3MicroTom Metabolic Network: Rewiring Tomato Metabolic Regulatory Network throughout the Growth Cycle显示文摘Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences between varieties and species,the dynamics of metabolic changes during the tomato growth cycle and the regulatory networks that underlie these changes are poorly understood.In this study,we integrated high-resolution spatio-temporal metabolome and transcriptome data to systematically explore the metabolic landscape across 20 major tomato tissues and growth stages.In the resulting MicroTom Metabolic Network,the 540 detected metabolites and their co-expressed genes could be divided into 10 distinct clusters based on their biological functions.Using this dataset,we constructed a global map of the major metabolic changes that occur throughout the tomato growth cycle and dissected the underlying regulatory network.In addition to verifying previously well-established regulatory networks for important metabolites,we identified novel transcription factors that regulate the biosynthesis of important secondary metabolites such as steroidal glycoalkaloids and flavonoids.Our findings provide insights into spatiotemporal changes in tomato metabolism and generate a valuable resource for the study of metabolic regulatory processes in model plants.Yan Li Yang Chen Lu Zhou Shengjie You Heng Deng Ya Chen Saleh Alseekh Yong Yuan Rao Fu Zixin Zhang Dan Su Alisdair R.Fernie Mondher Bouzayen Tao Ma Mingchun Liu Yang Zhang 2020Molecular Plant2020,13,8:10
4Restoration of electrical microenvironment enhances bone regeneration under diabetic conditions by modulating macrophage polarization显示文摘Macrophage-mediated inflammation compromises bone repair in diabetic patients.Electrical signaling cues are known to regulate macrophage functions.However,the biological effects of electrical microenvironment from charged biomaterials on the immune response for regulating osteogenesis under diabetic conditions remain to be elucidated.Herein the endogeneous electrical microenvironment of native bone tissue was recapitulated by fabricating a ferroelectric BaTiO_(3)/poly(vinylidene fluoridetrifluoroethylene)(BTO/P(VDF-TrFE))nanocomposite membrane.In vitro,the polarized BaTiO_(3)/poly(vinylidene fluoridetrifluoroethylene)(BTO/P(VDF-TrFE))nanocomposite membranes inhibited high glucose-induced M1-type inflammation,by effecting changes in cell morphology,M1 marker expression and pro-inflammatory cytokine secretion in macrophages.This led to enhanced osteogenic differentiation of human bone marrow mesenchymal stem cells(BM-MSCs).In vivo,the biomimetic electrical microenvironment recapitulated by the polarized nanocomposite membranes switched macrophage phenotype from the pro-inflammatory(M1)into the pro-healing(M2)phenotype,which in turn enhanced bone regeneration in rats with type 2 diabetes mellitus.Mechanistic studies revealed that the biomimetic electrical microenvironment attenuated pro-inflammatory M1 macrophage polarization under hyperglycemic conditions by suppressing expression of AKT2 and IRF5 within the PI3K-AKT signaling pathway,thereby inducing favorable osteo-immunomodulatory effects.Our study thus provides fundamental insights into the biological effects of restoring the electrical microenvironment conducive for osteogenesis under DM conditions,and offers an effective strategy to design functionalized biomaterials for bone regeneration therapy in diabetic patients.Xiaohan Dai Boon Chin Heng Yunyang Bai Fuping You Xiaowen Sun Yiping Li Zhangui Tang Mingming Xu Xuehui Zhang Xuliang Deng 2021Bioactive Materials2021,6,7:9

6Interaction between Schwann cells and other cells during repair of peripheral nerve injury显示文摘Peripheral nerve injury(PNI)is common and,unlike damage to the central nervous system injured nerves can effectively regenerate depending on the location and severity of injury.Peripheral myelinating glia,Schwann cells(SCs),interact with various cells in and around the injury site and are important for debris elimination,repair,and nerve regeneration.Following PNI,Wallerian degeneration of the distal stump is rapidly initiated by degeneration of damaged axons followed by morphologic changes in SCs and the recruitment of circulating macrophages.Interaction with fibroblasts from the