| 1 | Integrating TM and Ancillary Geographical Data with Classification Trees for Land Cover Classification of Marsh Area显示文摘The main objective of this research is to determine the capacity of land cover classification combining spec- tral and textural features of Landsat TM imagery with ancillary geographical data in wetlands of the Sanjiang Plain, Heilongjiang Province, China. Semi-variograms and Z-test value were calculated to assess the separability of grey-level co-occurrence texture measures to maximize the difference between land cover types. The degree of spatial autocorrelation showed that window sizes of 3×3 pixels and 11×11 pixels were most appropriate for Landsat TM im- age texture calculations. The texture analysis showed that co-occurrence entropy, dissimilarity, and variance texture measures, derived from the Landsat TM spectrum bands and vegetation indices provided the most significant statistical differentiation between land cover types. Subsequently, a Classification and Regression Tree (CART) algorithm was applied to three different combinations of predictors: 1) TM imagery alone (TM-only); 2) TM imagery plus image texture (TM+TXT model); and 3) all predictors including TM imagery, image texture and additional ancillary GIS in- formation (TM+TXT+GIS model). Compared with traditional Maximum Likelihood Classification (MLC) supervised classification, three classification trees predictive models reduced the overall error rate significantly. Image texture measures and ancillary geographical variables depressed the speckle noise effectively and reduced classification error rate of marsh obviously. For classification trees model making use of all available predictors, omission error rate was 12.90% and commission error rate was 10.99% for marsh. | 2 | Tracing the first hematopoietic stem cell generation in human embryo by single-cell RNA sequencing显示文摘Tracing the emergence of the first hematopoietic stem cells(HSCs)in human embryos,particularly the scarce and transient precursors thereof,is so far challenging,largely due to the technical limitations and the material rarity.Here,using single-cell RNA sequencing,we constructed the first genome-scale gene expression landscape covering the entire course of endothelial-to-HSC transition during human embryogenesis.The transcriptomically defined HSC-primed hemogenic endothelial cells(HECs)were captured at Carnegie stage(CS)12-14 in an unbiased way,showing an unambiguous feature of arterial endothelial cells(ECs)with the up-regulation of RUNX1,MYB and ANGPTL Importantly,subcategorizing CD34+CD45-ECs into a CD44+population strikingly enriched HECs by over 10-fold.We further mapped the developmental path from arterial ECs via HSC-primed HECs to hematopoietic stem progenitor cells,and revealed a distinct expression pattern of genes that were transiently over-represented upon the hemogenic fate choice of arterial ECs#including EMCN,PROCR and RUNX1T1.We also uncovered another temporally and molecularly distinct intra-embryonic HEC population,which was detected mainly at earlier CS 10 and lacked the arterial feature.Finally,we revealed the cellular components of the putative aortic niche and potential cellular interactions acting on the HSCprimed HECs.The cellular and molecular programs that underlie the generation of the first HSCs from HECs in human embryos,together with the ability to distinguish the HSC-primed HECs from others,will shed light on the strategies for the production of clinically useful HSCs from pluripotent stem cells. | Yang Zeng Jian He Zhijie Bai Zongcheng Li Yandong Gong Chen Liu Yanli Ni Junjie Du Chunyu Ma Lihong Bian Yu Lan Bing Liu | 2019 | Cell Research2019,29,11: | 14 |

| 5 | ENPP1/PC-1 Gene K121Q Polymorphism Is Associated with Obesity in European Adult Populations: Evidence from A Meta-Analysis Involving 24 324 Subjects显示文摘Objective Findings from the previous studies have suggested a relationship between ectonucleotide pyrophosphatase /phosphodiesterase 1 (ENPP‐1) or plasma cell membrane glycoprotein 1 (PC‐1) gene single nucleotide polymorphism (K121Q, rs1044498) and genetic susceptibility to obesity. However, such relationship is not reproduced by some currently available studies. In this context, the present study is aimed to quantitatively analyze the association of K121Q variant with obesity in all published case‐control studies in European adult populations. Methods Published literature from PubMed, EMBASE, and ISI web of science databases were retrieved. The studies evaluating the association of ENPP1/PC1 gene K121Q polymorphism with obesity were included, in which sufficient data were presented to calculate the odds ratio (OR) with 95% confidence intervals (CIs). Results Ten case‐control studies meeting the inclusion criteria identified a total of 24,324 subjects including 11,372 obese and 12,952 control subjects. The meta‐analysis results showed a statistically significant association of K121Q with obesity [OR (95%CI): 1.25 (1.04‐1.52) P=0.021] under a recessive model of inheritance (QQ vs. KK+KQ) without heterogeneity or publication bias. | 6 | Whole-genome sequencing of monozygotic twins discordant for