| 1 | Diabetes mellitus: The epidemic of the century显示文摘The epidemic nature of diabetes mellitus in different regions is reviewed. The Middle East and North Africa region has the highest prevalence of diabetes in adults(10.9%) whereas, the Western Pacific region has the highest number of adults diagnosed with diabetes and has countries with the highest prevalence of diabetes(37.5%). Different classes of diabetes mellitus, type 1, type 2, gestational diabetes and other types of diabetes mellitus are compared in terms of diagnostic criteria, etiology and genetics. The molecular genetics of diabetes received extensive attention in recent years by many prominent investigators and research groups in the biomedical field. A large array of mutations and single nucleotide polymorphisms in genes that play a role in the various steps and pathways involved in glucose metabolism and the development, control and function of pancreatic cells at various levels are reviewed. The major advances in the molecular understanding of diabetes in relation to the different types of diabetes in comparison to the previous understanding in this field are briefly reviewed here. Despite the accumulation of extensive data at the molecular and cellular levels, the mechanism of diabetes development and complications are still not fully understood. | 2 | Molecular detection of monocyte chemotactic protein-1 polymorphism in spontaneous bacterial peritonitis patients显示文摘AIM: To investigate the association of the functional monocyte chemotactic protein-1(MCP-1) promoter polymorphism(A-2518G) with spontaneous bacterial peritonitis(SBP).METHODS: Fifty patients with post-hepatitis C liver cirrhosis and ascites were categorized into two groups; group Ⅰ included 25 patients with SBP and group Ⅱ included 25 patients free from SBP. In addition, a group of 20 healthy volunteers were included. We assessed the MCP-1 gene polymorphism and gene expression as well as interleukin(IL)-10 levels in both blood and ascitic fluid. RESULTS: A significant MCP-1 gene polymorphism was detected in groups Ⅰ and Ⅱ(P = 0.001 and 0.02 respectively). Group Ⅰ was associated with a significantly higher frequency of AG genotype [control 8(40%) vs SBP 19(76.0%), P < 0.001], and group Ⅱ was associated with a significantly higher frequency of GG genotype when compared to healthy volunteers [control 1(5%) vs cirrhotic 16(64%), P < 0.001]. Accordingly, the frequency of G allele was significantly higher in both groups(Ⅰ and Ⅱ) [control 10(25%) vs SBP 27(54%), P < 0.001 and vs cirrhotic 37(74.0%), P < 0.001, respectively]. The total blood and ascetic fluid levels of IL-10 and MCP-1 gene expression were significantly higher in group Ⅰ than in group Ⅱ. Group Ⅰ showed significant reductions in the levels of MCP-1 gene expression and IL-10 in the whole blood and ascetic fluid after therapy. CONCLUSION: MCP-1 GG genotype and G allele may predispose HCV infected patients to a more progressive disease course, while AG genotype may increase the susceptibility to SBP. | 3 | GADD45β基因表达诱导对不同p53状态的肝癌细胞作用显示文摘目的:体外合成GADD45β基因全序列表达质粒后,研究GADD45β基因表达诱导对呈不同p53状态的肝癌细胞的作用及可能机制。