| 2 | Distinct distribution revealing multiple bacterial sources for 1-Omonoalkyl glycerol ethers in terrestrial and lake environments显示文摘The 1-O-monoalkyl glycerol ethers(MAGEs) were initially viewed as the biomarkers for sulfate-reducing bacteria(SRB) mediating anaerobic oxidation of methane in the marine environments. However, limited information is known about their distribution in terrestrial and other aquatic settings including soils, fresh water lakes, and cave sediments, which may obscure our understanding of their biological sources. Here we found that MAGEs were ubiquitous but differed obviously in distributional pattern among those environments. The surface soils are dominated generally by i C15:0-MAGE, followed by n C16:0-MAGE whereas the lake sediments show the opposite, resulting in significantly higher i C15:0/n C16:0 ratios in soils than in lake sediments. The cave deposits are characterized by considerably higher proportions of branched MAGEs than the former two environments. The logarithm of i C15:0/a C15:0 ratio shows a significant negative correlation with soil p H, likely reflecting an adaptation of microbial cell membrane to change in the ambient proton concentration. The MAGE profiles in cultured bacteria cannot fully explain the MAGE distribution in all the samples analyzed. Therefore, MAGEs in soil, lake sediments, and cave deposits likely have additional biological source(s) other than SRB and cultured MAGE-producing bacteria. Based on their own actual conditions, major colleges and universities have implemented different talent training modes, including talent training modes of running talent classes, '2+1+1', 'Chinese Medicine Research Practice Class', employment-oriented professional talent training mode, three-level three-type diversified talent training mode, and talent training mode oriented with adapting to the characteristics of ethnic regions. Various talent training modes have different training objectives, and corresponding training programs are formulated to meet the requirements of the National Standard for the Teaching Quality of Undergraduate Majors in Ordinary Colleges and Universities. | 12 | A telomere-to-telomere reference genome provides genetic insight into the pentacyclic triterpenoid biosynthesis in Chaenomeles speciosa显示文摘Chaenomeles speciosa(2n=34),a medicinal and edible plant in the Rosaceae,is commonly used in traditional Chinese medicine.To date,the lack of genomic sequence and genetic studies has impeded efforts to improve its medicinal value.Herein,we report the use of an integrative approach involving PacBio HiFi(third-generation)sequencing and Hi-C scaffolding to assemble a high-quality telomere-to-telomere genome of C.speciosa.The genome comprised 650.4 Mb with a contig N50 of 35.5 Mb.Of these,632.3 Mb were anchored to 17 pseudo-chromosomes,in which 12,4,and 1 pseudo-chromosomes were represented by a single contig,two contigs,and four contigs,respectively.Eleven pseudo-chromosomes had telomere repeats at both ends,and four had telomere repeats at a single end.Repetitive sequences accounted for 49.5%of the genome,while a total of 45515 protein-coding genes have been annotated.The genome size of C.speciosa was relatively similar to that of Malus domestica.Expanded or contracted gene families were identified and investigated for their association with different plant metabolisms or biological processes.In particular,functional annotation characterized gene families that were associated with the biosynthetic pathway of oleanolic and ursolic acids,two abundant pentacyclic triterpenoids in the fruits of C.speciosa.Taken together,this telomere-to-telomere and chromosome-level genome of C.speciosa not only provides a valuable resource to enhance understanding of the biosynthesis of medicinal compounds in tissues,but also promotes understanding of the evolution of the Rosaceae. | Shaofang He Duanyang Weng Yipeng Zhang Qiusheng Kong Keyue Wang Naliang Jing Fengfeng Li Yuebin Ge Hui Xiong Lei Wu De-Yu Xie Shengqiu Feng Xiaqing Yu Xuekui Wang Shaohua Shu Zhinan Mei | 2023 | Horticulture Research2023,10,10: | 0 |

