| 1 | Yu Gan Long Ameliorates Hepatic Fibrosis by Inhibiting PI3K/AKT,Ras/ERK and JAK1/STAT3 Signaling Pathways in CCl4-induced Liver Fibrosis Rats显示文摘Yu Gan Long(YGL)is a Chinese traditional herbal formula which has been reported to attenuate liver fibrosis for many years and we have explored its anti-fibrotic mechanism through blocking transforming growth factor(TGF-β)in the previous study.But the mechanisms associated with platelet-derived growth factor(PDGF)-BB remain obscure.In this study,we further investigated the mechanism of YGL reducing carbon tetrachloride(CCl4)-induced liver fibrosis in rats.Our results showed that YGL suppressed CCl4-induced upregulation of collagen IV(Col IV),type HI precollagen(PCHI),hyaluronuc acid(HA)and laminin(LN),which are implicated in liver fibrosis.Also,YGL reduced theα-smooth muscle actin(α-SMA)expression,which acts as the indicator of liver fibrosis.Furthermore,YGL decreased the serum levels of hepatic stellate cell(HSC)mitogen PDGF-BB and inflammation cytokines,including TNF-α,IL-1β,IL-6.Markers involved in liver fibrosis,such as Ras,p-Raf-1,p-ERK1/2,p-JNK,p-P38,p-PI3K,p-AKT,p-JAKl,p-STAT3 were downregulated significantly after treatment with YGL.Our results indicated that YGL ameliorated CCl4-induced liver fibrosis by reducing inflammation cytokines production,and suppressing Ras/ERK,PI3K/AKT,and JAK1/STAT3 signaling pathways,which provided further evidence towards elucidation of the anti-fibrotic mechanism of YGL. | Hou-gang LI Peng-tao YOU Yu XI Yu CAI Yi-jun TU Meng-heng WANG Wan-ci SONG Tai-min QUAN Hui-ying REN Yan-wen LIU Han-xiong DAN Shi-qing XU | 2020 | Current Medical Science2020,40,3: | 9 |
| 2 | Extending optical filaments with phase-nested laser beams显示文摘Extending the length of femtosecond laser filamentation has always been desired for practical applications. Here,we demonstrate that significant extending of a single filament in BK7 glass can be achieved by constructing phasenested beams. The filamentation and the following energy replenishment are assembled in a single phase-nested beam. The central part of the phase-nested beam is an apertured Gaussian beam, which is focused into one focal spot to produce a short filament. In contrast, the rest of the annular part converges gradually towards the central axis to continuously replenish the energy for supporting the regeneration of filaments. The common-path generating system ensures the stability of generated filaments and easily optimizes the beam parameters to obtain the longest high-quality filament due to its flexibility. | 5 | Suppression of RNA Interference Pathway in vitro by Grass Carp Reovirus显示文摘The means of survival of genomic dsRNA of reoviruses from dsRNA-triggered and Dicer-initiated RNAi pathway remains to be defined. The present study aimed to investigate the effect of Grass carp reovirus (GCRV) replication on the RNAi pathway of grass carp kidney cells (CIK). The dsRNA-triggered RNAi pathway was demonstrated unimpaired in CIK cells through RNAi assay. GCRV-specific siRNA was generated in CIK cells transfected with purified GCRV genomic dsRNA in Northern blot analysis; while in GCRV-infected CIK cells, no GCRV-specific siRNA could be detected. Infection and transfection experiments further indicated that replication of GCRV correlated with the increased transcription level of the Dicer gene and functional inhibition of in vitro synthesized egfp-siRNA in silencing the EGFP reporter gene. | 9 | Kinetic Characteristics of Neutralizing Antibody Responses Vary among Patients with COVID-19显示文摘Objective The coronavirus disease 2019(COVID-19) pandemic continues to present a major challenge to public health. Vaccine development requires an understanding of the kinetics of neutralizing antibody(NAb) responses to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Methods In total, 605 serum samples from 125 COVID-19 patients(from January 1 to March 14, 2020)varying in age, sex, severity of symptoms, and presence of underlying diseases were collected, and antibody titers were measured using a micro-neutralization assay with wild-type SARS-CoV-2.Results NAbs were detectable approximately 10 days post-onset(dpo) of symptoms