| 11 | Phylogenetic and chemotaxonomic resolution of the genus Annulohypoxylon(Xylariaceae)including four new species显示文摘This study deals with an extensive evaluation focusing on phylogenetic and chemotaxonomic infrageneric relationships of the genus Annulohypoxylon(Xylariaceae,Ascomycota),whose species are ubiquitously associated with seed plants as endophytes and saprotrophs in all forested areas of the world.Using evidence from phylogenetic,chemotaxonomic and morphological data,various varieties within the genus are raised to species level,leading to the new combinations for A.areolatum(:A.bovei var.microsporum),A.macrosporum(:A.leptascum var.macrosporum),and A.microdiscum(:A.moriforme var.microdiscum).Annulohypoxylon substygium nom.nov.is applied for A.stygium var.annulatumand the four new tropical and subtropical species A.massivum,A.violaceopigmentum,A.viridistratum and A.yungensis are introduced.Furthermore,A.leucadendri is excluded from the genus as its morphological characters disagree with the generic concept,the recently erected A.palmicola is synonymized with A.leptascum and A.austrobahiense has been reassigned to the genus Hypoxylon.In addition,the key taxa A.annulatum and A.truncatum have been reinvestigated and epitypes have been defined.For the first time,a dichotomous key to the genus is provided.A phylogenetic b-tubulin tree in conjunction with stromatal HPLC profiles clearly shows that Annulohypoxylon comprises two distinct lineages.The A.cohaerens/multiforme group might eventually warrant segregation into a new genus as further molecular data become available. | Eric Kuhnert Esteban B.Sir Christopher Lambert Kevin D.Hyde Adriana I.Hladki Andrea I.Romero Manfred Rohde Marc Stadler | 2017 | Fungal Diversity2017,,4: | 0 |
| 12 | Impact of intragastric balloon on blood pressure reduction:A retrospective study in Eastern North Carolina显示文摘BACKGROUND Obesity has evolved into a global pandemic.The prevalence of obesity and hypertension in eastern North Carolina are comparable,if not higher,than the national prevalence.In the United States,an estimated 34%of adults have hypertension,the most modifiable risk factor for heart disease and stroke.Lifestyle and pharmacological interventions often do not provide sustained weight loss in obese patients.Bariatric surgery offers an effective weight reduction with short-and long-term health improvements;however,a higher body mass index is associated with higher surgical morbidity and mortality,longer hospitalization,and increasing rates of 30-day readmission due to comorbidities.Intragastric balloon may bridge a critical gap in the treatment of obesity.The objective of this paper is to showcase the impact of endoscopic bariatric therapy on blood pressure reduction.AIM To investigate the impact of intragastric balloon on blood pressure reduction.METHODS A retrospective chart review was conducted from January 1,2016 to January 31,2019 of consecutive adults who received intragastric balloon therapy(IGBT)in a gastroenterology private practice in Eastern North Carolina.The balloon was introduced into the stomach under endoscopic guidance,and while in the region of the gastric body,inflation with saline was performed at increments of 50 mL until target volume between 500 to 650 mL of saline was attained depending on the patient's gastric capacity.No procedural complications were noted during endoscopic placement and removal of the balloon.A cohort study design was used for data analysis.A total of 172 patients had the Orbera^(■)intragastric balloon placed.Of the 172 patients who had IGBT at baseline,11 patients(6.4%)requested early balloon removal due to foreign body sensation(n=1),and/or intolerable gastrointestinal adverse events(n=10).The reported gastrointestinal adverse events were nausea,vomiting,abdominal pain,and diarrhea.Eventually,6-mo follow-up data were available for only 140 patients.As a result,only the 140 available at the 6-mo follow-up were included in the analysis.Univariate,bivariate,and multivariate statistical analyses were performed.Specifically,scatterplots were created to show the relationship between weight and blood pressure,and paired two-sample t-test was carried out to determine if there was a significant reduction in weight before and after the IGBT.Multiple regressions were also performed to examine the association between participants’total body weight and blood pressure.The outcome variables for the multiple regression were systolic and diastolic blood pressure measured as continuous variables.This was followed by logistic regression analyses to determine the association between total body weight and hypertension at 6-mo post-implantation.The outcome variables for the logistic regression were systolic blood pressure–nonhypertensive(140 mmHg or less)or hypertensive(greater than 140 mmHg),and diastolic blood pressure–non-hypertensive(90 mmHg or less)or hypertensive(greater than 90 mmHg).All authors had access to the study data and reviewed and approved the final manuscript.All statistical analyses were done using STATA 14®.RESULTS The study included 15%males and 85%females.50%of the patients were white and just over 22%were non-white,and about 27%declined to give their race.The average baseline patients’weight prior to IGBT was 231.61 Lbs.