lava was erupted at 439±3 Ma, much older than the 'Permian basalts' as previously thought. These rocks show arc-type trace element patterns (i.e., Nb-Ta depletion and light REE and large ion lithophile element enrichment) and unradiogenic Sr and highly radiogenic Nd and Hf isotope compositions. They can be subdivided into two petrogenetic groups: Group 1 basalts have relatively high TiO2, MgO and compatible elements and low Sr and Th, characterized by mid-oceanic ridge basalt (MORB)-type Sr-Nd-Hf isotope compositions (87Sr/86Sr(i)=0.7028―0.7032, εNd(t)=+9.8-+11.2, εHf(t)=+16.1- +18.4). Group 2 has lower TiO2, MgO and compatible elements and higher Sr and Th, and relatively evolved Sr-Nd-Hf isotope compositions (87Sr/86Sr(i)=0.7037-0.7038, εNd(t)=+5.7-+7.3, εHf(t)=+12.6- +13.0). Both groups were interpreted as melts derived from a metasomatized mantle wedge formed during the subduction of Paleo-Asian Ocean. The mantle source for Group 1 was probably a highly isotopically depleted oceanic mantle modified by predominant slab fluids; whereas subducted sediments had an important contribution to the melting source for Group 2. The petrogenesis of the Dashizhai basalts provides clear evidence for early Paleozoic subduction of the Paleo-Asian Ocean, and the highly radiogenic Nd and Hf compositions in these rocks suggest that these lavas and their possible intrusive counterparts were one of the important components for Phanerozoic crustal growth. | 8 | Chinese deserts and sand fields in Last Glacial Maximum and Holocene Optimum显示文摘The Last Glacial Maximum (LGM, c. 26-16 ka) and the Holocene Optimum (HO, c. 9-5 ka) were characterized by cold-dry and warm-wet climates respectively in the recently geological Earth. How Chinese deserts and sand fields responded to these distinctive climatic changes is still not clear, however. To reconstruct environments of the deserts and sand fields during the LGM and HO is helpful to understand the forcing mechanisms of environment change in this arid region, and to test paleoclimatic modeling results. Through our long-term field and laboratory investigations, 400 optically stimulated luminescence (OSL) ages and more than 100 depositional records in the Chinese deserts and sand fields were obtained; on the basis of these data, we reconstruct spatial distributions of the deserts and sand fields during the LGM and HO. Our results show that the sand fields of Mu Us, Hunshandake, Horqin and Hulun Buir in northern and northeastern China had expanded 25%, 37%, 38% and 270%, respectively, during the LGM; the sand fields of Gonghe in the northeastern Qinghai-Tibetan Plateau had expanded 20%, and the deserts of Badain Jaran, Tengger in central northern China had expanded 39% and 29% separately during the LGM; the deserts of Taklimakan, Gurbantünggüt and Kumtag in northwestern China had expanded 10%-20% respectively, compared to their modern areas. On the other hand, all of the sand fields were nearly completely covered by vegetation during the HO; the deserts in northwestern and central northern China were reduced by around 5%-20% in area during this time. Lakes in this arid region were probably expanded during the HO but this conclusion needs more investigation. Compared with the geological distributions of deserts and sand fields, human activity has clearly changed (expanded) the area of active sand dunes at the present time. | 11 | Positioning accuracy assessment for the 4GEO/5IGSO/2MEO constellation of COMPASS显示文摘Determined to become a new member of the well-established GNSS family,COMPASS(or BeiDou-2) is developing its capabilities to provide high accuracy positioning services.Two positioning modes are investigated in this study to assess the positioning accuracy of COMPASS' 4GEO/5IGSO/2MEO constellation.Precise Point Positioning(PPP) for geodetic users and real-time positioning for common navigation users are utilized.To evaluate PPP accuracy,coordinate time series repeatability and discrepancies with GPS' precise positioning are computed.Experiments show that COMPASS PPP repeatability for the east,north and up