| 1 | PK-tailored tertiary prophylaxis in patients with severe hemophilia A at beijing Children’s Hospital显示文摘Importance:Tertiary prophylaxis using a low-dose regimen is usually insufficient to prevent recurrent joint bleeding and deterioration in joint diseases in children with severe hemophilia A.Pharmacokinetic(PK)dosing is a useful approach to increase the precision and efficiency of prophylaxis.Objective:To explore the efficacy of PK-tailored tertiary prophylaxis in children with severe hemophilia A.Methods:We implemented a PK-tailored tertiary prophylaxis program for 15 boys with severe hemophilia A aged 5-16 years at Beijing Children’s Hospital.Following PK testing and a 6-month evaluation period(phase I),15 patients were divided in two groups according to individual PK data and actual bleeding:(1)a PK-tailored group[modified prophylaxis regimen according to PK data for the next 6 months(phase II);n=8]and(2)a maintenance group(continued the original regimen for the next 6 months;n=7).We compared the bleeding rate,infusion frequency,and factor VIII(FVIII)consumption between the two groups.results:In the PK-tailored group,the median annual joint bleeding rate was reduced from 7.8 in phase I to 1.4 in phase II,mean annual total factor consumption increased from 1619.0 IU/kg in phase I to 2401.9 IU/kg in phase II,and median infusion frequency for prophylaxis increased from 104 times/year in phase I to 156 times/year in phase II(P<0.05).Although the FVIII consumption increased,it remained at approximately half of the standard method.Interpretation:PK-tailored prophylaxis may represent a more efficient approach to individual prophylaxis in China,but further studies are required to verify this. | Peijing Li Zhenping Chen Xiaoling Cheng Yan Wang Ningning Zhang Yingzi Zhen Xinyi Wu Runhui Wu | 2019 | Pediatric Investigation2019,3,1: | 3 |
| 2 | Towards a Dynamic Controller Scheduling-Timing Problem in Software-Defined Networking显示文摘Controller vulnerabilities allow malicious actors to disrupt or hijack the Software-Defined Networking. Traditionally, it is static mappings between the control plane and data plane. Adversaries have plenty of time to exploit the controller's vulnerabilities and launch attacks wisely. We tend to believe that dynamically altering such static mappings is a promising approach to alleviate this issue, since a moving target is difficult to be compromised even by skilled adversaries. It is critical to determine the right time to conduct scheduling and to balance the overhead afforded and the security levels guaranteed. Little previous work has been done to investigate the economical time in dynamic-scheduling controllers. In this paper, we take the first step to both theoretically and experimentally study the scheduling-timing problem in dynamic control plane. We model this problem as a renewal reward process and propose an optimal algorithm in deciding the right time to schedule with the objective of minimizing the long-term loss rate. | 18 | miR-182-5p attenuates Schistosoma japonicum-induced hepatic fibrosis by targeting tristetraprolin显示文摘Egg granuloma formation in the liver is the main pathological lesion caused by Schistosoma japonicum infection,which generally results in liver fibrosis and may lead to death in advanced patients.MicroRNAs(miRNAs)regulate the process of liver fibrosis,but the putative function of miRNAs in liver fibrosis induced by S.japonicum infection is largely unclear.Here,we detect a new miRNA,miR-182-5p,which shows significantly decreased expression in mouse livers after stimulation by soluble egg antigen(SEA)of S.japonicum or S.japonicum infection.Knockdown or overexpression of miR-182-5p in vitro causes the increased or decreased expression of tristetraprolin(TTP),an important immunosuppressive protein in the process of liver fibrosis.Furthermore,knockdown of miR-182-5p in vivo upregulates TTP expression and significantly alleviates S.japonicum-induced hepatic fibrosis.Our data demonstrate that downregulation of miR-182-5p increases the expression of TTP in mouse livers following schistosome infection,which leads to destabilization of inflammatory factor mRNAs and attenuates liver fibrosis.Our results uncover fine-tuning of liver inflammatory reactions related to liver fibrosis caused by S.japonicum infection and provide new insights into the regulation of schistosomiasis-induced hepatic fibrosis. | Xuejun Zhao Zijie Xia Ziang Wang Mengsi Zhou Xuebing Qiu Cheng Wang Tian Xu Qian Fang Zhenping Ming Huifen Dong | 2022 | Acta Biochimica et Biophysica Sinica2022,54,10: | 0 |