schizophrenia indicates multiple genetic risk factors for schizophrenia显示文摘Schizophrenia is a common disorder with a high heritability, but its genetic architecture is still elusive.We implemented whole-genome sequencing(WGS) analysis of 8 families with monozygotic(MZ) twin pairs discordant for schizophrenia to assess potential association of de novo mutations(DNMs) or inherited variants with susceptibility to schizophrenia. Eight non-synonymous DNMs(including one splicing site) were identified and shared by twins, which were either located in previously reported schizophrenia risk genes(p.V24689 I mutation in TTN, p.S2506 T mutation in GCN1L1, IVS3+1G > T in DOCK1) or had a benign to damaging effect according to in silico prediction analysis. By searching the inherited rare damaging or loss-of-function(LOF) variants and common susceptible alleles from three classes of schizophrenia candidate genes, we were able to distill genetic alterations in several schizophrenia risk genes, including GAD1, PLXNA2, RELN and FEZ1. Four inherited copy number variations(CNVs; including a large deletion at 16p13.11) implicated for schizophrenia were identified in four families, respectively. Most of families carried both missense DNMs and inherited risk variants, which might suggest that DNMs, inherited rare damaging variants and common risk alleles together conferred to schizophrenia susceptibility. | 8 | Dahuang Fuzi decoction reduces inflammation levels and alleviates intestinal mucosal barrier damage in septic rats显示文摘Objective:To investigate the protective effect of Dahuang Fuzi Decoction(DHFZD),a traditional Chinese prescription,at alleviating sepsis-induced inflammation and gut barrier damage in rats.Methods:Forty clean-grade male Sprague-Dawley rats were divided randomly into three groups:normal control group(NCG,n?10),model control group(MCG,n?15)and DHFZD-treated group(DHFZDG,n?15).NCG rats were sham operated on and used as the controls,whereas MCG and DHFZDG rats were used to replicate the rat sepsis model using cecal ligation and puncture(CLP).The DHFZDG rats received DHFZD by gavage(4.5 mg/g of body weight)2 h prior to CLP and after its successful induction,while the NCG and MCG rats received equivalent amounts of sterilized water by gavage.All rat groups were starved and had free access to water.At 24 h post-experimental set up,the mortality of rats in each group was recorded,and peritoneal inflammation assessment and pathological changes related to the intestinal mucosal injury index(IMII)in the surviving rats were evaluated.D-lactic acid,tumor necrosis factor(TNF)-a,interleukin(IL)-6 and IL-10 peripheral blood concentrations,along with secretory immunoglobulin A(sIgA)in the intestinal mucosa were evaluated by enzyme-linked immunosorbent assays.Gut microbes were detected using 16S rRNA gene sequencing.Results:DHFZD reduced sepsis-related mortality in the rats.Moreover,it alleviated peritoneal inflammation and pathological changes according to the IMII.DHFZD reduced serum procalcitonin,TNF-a and IL-6 concentrations,but not the IL-10 concentration.It also reduced serum D-lactic acid and increased sIgA concentrations in intestinal mucosa.Notably,DHFZDG restored gut microbiota diversity and regulated the decrease in Bacteroidetes induced by sepsis,compared with the MCG rats.Conclusion:DHFZDG may play a protective role in sepsis by alleviating sepsis-induced inflammation and gut barrier damage in rats. | Fusheng Liu Chunyue Yu Jin Liu Nadia Johnson Yin Zhang Zeqi Su Chenchen Sun Xia Ding | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,1: | 3 |
| 9 | An ethanolic extract of Bailian(Radix Ampelopsis Japonicae):demonstration of colorectal cancer treatment efficacy via inhibition of β-catenin signaling in vitro显示文摘OBJECTIVE:To investigate the underlying mechanism of Bailian (Radix Ampelopsis Japonicae,BL) extract action on colorectal cancer (CRC).METHODS:We explored the involvement of β-catenin signaling on the anti-CRC effects of an BL ethanolic extract (BLE) in cell models by using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay,immunofluorescent staining,luciferase assay,Western blot analysis and real-time quantitative polymerase chain reaction analysis.Anti-CRC compounds were quantified by high performance liquid chromatography.RESULTS:The contents of gallic acid,catechin,and epicatechin in the BLE were 0.23,1.25,and 0.18 g/kg,respectively.BLE-mediated cytotoxic and apoptotic effects were accompanied by lowered β-catenin/Tcf transcriptional activity,reduced β-catenin nuclear localization,and downregulated protein and mRNA levels of both β-catenin and molecules regulated by β-catenin.CONCLUSION:The mechanism underpinning the anti-CRC effects of BLE may involve inhibition of β-catenin signaling.Further studies are necessary to establish the role of β-catenin signaling in the action of BLE-mediated anti-CRC effects. | Su Tao Wang Xinning Li Chunyu Bai Jingxuan Chi-Yan Cheng Fu Xiuqiong Li Ting Yu Zhiling | 2019 | Journal of Traditional Chinese Medicine2019,39,3: | 3 |