components of a receiver within China's Mainland is better than 2 cm,2 cm and 5 cm,respectively.Apparent systematic offsets of several centimeters exist between COMPASS precise positioning and GPS precise positioning,indicating errors remaining in the treatments of COMPASS measurement and dynamic models and reference frame differences existing between two systems.For common positioning users,COMPASS provides both open and authorized services with rapid differential corrections and integrity information available to authorized users.Our assessment shows that in open service positioning accuracy of dual-frequency and single-frequency users is about 5 m and 6 m(RMS),respectively,which may be improved to about 3 m and 4 m(RMS) with the addition of differential corrections.Less accurate Signal In Space User Ranging Error(SIS URE) and Geometric Dilution of Precision(GDOP) contribute to the relatively inferior accuracy of COMPASS as compared to GPS.Since the deployment of the remaining 1 GEO and 2 MEO is not able to significantly improve GDOP,the performance gap could only be overcome either by the use of differential corrections or improvement of the SIS URE,or both. | ZHOU ShanShi CAO YueLing ZHOU JianHua HU XiaoGong TANG ChengPan LIU Li GUO Rui HE Feng CHEN JunPing WU Bin | 2012 | Science China(Physics,Mechanics & Astronomy)2012,55,12: | 31 |

| 14 | Geochronology, geochemistry, and its geological significance of the Permian Mandula mafic rocks in Damaoqi, Inner Mongolia显示文摘The Hujierte-Chaganhadamiao volcanic-plutonic sequence is located in the Mandula area of northern Damaoqi, Inner Mongolia. It contains mainly mafic volcanics, with several ultramafic blocks in its central part and mafic-intermediate intrusions in the east. The zircon U-Pb ages of the gabbro and basalt samples are 278.5±3.0 Ma (MSWD=0.66) and 273.7±1.0 Ma (MSWD=0.36), respectively. These ages constrain the magmatism occurred in the Early Permian. The Mandula Permian mafic rocks are characterized by low abundances of REE and slightly LREE-enriched chondrite-normalized N-MORB-type REE patterns. And these mafic samples have high ( 143 Nd/ 144 Nd) i (0.51262-0.51270), relatively high positive ε Nd (t) (3.4-8.0), and high Mg # (49-54), suggesting derivation from the depleted asthenosphere mantle source. But they also show the enrichment in large ion lithophile elements (LILE, e.g. Rb, Ba and Sr), depletion in high field strength elements (HFSE); distinctly negative Nb, Ta anomalies and slightly negative P, Ti anomalies; high ( 87 Sr/ 86 Sr) i (0.70490-0.70537), low ( 206 Pb/ 204 Pb) i (17.39-17.93). All of these resemble the characteristics of the enriched mantle source or involvement of arc materials in the magma genesis. Furthermore, as shown in the correlation plots of ε Nd (t) vs ( 87 Sr/ 86 Sr) i , ( 206 Pb/ 204 Pb) i vs ( 207 Pb/ 204 Pb) i and ( 208 Pb/ 204 Pb) i , La/Ba La/Nb vs La/Nb, and La/Nb ratio, the magma source could have been experienced the contamination and metasomation from the previous subduction process. And the positive correlation between the selected major oxides and trace elements could be explained by the contamination from continental crust and arc materials during the magmatic emplacement. | 15 | Lean-non-alcoholic fatty liver disease increases risk for metabolic disorders in a normal weight Chinese population显示文摘AIM:To study the prevalence and clinical biochemical,blood cell and metabolic features of lean-non-alcoholic fatty liver disease(lean-NAFLD)and its association with other diseases.METHODS:Demographic,biochemical and blood examinations were conducted in all the subjects in this study.We classified the subjects into four groups according to their weight and NAFLD status:lean-control,lean-NAFLD[body mass index(BMI)<24 kg/m2],overweight-obese control and overweight-obese NAFLD.One-way analysis of variance(ANOVA)was used to compare the means of continuous variables(age,BMI,blood pressure,glucose,lipid,insulin,liver enzymes and blood cell counts)and theχ2 test was used to compare the differences in frequency of categorical variables(sex,education,physical