| 1 | A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. | 2 | Human Bop is a novel BH3-only member of the Bcl-2 protein family显示文摘One group of Bcl-2 protein family,which shares only the BH3 domain(BH3-only),is critically involved in the regulation of programmed cell death.Herein we demonstrated a novel human BH3-only protein(designated as Bop)which could induce apoptosis in a BH3 domain-dependent manner.Further analysis indicated that Bop mainly localized to mitochondria and used its BH3 domain to contact the loop regions of voltage dependent anion channel 1(VDAC1)in the outer mitochondrial membrane.In addition,purified Bop protein induced the loss of mitochondrial transmembrane potential(ΔΨm)and the release of cytochrome c.Furthermore,Bop used its BH3 domain to contact pro-survival Bcl-2 family members(Bcl-2,Bcl-XL,Mcl-1,A1 and Bcl-w),which could inhibit Bop-induced apoptosis.Bop would be constrained by pro-survival Bcl-2 proteins in resting cells,because Bop became released from phosphorylated Bcl-2 induced by microtubule-interfering agent like vincristine(VCR).Indeed,knockdown experiments indicated that Bop was partially required for VCR induced cell death.Finally,Bop might need to function through Bak and Bax,likely by releasing Bak from Bcl-XL sequestration.In conclusion,Bop may be a novel BH3-only factor that can engage with the regulatory network of Bcl-2 family members to process intrinsic apoptotic signaling. | Xiaoping Zhang Changjiang Weng Yuan Li Xiaoyan Wang Chunsun Jiang Xuemei Li Youli Xu Quan Chen Lei Pan Hong Tang | 2012 | Protein & Cell2012,3,10: | 2 |

| 15 | Bid BH3 peptide,but not its mutant form G^(94)E, induces permeability transition pore opening and cytochrome c release in vitro显示文摘It has been shown that Bid and its truncated form tBid could induce cytochrome c (cyt c) release without impacting on PTP. We first show that Bid BH3 peptide, but not its mutant form of Bid BH3 peptide G94E, which is unable to bind to Bcl-xL, induces permeability transition pore (PTP) opening in a dose dependent manner. Bid BH3 peptide also induces the reduction of mitochondria membrane potential (Ψm) and cyt c release from mitochondrial in vitro. PTP opening and the loss of Ψm, were inhibited by Bcl-xL, cyclosporin A and ruthenium red, and the latter was an inhibitor of Ca2+ uniporter in the mitochondrial membrane. | 19 | A smartphone-based automated fluorescence analysis system for point-of-care testing of Hg(Ⅱ)显示文摘This work demonstrates a smartphone-based automated fluorescence analysis system(SAFAS)for point-of-care testing(POCT)of Hg(Ⅱ).This system consists of three modules.The smartphone module is used to provide an excitation light source,and to collect and analyze fluorescent images.The dark box module is applied to integrate the desired optical elements and offers a dark environment.The cost of the integrated dark box mainly includes the upper cover,box body,lower bottom,¯xture and some optical elements which is about$109.The chip module is used for fluorescence sensing,which is composed of an upper plate,bottom plate and cloth-based chip.Due to the integration of multiple smartphone functions,the SAFAS eliminates the need for additional power sources,light sources and analysis systems.The dark box and upper and bottom plates are made by 3D printer.The cloth-based chip(about$0.005 for each chip)is fabricated using the wax screenprinting technique,with no need for expensive and complex fabrication equipments.To our knowledge,the cloth-based microfluidic fluorescence detection method combined with smartphone functions is first reported.By using optimal conditions,the designed system can realize the quantitative detection of Hg(Ⅱ),which has a linear range of 0.001–100μgmL^(-1)and a detection limit of 0.5 ngmL^(-1).Additionally,the SAFAS has been successfully applied for detecting Hg(Ⅱ)in actual water samples,with recoveries of 100.1%–111%,RSDs of 3.88%–9.74%,and fast detection time of about 1 min.Obviously,the proposed SAFAS has the advantages of high sensitivity,wide dynamic range,acceptable reproducibility,good stability and low cost.Therefore,it is believed that the presented SAFAS has great potential to perform the POCT of Hg(II)in different water samples. | Yafei Chen Ke Zhang Yuan Liu Chunsun Zhang | 2022 | Journal of Innovative Optical Health Sciences2022,15,5: | 0 |