injured nerve microenvironment also plays a role in nerve repair.The replication and migration of injury-induced dedifferentiated SCs are also important in repairing the nerve.In particular,SC migration stimulates axonal regeneration and subsequent myelination of regenerated nerve fibers.This mobility increases SC interactions with other cells in the nerve and the exogenous environment,which influence SC behavior post-injury.Following PNI,SCs directly and indirectly interact with other SCs,fibroblasts,and macrophages.In addition,the inter-and intracellular mechanisms that underlie morphological and functional changes in SCs following PNI still require further research to explain known phenomena and less understood cell-specific roles in the repair of the injured peripheral nerve.This review provides a basic assessment of SC function post-PNI,as well as a more comprehensive evaluation of the literature concerning the SC interactions with macrophages and fibroblasts that can influence SC behavior and,ultimately,repair of the injured nerve.Wen-Rui Qu Zhe Zhu Jun Liu De-Biao Song Heng Tian Bing-Peng Chen Rui Li Ling-Xiao Deng 2021Neural Regeneration Research2021,16,1:8

8Alterations of Alternative Splicing Patterns of Ser/Arg-Rich (SR) Genes in Response to Hormones and Stresses Treatments in Different Ecotypes of Rice (Oryza sativa)显示文摘Ser/Arg-rich (SR) genes encode proteins that play pivotal roles in both constitutive and alternative splicing of pre-mRNA. However, not much effort has been made to investigate the alternative splicing of their own pre-mRNA. In this study, we conducted comprehensive analyses of pre-mRNA splicing for 22 SR genes in three rice (Oryza sativa L.) ecotypes indica, japonica and javanica. Using different ecotypes we characterized the variations in expression and splicing patterns of rice SR genes in different tissues and at different developmental stages. In addition, we compared the divergence in expression and splicing patterns of SR genes from seedlings of different rice ecotypes in response to hormones application and environmental stresses. Our results revealed the complexity of alternative splicing of SR genes in rice. The splicing varies in different tissues, in different ecotypes, in response to stresses and hormones. 11Matrix stiffness modulates tip cell formation through the p-PXN-Rac1-YAP signaling axis显示文摘Endothelial tip cell outgrowth of blood-vessel sprouts marks the initiation of angiogenesis which is critical in physiological and pathophysiological procedures.However,how mechanical characteristics of extracellular matrix(ECM)modulates tip cell formation has been largely neglected.In this study,we found enhanced CD31 expression in the stiffening outer layer of hepatocellular carcinoma than in surrounding soft tissues.Stiffened matrix promoted sprouting from endothelial cell(EC)spheroids and upregulated expressions of tip cell-enriched genes in vitro.Moreover,tip cells showed increased cellular stiffness,more actin cytoskeleton organization and enhanced YAP nuclear transfer than stalk and phalanx ECs.We further uncovered that substrate stiffness regulates FAK and Paxillin phosphorylation in focal adhesion of ECs promoting Rac1 transition from inactive to active state.YAP is subsequently activated and translocated into nucleus,leading to increased tip cell specification.p-Paxillin can also loosen the intercellular connection which also facilitates tip cell specification.Collectively our present study shows that matrix stiffness modulates tip cell formation through p-PXN-Rac1-YAP signaling axis,shedding light on the role of mechanotransduction in tip cell formation.This is of special significance in biomaterial design and treatment of some pathological situations.Yaru Guo Feng Mei Ying Huang Siqin Ma Yan Wei Xuehui Zhang Mingming Xu Ying He Boon Chin Heng Lili Chen Xuliang Deng 2022Bioactive Materials2022,7,1:4