activity,smoking,alcohol consumption and prevalence of hypertension,hyperlipidemia,diabetes,metabolic syndrome central obesity and obesity).Both univariate and multivariate logistic regression models were adopted to calculate odds ratios(ORs)and predict hyperlipidemia,hypertension,diabetes and metabolic syndrome when we respectively set all controls,lean-control and overweightobese-control as references.In multivariate logistic regression models,we adjusted potential confounding factors,including age,sex,smoking,alcohol consumption and physical activity.RESULTS:The prevalence of NAFLD was very high in China.NAFLD patients were older,had a higher BMI,waist circumference,blood pressure,fasting blood glucose,insulin,blood lipid,liver enzymes and uric acid than the controls.Although lean-NAFLD patients had lower BMI and waist circumstance,they had significantly higher visceral adiposity index than overweightobese controls.Lean-NAFLD patients had comparable triglyceride,cholesterin and low-density lipoprotein cholesterin to overweight-obese NAFLD patients.In blood cell examination,both lean and overweightobese NAFLD was companied by higher white blood cell count,red blood cell count,hemoglobin and hematocrit value.All NAFLD patients were at risk of hyperlipidemia,hypertension,diabetes and metabolic syndrome(Met S).Lean-NAFLD was more strongly associated with diabetes(OR=2.47,95%CI:1.14-5.35),hypertension(OR=1.72,95%CI:1.00-2.96)and Met S(OR=3.19,95%CI:1.17-4.05)than overweight-obese-NAFLD(only OR for Met S was meaningful:OR=1.89,95%CI:1.29-2.77).NAFLD patients were more likely to have central obesity(OR=1.97,95%CI:1.38-2.80),especially in lean groups(OR=2.17,95%CI:1.17-4.05).CONCLUSION:Lean-NAFLD has unique results in demographic,biochemical and blood examinations,and adds significant risk for diabetes,hypertension and Met S in lean individuals. | Ren-Nan Feng Shan-Shan Du Cheng Wang Yan-Chuan Li Li-Yan Liu Fu-Chuan Guo Chang-Hao Sun | 2014 | World Journal of Gastroenterology2014,20,47: | 30 |
| 16 | The intracellular mechanism of alpha-fetoprotein promoting the proliferation of NIH 3T3 cells显示文摘AIM The existence and properties of alpha-fetoprotein (AFP) receptor on the surface of NIH 3T3 cells and the effects of AFP on cellular signal transduction pathway were investigated. METHODS The effect of AFP on the proliferation of NIH 3T3 cells was measured by incorporation of 3H-TdR. Receptor-binding assay of 125I-AFP was performed to detect the properties of AFP receptor in NIH 3T3 cells. The influences of AFP on the [cAMP]i and the activities of protein kinase A (PKA) were determined. Western blot was used to detect the change of K-ras P21 protein expression. RESULTS The proliferation of NIH 3T3 cells treated with 0-80 mg/L of AFP was significantly enhanced. The Scatchard analysis indicated that there were two classes of binding sites with KD of 2.722×10-9M (Bmax=12810 sites per cell) and 8.931× 10-SM (Bmax=l19700 sites per cell) respectively. In the presence of AFP (20 mg/L), the content of cAMP and activities of PKA were significantly elevated . The level of K-ras P21 protein was upregulated by AFP at the concentration of 20 mg/L. The monoclonal antibody against AFP could reverse the effects of AFP on the cAMP content, PKA activity and the expression of K-ras p21 gene. | 17 | K-Ar dating of late Mesozoic volcanism and geochemistry of volcanic gravels in the North Huaiyang Belt, Dabie orogen: Constraints on the stratigraphic framework and exhumation of the northern Dabie orthogneiss complex显示文摘Two eruption episodes are identified through systematic field investigations and K-Ar dating of the late Mesozoic volcanic rocks in the North Huaiyang belt (NHB), Dabie orogenic belt, of which the earlier volcanic suite termed Maotanchang Fm. (Fm.) occurring at Jinzhai, Xianhualing and Maotanchang, etc., was erupted from 149 Ma to 138 Ma. The other named Xiaotian Fm. mainly distributed at Xiaotian, Shucheng, etc., was formed between 132 Ma and 116 Ma. During the eruption gap of the two volcanic suites deposited a volcano-sedimentary conglomerate layer, which