| 2 | Artemisinin, the Magic Drug Discovered from Traditional Chinese Medicine显示文摘Artemisinin and its derivatives represent the most important and influential class of drugs in the fight against malaria. Since the discovery of artemisinin in the early 1970s, the global community has made great strides in characterizing and understanding this remarkable phytochemical and its unique chemical and pharmacological properties. Today, even as artemisinin continues to serve as the foundation for antimalarial therapy, numerous challenges have surfaced in the continued application and development of this family of drugs. These challenges include the emergence of delayed treatment responses to artemisinins in malaria and efforts to apply artemisinins for non-malarial indications. Here, we provide an overview of the story of artemisinin in terms of its past, present, and future. | 4 | IGHG1 promotes motility likely through epithelial-mesenchymal transition in ovarian cancer显示文摘Objective: Ovarian cancer(OC) is one of the leading causes of death for female cancer patients. COC166-9 is an OC-specific monoclonal antibody and we have identified immunoglobulin γ-1 heavy chain constant region(IGHG1) as its antigen. We explore the function of IGHG1 in proliferation, apoptosis and motility of OC cells further in this research.Methods: IGHG1 expression in OC specimens was detected through immunohistochemistry. Real-time quantitative polymerase chain reaction(RT-q PCR) or western blotting assay was used to test IGHG1 expression in OC cells. Viability of OC cells was tested by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay. Flow cytometry or western blotting assay was used to detect cell cycle and apoptosis. Cellular motility was analyzed by using transwell assay and the markers of epithelial-mesenchymal transition(EMT) were tested through immunoblots.Results: Although it exerts negligible effect on the viability and apoptosis of OC cells, IGHG1 could promote migration and invasion of malignant cells in vitro. Mechanistically, IGHG1 increases the expression of N-cadherin and Vimentin while decreases E-cadherin expression. Additionally, IGHG1 expression in OC specimens is higher relative to the paired normal counterparts. Further analysis demonstrates that the increased IGHG1 expression correlates positively with the lymph node metastasis of OC.Conclusions: IGHG1 promotes the motility of OC cells likely through executing the EMT program. | 6 | Factors that determine catastrophic expenditure for tuberculosis care: a patient survey in China显示文摘Background:Tuberculosis(TB)often causes catastrophic economic effects on both the individual suffering the disease and their households.A number of studies have analyzed patient and household expenditure on TB care,but there does not appear to be any that have assessed the incidence,intensity and determinants of catastrophic health expenditure(CHE)relating to TB care in China.That will be the objective of this paper.Methods:The data used for this study were derived from the baseline survey of the China Government–Gates Foundation TB Phase Ⅱ program.Our analysis included 747 TB cases.Catastrophic health expenditure for TB care was estimated using two approaches,with households defined as experiencing CHE if their annual expenditure on TB care:(a)exceeded 10%of total household income;and(b)exceeded 40%of their non-food expenditure(capacity to pay).Chi-square tests were used to identify associated factors and logistic regression analysis to identify the determinants of CHE.Results:The incidence of CHE was 66.8%using the household income measure and 54.7%using non-food expenditure(capacity to pay).An inverse association was observed between CHE rates and household income level.Significant determinants of CHE were:age,household size,employment status,health insurance status,patient income as a percentage of total household income,hospitalization and status as a minimum living security household.Factors including gender,marital status and type of TB case had no significant associations with CHE.Conclusions:Catastrophic health expenditure incidence from TB care is high in China.An integrated policy expanding the free treatment package and ensuring universal coverage,especially the height of UHC for TB patients,is needed.Financial and social protection interventions are essential for identified at-risk groups. | Chengchao Zhou Qian Long Jiaying Chen Li Xiang Qiang Li Shenglan Tang Fei Huang Qiang Sun Henry Lucas | 2016 | Infectious Diseases of Poverty2016,5,1: | 6 |

| 12 | Environmentally responsive dual-targeting nanotheranostics for overcoming cancer multidrug resistance显示文摘The development of multiple drug resistance(MDR) to chemotherapy and subsequent treatment failures are major obstacles in cancer therapy. An attractive option for combating MDR is inhibiting the expression of P-glycoprotein(P-gp) in tumor cells. Here, we report a novel chemosensitizing agent, XMD8-92,which can down-regulate P-gp. To enhance the specificity of MDR chemotherapy, a promising nanotheranostic micelle system based on poly(ethylene glycol)-blocked-poly(L-leucine)(PEG-b-Leu) was developed to simultaneously carry the anticancer drug doxorubicin, chemosensitizing agent XMD8-92, and superparamagnetic iron oxide nanoparticles(SPIOs). Featured with MDR environmentally responsive dual-targeting capability, controllable drug delivery, and efficient magnetic resonance(MR) imaging characteristics, the