12Diagnostic and Prognostic Value of Plasma Factor V Activity and Parameters in Thrombin Generation for Disseminated Intravascular Coagulation in Patients with Hematological Malignancies显示文摘In this study,we used plasma factor V activity and parameters of the thrombin generation test to discuss their diagnostic and prognostic value for disseminated intravascular coagulation (DIC) in patients with hematological malignancies.A total of 164 patients who were diagnosed with hematological malignancies in the Department of Hematology,Union Hospital,between Apr 2014 and Dec.2014 were enrolled in this study.There were 131 patients in the study group and 33 patients in the control group in terms of the laboratory results for DIC.The patients in the study group were divided into a DIC subgroup (n=59) and a non-DIC subgroup (n=72) based on the International Society of Thrombosis and Hemostasis (ISTH) Integral System,and they were divided into four subgroups [score ≤3 (n=35),score=4 (n=37),score=5 (n=47),and score >6 (n=12)] according to ISTH scores.Using 28-day mortality as the endpoint,the patients in the study group were divided into a survival subgroup (n=111) and a non-survival subgroup (n=20).The results showed that the plasma factor V activity was significantly weaker,and lag time and time to peak were significantly shorter in the study group than in the control group (P<0.01).The factor V activity,peak and endogenous thrombin potential (ETP) were significantly decreased in the DIC subgroup as compared with those in the non-DIC subgroup (P<0.01).Among factor V activity,lag time,peak,ETP,and ttPeak,only the factor V activity was significantly decreased in the nonsurvival subgroup compared with the survival subgroup (P<0.01).With the increase in ISTH score,the ETP and peak decreased gradually.The binary logistic regression analysis revealed that PLT,D-dimer,factor V activity and ETP had linear relationship with DIC diagnosed by ISTH Integral System.Using DIC diagnosed by ISTH Integral System as the endpoint,the area under curve (AUC) of factor V activity was found to be similar to that of blood platelet count (PLT) and prothrombin time (PT).In conclusion,factor V activity,ETP and peak had diagnostic value for DIC in patients with hematological malignancies,and only factor V activity had limited prognostic value.Hai-ming K0U Xiao-ping ZHANG Man-zhi WANG Jun DENG Heng MEI Yu HU 2019Current Medical Science2019,39,4:3
13The membrane-associated E3 ubiquitin ligase MARCH3 downregulates the IL-6 receptor and suppresses colitis-associated carcinogenesis显示文摘The IL-6-STAT3 axis is critically involved in inflammation-associated carcinogenesis(IAC).How this axis is regulated to modulate IAC remains unknown.Here,we show that the plasma membrane-associated E3 ubiquitin ligase MARCH3 negatively regulates STAT3 activation triggered by IL-6,as well as another IL-6 subfamily member,Oncostatin M(OSM).MARCH3 is associated with the IL-6 receptorα-chain(IL-6Rα)and its coreceptor gp130.Biochemical experiments indicated that MARCH3 mediates the polyubiquitination of IL-6Rαat K401 and gp130 at K849 following IL-6 stimulation,leading to their translocation to and degradation in lysosomes.MARCH3 deficiency increases IL-6-and OSM-triggered activation of STAT3 and induction of downstream effector genes in various cell types.MARCH3 deficiency enhances dextran sulfate sodium(DSS)-induced STAT3 activation,increases the expression of inflammatory cytokines,and exacerbates colitis,as well as azoxymethane(AOM)/DSS-induced colitis-associated cancer in mice.In addition,MARCH3 is downregulated in human colorectal cancer tissues and associated with poor survival across different cancer types.Our findings suggest that MARCH3 is a pivotal negative regulator of IL-6-induced STAT3 activation,inflammation,and inflammation-associated carcinogenesis.Heng Lin Lu Feng Kai-Sa Cui Lin-Wen Zeng Deng Gao Long-Xiang Zhang Wen-Hua Xu Yu-Hao Sun Hong-Bing Shu Shu Li 2021Cellular & Molecular Immunology2021,18,12:3

15Basement Structure and Properties of the Western Junggar Basin, China显示文摘A comprehensive geophysical profile stretching from Qingyijing at the southern edge of the Junggar Basin to Ubara on the northern margin of the Junggar Basin was conducted in an attempt to probe the crustal structure of the western Junggar Basin(hereafter referred to simply as ‘the Basin'), and, in particular, the structure and property of the Basin's crystalline basement. A survey using seismically converted waves was conducted along this profile to determine the characteristics of the P-and Swave velocities typical of the crust and uppermost mantle. A joint inversion