1Construction, expression and characterization of the engineered antibody against tumor surface antigen, p185^(c-erbB-2)显示文摘The c-erbB-2 proto-oncogene encodes a 185kDa protein p185, which belongs to epidermal growth factorreceptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis forcertain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibitsproliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. Inorder to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed thesingle-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichiacoli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cellimmunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain(ECD) of p185. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion moleculewith a homodimer form and the recombinant product was expressed in mammalian cells. In a series ofsubsequent analysis this fusion protein showed identical antigen binding site and activity with the parentantibody. 2Infection with novel coronavirus(SARS-CoV-2)causes pneumonia in Rhesus macaques显示文摘The 2019 novel coronavirus(SARS-CoV-2)outbreak is a major challenge for public health.SARS-CoV-2 infection in human has a broad clinical spectrum ranging from mild to severe cases,with a mortality rate of^6.4%worldwide(based on World Health Organization daily situation report).However,the dynamics of viral infection,replication and shedding are poorly understood.Here,we show that Rhesus macaques are susceptible to the infection by SARS-CoV-2.After intratracheal inoculation,the first peak of viral RNA was observed in oropharyngeal swabs one day post infection(1 d.p.i.),mainly from the input of the inoculation,while the second peak occurred at 5 d.p.i.,which reflected on-site replication in the respiratory tract.Histopathological observation shows that SARS-CoV-2 infection can cause interstitial pneumonia in animals,characterized by hyperemia and edema,and infiltration of monocytes and lymphocytes in alveoli.We also identified SARS-CoV-2 RNA in respiratory tract tissues,including trachea,bronchus and lung;and viruses were also re-isolated from oropharyngeal swabs,bronchus and lung,respectively.Furthermore,we demonstrated that neutralizing antibodies generated from the primary infection could protect the Rhesus macaques from a second-round challenge by SARS-CoV-2.The non-human primate model that we established here provides a valuable platform to study SARS-CoV-2 pathogenesis and to evaluate candidate vaccines and therapeutics.Chao Shan Yan-Feng Yao Xing-Lou Yang Yi-Wu Zhou Ge Gao Yun Peng Lian Yang Xue Hu Jin Xiong Ren-Di Jiang Hua-Jun Zhang Xiao-Xiao Gao Cheng Peng Juan Min Ying Chen Hao-Rui Si Jia Wu Peng Zhou Yan-Yi Wang Hong-Ping Wei Wei Pang Zheng-Fei Hu Long-Bao Lv Yong-Tang Zheng Zheng-Li Shi Zhi-Ming Yuan 2020Cell Research2020,30,8:19
3Chinese Medicine Sequential Therapy Improves Pregnancy Outcomes after Surgery for Endometriosis-Associated Infertility: A Multicenter Randomized Double-blind Placebo Parallel Controlled Clinical Trial显示文摘Objective:To evaluate the efficacy and safety of Chinese medicine(CM)improving pregnancy outcomes after surgery for endometriosis-associated infertility.Methods:A multicenter,randomized,doubleblind placebo parallel controlled clinical trial was designed.A total of 202 patients who had laparoscopy for endometriosis-associated infertility with qi stagnation and blood stasis syndrome were included and randomly divided into the CM treatment group and placebo control group at a ratio of 1:1 using a central block randomization from May 2014 to September 2017,101 patients in each group.The two groups received continuous intervention at 1–5 days after surgery,for 6 menstrual cycles.Before ovulation,the CM group was treated Huoxue Xiaoyi Granule(活血消异颗粒);after ovulation,Bushen Zhuyun Granule(补肾助孕颗粒)was involved.The control group was treated with placebo.Transvaginal ultrasonography was performed every menstrual cycle during the treatment,and female hormone levels in the follicular and luteal phases were measured during the 1 st,3 rd and 6 th menstrual cycles.The analysis was continued until pregnancy.The primary outcomes were clinical pregnancy rate and pregnancy outcome,and the secondary outcomes were follicular development and endometrial receptivity.Safety evaluations were performed before and after treatment.Results:(1)Clinical pregnancy and live birth rates:the clinical pregnancy and live birth rates of the CM group were significantly higher than those of the placebo group[44.6%(45/101)vs.29.7%(30/101),34.7%(35/101)vs.20.8%(21/101),both P<0.05].(2)Follicle development:the incidence of dominant follicles,rate of cumulative cycle ovulation,and rate of cumulative cycle mature follicle ovulation were significantly higher in the CM group than those in the placebo group[93.8%(350/373)vs.89.5%(341/381),80.4%(275/342)vs.69.1%(253/366),65.8%(181/275)vs 56.1%(142/253),P<0.05 or P<0.01]).The incidence of cumulative cycle luteinized unruptured follicle syndrome was significantly lower in the CM group than in the placebo group[11.7%(40/342)vs.17.8%(65/366),P<0.05).