and peaked at approximately 20 dpo. The NAb levels were slightly higher in young males and severe cases, while no significant difference was observed for the other classifications. In follow-up cases, the NAb titer had increased or stabilized in 18 cases, whereas it had decreased in 26 cases, and in one case NAbs were undetectable at the end of our observation. Although a decreasing trend in NAb titer was observed in many cases, the NAb level was generally still protective.Conclusion We demonstrated that NAb levels vary among all categories of COVID-19 patients. | 10 | Mechanistic analysis for the origin of diverse diterpenes in Tripterygium wilfordii显示文摘Tripterygium wilfordii is a valuable medicinal plant rich in biologically active diterpenoids,but there are few studies on the origins of these diterpenoids in its secondary metabolism.Here,we identified three regions containing tandemly duplicated diterpene synthase genes on chromosomes(Chr) 17 and 21 of T. wilfordii and obtained 11 diterpene synthases with different functions.We farther revealed that these diterpene synthases underwent duplication and rearrangement at approximately 2.3-23.7 million years ago(MYA) by whole-genome triplication(WGT),transposon mediation,and tandem duplication,followed by functional divergence.We first demonstrated that four key amino acids in the sequences of TwCPS3,TwCPS5,and TwCPSS were altered during evolution,leading to their functional divergence and the formation of diterpene secondary metabolites.Then,we demonstrated that the functional divergence of three TwKSLs was driven by mutations in two key amino acids.Finally,we discovered the mechanisms of evolution and pseudogenization of miltiradiene synthases in T.wilfordii and elucidated that the new function in TwMS1/2 from the terpene synthase(TPS)-b subfamily was caused by progressive changes in multiple amino acids after the WGT event.Our results provide key evidence for the formation of diverse diterpenoids during the evolution of secondary metabolites in T.wilfordii. | Lichan Tu Xinbo Cai Yifeng Zhang Yuru Tong Jian Wang Ping Su Yun Lu Tianyuan Hu Yunfeng Luo Xiaoyi Wu Dan Li Luqi Huang Wei Gao | 2022 | Acta Pharmaceutica Sinica B2022,12,6: | 1 |

| 15 | Collusion Potential Assessment in Electricity Markets Considering Generation Flexibility显示文摘The collusion among various generating units has been a problematic issue affecting the fairness and transparency of electricity markets.Therefore,it is of great significance to assess the potential of such collusion in the electricity market.However,the previous assessment studies primarily focused on the bidding behaviors of collusive generating units,without considering the influences of generation flexibility,such as ramp rates.In this paper,a novel assessment method is proposed to evaluate the collusion potential in the electricity market considering generation flexibility.First,a bi-level optimization model is developed to simulate the collusive strategies of dif-ferent generating units,including the withholding of generation capacities and ramp rates,as well as the uplifting of minimum outputs and bidding prices.In the upper-level problem,collusive generating units optimize their offering strategies to optimize the generation profits without violating the regulatory laws.The lower-level problem is a day-ahead economic dispatch model which minimizes the dispatching costs.Based on the optimal collusive strategies determined by the bi-level model,a framework is then proposed to assess the collusion potential in electricity markets.Moreover,price-based and profit-based indices are proposed to quantitatively evaluate the collusion potential of different generating units.Finally,the proposed assessment method is validated on a modified IEEE 39-node system.The numerical results demonstrated that generation flexibility can be exploited collusively for making excessive profits,particularly during load peaks and valleys.Index Terms-Collusionpotential,economic cdispatch,generation flexibility,ramp rates,strategic withholding. | Teng Tu Yi Ding Peng Ji Minglei Bao Nan Shang Yonghua Song Dan Zeng | 2023 | CSEE Journal of Power and Energy Systems2023,9,4: | 1 |