are composed of the multi-compositional gravels, including the North Dabie orthogneiss complex (NDOC), volcanic gravels, etc. These volcanic gravels in the conglomerate layer show identical geochemical and isotopic compositions (87Sr/86Sr(t) =0.7084-0.7092,εNd(t) =-21.8--24.4) to the Maotanchang Fm. volcanic rocks (87Sr/86Sr = 0.7086-0.7102,εNd = -19.2-24.4), but significantly distinct from those of Xiaotian Fm. | 18 | Germ cell apoptosis induced by Ureaplasma urealyticum infection显示文摘Aim: To study the effect of Ureaplasma urealyticum (UU) infection on germ cell apoptosis of male rats. Methods: Male rats were infected artificially with UU serotype 8 (T960) . Morphological changes of germ cells in the seminiferous tubules and the lumen of the epididymides were observed under the light microscope. Fluorescence-conjugated polyclonal antibodies to Fas and Fas ligand (FasL) were used to localize Fas and FasL. TUNEL staining of germ cells and Sertoli cells was performed by the AKPase method. TUNEL-positive rate ( % positive cells) and TUNEL-positive area (area occupied by stained cells) were analysed by KS400 Image Analysis System. The DNA laddering analysis was performed by agarose gels electrophoresis. Results: In those rats infected with UU: (1) Exfoliated germ cells were dramatically increased. Many multinucleated giant cells were found in the seminiferous tubules and the lumen of the epididymides. (2) The number of TUNEL-positive cells and the TUNEL-positive area were significantly increased.(3) The expression of Fas and FasL in germ cells and Sertoli cells was up-regulated. (4) Discrete bands of fragmented DNA were found in the testicular cells. | 20 | The crustal structures of the central Longmenshan along and its margins as related to the seismotectonics of the 2008 Wenchuan Earthquake显示文摘In 2010,a 500-km-long wide-angle reflection/refraction seismic profile was completed,running northwest from the central Sichuan Basin.This profile orthogonally crosses the meizoseismal area of great Wenchuan earthquake of 12 May 2008,which occurred in the central part of the Longmenshan.The profile also passes through the northwestern Sichuan Plateau,along which a new deep seismic sounding observation system was set up that was much improved over previous datasets and enabled abundant observations to be recorded.Seismic wave phase records that reflect the structural characteristics of different tectonic blocks,especially the complicated phase features associated with the Wenchuan earthquake,were calculated and analyzed in detail.A 2D crustal P-wave velocity model for the orogenic belt in the central Longmenshan and its margins was determined,and crustal structure differences between the stable Sichuan Basin and the thickened northwestern Sichuan Plateau were characterized.Lithological variations within the upper and lower crust in the interior of the plateau,especially a great velocity decrease and plastic rheological properties associated with strong lithologic weakening in lower crust,were detected.From west to east in the lower crust beneath the orogenic belt lying between the Sichuan Basin and the northwestern Sichuan Plateau,a giant shovel-like upwelling is observed that dips gently in the lower part and at higher angles in the upper part;this is inferred to be related to the fault systems in the central Longmenshan.An upwelling in the upper-middle crust along the eastern margin of the orogenic belt is associated with steeply dipping thrusts that strongly uplift the upper crust and crystalline basement beneath a central fault system in the Longmenshan.The data,combined with an understanding of the regional tectonic stress field and previous geological results,enable a discussion of basin-and-range coupling,orogenic tectonics,the crustal fault system,and the seismogenic tectonic environment of the central Longmenshan along the eastern margin of the Qinghai-Tibet Plateau. | JIA ShiXu LIU BaoJin XU ZhaoFan LIU Zhi FENG ShaoYing ZHANG JianShi LIN JieYan TIAN XiaoFeng LIU QiaoXia GUO WenBin | 2014 | Science China Earth Sciences2014,57,4: | 26 |
