| 1 | Differing coping mechanisms, stress level and anorectal physiology in patients with functional constipation显示文摘AIM: To investigate coping mechanisms, constipation symptoms and anorectal physiology in 80 constipated subjects and 18 controls.METHODS: Constipation was diagnosed by Rome Ⅱ criteria.Coping ability and anxiety/depression were assessed by validated questionnaires. Transit time and balloon distension test were performed.RESULTS: 34.5% patients were classified as slow transit type of constipation. The total colonic transit time (56 h vs 10 h, P<0.0001) and rectal sensation including urge sensation (79 mL vs 63 mL, P = 0.019) and maximum tolerable volume (110 mL vs95 mL, P = 0.03) differed in patients and controls. Constipated subjects had significantly higher anxiety and depression scores and lower SF-36 scores in all categories. They also demonstrated higher scores of'monitoring' coping strategy (14+6 vs9+3, P = 0.001),which correlated with the rectal distension sensation (P = 0.005), urge sensation (P=0.002), and maximum tolerable volume (P = 0.035). | 3 | Reappraisal of endoscopic papillary balloon dilation for the management of common bile duct stones显示文摘Although endoscopic sphincterotomy(EST) is still considered as a gold standard treatment for common bile duct(CBD) stones in western guideline, endoscopic papillary balloon dilation(EPBD) is commonly used by the endoscopists in Asia as the first-line treatment for CBD stones. Besides the advantages of a technical easy procedure, endoscopic papillary large balloon dilation(EPLBD) can facilitate the removal of large CBD stones.The indication of EPBD is now extended from removal of the small stones by using traditional balloon, to removal of large stones and avoidance of lithotripsy by using large balloon alone or after EST. According to the reports of antegrade papillary balloon dilatation, balloon dilation itself is not the cause of pancreatitis. On the contrary, adequate dilation of papillary orifice can reduce the trauma to the papilla and pancreas by the basket or lithotripter during the procedure of stone extraction. EPLBD alone is as effective as EPLBD with limited EST. Longer ballooning time may be beneficial in EPLBD alone to achieve adequate loosening of papillary orifice. The longer ballooning time does not increase the risk of pancreatitis but may reduce the bleeding episodes in patients with coagulopathy. Slowly inflation of the balloon, but not exceed the diameter of bile duct and tolerance of the patients are important to prevent the complication of perforation. | 4 | Aberrant crypt loci as microscopic precursors of colorectal cancer显示文摘Since the first detection of aberrant crypt foci (ACF) in carcinogen-treated mice, there have been numerous studies focusing on these microscopically visible lesions both in rodents and in humans. ACF have been generally accepted as precancerous lesions in regard to histopathological characteristics, biochemical and immunohistochemical alterations, and genetic and epigenetic alterations. ACF show variable histological features, ranging from hyperplasia to dysplasia. ACF in human colon are more frequently located in the distal parts than in the proximal parts, which is in accordance with those in colorectal cancer (CRC). The immunohistochemical expressions of carcinoembryonic antigen (CEA), β-catenin, placental cadherin (P-cadherin),epithelial cadherin (E-cadherin), inducible nitric oxide synthase (iNOS), cyclooxygenase (COX-2), and P16INK4a are found to be altered. Genetic mutations of K-ras, APC and p53, and the epigenetic alterations of CpG island methylation of ACF have also been demonstrated. Genomic instabilities due to the defect of mismatch repair (MMR) system are detectable in ACF Two hypotheses have been proposed.One is the 'dysplasia ACF-adenoma-carcinoma sequence',the other is 'heteroplastic ACF-adenoma-carcinoma sequence'. The malignant potential of ACF, especially dyspiastic ACF, makes it necessary to reveal the nature of these lesions, and to prevent CRC from the earliest possible stage. | 6 | Protective effects and mechanisms of Baicalin