| 2 | OsWRKY03, a rice transcriptional activator that functions in defense signaling pathway upstream of OsNPR1显示文摘WRKY family proteins are a class of plant specific transcription factors that involve in many stress response pathways. It has been shown that one Arabidopsis WRKY protein, AtWRKY29/22, is activated by MAP kinase signaling cascade and confers resistance to both bacterial and fungal pathogens. However, little is known about the biological roles of WRKY proteins in rice. In this study, we investigated the expression patterns of rice AtWRKY29/22 homolog, OsWRKY03, under different conditions, and also its possible role involved in plant defense. Our results showed that OsWRKY03 was up-regulated by several defense signaling molecules or different treatments. Further analysis revealed that the expres- sion of OsWRKY03 was light dependent. Transcriptional activation activity of OsWRKY03 was also demonstrated by yeast functional assay. Transient expression of OsWRKY03-GFP fusion protein in onion epidermis cells showed that OsWRKY03 was a nuclear localized protein. OsNPR1 as well as several other pathogenesis-related genes, such as OsPR1b, phenylalanine ammonia-lyase (ZB8) and peroxidase (POX22.3), were induced in OsWRKY03-overexpressing transgenic plants. | 3 | Methanation of carbon dioxide on Ni/ZrO_2-Al_2O_3 catalysts:Effects of ZrO_2 promoter and preparation method of novel ZrO_2-Al_2O_3 carrier显示文摘The novel nickel-based catalysts with a nickel content of 12 wt% were prepared with the zirconia-alumina composite as the supports. The new carriers, ZrO2 improved alumina, were synthesized by three methods, i.e., impregnation-precipitation, co-precipitation, and impregnation method. The catalytic properties of these catalysts were investigated in the methanation of carbon dioxide, and the samples were characterized by X-ray diffraction (XRD), X-ray photoelectron spectroscope (XPS), temperature-programmed reduction (TPR) and temperature-programmed desorption (TPD) techniques. The new catalysts showed higher catalytic activity and better stability than Ni/γ-Al2O3. Furthermore, as a support for new nickel catalyst, the ZrO2-Al2O3 composite prepared by the impregnation-precipitation method was more efficient than the other supports in the methanation of carbon dioxide. | 8 | Clinical analysis of multiple primary malignancies in the digestive system: A hospital-based study显示文摘AIM: To analyze the characteristics of multiple primary malignancies (MPMs) of digestive system; including incidence, types of tumor combinations, time intervals between development of multiple tumors, clinical course,and prognostic factors affecting survival and mortality.METHODS: Data from a total of 129 patients treated from January 1991 to December 2000 for pathologically proved MPMs, including at least one originating from the digestive system, were reviewed retrospectively.RESULTS: Among 129 patients, 120 (93.02%) had two primary cancers and 9 (6.98%) had three primary cancers. The major sites of MPMs of the digestive system were large intestine, stomach, and liver. Associated nondigestive cancers included 40 cases of gynecological cancers, of which 31 were carcinoma of cervix and 10cases of genitourinary cancers, of which 5 were bladder cancers. Other cancers originated from the lung, breast,nasopharynx, larynx, thyroid, brain, muscle, and skin.Reproductive tract cancers, especially cervical, ovarian,bladder, and prostate cancers were the most commonly associated non-GI cancers, followed by cancer of the lung and breasts. Forty-three cases were synchronous, while the rest (86 cases) were metachronous cancers. Staging of MPMs and treatment regimes correlated with the prognosis between survival and non-survival groups.CONCLUSION: As advances in cancer therapy bring about a progressively larger percentage of long-term survivors, the proportion of patients with subsequent primary lesions will increase. | 9 | Detection and fine-mapping of SC7 resistance genes via linkage and association analysis in soybean显示文摘Soybean mosaic virus(SMV) disease is one of the most serious and broadly distributed soybean(Glycine max(L.) Merr.) diseases. Here, we combine the advantages of association and linkage analysis to identify and fine-map the soybean genes associated with resistance to SMV strain SC7.A set of 191 soybean accessions from different geographic origins and 184 recombinant inbred lines(RILs) derived from Kefeng No.1(resistant) Nannong 1138-2(susceptible) were used in this study. The SC7 resistance genes were previously mapped to a 2.65 Mb region on chromosome 2 and a 380 kb region on chromosome 13. Among 19 single nucleotide polymorphisms(SNPs) detected via association analysis in the study, the SNP BARC-021625-04157 was located in the2.65 Mb region, and the SNP BARC-041671-08065 was located near the 380 kb region; three genes harboring the SNPs were probably related to SC7 resistance. The resistance gene associated with BARC-021625-04157 was then finemapped to a region of approximately 158 kb on chromosome2 using 184 RILs. Among the 15 genes within this region, one NBS-LRR type gene, one HSP40 gene and one serine carboxypeptidase-type gene might be candidate SC7 resistance genes. | 12 | Uncertainty analysis of hydrological model parameters based on the bootstrap method:A case study of the SWAT model applied to the Dongliao River Watershed,Jilin Province,Northeastern China显示文摘As an important tool for the description and analysis of hydrological processes,the watershed