| 14 | Spectral Characteristics and Quantitative Analysis of Troxerutin-Cu^2+ Complex显示文摘[Objectives]This study aims to establish a simple,convenient,fast,low-cost,low-consumption and efficient method for quality control of troxerutin.[Methods]In NaAc-phosphate buffer at pH=8,troxerutin could react with Cu^2+to form complex,which had a characteristic absorption peak at 408 nm in the ultraviolet spectrum.The content of troxerutin could be determined indirectly by measuring the absorbance of the complex.[Results]Within the linear range of 27.82-74.19μg/mL,the absorbance of the complex has a good linear relationship with the concentration of troxerutin.The regression equation is A=0.0119C-0.1384(r=0.9991),and the detection limit is 1.59μg/mL.This method used to determine the content of troxerutin in samples from different manufacturers,and the recovery is 100.9%-102.7%.[Conclusions]The ultraviolet spectrophotometry is used in this experiment to indirectly determine the content of troxerutin by measuring the absorbance of troxerutin-Cu^2+complex,and the results are accurate,reproducible and highly precise.This method can be used for quantitative analysis of troxerutin. | Yu LIN Liujun LIANG Wen LIU Zhenzhen PAN Fengfeng XIE | 2020 | Asian Agricultural Research2020,12,3: | 0 |
| 15 | Safety Limit Test of Heavy Metals and Pesticide Residues in the Zhuang Ethnic Medcine Herba Polygoni Chinensis from 13 Producing Areas显示文摘[Objectives]This study was conducted to determine the contents of such five heavy metals as lead(Pb),cadmium(Cd),arsenic(As),mercury(Hg)and copper(Cu)and the contents of nine kinds of organochlorine pesticide residues including total BHC(α-BHC,β-BHC,γ-BHC,δ-BHC)and total DDT(pp-DDE,pp-DDD,op-DDT,pp-DDT)and pentachloronitrobenzene in the Zhuang medicine Herba Polygoni Chinensis from 13 producing areas.[Methods]The five heavy metals were determined in accordance with the second method of general rule 2341,the inductively coupled plasma mass spectrometry method,in part 4 of the 2020 edition of Chinese Pharmacopoeia;and the nine organochlorines were determined in accordance with the first method of general rule 2341,organochlorine pesticide residue determination method(chromatography),in part 4 of the 2020 edition of Chinese Pharmacopoeia.[Results]In the Herba Polygoni Chinensis from 13 origins,the lead contents were in the range of 0.50-3.28 mg/kg;the cadmium contents were in the range of 0.06-4.21 mg/kg;the arsenic contents were in the range of 0.16-6.9 mg/kg;the copper contents were in the range of 10.5-45.1 mg/kg;the mercury contents were in the range of 0.00-0.003 mg/kg;and the nine organochlorines including the total benzene hexachloride(α-BHC,β-BHC,γ-BHC,δ-BHC),total DDT(pp-DDE,pp-DDD,op-DDT,pp-DDT)and pentachloronitrobenzene were all not detected.Referring to part 4 of the 2020 edition of Chinese Pharmacopoeia,the Cu and As of the Herba Polygoni Chinensis produced in Hezhou exceeded the standards,the Cu of the Herba Polygoni Chinensis produced in Wuzhou exceeded the standard,and the As of the Herba Polygoni Chinensis produced in Yulin City and Guilin City exceeded the standard.[Conclusions]This study provides safety limit test data for the development and utilization of Herba Polygoni Chinensis of the Zhuang medicine. | Fengfeng XIE Anda WEI Liba XU Hua ZHU Guodong HUANG Shiyan HE Linmiao LI | 2021 | Agricultural Biotechnology2021,10,3: | 0 |

[Methods] The M. convoluta genomic DNA was extracted by improved CTAB method,and the SSR primers for M. convoluta were selected through digestion,ligation,suppression PCR,pre-amplification,SAM method and PCR. [Results] The improved CTAB method extracted a high purity of DNA; the SAM PCR method gave clear bands; and 100 positive clones were sequenced,obtaining 86 sequences.