(3)Endometrial receptivity:after treatment,both endometrial types and endometrial blood flow types in the CM group were mainly types A and B,while those in the placebo group were mainly types B and C,with a significant difference between the two groups(both P<0.05).(4)Adverse events:the incidence of adverse events between the two groups was not significantly different(P>0.05).Conclusion:Strategies for activating blood circulation-regulating Gan(Liver)-tonifying Shen(Kidney)sequential therapy can effectively improve the clinical pregnancy rate and live birth rate of endometriosis-associated infertility with qi stagnation and blood stasis after laparoscopy,improve follicular development,promote ovulation,improve endometrial receptivity,while being a safe treatment option.(Trial registration No.NCT02676713).ZHAO Rui-hua LIU Yong LU Dan WU Ying WANG Xiao-yun LI Wei-li ZENG Cheng MENG Qing-wei LIAN Feng-mei ZHOU Jun SHI Yun SUN Wei-wei HAN Qian TANG Yi SHI Guang 2020Chinese Journal of Integrative Medicine2020,26,2:13
4Growth and characterization of 2-inch high quality β-Ga2O3 single crystals grown by EFG method显示文摘β-Ga_2O_3 is an ultra-wide band-gap semiconductor with promising applications in UV optical detectors,Schottky barrier diodes, field-effect transistors and substrates for light-emitting diodes. However, the preparation of large β-Ga_2O_3 crystals is undeveloped and many properties of this material have not been discovered yet. In this work, 2-inch β-Ga_2O_3 single crystals were grown by using an edge-defined film-fed growth method. The high quality of the crystal has been proved by high-resolution X-ray diffraction with 19.06 arcsec of the full width at half maximum. 5A new dynamic pushback control method for reducing fuel-burn costs: Using predicted taxi-out time显示文摘Long departure-taxi-out time leads to significant airport surface congestion, fuel-burn costs, and excessive emissions of greenhouse gases. To reduce these undesirable effects, a Predicted taxi-out time-based Dynamic Pushback Control(PDPC) method is proposed. The implementation of this method requires two steps: first, the taxi-out times for aircraft are predicted by the leastsquares support-vector regression approach of which the parameters are optimized by an introduced improved Firefly algorithm. Then, a dynamic pushback control model equipped with a linear gate-hold penalty function is built, along with a proposed iterative taxiway queue-threshold optimization algorithm for solving the model. A case study with data obtained from Beijing International airport(PEK) is presented. The taxi-out time prediction model achieves predictive accuracy within 3 min and 5 min by 84.71% and 95.66%, respectively. The results of the proposed pushback method show that total operation cost and fuel-burn cost achieve a 14.0% and 21.1%reduction, respectively, as compared to the traditional K-control policy.(3) From the perspective of implementation, using PDPC policy can significantly reduce the queue length in taxiway and taxi-out time. The total operation cost and fuel-burn cost can be curtailed by 37.2% and 52.1%,respectively, as compared to the non-enforcement of any pushback control mechanism. 6Oxidative Stress before and after Operation in Patients with Chronic Cholecystitis Containing Gallstone显示文摘To study the relationship of oxidative, antioxidative constituents and antioxidases in blood with chronic cholecystitis containing gallstone, levels of lipoperoxides (LPO), nitric oxide (NO), vitamin C(VC), vitamin E (VE) and β carotene (β CAR) in plasma as well as level of LPO, activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH Px) in erythrocytes were investigated by spectrophotometric assay in 107 patients with this condition (PCg) and 100 healthy volunteers (HVs). Compared with HVs group, the average value of LPO and NO in plasma and that of LPO in erythrocytes of PCg group were significantly increased ( P <0.0001), while that of VC, VE and β CAR in plasma and the average activities of SOD, CAT and GSH Px in erythrocytes were significantly decreased ( P <0.0001). Linear regression and correlation analysis for 107 preoperative PCg showed that the value of LPO and NO in plasma and that of LPO in erythrocytes of PCg gradually increased ( P <0.0001), representing a significant linear positive correlation. The value of VC, VE and β CAR in plasma and that of SOD, CAT and GSH Px in erythrocytes of PCg gradually decreased ( P <0.0001), representing a significant linear negative correlation. Stepwise regression and correlation analysis for 107 preoperative PCg suggested that the closest correlation was observed between the course of disease and the value of NO and VC in plasma and that of SOD, GSH Px and LPO in erythrocytes, r =0.7306, F =32.1408, P <0.0001. Compared with the preoperative PCg group, the average value of LPO and NO in plasma and that of LPO in erythrocytes of the postoperative PCg group were significantly decreased ( P <0.0001). Furthermore, the average value of VC in plasma and that of SOD, CAT and GSH Px in erythrocytes of the postoperative PCg group were significantly increased ( P <0.0001), whereas no significant difference was found between their average value of VE and β CAR in plasma. These findings suggested that oxidative stress was an aggravating pathological condition in PCg group. 