| 1 | Folate-chitosan-gemcitabine core-shell nanoparticles targeted to pancreatic cancer显示文摘Objective:Human pancreatic cancer is one of the most common clinical malignancies.The effect of comprehensive treatment based on surgery is general.The effects of chemotherapy were not obvious mainly because of lack of targeting and chemoresistance in pancreatic cancer.This study aimed to investigate the effects of folate receptor(FR)-mediated gemcitabine FA-Chi-Gem nanoparticles with a core-shell structure by electrostatic spray on pancreatic cancer.Methods:In this study,the levels of expression of FR in six human pancreatic cancer cell lines were studied by immunohistochemical analysis.The uptake rate of isothiocyanate-labeled FA-Chi nanoparticles in FR high expression cell line COLO357 was assessed by fluorescence microscope and the inhibition rate of FAChi-Gem nanoparticles on COLO357 cells was evaluated by MTT assay.Moreover,the biodistribution of PEG-FA-ICGDER02-Chi in the orthotopic pancreatic tumor model was observed using near-infrared imaging and the human pancreatic cancer orthotopic xenografts were treated with different nanoparticles and normal saline control.Results:The expression of FR in COLO357 was the highest among the six pancreatic cancer cell lines.The FR mainly distributed on cell membrane and fewer in the cytoplasm in pancreatic cancer.Moreover,the absorption rate of the FA-Chi-Gem nanoparticles was more than the Chi nanoparticles without FA modified.The proliferation of COLO357 was significantly inhibited by FA-Chi-Gem nanoparticles.The PEG-FAICGDER02-Chi nanoparticles were enriched in tumor tissue in human pancreatic cancer xenografts,while non-targeted nanoparticles were mainly in normal liver tissue.PEG-FA-Gem-Chi significantly inhibited the growth of human pancreatic cancer xenografts(PEG-FA-Gem-Chi vs.Gem,t=22.950,P=0.000).Conclusions:PEG-FA-FITC-Chi nanoparticles might be an effective targeted drug for treating human FR-positive pancreatic cancer. | Jiahua Zhou Junying Wang Qian Xu Shi Xu Jin Wen Zeqian Yu Detong Yang | 2013 | Chinese Journal of Cancer Research2013,25,5: | 5 |

| 4 | Three-dimensional numerical modeling of nearshore circulation显示文摘A three-dimensional nearshore circulation model was developed by coupling CH3D, a three-dimensional hydrodynamic model and REF/DIF, a nearshore wave transformation model. The model solves the three-dimensional wave-averaged equations of motion. Wave-induced effects on circulation were introduced in the form of radiation stresses, wave-induced mass transport, wave-induced enhancement of bottom friction and wave-induced turbulent mixing. Effects of breaking waves were considered following Svendsen (1984a and 1984b) and Stive and Wind (1986). The model was successfully tested against the analytical solution of longshore currents by Longuet and Higgins (1970). The model successfully simulated the undertow as observed in a laboratory experiment by Stive and Wind (1982). In addition, the model was applied to a physical model by Mory and Hamm (1997) and successfully reproduced the eddy behind a detached breakwater as well as the longshore current on the open beach and the contiguous eddy in the open area of the wave tank. While the qualitative agreement between model results and experimental observations was very good, the quantitative agreement needs to be further improved. Albeit difficult to explain every discrepancy between the model results and observations, in general, sources of errors are attributed to the lack of understanding and comprehensive description of following processes: (1)the horizontal and vertical distribution of radiation stress, especially for breaking waves;(2)the detailed structure of turbulence;(3)Wave-current interaction (not included at this moment); and (4)the wave-current boundary layer and the resulting bottom shear stress. | SUN Detong | 2008 | Acta Oceanologica Sinica2008,27,z1: | 2 |
| 5 | A Dwindling Filter Line Search Algorithm for Nonlinear Equality Constrained Optimization显示文摘This paper proposes a dwindling filter line search algorithm for nonlinear equality constrained optimization. A dwindling filter, which is a modification of the traditional filter, is employed in the algorithm. The envelope of the dwindling filter becomes thinner and thinner as the step size approaches zero. This new algorithm has more flexibility for the acceptance of the trial step and requires less computational costs compared with traditional filter algorithm. The global and local convergence of the proposed algorithm are given under some reasonable conditions. | 7 | CONVERGENCE PROPERTIES OF PROJECTED GRADIENT METHODS WITH NONMONOTONIC BACK TRACKING TECHNIQUE FOR CONVEX CONSTRAINED OPTIMIZATION显示文摘This paper proposes projected gradient algorithms in association with using both trust region and line search techniques for convex constrained optimization problems. The mixed strategy is adopted which switches to back tracking steps when a trial projected gradient step produced by the trust region subproblem is unacceptable. A nonmonotone criterion is used to speed up the convergence progress in some curves with large curvature. A theoretical analysis is given which proves that the proposed algorithms are globally convergent and have local superlinear convergence rate under some reasonable conditions.