of gravitation and aeromagnetic data was also performed to acquire the density and magnetization intensity values found beneath the western Basin. This research revealed that the Basin is composed of the so-called Manasi terrain in the south, and the Wulungu terrain in the north. Their boundary is located along the WNW-trending Dishuiquan-Sangequan suture, linking the NE-striking Da'erbute suture(DS) in the west, and the WNWtrending Kalameili suture(KS) in the east. In its northern part, the Wulungu-type terrain has a doublelayered basement, of which the upper layer is a folded basement of Hercynian orogenic origin, and the lower layer is a crystalline basement of Middle–Upper Proterozoic age. The southern part of the Basin, i.e., the Manasi terrain, has a single-layered crystalline basement. 17Generation of a series of mutant lines resistant to imidazolinone by screening an EMS-based mutant library in common wheat显示文摘The breeding of herbicide-resistant wheat varieties has helped control weeds in wheat fields economically and effectively.Imidazolinone (IMI) herbicides are popular as they have low toxicity in mammals,are effective at small doses,and exhibit broad-spectrum herbicidal action in the field.Therefore,the isolation and genetic and molecular characterization of IMI-resistant wheat mutants will enhance weed management in wheat fields.In the present study,352 IMI-resistant plants were isolated by genetic screening from a mutant pool prepared by EMS-based random mutagenesis.Cloning of the mutated genes from the IMI-resistant plants indicated that ten taals alleles had been isolated,and mutation in one of three Ta ALS homolog genes conferred IMI resistance,and such a mutation is a dominant trait.Further analysis showed that taals-d exhibited the greatest IMI resistance,whereas taals-b exhibited the weakest resistance to IMI among three homologous taals mutants.In terms of IMI resistance,the taals triple mutant was stronger than the taals double mutants,and the taals double mutants were stronger than the single mutants,indicating a dose-dependent effect of the Ta ALS mutation on IMI resistance in wheat.Biochemical analysis indicated that the mutation in Ta ALS increased the tolerance of Ta ALS to inhibition by IMI.Our work details the genetic and molecular characterization of als wheat mutants,provides a foundation for understanding IMI resistance and breeding wheat varieties with herbicide resistance,and indicates that genetic screening using a mutagenized pool is an effective and important means of breeding crops with additional desired agricultural traits.Zhuo Chen Zheng Wang Yanfang Heng Jian Li Jiawei Pei Ying Cao Xing Wang Deng Ligeng Ma 2021The Crop Journal2021,9,5:3

20Digital radiography of crush thoracic trauma in the Sichuan earthquake显示文摘AIM:To investigate the features of crush thoracic trauma in Sichuan earthquake victims using chest digital radiography(CDR).METHODS:We retrospectively reviewed 772 CDR of 417 females and 355 males who had suffered crush thoracic trauma in the Sichuan earthquake.Patient age ranged from 0.5 to 103 years.CDR was performed between May 12,2008 and June 7,2008.We looked for injury to the thoracic cage,pulmonary parenchyma and the pleura.obtained in 349 patients,the remaining 423 patients underwent only AP CDR.Thoracic cage fractures,pulmonary contusion and pleural injuries were noted in 331(42.9%;95% CI:39.4%-46.4%),67 and 135 patients,respectively.Of the 256 patients with rib fractures,the mean number of fractured ribs per patient was 3.Rib fractures were mostly distributed from the 3rd through to the 8th ribs and the vast majority involved posterior and lateral locations along the rib.Rib fractures had a significant positive association with non-rib thoracic fractures,pulmonary contusion and pleural injuries(P < 0.001).The number of rib fractures and pulmonary contusions were significant factors associated with patient death.CONCLUSION:Earthquake-related crush thoracic trauma has the potential for multiple fractures.The high number of fractured ribs and pulmonary contusions were significant factors which needed appropriate medical treatment.Zhi-Hui Dong Heng Shao Tian-Wu Chen Zhi-Gang Chu Wen Deng Si-Shi Tang Jing Chen Zhi-Gang Yang 2011World Journal of Radiology2011,3,11:2
