| 1 | Intrinsic apoptotic pathway and G2/M cell cycle arrest involved in tubeimoside I-induced EC109 cell death显示文摘Objective: Squamous esophageal carcinoma is highly prevalent in developing countries, especially in China. Tu Bei Mu (TBM), a traditional folk medicine, has been used to treat esophageal squamous cell carcinoma (ESCC) for a long term. tubeimoside I (TBMS1) is the main component of TBM, exhibiting great anticancer potential. In this study, we investigated the mechanism of TBMS1 cytotoxic effect on EC109 cells. Methods: Comparative nuclear proteomic approach was applied in the current study and we identified several altered protein spots. Further biochemical studies were carried out to detect the mitochondrial membrane potential, cell cycle and corresponding proteins' expression and location. Results: Subcellular proteomic study in the nucleus from EC109 cells revealed that altered proteins were associated with mitochondrial function and cell proliferation. Further biochemical studies showed that TBMS1-induced molecular events were related to mitochondria-induced intrinsic apoptosis and P21-cyclin B1/cdc2 complex-related G2/M cell cycle arrest. | 2 | Current perspectives on in vitro maturation and its effects on oocyte genetic and epigenetic profiles显示文摘In vitro maturation(IVM), the maturation in culture of immature oocytes, has been used in clinic for more than 20 years.Although IVM has the specific advantages of low cost and minor side effects over controlled ovarian stimulation, the prevalence of IVM is less than 1% of routine in vitro fertilization and embryo transfer techniques in many reproductive centers. In this review, we searched the MEDLINE database for all full texts and/or abstract articles published in English with content related to oocyte IVM mainly between 2000 and 2016. Many different aspects of the IVM method may influence oocyte potential,including priming, gonadotrophin, growth factors, and culture times. The culture conditions of IVM result in alterations in the oocyte or cumulus cell transcriptome that are not observed under in vivo culture conditions. Additionally, epigenetic modifications, such as DNA methylation or acetylation, are also different between in vitro and in vivo cultured oocytes. In sum,current IVM technique is still not popular and requires more systematic and intensive research to improve its effects and applications. | 3 | Granulosa cell proliferation differentiation and its role in follicular development显示文摘Granulosa cells (GCs) are the most important cells in the ovary that undergo serious changes morphologi- cally and physiologically during the processes of follicular proliferation, differentiation, ovulation, lutenization and atresia. Oocyte (OC) directs GC proliferation and differen- tiation, while GCs influence OC maturation. Many ovarian factors are involved in the regulation of these processes via different molecular mechanisms and signal pathways. P38 MAPK can selectively regulate steroidogenesis in GCs con- trolled by FSH; Transcript factors LRH-1 and DAX-1 play an important role in this process; FSH induces GC prolifera- tion and differentiation by stimulating PCNA and StAR ex- pression and steroidogenesis. Activated ERK1/2 signal pathway may be involved in the FSH-regulated GC prolif- eration and differentiation. Therefore,GC is an ideal model for studying cell proliferation, differentiation and interaction as well as signal transduction. | 11 | Screening of abiotic stress-responsive cotton genes using a cotton full-length cDNA overexpressing Arabidopsis library显示文摘Cotton(Gossypium hirsutum L.)is a major crop and the main source of natural fiber worldwide.Because various abiotic and biotic stresses strongly influence cotton fiber yield and quality,improved stress resistance of this crop plant is urgently needed.In this study,we used Gateway technology to construct a normalized full-length cDNA overexpressing(FOX)library from upland cotton cultivar ZM12 under various stress conditions.The library was transformed into Arabidopsis to produce a cotton-FOX-Arabidopsis library.Screening of this library yielded 6,830 transgenic Arabidopsis lines,of which 757 were selected for sequencing to ultimately obtain 659 cotton ESTs.GO and KEGG analyses mapped most of the cotton ESTs to plant biological process,cellular compo-nent,and molecular function categories.Next,156potential stress-responsive cotton genes were identified from the cotton-FOX-Arabidopsis library under drought,salt,ABA,and other stress conditions.Four stress-related genes identified from the library,designated as GhCAS,GhAPX,GhSDH,and GhPOD,were cloned from cotton complementary DNA,and their expression patterns under stress were analyzed.Phenotypic experiments in-dicated that overexpression of these cotton genes in Arabidopsis affected the response to abiotic stress.The method developed in this study lays a foundation for high-throughput cloning and rapid identification of cotton functional genes. | Shengting Li Hao Chen Zhi Hou Yu Li Cuiling Yang Daojie Wang Chun-Peng Song | 2020 | Journal of Integrative Plant Biology2020,62,7: | 0 |