prepared nanotheranostics(DXS@NPs) showed outstanding in vitro cytotoxicity on MDR cells(SCG 7901/VCR) with only 53% of cells surviving compared to 90% of DOX-treated cells.Furthermore, efficient tumor inhibition and highly reduced systemic toxicity were exhibited by MDR tumor-bearing mice treated with DXS@NPs. | 15 | Responses of calcareous sand foundations to variations of groundwater table and applied loads显示文摘The long-term settlement of calcareous sand foundations caused by daily periodic fluctuations has become a significant geological hazard,but effective monitoring tools to capture the deformation profiles are still rarely reported.In this study,a laboratory model test and an in situ monitoring test were conducted.An optical frequency domain reflectometer(OFDR)with high spatial resolution(1 mm)and high accuracy(10-6)was used to record the soil strain responses to groundwater table and varied loads.The results indicated that the fiber-optic measurements can accurately locate the swelling and compressive zones.During the loading process,the interlock between calcareous sand particles was detected,which increased the internal friction angle of soil.The foundation deformation above the sliding surface was dominated by compression,and the soil was continuously compressed beneath the sliding surface.After 26e48 h,calcareous sand swelling occurred gradually above the water table,which was primarily dependent on capillary water.The swelling of the soil beneath the groundwater table was completed rapidly within less than 2 h.When the groundwater table and load remain constant,the compression creep behavior can be described by the Yasong-Wang model with R2¼0.993.The daily periodically varying in situ deformation of calcareous sand primarily occurs between the highest and lowest groundwater tables,i.e.4.2e6.2 m deep.The tuff interlayers with poor water absorption capacity do not swell or compress,but they produce compressive strain under the influence of deformed calcareous sand layers. | Dingfeng Cao Sanjay Kumar Shukla Linqing Yang Chengchao Guo Jinghong Wu Fuming Wang | 2022 | Journal of Rock Mechanics and Geotechnical Engineering2022,14,4: | 2 |

| 18 | Regulation of the stability and ABA import activity of NRT1.2/NPF4.6 by CEPR2-mediated phosphorylation in Arabidopsis显示文摘Abscisic acid(ABA)transport plays an important role in systemic plant responses to environmental factors.However,it remains largely unclear about the precise regulation of ABA transporters in plants.In this study,we show that the C-terminally encoded peptide receptor 2(CEPR2)directly interacts with the ABA transporter NRT1.2/NPF4.6.Genetic and phenotypic analyses revealed that NRT1.2/NPF4.6 positively regulates ABA response and that NRT1.2/NPF4.6 is epistatically and negatively regulated by CEPR2.Further biochemical assays demonstrated that CEPR2 phosphorylates NRT1.2/NPF4.6 at serine 292 to promote its degradation under normal conditions.However,ABA treatment and non-phosphorylation at serine 292 prevented the degradation of NRT1.2/NPF4.6,indicating that ABA inhibits the phosphorylation of this residue.Transport assays in yeast and Xenopus oocytes revealed that non-phosphorylated NRT1.2/NPF4.6 had high levels of ABA import activity,whereas phosphorylated NRT1.2/NPF4.6 did not import ABA.Analyses of complemented nrt1.2 mutants that mimicked non-phosphorylated and phosphorylated NRT1.2/NPF4.6 confirmed that non-phosphorylated NRT1.2S292A had high stability and ABA import activity in planta.Additional experiments showed that NRT1.2/NPF4.6 was degraded via the 26S proteasome and vacuolar degradation pathways.Furthermore,we found that three E2 ubiquitin-conjugating enzymes,UBC32,UBC33,and UBC34,interact with NRT1.2/NPF4.6 in the endoplasmic reticulum and mediate its ubiquitination.NRT1.2/NPF4.6 is epistatically and negatively regulated by UBC32,UBC33,and UBC34 inplanta.Taken together,these results suggest that the stability and ABA import activity of NRT1.2/NPF4.6 are precisely regulated by its phosphorylation and degradation in response to environmental stress. | Lei Zhang Zipeng Yu Yang Xu Miao Yu Yue Ren Shizhong Zhang Guodong Yang Jinguang Huang Kang Yan Chengchao Zheng Changai Wu | 2021 | Molecular Plant2021,14,4: | 2 |
| 19 | Discovery and repurposing of artemisinin显示文摘Malaria is an ancient infectious disease that threatens millions of lives globally even today.The discovery of artemisinin,inspired by traditional Chinese medicine(TCM),has brought in a paradigm shift and been recognized as the“best hope for the treatment of malaria”by World Health Organization.With its high potency and low toxicity,the wide use of artemisinin effectively treats the otherwise drug-resistant parasites and helps many countries,including China,to eventually eradicate malaria.Here,we will first review the initial discovery of artemisinin,an extraordinary journey that was in stark contrast with many drugs in western medicine.We will then discuss how artemisinin and its derivatives could be repurposed to treat cancer,inflammation,immunoregulation-related diseases,and COVID-19.Finally,we will discuss the implications of the“artemisinin story”and how that can better guide the development of TCM today.We believe that artemisinin is just a starting point and TCM will play an even bigger role in healthcare in the 21st century. | Qiaoli Shi Fei Xia Qixin Wang Fulong Liao Qiuyan Guo Chengchao Xu Jigang Wang | 2022 | Frontiers of Medicine2022,16,1: | 2 |
