1Mammalian target of rapamycin pathway inhibition enhances the effects of 5-aza-dC on suppressing cell proliferation in human gastric cancer cell lines显示文摘The present study aimed to evaluate the relationship between mTOR signaling pathway and DNA methylation in cell survival,cell cycle,gene expression and protein level on human gastric cancer cells. Human gastric cancer cell lines,MKN45 and SGC7901 were treated with 5-aza-dC,rapamycin and/or LY294002.Cell viability was analyzed by MTT.Cell cycle distribution was evaluated by flow cytometry (FCM).The transcription level of PTEN and p27 Kip1 genes was detected by using real-time PCR.Protein expressions were detected by Western blotting.We found that cell viability was moderately reduced when treated with 5-aza-dC alone,but remarkably reduced when mTOR pathway was inhibited together (P<0.01).mTOR inhibition enhances the effects of 5-aza-dC on arresting cell cycle at G2 phase in human gastric cancer cell lines.The expression of PTEN and p27 Kip1 mRNA was remarkably increased in the gastric cancer cells treated with combind drugs(P<0.01).Phosphorylation of Akt,p70S6K and 4E-BP1 were significantly reduced in the cells treated with LY294002 or RAPA(P<0.01),but we failed to find that 5-aza-dC enhance these effects.We suggested that mTOR inhibition could enhance the effects of 5-aza-dC on suppressing cell proliferation and arresting cell cycle in human gastric cancer cell lines, which might be a potential target for tumor therapy.SUN DanFeng,TIAN XiaoQing,ZHANG YanJie,CHEN YingXuan,LU Rong,WANG Xia &FANG JingYuan Shanghai Jiao-Tong University Sc0hool of Medicine Renji Hospital,Shanghai Institute of Digestive Disease,Shanghai 200001, China 2008Science China(Life Sciences)2008,51,7:12
2Percent free prostate-specific antigen for prostate cancer diagnosis in Chinese men with a PSA of 4.0-10.0 ng/mL:Results from the Chinese Prostate Cancer Consortium显示文摘Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort with a PSA level of 4.0e10.0 ng/mL in China.Methods:Consecutive patients with a PSA of 4.0-10.0 ng/mL who underwent transrectal ultrasound-guided biopsy were enrolled at 16 Chinese medical centers from January 1st,2010 to December 31st,2013.Total and free serum PSA determinations were performed using three types of electro-chemiluminescence immunoassays recalibrated to the World Health Organization(WHO)standard.The diagnostic accuracy of PSA,%fPSA,and %fPSA in combination with PSA(%fPSA t PSA)was determined using the area under the receiver operating characteristic(ROC)curve(AUC).Results:A total of 2310 consecutive men with PSA levels between 4.0 and 10.0 ng/mL were included,and the detection rate of PCa was 25.1%.The AUC of%fPSA and %fPSA t PSA in predicting any PCa was superior to PSA alone in men aged≥60 years(0.623 vs.0.534,p<0.0001)but not in men aged 40e59 years(0.517 vs.0.518,p=0.939).Similar result was yield in predicting HGPCa.Conclusion:In a clinical setting of Chinese men with 4.0e10.0 ng/mL PSA undergoing initial prostate biopsy,adding %fPSA to PSA can moderately improve the diagnostic accuracy for any PCa and HGPCa compared with PSA alone in patients≥60 but not in patients aged 40-59 years.Rui Chen Liping Xie Xiaobing Cai Yiran Huang Liqun Zhou Lulin Ma Xu Gao Chuanliang Xu Shancheng Ren Pengfei Shao Danfeng Xu Kexin Xu Zhangqun Ye Chunxiao Liu Dingwei Ye Li Lu Qiang Fu Jianquan Hou Jianlin Yuan Dalin He Tie Zhou Fubo Wang Biming He Yinghao Sun 2015Asian Journal of Urology2015,2,2:8
3High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat.Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu 2020Molecular Plant2020,13,9:8
