| 1 | The exploitation of epichloae endophytes for agricultural benefit显示文摘Epichloae endophytes of family Clavicipitaceae(comprising genera Epichloëand Neotyphodium)are fungal symbionts of Pooideae grasses.The associations formed,range from mutually beneficial to antagonistic and the nature of this relationship is dependent upon the importance of vertical(via host seeds)versus horizontal(ascospore mediated)transmission of the fungus.These endophytes can enhance their hosts’survival through protection from abiotic and biotic stresses and can thus be utilized in an agricultural context.Animal-safe grass-endophyte associations that confer bio-protective properties for increased pasture persistence and productivity have been developed and commercialized.One of the crucial drivers underpinning the selection of epichloae strains for commercial development is endophyte derived bioactivity.The potential of next generation endophytes is determined by testing a number of attributes such as agronomic fitness,animal and food safety as well as compatibility with host plants of interest.Strategic research supports these activities by focusing on elucidating mechanisms of compatibility between host and fungal symbiont,as well as investigating other molecular drivers of symbiosis such as siderophore mediated iron-uptake,fungal signalling,fungal growth in host plants and fungal secondary metabolism.This review weaves together the different strands of multidisciplinary research aimed at ultimately exploiting epichloae endophytes for increased pasture performance. | Linda J.Johnson Anouck C.M.de Bonth Lyn R.Briggs John R.Caradus Sarah C.Finch Damien J.Fleetwood Lester R.Fletcher David E.Hume Richard D.Johnson Alison J.Popay Brian A.Tapper Wayne R.Simpson Christine R.Voisey Stuart D.Card | 2013 | Fungal Diversity2013,,3: | 5 |

| 19 | Differential subcellular distribution renders HAI-2 a less effective protease inhibitor than HAI-1 in the control of extracellular matriptase proteolytic activity显示文摘The integral membrane,Kunitz-type serine protease inhibitors HAI-1 and HAI-2,can suppress the proteolytic activity of the type 2 transmembrane serine protease matriptase with high specificity and potency.High levels of extracellular matriptase proteolytic activity have,however,been observed in some neoplastic B-cells with high levels of endogenous HAI-2,indicating that HAI-2 may be an ineffective matriptase inhibitor at the cellular level.The different effectiveness of the HAIs in the control of extracellular matriptase proteolytic activity is examined here.Upon inducing matriptase zymogen activation in the HAI Teton Daudi Burkitt lymphoma cells,which naturally express matriptase with very low levels of HAI-2 and no HAI-1,nascent active matriptase was rapidly inhibited or shed as an enzymatically active enzyme.With increasing HAI-1 expression,cellular matriptase-HAI-1 complex increased,and extracellular active matriptase decreased proportionally.Increasing HAI-2 expression,however,resulted in cellular matriptase-HAI-2 complex levels reaching a plateau,while extracellular active matriptase remained high.In contrast to this differential effect,both HAI-1 and HAI-2,even at very low levels,were shown to promote the expression and cell-surface translocation of endogenous matriptase.The difference in the suppression of extracellular active matriptase by the two closely related serine protease inhibitors could result from the primarily cell surface expression of HAI-1 compared to the mainly intracellular localization of HAI-2.The HAIs,therefore,resemble one another with respect to promoting matriptase expression and surface translocation but differ in their effectiveness in the control of extracellular matriptase enzymatic activity. | Yi-Lin Chiu Yi-Ying Wu Robert B.Barndt Yu-Wen Lin Hou-Ping Sytwo Amy Cheng Kacy Yang Khee-Siang Chan Jehng-Kang Wang Michael D.Johnson Chen-Yong Lin | 2022 | Genes & Diseases2022,9,4: | 0 |
| 20 | Design of refractory multi-principal-element alloys for high-temperature applications显示文摘Refractory multi-principal-element alloys(RMPEAs)exhibit high specific strength at elevated temperatures(T).However,current RMPEAs lack a balance of room-temperature(RT)ductility,high-T strength,and high-T creep resistance.Using density-functional theory methods,we scanned composition space using four criteria:(1)formation energies for operational stability:-150250 GPa;(3)inverse Pugh ratio for ductility:G/B<0.57;and(4)high melting points:T_(m)>2500°C.Using rapid bulk alloy synthesis and characterization,we validated theory and down-selected promising alloy compositions and discovered Mo72.3W12.8Ta10.0Ti2.5Zr2.5 having well-balanced RT and high-T mechanical properties.This alloy has comparable high-T compressive strength to well-known MoNbTaW but is more ductile and more creep resistant.It is also superior to a commercial Mo-based refractory alloy and a nickel-based superalloy(Haynes-282)with improved high-T tensile strength and creep resistance. | Gaoyuan Ouyang Prashant Singh Ranran Su Duane D.Johnson Matthew J.Kramer John H.Perepezko Oleg N.Senkov Daniel Miracle Jun Cui | 2023 | npj Computational Materials2023,,1: | 0 |