hydrological model has been increasingly applied to watershed hydrological simulations and water resource management.However,in most cases,model parameters are only determined in a calibration scheme which fits the modeled data to observations,thus significant uncertainties exist in the model parameters.How to quantitatively evaluate the uncertainties in model parameters and the resulting uncertainty impacts on model simulations has always been a question which has attracted much attention.In this study,two methods based on the bootstrap method(specifically,the model-based bootstrap and block bootstrap)are used to analyze the parameter uncertainties in the case of the SWAT(Soil and Water Assessment Tool)model applied to a hydrological simulation of the Dongliao River Watershed.Then,the uncertainty ranges of five sensitivity parameters are obtained.The calculated variation coefficients and the variable parameter contributions show that,among the five parameters,ESCO and CN2 have relatively high uncertainties:the variation coefficients and contribution rates are 23.98 and 70%,14.43 and 18%,respectively.The three remaining parameters have relatively low uncertainties.We compare the two uncertainty ranges of parameters acquired by the two bootstrap methods,and find that the uncertainty ranges of parameters acquired by the block bootstrap are narrower than those acquired by the model-based bootstrap.Further analysis of the effects of parameter uncertainties on the model simulation reveals that the parameter uncertainties have great impacts on results of the model simulation,and in the model calibration stage 60%70%of runoff observations were within the corresponding 95%confidence interval.The uncertainty in the model simulation during the flood season(i.e.the wet period)is relatively higher than that during the dry season. | ZHANG Zheng LU WenXi CHU HaiBo CHENG WeiGuo ZHAO Ying | 2014 | Science China(Technological Sciences)2014,57,1: | 9 |

| 15 | Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. | 16 | Time-domain solution for transient dynamic response of a large-diameter thin-walled pipe pile显示文摘The propagation of stress waves in a large-diameter pipe pile for low strain dynamic testing cannot be explained properly by traditional 1D wave theories. A new computational model is established to obtain a wave equation that can describe the dynamic response of a large-diameter thin-walled pipe pile to a transient point load during a low strain integrity test. An analytical solution in the time domain is deduced using the separation of variables and variation of constant methods. The validity of this new solution is verifi ed by an existing analytical solution under free boundary conditions. The results of this time domain solution are also compared with the results of a frequency domain solution and fi eld test data. The comparisons indicate that the new solution agrees well with the results of previous solutions. Parametric studies using the new solution with reference to a case study are also carried out. The results show that the mode number affects the accuracy of the dynamic response. A mode number greater than 10 is required to enable the calculated dynamic responses to be independent of the mode number. The dynamic response is also greatly affected by soil properties. The larger the side resistance, the smaller the displacement response and the smaller the refl ected velocity wave crest. The displacement increases as the stress waves propagate along the pile when the pile shaft is free. The incident waves of displacement and velocity responses of the pile are not the same among different points in the circumferential direction on the pile top. | 20 | A Tailored High-Efficiency Sample Pretreatment Method for Simultaneous Quantification of 10 Classes of Known Endogenous Phytohormones显示文摘One of the hottest topics in plant hormone biology is the crosstalk mechanisms,whereby multiple classes of phytohormones interplay with each other through signaling networks.To better understand the roles of hormonal crosstalks in their complex regulatory networks,it is of high significance to investigate the spatial and temporal distributions of multiple-phytohormones simultaneously from one plant tissue sample.In this study,we develop a high-sensitivity and high-throughput method for the simultaneous quantitative analysis of 44 phytohormone compounds,covering currently known 10 major classes of phytohormones(strigolactones,brassinosteroids,gibberellins,auxin,abscisic acid,jasmonic acid,salicylic acid,cytokinins,ethylene,and polypeptide hormones[e.g.,phytosulfokine])from only 100 mg of plant sample.These compounds were grouped and purified separately with a tailored solid-phase extraction procedure based on their physicochemical properties and then analyzed by LC–MS/MS.The recoveries of our method ranged from 49.6%to 99.9%and the matrix effects from 61.8%to 102.5%,indicating that the overall sample pretreatment design resulted in good purification.The limits of quantitation(LOQs)of our method ranged from 0.06 to 1.29 pg/100 mg fresh weight and its precision was less than 13.4%,indicating high sensitivity and good reproducibility of the method.Tests of our method in different plant matrices demonstrated its wide applicability.Collectively,these advantages will make our method helpful in clarifying the crosstalk networks of phytohormones. | Peiyong Xin Qiuhuan Guo Bingbing Li Shujing Cheng Jijun Yan Jinfang Chu | 2020 | Plant Communications2020,1,3: | 7 |
