| 1 | High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat. | Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu | 2020 | Molecular Plant2020,13,9: | 8 |
| 2 | Uncommon EGFR mutations in a cohort of Chinese NSCLC patients and outcomes of first-line EGFR-TKIs and platinum-based chemotherapy显示文摘Objective: Data on the clinical activity of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in patients with non-small-cell lung cancer(NSCLC) and uncommon EGFR mutations remain insufficient.This study aimed to investigate the effect of first-line EGFR-TKIs or platinum-based chemotherapy in NSCLC patients with uncommon EGFR mutations.Methods: We retrospectively enrolled 504 patients with EGFR-mutant NSCLC.The clinical characteristics and treatment outcomes were collected and compared between patients with common and uncommon EGFR-mutant NSCLC.Results: Seventy patients(13.9%) harboring uncommon EGFR mutations were included.Thirty of these patients received EGFR-TKIs and 40 received platinum-based chemotherapy as first-line therapy.The objective response rate(ORR) and median progression-free survival(m PFS) of patients treated with TKIs in the uncommon mutation group was significantly inferior to that in the common mutation group(ORR: 23.3% vs.51.8%,P=0.003; m PFS:7.1 vs.10.9 months,P<0.001).In the uncommon group,m PFS was similar between first-line EGFR-TKIs treatment and platinum-based chemotherapy(7.1 vs.6.1 months,P=0.893).In patients with EGFR G719 X or L861 Q mutations,the m PFS was longer in the first-line EGFR-TKIs treatment group than in the chemotherapy group,but the difference was not statistically significant(G719 X: 8.2 vs.5.8 months,P=0.061; L861 Q: 7.6 vs.4.1 months,P=0.872).Multivariate analyses identified adenocarcinoma(P=0.003) as the independent predictive factor for PFS in patients with uncommon EGFR mutations who were treated with first-line EGFR-TKIs.Conclusions: The current study demonstrated that the effect of first-line EGFR-TKIs was similar to that of platinum-based chemotherapy in patients with uncommon EGFR-mutant NSCLC.Adenocarcinoma was the independent predictive factor for PFS in uncommon EGFR-mutant NSCLC patients treated with first-line EGFRTKIs. | Jinpeng Shi Hui Yang Tao Jiang Xuefei Li Chao Zhao Limin Zhang Sha Zhao Xiaozhen Liu Yijun Jia Yan Wang Lei Xi Shijia Zhang Chunxia Su Shengxiang Ren Caicun Zhou | 2017 | Chinese Journal of Cancer Research2017,29,6: | 6 |

| 6 | Favorable clinical outcomes of checkpoint inhibitorbased combinations after progression with immunotherapy in advanced non-small cell lung cancer显示文摘Aim:Immune checkpoint inhibitors(ICIs)have dramatically changed the treatment paradigm in patients with non-small-cell lung cancer(NSCLC).However,progression patterns with immunotherapy are currently unclear and therapeutic options beyond resistance remain challenging.Methods:We reviewed advanced NSCLC patients between January 2016 and December 2019 who were treated with anti-PD-1/PD-L1 inhibitors in our center and identified those who developed disease progression.Later-line treatment strategies were collected and objective response rate,progression-free survival(PFS),and overall survival(OS)were assessed.Results:Of the 118 patients,46(39.0%)showed oligoprogression and 72(61.0%)showed systemic progression.No difference in progression patterns was observed between monotherapy and combination therapy.Systemic progression was strongly associated with never-smokers(51.4%vs.21.7%,P=0.001)and ECOG PS=2(13.9%vs.2.2%,P=0.048)at baseline.The distribution of progression sites was roughly similar between oligoprogression and systemic progression,and the most commonly affected anatomic site was lung(66.9%),followed by bone(12.7%)and lymph nodes(11.0%).For patients beyond first disease progression,checkpoint inhibitor-based combinations could lead to a significantly longer PFS2 compared with ICIs monotherapy(9.63 months vs.4.23 months,P=0.004,HR=0.394,95%CI:0.174-0.893)and other therapy(9.63 months vs.4.07 months,P=0.046,HR=0.565,95%CI:0.326-0.980).Median OS of the ICIs combination group was not reached but was significantly longer than other therapy group(NR vs.14.37 months,P=0.010,HR=0.332,95%CI:0.167-0.661).Conclusion:Systemic progression occurs more frequently among NSCLC patients receiving ICIs.Checkpoint inhibitor-based combinations show favorable outcomes as subsequent treatment strategies after the failure of previous ICIs treatment. | Xin Yu Xiangling Chu Yan Wu Juan Zhou Jing Zhao Fei Zhou Chaonan Han Chunxia Su | 2021 | Cancer Drug Resistance2021,4,3: | 0 |