(SD=46.53 Lbs.).However,the average patients’weight after IGBT at the 6-mo follow-up was 203.88 Lbs.(SD=41.04 Lbs.).Hence,on average,the percent total body weight loss at 6-mo is 11.97 after IGBT.The logistic regression performed revealed that weight(β=0.0140,P<0.000)and age(β=0.0534,P<0.000)are important factors in determining systolic blood pressure after IGBT.None of the other demographic characteristics or indicated comorbidities were found to be significant.CONCLUSION IGBT can be an effective short-term weight reduction modality with a relatively little risk of adverse event.Due to its improvement on systolic blood pressure,IGBT may help reduce cardiovascular risk. | Gbeminiyi Olanrewaju Samuel Karissa Lambert Elijah Asagbra Glenn Harvin Eric Ibegbu | 2021 | World Journal of Gastrointestinal Endoscopy2021,13,5: | 0 |
| 13 | Physiological and druggable skipping of immunoglobulin variable exons in plasma cells显示文摘The error-prone V(D)J recombination process generates considerable amounts of nonproductive immunoglobulin(Ig)pre-mRNAs.We recently demonstrated that aberrant Ig chains lacking variable(V)domains can be produced after nonsense-associated altered splicing(NAS)events.Remarkably,the expression of these truncated Ig polypeptides heightens endoplasmic reticulum stress and shortens plasma cell(PC)lifespan.Many questions remain regarding the molecular mechanisms underlying this new truncated Ig exclusion(TIE-)checkpoint and its restriction to the ultimate stage of B-cell differentiation.To address these issues,we evaluated the extent of NAS of Ig pre-mRNAs using an Ig heavy chain(IgH)knock-in model that allows for uncoupling of V exon skipping from TIE-induced apoptosis.We found high levels of V exon skipping in PCs compared with B cells,and this skipping was correlated with a biallelic boost in IgH transcription during PC differentiation.Chromatin analysis further revealed that the skipped V exon turned into a pseudo-intron.Finally,we showed that hypertranscription of Ig genes facilitated V exon skipping upon passive administration of splice-switching antisense oligonucleotides(ASOs).Thus,V exon skipping is coupled to transcription and increases as PC differentiation proceeds,likely explaining the late occurrence of the TIE-checkpoint and opening new avenues for ASO-mediated strategies in PC disorders. | Mohamad Omar Ashi Nivine Srour Jean-Marie Lambert Anne Marchalot Ophelie Martin Sandrine Le Noir Eric Pinaud Maria Victoria Ayala Christophe Sirac Jerome Sauliere Jerome Moreaux Michel Cogne Laurent Delpy | 2019 | Cellular & Molecular Immunology2019,16,10: | 0 |
| 14 | High quality genome sequences of thirteen Hypoxylaceae(Ascomycota)strengthen the phylogenetic family backbone and enable the discovery of new taxa显示文摘The Hypoxylaceae(Xylariales,Ascomycota)is a diverse family of mainly saprotrophic fungi,which commonly occur in angiosperm-dominated forests around the world.Despite their importance in forest and plant ecology as well as a prolific source of secondary metabolites and enzymes,genome sequences of related taxa are scarce and usually derived from envi-ronmental isolates.To address this lack of knowledge thirteen taxonomically well-defined representatives of the family and one member of the closely related Xylariaceae were genome sequenced using combinations of Illumina and Oxford nanopore technologies or PacBio sequencing.The workflow leads to high quality draft genome sequences with an average N50 of 3.0 Mbp.A backbone phylogenomic tree was calculated based on the amino acid sequences of 4912 core genes reflecting the current accepted taxonomic concept of the Hypoxylaceae.A Percentage of Conserved Proteins(POCP)analysis revealed that 70%of the proteins are conserved within the family,a value with potential application for the definition of family boundaries within the order Xylariales.Also,Hypomontagnella spongiphila is proposed as a new marine derived lineage of Hypom.monticulosa based on in-depth genomic comparison and morphological differences of the cultures.The results showed that both species share 95%of their genes corresponding to more than 700 strain-specific proteins.This difference is not reflected by standard taxonomic assessments(morphology of sexual and asexual morph,chemotaxonomy,phylogeny),preventing species delimitation based on traditional concepts.Genetic changes are likely to be the result of environmental adaptations and selective pressure,the driving force of speciation.These data provide an important starting point for the establishment of a stable phylogeny of the Xylariales;they enable studies on evolution,ecological behavior and biosynthesis of natural products;and they significantly advance the taxonomy of fungi. | Daniel Wibberg Marc Stadler Christopher Lambert Boyke Bunk Cathrin Spröer Christian Rückert Jörn Kalinowski Russell JCox Eric Kuhnert | 2021 | Fungal Diversity2021,,1: | 0 |