8Sini powder ameliorates the inflammatory response in rats with stress-induced non-alcoholic fatty liver disease by inhibiting the nuclear factor kappa-B/pyrin domain-containing protein 3 pathway显示文摘OBJECTIVE: To evaluate the effectiveness of Sini powder for the treatment of non-alcoholic fatty liver disease(NAFLD) in rats and the molecular mechanisms involved.METHODS: A rat model of stress-induced NAFLD was established by a combination of long-term tethering and feeding of a high-fat, high-calorie diet. These rats were then intragastrically administered with either simvastatin, Sini powder, or vehicle for 1 week. The body mass and field test scores for each group were recorded weekly. Serum aspartate aminotransferase and alanine aminotransferase activities, and triglyceride, total cholesterol,and free fatty acid concentrations were measured.Liver tissue histopathology was examined on hematoxylin and eosin-stained paraffin sections and oil red O-stained frozen sections. The hepatic m RNA expression of nuclear factor kappa-B(NF-κB),NOD-, LRR-, and pyrin domain-containing protein 3(NLRP3), apoptosis-associated speck-like protein containing CARD(ASC), and caspase-1 were measured by reverse transcription-polymerase chain reaction(RT-PCR). The hepatic protein concentrations of NF-κB and NLRP3, ASC, caspase-1, interleukin-1β(IL-1β), interleukin-6(IL-6), and the serum concentrations of IL-1β and IL-6 were measured by enzyme-linked immunosorbent assay.RESULTS: Compared with the Blank group, rats in the Compound model group showed significant pathologic manifestations of NAFLD, and the expression of NF-κB, NLRP3, ASC, caspase-1, IL-1βand IL-6 were significantly higher(all P < 0.01). Both simvastatin and Sini powder significantly ameliorated the NAFLD pathology and the abnormal expression of NF-κB, NLRP3, ASC, caspase-1, IL-1β, and IL-6(all P < 0.01).CONCLUSION: Sini powder inhibits the inflamma-tory response in rats with NAFLD, which is mediated by NF-κB/NLRP3, IL-1β, and IL-6, reduces the effects of psychological stress, and improves lipid metabolism. 11Microstructure of Steel 5Cr21Mn9Ni4N Alloyed by Rare Earth显示文摘The microstructure, composition and shape of precipitated piase under as-cast and finished product state of 5Cr21Mn9Ni4N steel with different rare earth (RE) amount were studied. Mechanical properties of 5Cr21Mn9Ni4N steels withont RE addition and with RE added by 0.2% in mass percent were tested respectively. The results indicate that the solid solution amount of RE is about 10 -6-10 -5 order of magnitude in 5Cr21Mn9Ni4N steel. Dendrite of as-cast condition is refined obviously and dimension of interstitial phase is shortened when RE is added by 0.10%-0.20%. But the microstructure will be coarser if surplus RE is added. 14Coral community changes in response to a high sedimentation event:A case study in southern Hainan Island显示文摘A high sedimentation event caused by dredging and dumping of sediment was recorded on Xiaodonghai Reef in Yulin Bay,southern Hainan Island,China.Significantly high sedimentation and constant light shading were observed during the sediment dumping event(SD Event).Using long-term video transects,we quantified coral community changes and responses to the SD Event between 2008 and 2010.The SD Event caused severe coral mortality on Xiaodonghai Reef at a depth of 6 m,while corals at 3 m were less affected.Total live coral cover at 6 m decreased from 54.3% to 14.8%,and Diploastrea heliopora replaced Galaxea fascicularis as the dominant coral species at 6 and 9 m.The density of juvenile corals also declined after the SD Event,especially for the genera Galaxea and Platygyra.However,the density of juvenile Porites and Pocillopora spp.slightly increased.Monitoring for 11 months after the SD Event indicated no recovery of coral communities on Xiaodonghai Reef.Long-term video transect data also revealed that mean live coral cover dramatically declined,from 30.5% in 2008 to 9% in 2010,while the dominant corals in Yulin Bay shifted to more tolerant coral species,such as massive Porites spp.and D.heliopora.The rapid coral community degradation in Yulin Bay between 2008 and 2010 was probably caused by high sediment deposition resulting from intensive dredging and land-clearing activities.These results highlight the necessity for an integrated watershed management to control sediment deposition on near-shore coral reefs.LI XiuBao HUANG Hui LIAN JianSheng YANG JianHui YE Cheng CHEN YongQiang HUANG LiangMin 2013Chinese Science Bulletin2013,58,9:7