4Construction of heparin-based hydrogel incorporated with Cu_(5.4)O ultrasmall nanozymes for wound healing and inflammation inhibition显示文摘Excessive production of inflammatory chemokines and reactive oxygen species(ROS)can cause a feedback cycle of inflammation response that has a negative effect on cutaneous wound healing.The use of wound-dressing materials that simultaneously absorb chemokines and scavenge ROS constitutes a novel‘weeding and uprooting’treatment strategy for inflammatory conditions.In the present study,a composite hydrogel comprising an amine-functionalized star-shaped polyethylene glycol(starPEG)and heparin for chemokine sequestration as well as Cu_(5.4)O ultrasmall nanozymes for ROS scavenging(Cu_(5.4)O@Hep-PEG)was developed.The material effectively adsorbs the inflammatory chemokines monocyte chemoattractant protein-1 and interleukin-8,decreasing the migratory activity of macrophages and neutrophils.Furthermore,it scavenges the ROS in wound fluids to mitigate oxidative stress,and the sustained release of Cu_(5.4)O promotes angiogenesis.In acute wounds and impaired-healing wounds(diabetic wounds),Cu_(5.4)O@Hep-PEG hydrogels outperform the standard-of-care product Promogram®in terms of inflammation reduction,increased epidermis regeneration,vascularization,and wound closure.Yuan Peng Danfeng He Xin Ge Yifei Lu Yuanhao Chai Yixin Zhang Zhengwei Mao Gaoxing Luo Jun Deng Yan Zhang 2021Bioactive Materials2021,6,10:7
5Cytoplasmic and nuclear genome variations of rice hybrids and their parents inform the trajectory and strategy of hybrid rice breeding显示文摘The male sterility(MS)line is a prerequisite for efficient production of hybrid seeds in rice,a self-pollinating species.MS line breeding is pivotal for hybrid rice improvement.Understanding the historical breeding tra-jectory will help to improve hybrid rice breeding strategies.Maternally inherited cytoplasm is an appro-priate tool for phylogenetic reconstruction and pedigree tracing in rice hybrids.In this study,we analyzed the cytoplasmic genomes of 1495 elite hybrid rice varieties and identified five major types of cytoplasm,which correspond to different hybrid production systems.As the cytoplasm donors for hybrids,461 MS lines were also divided into five major types based on cytoplasmic and nuclear genomic architecture.Spe-cific core accessions cooperating with different fertility-associated genes drove the sequence divergence of MS lines.Dozens to hundreds of convergent and divergent selective sweeps spanning several agro-nomic trait-associated genes were identified among different types of MS lines.We further analyzed the cross patterns between different types of MS lines and their corresponding restorers.This study system-atically analyzed the cytoplasmic genomes of rice hybrids revealed their relationships with nuclear ge-nomes of MS lines,and illustrated the trajectory of hybrid rice breeding and the strategies for breeding different types of MS lines providing new insights for future improvement of hybrid rice.Zhoulin Gu Zhou Zhu Zhen Li Qilin Zhan Qi Feng Congcong Zhou Qiang Zhao Yan Zhao Xiaojian Peng Bingxin Dai Rongrong Sun Yan Li Hengyun Lu Lei Zhang Tao Huang Junyi Gong Danfeng Lv Xuehui Huang Bin Han 2021Molecular Plant2021,14,12:2

15Portable visual and electrochemical detection of hydrogen peroxide release from living cells based on dual-functional Pt-Ni hydrogels显示文摘It is important to monitor the intra-/extracellular concentration of hydrogen peroxide(H_(2)O_(2))in biological processes.However,miniaturized devices that enable portable and accurate H_(2)O_(2)measurement are still in their infancy because of the difficulty of developing facile sensing strategies and highly integrated sensing devices.In this work,portable H_(2)O_(2)sensors based on Pt-Ni hydrogels with excellent peroxidase-like and electrocatalytic activities are demonstrated.Thus,simple and sensitive H_(2)O_(2)sensing is achieved through both colorimetric and electrochemical strategies.The as-fabricated H_(2)O_(2)sensing chips exhibit favorable performance,with low detection limits(0.030μM&0.15μM),wide linearity ranges(0.10μM-10.0 mM&0.50μM-5.0 mM),outstanding long-term stability(up to 60 days),and excellent selectivity.With the aid of an M5stack development board,portable visual and electrochemical H_(2)O_(2)sensors are successfully constructed without complicated and expensive equipment or professional operators.When applied to the detection of H_(2)O_(2)released from HeLa cells,the results obtained by the developed sensors are in good agreement with those from an ultraviolet‒visible spectrophotometer(UV‒vis)(1.97μM vs.2.08μM)and electrochemical station(1.77μM vs.1.84μM).Guanglei Li Yao Chen Fei Liu Wenhua Bi Chenxin Wang Danfeng Lu Dan Wen 2023Microsystems & Nanoengineering2023,9,6:0