| 8 | Seasonality and Sex-Biased Fluctuation of Birth Weight in Tibetan Populations显示文摘Birth weight(BW)is a key determinant of infant mortality.Previous studies have reported seasonal fluctuation of BW.However,the responsible environmental factors remain disputable.High-altitude environment provides a great opportunity to test the current hypotheses due to its distinctive climate conditions.We collected BW data of~9000 Tibetan singletons born at Lhasa(elevation:3660 m)from 2014 to 2018.Using regression models,we analyzed BW seasonality of highland Tibetans.Multivariate models with meteorological factors as independent variables were employed to examine responsible environmental factors accounting for seasonal variation.We compared BW,low-BW prevalence and sex ratio between high-land and lowland populations,and we observed a significant seasonal pattern of BW in Tibetans,with a peak in winter and a trough in summer.Notably,there is a marked sex-biased pattern of BW seasonality(more striking in males than in females).Sunlight exposure in the 3rd trimester and barometric pressure exposure in the 2nd trimester are significantly correlated with BW,and the latter can be explained by seasonal change of oxygen partial pressure.In particular,due to the male-biased BW seasonality,we found a more serious BW reduction and higher prevalence of low-BW in males,and a skewed sex ratio in highlanders.The infant BW of highland Tibetans has a clear pattern of seasonality.The winter BW is larger than the summer BW,due to the longer sunlight exposure during the late-trimester.Male infants are more sensitive to hypoxia than female infants during the 2nd trimester,leading to more BW reduction and higher mortality. | Yaoxi He Jun Li Tian Yue Wangshan Zheng Yongbo Guo Hui Zhang Li Chen Chunxia Li Hongyan Li Chaoying Cui Ouzhuluobu Xuebin Qi Bing Su | 2022 | Phenomics2022,2,1: | 0 |

| 12 | Development of an intelligent heterojunction fenton catalyst for chemodynamic/starvation synergistic cancer therapy显示文摘Chemodynamic therapy(CDT)as an emerging modality in cancer treatment,its implementation remains a daunting challenge by the lack of smart Fenton catalyst under acidic tumor microenvironments.Herein,we have successfully constructed a Fe_(3)O_(4)@MIL-100 heterojunction by growing Fe-based metal-organic framework(MIL-100)onto the surface of Fe_(3)O_(4) nanoparticles.The as-made heterojunction after encapsu-lating glucose oxidase(termed FMG)is demonstrated as a pH-responsive intelligent Fenton nanosystem with the synergistic effect of starvation therapy(ST).Density functional theory(DFT)calculations reveal that such heterojunction could greatly reduce the energy barrier of the Fenton reaction,which better ex-plains the mechanism of Fenton performance improvement.Moreover,the encapsulated glucose oxidase has successfully activated the ST process,in which its generated H_(2)O_(2) and gluconic acid further improve the CDT efficiency.More O_(2) from the enhanced CDT in turn promotes the enzymatic reaction of glucose oxidase.The Fenton/cascade enzymatic reaction operates in a self-feedback manner as proposed.In vitro and in vivo experiments demonstrate that such intelligent Fenton nanoreactors provide a powerful anti-cancer mechanism for effective tumor ablation with enough safety.This work provides insights into the developments of MOF-based heterojunctions as powerful anticancer treatment nanoreactors. | Weishu Ni Ke Jiang Qiaomei Ke Jia Su Xianying Cao Ling Zhang Chunxia Li | 2023 | Journal of Materials Science & Technology2023,,10: | 0 |
