1Cross-talk between NMDA and GABA_A receptors in cultured neurons of the rat inferior colliculus显示文摘Neuronal ion channels of different types often do not function independently but will inhibit or potentiate the activity of other types of channels,a process called cross-talk.The N-methyl-D-aspartate receptor (NMDA receptor) and the γ-aminobutyric acid type A receptor (GABAA receptor) are important excitatory and inhibitory receptors in the central nervous system,respectively.Currently,cross-talk between the NMDA receptor and the GABAA receptor,particularly in the central auditory system,is not well understood.In the present study,we investigated functional interactions between the NMDA receptor and the GABAA receptor using whole-cell patch-clamp techniques in cultured neurons from the inferior colliculus,which is an important nucleus in the central auditory system.We found that the currents induced by aspartate at 100 μmol L-1 were suppressed by the pre-perfusion of GABA at 100 μmol L-1,indicating cross-inhibition of NMDA receptors by activation of GABAA receptors.Moreover,we found that the currents induced by GABA at 100 μmol L-1 (IGABA) were not suppressed by the pre-perfusion of 100 μmol L-1 aspartate,but those induced by GABA at 3 μmol L-1 were suppressed,indicating concentration-dependent cross-inhibition of GABAA receptors by activation of NMDA receptors.In addition,inhibition of IGABA by aspartate was not affected by blockade of voltage-dependent Ca2+ channels with CdCl2 in a solution that contained Ca2+,however,CdCl2 effectively attenuated the inhibition of IGABA by aspartate when it was perfused in a solution that contained Ba2+ instead of Ca2+ or a solution that contained Ca2+ and 10 mmol L-1 BAPTA,a membrane-permeable Ca2+ chelator,suggesting that this inhibition is mediated by Ca2+ influx through NMDA receptors,rather than voltage-dependent Ca2+ channels.Finally,KN-62,a potent inhibitor of Ca2+/calmodulin-dependent protein kinase II (CaMKII),reduced the inhibition of IGABA by aspartate,indicating the involvement of CaMKII in this cross-inhibition.Our study demonstrates a functional interaction between NMDA and GABAA receptors in the inferior colliculus of rats.The presence of cross-talk between these receptors suggests that the mechanisms underlying information processing in the central auditory system may be more complex than previously believed.CONG DanNi TANG ZhengQuan LI LongZhu HUANG YiNa WANG Jun CHEN Lin 2011Science China(Life Sciences)2011,54,6:9

3Regulation of extracellular bioactive cations in bone tissue microenvironment induces favorable osteoimmune conditions to accelerate in situ bone regeneration显示文摘The design of orthopedic biomaterials has gradually shifted from“immune-friendly”to“immunomodulatory,”in which the biomaterials are able to modulate the inflammatory response via macrophage polarization in a local immune microenvironment that favors osteogenesis and implant-to-bone osseointegration.Despite the well-known effects of bioactive metallic ions on osteogenesis,how extracellular metallic ions manipulate immune cells in bone tissue microenvironments toward osteogenesis and subsequent bone formation has rarely been studied.Herein,we investigate the osteoimmunomodulatory effect of an extracellular bioactive cation(Mg^(2+))in the bone tissue microenvironment using custom-made poly lactic-co-glycolic acid(PLGA)/MgO-alendronate microspheres that endow controllable release of magnesium ions.The results suggest that the Mg^(2+)-controlled tissue microenvironment can effectively induce macrophage polarization from the M0 to M2 phenotype via the enhancement of anti-inflammatory(IL-10)and pro-osteogenic(BMP-2 and TGF-β1)cytokines production.It also generates a favorable osteoimmune microenvironment that facilitates the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells.The in vivo results further verify that a large amount of bony tissue,with comparable bone mineral density and mechanical properties,has been generated at an early post-surgical stage in rat intramedullary bone defect models.This study demonstrates that the concept of in situ immunomodulated osteogenesis can be realized in a controlled magnesium tissue microenvironment.Zhengjie Lin Danni Shen Weixiao Zhou Yufeng Zheng Tiantian Kong Xuanyong Liu Shuilin Wu Paul K.Chu Ying Zhao Jun Wu Kenneth M.C.Cheung Kelvin W.K.Yeung 2021Bioactive Materials2021,6,8:5
4Phospholipase C Delta 3 inhibits apoptosis and promotes proliferation,migration,and invasion of thyroid cancer cells via Hippo pathway显示文摘In recent decades,the incidence of thyroid cancer(TC)has rapidly increased,leading us to explore the complex underlying mechanisms.We identified the gene Phospholipase C Delta 3(PLCD3)as a potential oncogene in TC by conducting the whole transcriptome sequencing.Our study is to understand the oncogenic role of PLCD3 in TC.We verified the overexpression of PLCD3 in TC from The Cancer Genome Atlas,Gene Expression Omnibus databases,and a locally validated cohort.Clinical correlation analysis showed that PLCD3 expression was related to histological type,T stage,lymph node metastasis(LNM),and disease stage.The high expression of PLCD3 could be a distinguishing factor for TC and its LNM.The biological function was examined using small interfering RNA-transfected TC cell lines.Silenced PLCD3 could inhibit colony formation,migration,and invasion ability and promote apoptosis of TC cell lines.PLCD3 silencing reversed the epithelial-mesenchymal transition but induced the apoptotic progress.Further exploration revealed that PLCD3 might be associated with critical genes of the Hippo pathway.The expressions of RHOA,YAP1/TAZ,and their downstream targets were decreased significantly when PLCD3 was down-regulated.YAP1 overexpression rescued the tumor-suppressive effect caused by PLCD3 silencing.This study demonstrates that PLCD3 is an oncogene that supports tumorigenesis and progression in TC,and PLCD3 may be a potential target gene for TC treatment.Lizhi Lin Jialiang Wen Bangyi Lin Hao Chen Adheesh Bhandari Yufeng Qi Danni Zheng Ouchen Wang 2021Acta Biochimica et Biophysica Sinica2021,53,4:4