and octreotide on renal injury of rats with severe acute pancreatitis显示文摘AIM: To investigate the protective effects and mechanisms of Baicalin and octreotide on renal injury of rats with severe acute pancreatitis (SAP). METHODS: One hundred and eighty SD rats were randomly assigned to the model group, Baicalin-treated group, octreotide-treated group and sham operation group. The mortality, plasma endotoxin level, contents of blood urea nitrogen (BUN), creatinine (CREA), phospholipase A2 (PLA2), nitrogen monoxide (NO), tumor necrosis factor (TNF)-α, IL-6 and endothelin-1 (ET-1) in serum, expression levels of renal Bax and Bcl-2 protein, apoptotic indexes and pathological changes of kidney were observed at 3, 6 and 12 h after operation. RESULTS: The renal pathological changes were milder in treated group than in model group. The survival at 12 h and renal apoptotic indexes at 6 h were significantly (P < 0.05) higher in treated group than in model group [66.67% vs 100%; 0.00 (0.02)% and 0.00 (0.04)% vs 0.00 (0.00)%, respectively]. The serum CREA content was markedly lower in octreotide-treated group than in model group at 3 h and 6 h (P < 0.01, 29.200 ± 5.710 μmol/L vs 38.400 ± 11.344 μmol/L; P < 0.05, 33.533 ± 10.106 μmol/L vs 45.154 ± 17.435 μmol/L, respectively). The expression level of renal Bax protein was not significantly different between model group and treated groups at all time points. The expression level of renal Bcl-2 protein was lower in Baicalin-treated group than in model group at 6 h [P < 0.001, 0.00 (0.00) grade score vs 3.00 (3.00) grade score]. The Bcl-2 expression level was lower in octreotide-treated group than in model group at 6 h and 12 h [P < 0.05, 0.00 (0.00) grade score vs 3.00 (3.00) grade score; 0.00 (0.00) grade score vs 0.00 (1.25) grade score, respectively]. The serum NO contents were lower in treated groups than in model group at 3 h and 12 h [P < 0.05, 57.50 (22.50) and 52.50 (15.00) μmol/L vs 65.00 (7.50) μmol/L; P < 0.01, 57.50 (27.50) and 45.00 (12.50) μmol/L vs 74.10 (26.15) μmol/L, respectively]. The plasma endotoxin content and serum BUN content (at 6 h and 12 h) were lower in treated groups than in model group. The new method is an extension of the progressive interpolation technique for B-splines. Given a triangular mesh M, the idea is to iteratively upgrade the vertices of M to generate a new control mesh M such that limit surface of M would interpolate M. It can be shown that the iterative process is convergent for Loop subdivision surfaces. Hence, the method is well-defined. The new method has the advantages of both a local method and a global method, i.e., it can handle meshes of any size and any topology while generating smooth interpolating subdivision surfaces that faithfully resemble the shape of the given meshes. | 8 | Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. | 10 | Effect of temperature on the development of sclerotia in Villosiclava virens显示文摘The sclerotia of Villosiclava virens were found commonly in high altitude and the temperate regions, where the temperatures are relatively low in rice filling stage. To make sure if low temperature induce the sclerotial formation in V. virens, the inoculated rice panicles in laboratory and the diseased rice panicles cut from paddy fields were treated under different temperatures. The results showed that 3 days of night temperature at 15°C were enough to induce the sclerotial formation. The low temperature was much more effective for young balls with intact membranes. After appearance of chlamydospores on the ball surfaces, the sclerotium could not differentiate anymore. The sclerotia began to differentiate below the chlamydospore layer and gradually grew onto the ball surfaces. | 11 | Effect of artesunate supplementation on bacterial translocation and dysbiosis of gut microbiota in rats with liver cirrhosis显示文摘AIM: To evaluate the effect of artesunate(AS) supplementation on bacterial translocation(BT) and gut microbiota in a rat model of liver cirrhosis. METHODS: Fifty-four male Sprague-Dawley rats were randomly divided into a normal control group(N), a liver