| 10 | Characteristics of Starch synthesis,Yield and Quality Provide Insights into Rice-Tartary Buckwheat Utilization and Improvement显示文摘[Objectives] Rice-Tartary buckwheat is a special type of Tartary buckwheat with easy hulling thin shell.This study was conducted to characterize the carbohydrate metabolism,quality and yield and provide a basis for food processing and breeding of rice-Tartary buckwheat.[Methods]The local rice-Tartary buckwheat‘cv.Xiaomiqiao’(XMQ) was used to explore the characteristics of photosynthesis,starch synthesis enzyme activities,traits of yield and quality,as compared to Tartary buckwheat‘cv.Jinqiao 2’(JQ2).[Results]XMQ showed significantly lower values of net photosynthetic rate,ADPGase activity and starch synthase activity at one or more periods during grain filling than JQ2.The effective branch number and grain number per plant of XMQ were similar to that of JQ2 regardless of different years,but the 1 000-grain weight was significantly lighter that resulted in the significantly lower yield.XMQ accumulated similar contents of starch(amylose,amylopectin) and protein(glutelin,albumin,prolamin,globulin) to those of JQ2,but exhibited significantly lower flavonoid content.The values of peak viscosity and hot paste viscosity in XMQ were similar to those of JQ2,but the values of cool paste viscosity,breakdown and setback were significantly lower.In conclusion,XMQ showed low yield due to the insufficient of photosynthesis and starch synthesis.XMQ exhibited high nutritional quality as well as normal Tartary buckwheat and stored 18.88 mg/g of flavonoid,that might be consumed as daily diet.[Conclusions]The present results will provide a basis for utilization and improvement of rice-Tartary buckwheat. | Kai LIAO Yan WANG Wujuan YU Chunyu WEI Chenggang LIANG | 2020 | Agricultural Biotechnology2020,9,2: | 2 |
| 11 | Degradation of tetracycline hydrochloride by ultrafine TiO_(2) nanoparticles modified g-C_(3)N_(4) heterojunction photocatalyst:Influencing factors,products and mechanism insight显示文摘The unique heterojunction photocatalyst of graphite carbon nitride (g-C_(3)N_(4)) modified ultrafine TiO_(2) (g-C_(3)N_(4)/TiO_(2)) was successfully fabricated by electrochemical etching and co-annealing method. However, the effects of various environmental factors on the degradation of TC by g-C_(3)N_(4)/TiO_(2) and the internal reaction mechanism are still unclear. In this study, the effects of initial pH, anions, and cations on the photocatalytic degradation of tetracycline hydrochloride (TC) by g-C_(3)N_(4)/TiO_(2) were systematically explored, and the scavenging experiment and intermediate detection were conducted to better reveal the mechanism on photocatalytic degradation of TC. The results showed that the removal efficiency of photocatalytic degradation of TC by g-C_(3)N_(4)/TiO_(2) could reach 99.04% under Xenon lamp irradiation within 120 min. The unique g-C_(3)N_(4)/TiO_(2) heterojunction photocatalyst showed excellent photocatalytic performance for the degradation of TC at pH 3~7, and possesses outstanding anti-interference ability to NO_(3)^(-), Cl^(-), Na^(+), Ca^(2+) and Mg^(2+) ions in natural waters during the photocatalytic degradation TC process. Superoxide radicals (O_(2)^(·-)) and hydroxyl radicals (^(·)OH) were proved as the main reactive species for TC degradation, and the possible mechanism of the unique photocatalytic system for g-C_(3)N_(4)/TiO_(2) was also proposed. | 18 | Association of immunoglobulin G4-related disease with a family history of malignancy:A retrospective study of 168 cases显示文摘Objective:Immunoglobulin G4-related disease(IgG4-RD)is a systemic immune-mediated fibroinflammatory condition.Previous studies have indicated relationships between malignancy and autoimmunity.This retrospective cohort study aimed to determine IgG4-RD incidence and clinical features in patients with a family history of malignancy.Methods:To analyze the relationship between IgG4-RD and family history of malignancy,we reviewed IgG4-RD patients with a family history of malignancy diagnosed in various departments in West China Hospital,Sichuan University from December 2012 to September 2019.Clinical data and laboratory features were compared between IgG4-RD patients with and without a family history of malignancy.Results:Among 168 enrolled patients with IgG4-RD,22(13.1%)had a family history of malignancy.The most frequently involved system in family members with malignancy was the digestive system(38.5%).Among patients with a family history of malignancy,the most frequently involved organs in IgG4-RD were the pancreas(31.9%)and lymph nodes(31.9%).Age at symptom onset was older in IgG4-RD patients with a family history of malignancy than in patients without a family history of malignancy(50–59 years:36.3%vs.16.4%,p=0.0393).Hemoglobin(p=0.0172)and albumin(p=0.0247)levels were higher and globulin(p=0.0232)levels were lower in patients with a family history of malignancy.Conclusions:Our findings suggest that a family history of malignancy may be associated with IgG4-RD development.We hypothesize that genetic susceptibility may be involved in the pathogenesis of IgG4-RD. | Diping Hu Yanhong Li Yu Zhou Yubin Luo Lu Cheng Chunyu Tan Qiuping Zhang Yi Zhao Yi Liu | 2021 | Rheumatology & Autoimmunity2021,1,2: | 1 |