方法:采用RT-PCR法获得GADD45β基因全序列,插入pDrive穿梭克隆载体和pIRES2-EGFP荧光表达载体后大量扩增获得DNA,结合p53全基因表达质粒pp53-EGFP转染HepG2、Hep3B细胞后,以[3H]胸腺嘧啶脱氧核苷掺入法(3H-Tdr)和细胞克隆形成法分析DNA合成变化及细胞生长能力;以双抗体夹心ELISA法测定TGF-β1表达变化。结果:成功合成GADD45β基因全序列和表达质粒,通过流式细胞仪收集转染阳性的荧光表达细胞能显著提高转染效率;转染GADD45β后,具有野生型p53基因的HepG2细胞的细胞克隆形成能力和DNA合成能力明显受到抑制,细胞凋亡明显增加,TGF-β1的表达亦明显受抑。与之相反,缺失p53基因的Hep3B需要同时共转染p53基因后,方出现抑制效应。结论:GADD45β基因能够有效抑制肝癌细胞的生长,其功能需要完整p53基因的辅助和(或)调控。GADD45表达和(或)功能异常,导致p53介导的DNA损伤修复途径异常或阻断,是肝脏细胞恶性转化及形成肿瘤的可能机制。 | 邱伟华 Dana Darwish Peiguo G.Chu Frank Luh 陈皓 邓侠兴 沈柏用 杨卫平 李宏为 Yun Yen | 2005 | 外科理论与实践2005，10，2： | 6 |
| 4 | STAT3在SAMe抑制HepG2细胞VEGF表达中的作用显示文摘目的研究肝癌细胞(HepG2)和正常胎肝细胞(CL-48)血管内皮生长因子(VEGF)和信号转导与转录激活因子3(STAT3)的表达状态,以及S-腺苷甲硫氨酸(SAM e)对其的干预作用,为HepG2和CL-48的不同调控途径提供功能性依据。方法在对数生长期的CL-48和HepG2细胞中加入不同浓度的SAM e(0、0.5、1.0 mmol/L)作用72 h,分别采用Northern印迹法、实时荧光定量PCR、W estern印迹法以及[3H]胸腺嘧啶脱氧核苷掺入法(3H-Tdr)和细胞克隆形成法,观察VEGF和STAT3表达的变化趋势,分析DNA合成及细胞生长能力的变化。结果SAM e作用前(SAM e 0 mmol/L),HepG2和CL-48均呈现VEGF和STAT3高表达状态。SAM e作用后:HepG2的VEGF和STAT3表达明显降低,并呈现剂量-效应关系,DNA合成和细胞生长能力明显受限;CL-48的STAT3表达虽然受到抑制,但VEGF的高表达状态无明显降低趋势,且DNA合成和细胞生长能力无显著变化。结论虽然HepG2和CL-48均呈现VEGF和STAT3的高表达,但二者具有不同的调控途径;SAM e通过抑制STAT3的组成性活化而降低VEGF的高表达状态,可能是SAM e的肝脏保护机制之一。 | 邱伟华 Bingsen Zhou Dana Darwish Peiguo G.Chu Frank Luh 陈皓 杨卫平 李宏为 Yun Yen | 2006 | 上海交通大学学报(医学版)2006，26，6： | 6 |

| 6 | Geochemical Stream Sediment Survey in the Wadi Umm Rilan Area,South Eastern Desert,Egypt:A New Occurrence for Gold Mineralization显示文摘The present paper investigates the stream sediment geochemistry,behavior of gold and associated elements,delineates Au-anomalous sites and defines related sources in the Wadi Umm Rilan area.Fifty three major and trace elements were analyzed using ICP-MS and treated applying various statistical and mapping techniques.The results showed a significant difference of mean and median Au and most chemical elements in the three portions of the area,and higher values were recorded in the western portion.Furthermore,Au-anomalous sites appeared in upper and lower parts of the Wadi Umm Rilan,along the tributaries of metavolcanic unit and near granitoid contacts.This indicates the main source of Au mineralization is related to emplacement of granitoid plutons and accompanying hydrothermal solutions.There are significant indications for the presence of more than one mineralization event forming a probable single major episode of mineralization in the area,involving Au,Pb and U mineralizations.Geology,geomorphologic aspects and weathering processes could control stream sediment geochemistry,anomalies of Au and associated elements,elemental association and their dispersion patterns.Therefore,the area is fruitful and regarded as a promising target for Au exploration,using Ag,As,Sb,Cd,Cs,and Tl as pathfinder elements. | Mohamed Abdallah Gad DARWISH | 2017 | Acta Geologica Sinica(English Edition)2017，91，3： | 2 |