6Magnetic resonance (MR) safety and compatibility of a novel iron bioresorbable scaffold显示文摘Fully bioresorbable scaffolds have been designed to overcome the limitations of traditional drug-eluting stents(DESs),which permanently cage the native vessel wall and pose possible complications.The ultrathin-strut designed sirolimus-eluting iron bioresorbable coronary scaffold system(IBS)shows comparable mechanical properties to traditional DESs and exhibits an adaptive degradation profile during target vessel healing,which makes it a promising candidate in all-comers patient population.For implanted medical devices,magnetic resonance(MR)imaging properties,including MR safety and compatibility,should be evaluated before its clinical use,especially for devices with intrinsic ferromagnetism.In this study,MR safety and compatibility of the IBS scaffold were evaluated based on a series of well-designed in-vitro,ex-vivo and in-vivo experiments,considering possible risks,including scaffold movement,over-heating,image artifact,and possible vessel injury,under typical MR condition.Traditional ASTM standards for MR safety and compatibility evaluation of intravascular devices were referred,but not only limited to that.The unique time-relevant MR properties of bioresorbable scaffolds were also discussed.Possible forces imposed on the scaffold during MR scanning and MR image artifacts gradually decreased along with scaffold degradation/absorption.Rigorous experiments designed based on a scientifically based rationale revealed that the IBS scaffold is MR conditional,though not MR compatible before complete absorption.The methodology used in the present study can give insight into the MR evaluation of magnetic scaffolds(bioresorbable)or stents(permanent).Dong Bian Li Qin Wenjiao Lin Danni Shen Haiping Qi Xiaoli Shi Gui Zhang Hongwei Liu Han Yang Jin Wang Deyuan Zhang Yufeng Zheng 2020Bioactive Materials2020,5,2:2

11Atomically Dispersed Fe-N_4 Modified with Precisely Located S for Highly Efficient Oxygen Reduction显示文摘Immobilizing metal atoms by multiple nitrogen atoms has triggered exceptional catalytic activity toward many critical electrochemical reactions due to their merits of highly unsaturated coordination and strong metal-substrate interaction.Herein,atomically dispersed Fe-NC material with precise sulfur modification to Fe periphery(termed as Fe-NSC) was synthesized,X-ray absorption near edge structure analysis confirmed the central Fe atom being stabilized in a specific configuration of Fe(N3)(N-C-S).By enabling precisely localized S doping,the electronic structure of Fe-N4 moiety could be mediated,leading to the beneficial adjustment of absorption/desorption properties of reactant/intermediate on Fe center.Density functional theory simulation suggested that more negative charge density would be localized over Fe-N4 moiety after S doping,allowing weakened binding capability to *OH intermediates and faster charge transfer from Fe center to O species.Electrochemical measurements revealed that the Fe-NSC sample exhibited significantly enhanced oxygen reduction reaction performance compared to the S-free Fe-NC material(termed as Fe-NC),showing an excellent onset potential of 1.09 V and half-wave potential of 0.92 V in 0.1 M KOH.Our work may enlighten relevant studies regarding to accessing improvement on the catalytic performance of atomically dispersed M-NC materials by managing precisely tuned local environments of M-Nx moiety.Yin Jia Xuya Xiong Danni Wang Xinxuan Duan Kai Sun Yajie Li Lirong Zheng Wenfeng Lin Mingdong Dong Guoxin Zhang Wen Liu Xiaoming Sun 2020Nano-Micro Letters2020,12,9:1
