| 1 | Differential control of growth,apoptotic activity and gene expression in human colon cancer cells by extracts derived from medicinal herbs,Rhazya stricta and Zingiber officinale and their combination显示文摘AIM:To investigate the effects of extracts from Rhazya stricta(R.stricta) and Zingiber officinale(Z.officinale) on human colorectal cancer cells.METHODS:Human colorectal cancer cells(HCT116) were subjected to increasing doses of crude alkaloid extracts from R.stricta(CAERS) and crude flavonoid extracts from Z.officinale(CFEZO).Cells were then harvested after 24,48 or 72 h and cell viability was examined by trypan blue exclusion dye test; clonogenicity and soft agar colony-forming assays were also carried out.Nuclear stain(Hoechst 33342),acridine orange/ethidium bromide double staining,agarose gel electrophoresis and comet assays were performed to assess pro-apoptotic potentiality of the extracts.Quantitative reverse transcription-polymerase chain reaction(qRT-PCR),using gene-specific primers and Western blot analyses were performed to assess the impact of CAERS and CFEZO on the expression levels of key regulatory proteins in HCT116 cells.RESULTS:Treatment with a combination of CAERS and CFEZO synergistically suppressed the proliferation,colony formation and anchorage-independent growth of HCT116 cells.Calculated IC50,after 24,48 and 72 h,were 70,90 and 130 μg/mL for CAERS,65,85 and 120 μg/mL for CFEZO and 20,25 and 45 μg/mL for both agents,respectively.CAERS- and CFEZO-treated cells exhibited morphologic and biochemical features of apoptotic cell death.The induction of apoptosis was associated with the release of mitochondrial cytochrome c,an increase in the Bax/Bcl-2 ratio,activation of caspases 3 and 9 and cleavage of poly ADP-ribose polymerase.CAERS and CFEZO treatments downregulated expression levels of anti-apoptotic proteins including Bcl-2,Bcl-X,Mcl-1,survivin and XIAP,and upregulated expression levels of proapoptotic proteins such as Bad and Noxa.CAERS and CFEZO treatments elevated expression levels of the oncosuppressor proteins,p53,p21 and p27,and reduced levels of the oncoproteins,cyclin D1,cyclin/cyclin-dependent kinase-4 and c-Myc.CONCLUSION:These data suggest that a combination of CAERS and CFEZO is a promising treatment for the prevention of colon cancer. | Ayman I Elkady Rania Abd El Hamid Hussein Osama A Abu-Zinadah | 2014 | World Journal of Gastroenterology2014，20，41： | 12 |
| 2 | Incidence and characteristics of HBV reactivation in hematological malignant patients in south Egypt显示文摘AIM:To investigate characteristics of hepatitis B virus(HBV)implicated in HBV reactivation in patients with hematological malignancies receiving immunosuppressive therapy.METHODS:Serum samples were collected from 53 patients with hematological malignancies negative for hepatitis B surface antigen(HBsAg)before the start of and throughout the chemotherapy course.HBV reactivation was diagnosed when the HBsAg status changed from negative to positive after the initiation of chemotherapy and/or when HBV DNA was detected by realtime detection polymerase chain reaction(RTD-PCR).For detecting the serological markers of HBV infection,HBsAg as well as antibodies to the core antigen(antiHBc)and to the surface antigen were measured in the sera by CEIA.Nucleic acids were extracted from sera,and HBV DNA sequences spanning the S gene were amplified by RTD-PCR.The extracted DNA was further subjected to PCR to amplify the complete genome as well as the specific genomic sequences bearing the enhancerⅡ/core promoter/pre-core/core regions(nt1628-2364).Amplicons were sequenced directly.RESULTS:Thirty-five(66%)of the 53 HBsAg-negative patients were found to be negative serologically for antiHBc,and the remaining 18(34%)patients were positive for anti-HBc.Five of the 53(9.4%)patients with hematologic malignancies experienced HBV reactivation.Genotype D1 was detected in all five patients.Four types of mutant strains were detected in the S gene product of HBV strains and were isolated from 3 patients with HBV reactivation:T/S120,L143,and I126.HBV DNA was detected in the pretreatment HBsAg-negative samples in one of the five patients with HBV reactivation.In this patient,sequences encompassing the HBV full genome obtained from sera before the start of chemotherapy and at the time of de novo HBV hepatitis were detected and it showed 100%homology.Furthermore,in the phylogenetic tree,the sequences were clustered together,thereby indicating that this patient developed reactivation from an occult HBV infection.CONCLUSION:Past infection with HBV is a risk factor for HBV reactivation in Egypt.Mandatory anti-HBc screening prior to chemotherapy in patients with hematological malignancies is recommended. | Abeer Elkady Sahar Aboulfotuh Elsayed Mostafa Ali Douaa Sayed Nashwa M Abdel-Aziz Amany M Ali Shuko Murakami Sayuki Iijima Yasuhito Tanaka | 2013 | World Journal of Gastroenterology2013，19，37： | 7 |
| 3 | Characteristics of escape mutations from occult hepatitis B virus infected patients with hematological malignancies in South Egypt显示文摘AIM To investigate the prevalence and virological characteristics of occult hepatitis B virus(HBV) infections in patients with hematological malignancies in South Egypt.METHODS Serum samples were collected from 165 patients with hematological malignancies to monitor titers of HBV DNA, hepatitis B surface antigen(HBs Ag), and antibodies to HBV core(anti-HBc) and surface antigens. Serum samples negative for HBs Ag and positive for anti-HBc were subjected to nucleic acid extraction and HBV DNA detection by real-time polymerase chain reaction. DNA sequences spanning the S region were analyzed in cases with occult HBV infection. In vitro comparative study of constructed 1.24-fold wild type and S protein mutant HBV genotype D clones was further performed. RESULTS HBV DNA was detected in 23(42.6%) of 54 patients with hematological malignancies who were HBsA g negative, but anti-HBc positive, suggesting the presence of occult HBV infection. The complete HBV genome was retrieved from 6 occult HBV patients, and P120 T and S143 L were detected in 3 and 2 cases, respectively. Site directed mutagenesis was done to produce 1.24-fold genotype D clones with amino acid mutations T120 and L143. The in vitro analyses revealed that a lower level of extracellular HBsA g was detected by chemiluminescence enzyme immunoassay(CLEIA) with the clone containing T120 mutation, compared with the wild type or the clone with S143 L mutation despite the similar levels of extracellular and intracellular HBs Ag detected by Western blot. Southern blot experiments showed that the levels of intracellular HBV DNA were not different between these clones. | 14 | Improved reversed phase liquid chromatographic method with pulsed electrochemical detection for tobramycin in bulk and pharmaceutical formulation显示文摘Tobramycin is one of the aminoglycoside antibiotics that lack a UV absorbing chromophore. However, the application of pulsed electrochemical detection (PED) has been used successfully for the analysis of this and similar antibiotics. This work describes an improved liquid chromatographic (LC) method combined with PED, which is able to separate much more impurities than before. Using a Discovery C-18 RP column (250 mm 4.6 mm i.d., 5 mm), isocratic elution was carried out with a mobile phase, containing sodium sulfate (35 g/L), sodium octanesulphonic acid (1 g/L), tetrahydrofuran (14 mL/L) and 0.2 M phosphate buffer pH 3.0 (50 mL/L). Using these experimental conditions, the limit of quantification (LOQ, S/N 10) was 5 ng. The linearity was examined in the range LOQ-60 mg/mL and the coefficient of determination was 0.998. The method also proved to be repeatable and the recovery was close to 100%. The influence of the different chromatographic parameters on the separation was investigated by means of an experimental design.
