| 1 | Current perspectives on in vitro maturation and its effects on oocyte genetic and epigenetic profiles显示文摘In vitro maturation(IVM), the maturation in culture of immature oocytes, has been used in clinic for more than 20 years.Although IVM has the specific advantages of low cost and minor side effects over controlled ovarian stimulation, the prevalence of IVM is less than 1% of routine in vitro fertilization and embryo transfer techniques in many reproductive centers. In this review, we searched the MEDLINE database for all full texts and/or abstract articles published in English with content related to oocyte IVM mainly between 2000 and 2016. Many different aspects of the IVM method may influence oocyte potential,including priming, gonadotrophin, growth factors, and culture times. The culture conditions of IVM result in alterations in the oocyte or cumulus cell transcriptome that are not observed under in vivo culture conditions. Additionally, epigenetic modifications, such as DNA methylation or acetylation, are also different between in vitro and in vivo cultured oocytes. In sum,current IVM technique is still not popular and requires more systematic and intensive research to improve its effects and applications. | 3 | Granulosa cell proliferation differentiation and its role in follicular development显示文摘Granulosa cells (GCs) are the most important cells in the ovary that undergo serious changes morphologi- cally and physiologically during the processes of follicular proliferation, differentiation, ovulation, lutenization and atresia. Oocyte (OC) directs GC proliferation and differen- tiation, while GCs influence OC maturation. Many ovarian factors are involved in the regulation of these processes via different molecular mechanisms and signal pathways. P38 MAPK can selectively regulate steroidogenesis in GCs con- trolled by FSH; Transcript factors LRH-1 and DAX-1 play an important role in this process; FSH induces GC prolifera- tion and differentiation by stimulating PCNA and StAR ex- pression and steroidogenesis. Activated ERK1/2 signal pathway may be involved in the FSH-regulated GC prolif- eration and differentiation. Therefore,GC is an ideal model for studying cell proliferation, differentiation and interaction as well as signal transduction. | 8 | Effects of Different Enucleation Methods on Developmental Potency of Pig Handmade Clone Reconstructed Embryos显示文摘Effects of different enucleation methods on developmental potency of pig handmade clone(HMC)reconstructed embryo was investigated in the paper.We compared enucleation efficiency of blind cut method,first polar body(Pb1)positioning method and demecolcine(DM)assisted enucleation,as well as their effects on development of HMC reconstructed embryos.The results showed that overall enucleation efficiency of Pb1 positioning method was significantly higher than that of blind cut method(P<0.05).The protuberance rate and overall enucleation efficiency of 0.4μg/mL DM treated group for 60 min was significantly higher than that of other concentrations and time treatment groups(P<0.05).For effects on development of HMC reconstructed embryos,there was no significant difference between DM-assisted enucleation and Pb1 positioning method.In conclusion,appropriate addition of DM could enhance enucleation efficiency of HMC,which had no significant influence on developmental potency of reconstructed embryos. | Lv Lingyan Lu Xingrong Sun Junming Pan Cuiling Lan Haien Wang Yanling Cui Kuiqing Xie Bingkun | 2016 | Animal Husbandry and Feed Science2016,8,3: | 0 |