17High-affinity mutant Interleukin-13 targeted CAR T cells enhance delivery of clickable biodegradable fluorescent nanoparticles to glioblastoma显示文摘Glioblastoma(GBM),the deadliest form of brain cancer,presents long-standing problems due to its localization.Chimeric antigen receptor(CAR)T cell immunotherapy has emerged as a powerful strategy to treat cancer.IL-13-receptor-α2(IL13Rα2),present in over 75%of GBMs,has been recognized as an attractive candidate for antiglioblastoma therapy.Here,we propose a novel multidisciplinary approach to target brain tumors using a combination of fluorescent,therapeutic nanoparticles and CAR T cells modified with a targeted-quadruplemutant of IL13(TQM-13)shown to have high binding affinity to IL13Rα2-expressing glioblastoma cells with low off-target toxicity.Azide-alkyne cycloaddition conjugation of nanoparticles to the surface of T cells allowed a facile,selective,and high-yielding clicking of the nanoparticles.Nanoparticles clicked onto T cells were retained for at least 8 days showing that the linkage is stable and promising a suitable time window for in vivo delivery.T cells clicked with doxorubicin-loaded nanoparticles showed a higher cytotoxic effect in vitro compared to bare T cells.In vitro and in vivo T cells expressing TQM-13 served as delivery shuttles for nanoparticles and significantly increased the number of nanoparticles reaching brain tumors compared to nanoparticles alone.This work represents a new platform to allow the delivery of therapeutic nanoparticles and T cells to solid tumors.Gloria B.Kim Virginia Aragon-Sanabria Lauren Randolph Hali Jiang Joshua A.Reynolds Becky S.Webb Achuthamangalam Madhankumar Xiaojun Lian James R.Connor Jian Yang Cheng Dong 2020Bioactive Materials2020,5,3:6
18Copper single-atom catalysts with photothermal performance and enhanced nanozyme activity for bacteria-infected wound therapy显示文摘Nanozymes have become a new generation of antibiotics with exciting broad-spectrum antibacterial properties and negligible biological toxicity.However,their inherent low catalytic activity limits their antibacterial properties.Herein,Cu single-atom sites/N doped porous carbon(Cu SASs/NPC)is successfully constructed for photothermal-catalytic antibacterial treatment by a pyrolysis-etching-adsorption-pyrolysis(PEAP)strategy.Cu SASs/NPC have stronger peroxidase-like catalytic activity,glutathione(GSH)-depleting function,and photothermal property compared with non-Cu-doped NPC,indicating that Cu doping significantly improves the catalytic performance of nanozymes.Cu SASs/NPC can effectively induce peroxidase-like activity in the presence of H2O2,thereby generating a large amount of hydroxyl radicals(•OH),which have a certain killing effect on bacteria and make bacteria more susceptible to temperature.The introduction of near-infrared(NIR)light can generate hyperthermia to fight bacteria,and enhance the peroxidase-like catalytic activity,thereby generating additional•OH to destroy bacteria.Interestingly,Cu SASs/NPC can act as GSH peroxidase(GSH-Px)-like nanozymes,which can deplete GSH in bacteria,thereby significantly improving the sterilization effect.PTT-catalytic synergistic antibacterial strategy produces almost 100%antibacterial efficiency against Escherichia coli(E.coli)and methicillin-resistant Staphylococcus aureus(MRSA).In vivo experiments show a better PTT-catalytic synergistic therapeutic performance on MRSA-infected mouse wounds.Overall,our work highlights the wide antibacterial and anti-infective bio-applications of Cu single-atom-containing catalysts.Xianwen Wang Qianqian Shi Zhengbao Zha Dongdong Zhu Lirong Zheng Luoxiang Shi Xianwen Wei Lian Lian Konglin Wu Liang Cheng 2021Bioactive Materials2021,6,12:6
19Multiscale modeling of electrolytes in porous electrode:From equilibrium structure to non-equilibrium transport显示文摘Understanding the mechanisms and properties of various transport processes in the electrolyte,porous electrode,and at the interface between electrode and electrolyte plays a crucial role in guiding the improvement of electrolytes,materials and microstructures of electrode.Nanoscale equilibrium properties and nonequilibrium ion transport are substantially different to that in the bulk,which are difficult to observe from experiments directly.In this paper,we introduce equilibrium and no-equilibrium thermodynamics for electrolyte in porous electrodes or electrolyte-electrode interface.The equilibrium properties of electrical double layer(EDL)including the EDL structure and capacitance are discussed.In addition,classical non-equilibrium thermodynamic theory is introduced to help us understand the coupling effect of different transport processes.We also review the recent studies of nonequilibrium ion transport in porous electrode by molecular and continuum methods,among these methods,dynamic density functional theory(DDFT)shows tremendous potential as its high efficiency and high accuracy.Moreover,some opportunities for future development and application of the non-equilibrium thermodynamics in electrochemical system are prospected.Haolan Tao Cheng Lian Honglai Liu 2020Green Energy & Environment2020,5,3:5