cirrhosis group(M) and a liver cirrhosis group intervened with AS(MA). Each group was sampled at 4, 6 and 8 wk. Liver cirrhosis was induced by injection of carbon tetrachloride(CCl4), intragastric administration of 10% ethanol, and feeding a high fat diet. Rats in the MA group were intragastrically administered with AS(25 mg/kg body weight, once daily). Injuries of the liver and intestinal mucosa were assessed by hematoxylineosin or Masson's trichrome staining. Liver index was calculated as a ratio of the organ weight(g) to body weight(g). The gut microbiota was examined by automated ribosomal intergenic-spacer analysis of fecal DNA. BT was assessed by standard microbiological techniques in the blood, mesenteric lymph nodes(MLNs), liver, spleen, and kidney. RESULTS: Compared to group N, the body weight was reduced significantly in groups M and MA due to the development of liver cirrhosis over the period of 8 wk. The body weight was higher in group MA than in group M. The liver indices were significantly elevated at 4, 6 and 8 wk in groups M and MA compared to group N. AS supplementation partially decreased the liver indices in group MA. Marked histopathologic changes in the liver and small intestinal mucosa in group M were observed, which were alleviated in group MA. Levels of pro-inflammatory interleukin-6 and tumor necrosis factor-α were significantly elevated at 8 wk in ileal homogenates in group M compared to group N, which were decreased after AS supplementation in group MA. The dysbiosis of gut microbiota indicated by the mean diversity(Shannon index) and mean similarity(Sorenson index) was severe as the liver cirrhosis developed, and AS supplementation had an apparent intervention effect on the dysbiosis of gut microbiota at 4 wk. The occurrence of BT was increased in the liver of group M compared to that of group N. AS supplementation reduced BT in group MA at 8 wk. BT also occurred in the MLNs, spleen, and kidney, which was reduced by AS supplementation. BT was not detected in the blood in any group.CONCLUSION: Dysbiosis of gut microbiota, injury of intestinal mucosal barrier and BT occurred as liver cirrhosis progressed, which might enhance inflammation and aggravate liver injury. | 12 | Thermodynamic conditions of framework grain dissolution of clastic rocks and its application in Kela 2 gas field显示文摘Feldspar and clastic debris are the most important constituent framework grains of sedimentary clastic rocks and their chemical dissolution plays an essential role in the formation and evolution of the secondary pore in the reservoir rocks. On the basis of thermodynamic phase equilibrium, this study investigates the chemical equilibrium relationships between fluid and various plagioclase and K-feldspar in diagenesis of the sediments, particularly, the impact of temperature and fluid compositions (pH, activity of K+, Na+, Ca2+ and so on) on precipitation and dissolution equilibria of feldspars. Feldspar is extremely easily dissolved in the acid pore water with a low salinity when temperature decreases. The dissolution of anorthite end-member of plagioclase is related to the Ca content of the mineral and the fluid, higher Ca either in the mineral or in the fluid, easier dissolution of the feldspar. Moreover, the dissolution of albite end-member of plagioclase is related to Na of both the mineral and fluid, lower Na out of both the mineral and fluid, easier dissolution of the mineral. Similarly, lower-K fluid is more powerful to dissolve K-feldspar than the higher. The anorthite component of plagioclase is most easily dissolved in ground water-rock system, the albite is the secondary, and K-feldspar is the most stable. Selective dissolution of plagioclase occurs in diagenesis because of the plagioclase solid solution that is mainly composed of albite and anorthite end-members, Ca-rich part of which is preferentially dissolved by the pore water, in contrast to the Na-rich parts. Based on investigation of reservoir quality, secondary pore, dissolution structures of feldspar, clay minerals and ground water chemistry of the Kela 2 gas field of Kuqa Depression in the Tarim basin of Western China, we discovered that the secondary pore is very well developed in the highest quality section of the reservoir, and the plagioclase of which was obviously selectively dissolved, in contrast to the overgrowth of K-feldspar. Chemistry of the ground water out of the highest quality section of the reservoir is consistent with the complete dissolution field of anorthite, partial dissolution field of albite component of plagioclase, and the precipitation of K-feldspar on the temperature-logK phase equilibrium diagram drawn from this study. The thermodynamic modeling calculation gave us an insight into formation of the secondary pore within the reservoir, and the dissolution of plagioclase an important cause of the secondary pore. | 13 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 15 | Analysis of Disequilibrium Hybridization in Hybrid and Backcross Progenies of Saccharum officinarum × Erianthus arundinaceus显示文摘Erianthus arundinaceus is an important, closely related genus of Saccharum officinarum L. It is therefore important to understand how the chromosomes are transmitted when it hybridizes with sugarcane. The hybrids and backcross progenies of S. officinarum and E. arundinaceus and their parents were used for Karyotype analysis and to study the law of chromosome transmission. The results showed that the somatic chromosome number of both of the E. arundinaceus Hainan92-105 and Hainan92-77 were 2n = 60 = 60sm, belonging to type 1A, and the BC1 YC01-21 was 2n = 104 = 100m + 4sm, belonging to type 2C. The other six tested clones belonged to type 2B. The both F1s YC96-66 and YC96-40 that originated from Badila (2n = 80 = 70m + 10sm) with E. Arundinaceus were 2n = 70 = 68m + 2sm, which suggests an n + n transmission. The cross between YC96-66 (female parent) and CP84-1198 (male parent, 2n = 120 = 114m + 6sm) also followed the same genetic law and the somatic chromosome number of their progeny, YC01-3 (2n = 105 = 95m + 10sm). The cross derived from YC96-40 (female) and CP84-1198 (male), YC01-21 had 2n = 104 = 100m + 4sm chromosomes, following the same genetic law of n + n. However, YC01-36 had 2n = 132 = 130m + 2sm chromosomes, which suggests a 2n + n chromosome transmission. It can be inferred that the inheritance of chromosomes was very complex in the BC1. The difference in chromosome number between clones was as high as 28. This could be explained by the 2n + n transmission of chromosomes. | 17 | Metabolomic alterations and chromosomal instability status in gastric cancer显示文摘AIM To explore the correlation of metabolomics profiles ofgastric cancer(GC) with its chromosomal instability(CIN) status.METHODS Nineteen GC patients were classified as CIN and nonCIN type by The Cancer Genome Atlas Research Group system, based on 409 oncogenes and tumor suppressor genes sequenced. The aqueous metabolites of the GC tumor and its surrounding adjacent healthy tissues were identified through liquid chromatographymass spectrometry. Groups were compared by defining variable importance in projection score of > 1.2, a fold change value or its reciprocal of > 1.2, and a P value of < 0.05 as a significant difference.RESULTS In total,twelve men and seven women were enrolled, with a median age of 66 years(range, 47-87 years). The numbers of gene alterations in the CIN GC group were significantly higher than those in the non-CIN GC(32-218 vs 2-17; P < 0.0005). Compared with the adjacent healthy tissues, GC tumors demonstrated significantly higher aspartic acid, citicoline, glutamic acid, oxidized glutathione, succinyladenosine, and uridine diphosphate-Nacetylglucosamine levels, but significantly lower butyrylcarnitine, glutathione hydroxyhexanoycarnitine, inosinic acid, isovalerylcarnitine, and threonine levels(all P < 0.05). CIN tumors contained significantly higher phosphocholine and uridine 5'-monophosphate levels but significantly lower beta-citryl-L-glutamic acid levels than did non-CIN tumors(all P < 0.05). | 18 | Suppressing secondary reactions of coal pyrolysis by reducing pressure and mounting internals in fixed-bed reactor显示文摘Pyrolysis of Shenmu coal was performed in fixed-bed reactors indirectly heated by reducing operating pressure and mounting internals in the reactor to explore their synergetic effects on coal pyrolysis. Mounting internals particularly designed greatly improved the heat transfer inside coal bed and raised the yield of tar production.Reducing pressure further facilitated the production of tar through its suppression of secondary reactions occurring in the reactor. The absolute increase in tar yield reached 3.33 wt% in comparison with the pyrolysis in the reactor without internals under atmospheric pressure. The obtained tar yield in the reactor with internals under reduced pressure was even higher than the yield of Gray–King assay. Through experiments in a laboratory fixed bed reactor, it was also clarified that the effect of reducing pressure is related to volatile release rate in pyrolysis. It did not obviously vary tar yield at pyrolysis temperatures below 600 °C, while the effect was evident at 650 and 700 °C but became limited again above 800 °C. | 19 | Resveratrol induces AMPK and mTOR signaling inhibition-mediated autophagy and apoptosis in multiple myeloma cells显示文摘Resveratrol,a natural compound extracted from the skins of grapes,berries,or other fruits,has been shown to have anti-tumor effects against multiple myeloma(MM)via promoting apoptosis and inhibiting cell viability.In addition to apoptosis,autophagy also plays a significant role in anti-tumor effects.However,whether autophagy is involved in anti-MM activity of resveratrol remains unclear.In this study,human MM cell lines U266,RPMI-8226,and NCI-H929 were treated with resveratrol.Cell Counting Kit-8 assay and colony formation assay were used to measure cell viability.Western blot analysis was used to detect apoptosis-and autophagy-associated proteins.3-Methyladenine(3-MA)was applied to inhibit autophagy.Results showed that resveratrol inhibited cell viability and colony formation via promoting apoptosis and autophagy in MM cell lines U266,RPMI-8226,and NCI-H929.Resveratrol promoted apoptosis-related proteins,Caspase-3 activating poly-ADP-ribose polymerase and Caspase-3 cleavage,and decreased the protein level of Survivin in a dose-dependent manner.Additionally,resveratrol upregulated the levels of LC3 and Beclin1 in a dose-dependent way,indicating that autophagy might be implicated in anti-MM effect of resveratrol.Furthermore,3-MA relieved the cytotoxicity of resveratrol by blocking the autophagic flux.Resveratrol increased the phosphorylation of adenosine monophosphate(AMP)-activated protein kinase and decreased the phosphorylation of mammalian target of rapamycin(mTOR)and its downstream substrates p70S6K and 4EBP1 in a dose-dependent manner,leading to autophagy.Therefore,our results suggest that resveratrol exerts anti-MM effects through apoptosis and autophagy,which can be used as a new therapeutic strategy for MM in clinic. | Ruye Ma Dandan Yu Yu Peng Hongfei Yi Yingcong Wang Taofang Cheng Bingqing Shi Guang Yang Weiming Lai Xiaosong Wu Ye Lu Jumei Shi | 2021 | Acta Biochimica et Biophysica Sinica2021,53,6: | 7 |

Several beneficial microbes have been investigated and have yielded remarkable results. The endophytic fungus, Piriformospora indica, is one microbe group with the potential to offer numerous benefits and opportunities in modern horticultural crop production. This plant growth-promoting mycorrhizal fungus is of particular interest due to its beneficial implications in plant growth and development. The fungus has been experimentally proven to significantly improve water and nutrient/mineral absorption, early flowering, seed production/germination and plant photosynthetic capability, growth rates;especially in nutrient-deprived soils, alter the production of secondary metabolites, and promote adaptation, tolerance and/or resistance to biotic and abiotic stressors. Its unique capability to be axenically cultured makes it feasible for research and application. These attributes in combinations with its broad host range, offer immense potential for this fungus for research in horticultural plant improvement.
