2RANTES gene single nucleotide polymorphisms and expression in patients with chronic hepatitis B virus infection显示文摘Background Regulated on activation, normal T-cell expressed and secreted (RANTES) plays a critical role in T-lymphocyte activation and proliferation. The process is involved in both acute and chronic phases of inflammation. The present study was to ascertain the possible correlations between chronic hepatitis B virus (HBV) infection and the RANTES gene polymorphisms and their expression. Methods The study included 130 HBV negative healthy donors and 152 patients with chronic hepatitis B (CHB) virus infection. The polymerase chain reaction (PCR) and restriction fragment length polymorphisms (RFLPs) were used to detect RANTES gene single nucleotide polymorphisms (SNPs). RANTES levels in the platelet depleted plasma were detected by enzyme linked immunosorbent assay (ELISA). Results RANTES alleles -403G, -28C and In1.1T were the predominant alleles in the subjects studied. No significant correlation was found between CHB infection and the RANTES alleles, while a significant correlation was found between CHB infection and increased RANTES expression in platelet depleted plasma (P<0.05). Conclusions SNPs in RANTES gene do not affect chronic HBV infection or the outcome of interferon-α treatment in patients positive for HBV “e” antigen (HBeAg+). 3Temperature action and effect of concrete-filled steel tubular bridges:A review显示文摘Concrete-filled steel tube(CFST)has been widely applied in bridges due to its excellent structural and constructional performance.In CFST bridges,temperature action is not only an important but also a special load that must be considered in bridge design,construction and structural evaluation.This paper presents an overview on current researches of the temperature action and effect of CFST bridges.The historical and spatial characteristics as well as corresponding influencing factors of temperature distributions are summarized,and classifications of temperature actions are also reviewed.Results show that in-depth investigations of the contributions of temperature influencing factors can help to effectively reduce the adverse temperature effects on CFST bridges in the design stage.Due to the lack of long-term measurements and insufficient use of historical meteorological data,research on the representative value along with correlation analysis of CFST temperature actions and meteorological parameters have to be further carried out.In the meanwhile,a decomposition method of temperature distribution is proposed based on structural effects of CFST bridges in this paper.The decomposed temperature actions can be used for more accurate effect calculation than traditional temperature action classification method,especially for CFST truss bridges.At last,specific temperature action patterns still need to be investigated in some of the special problems,such as longitudinal temperature differences along the CFST arch rib and thermal debonding in the CFST interface.Further investigations are required on the accurate simulation of temperature distribution of CFST bridges considering the shading effects of components and terrain.Jiang Liu Yongjian Liu Chenyu Zhang Qiuhong Zhao Yi Lyu Lei Jiang 2020Journal of Traffic and Transportation Engineering(English Edition)2020,7,2:15
4Three-dimensional bioprinting of multicell-laden scaffolds containing bone morphogenic protein-4 for promoting M2 macrophage polarization and accelerating bone defect repair in diabetes mellitus显示文摘Critical-sized bone defect repair in patients with diabetes mellitus remains a challenge in clinical treatment because of dysfunction of macrophage polarization and the inflammatory microenvironment in the bone defect region.Three-dimensional(3D)bioprinted scaffolds loaded with live cells and bioactive factors can improve cell viability and the inflammatory microenvironment and further accelerating bone repair.Here,we used modified bioinks comprising gelatin,gelatin methacryloyl(GelMA),and 4-arm poly(ethylene glycol)acrylate(PEG)to fabricate 3D bioprinted scaffolds containing BMSCs,RAW264.7 macrophages,and BMP-4-loaded mesoporous silica nanoparticles(MSNs).Addition of MSNs effectively improved the mechanical strength of GelMA/gelatin/PEG scaffolds.Moreover,MSNs sustainably released BMP-4 for long-term effectiveness.In 3D bioprinted scaffolds,BMP-4 promoted the polarization of RAW264.7 to M2 macrophages,which secrete anti-inflammatory factors and thereby reduce the levels of pro-inflammatory factors.BMP-4 released from MSNs and BMP-2 secreted from M2 macrophages collectively stimulated the osteogenic differentiation of BMSCs in the 3D bioprinted scaffolds.Furthermore,in calvarial critical-size defect models of diabetic rats,3D bioprinted scaffolds loaded with MSNs/BMP-4 induced M2 macrophage polarization and improved the inflammatory microenvironment.And 3D bioprinted scaffolds with MSNs/BMP-4,BMSCs,and RAW264.7 cells significantly accelerated bone repair.In conclusion,our results indicated that implanting 3D bioprinted scaffolds containing MSNs/BMP-4,BMSCs,and RAW264.7 cells in bone defects may be an effective method for improving diabetic bone repair,owing to the direct effects of BMP-4 on promoting osteogenesis of BMSCs and regulating M2 type macrophage polarization to improve the inflammatory microenvironment and secrete BMP-2.Xin Sun Zhenjiang Ma Xue Zhao Wenjie Jin Chenyu Zhang Jie Ma Lei Qiang Wenhao Wang Qian Deng Han Yang Jinzhong Zhao Qianqian Liang Xiaojun Zhou Tao Li Jinwu Wang 2021Bioactive Materials2021,6,3:12
5On the Key Dynamical Processes Supporting the 21.7 Zhengzhou Record-breaking Hourly Rainfall in China显示文摘An extremely heavy rainfall event occurred in Zhengzhou,China,on 20 July 2021 and produced an hourly rainfall rate of 201.9 mm,which broke the station record for China's Mainland.Based on radar observations and a convection-permitting simulation using the WRF-ARW model,this paper investigates the multiscale processes,especially those at the mesoscale,that support the extreme observed hourly rainfall.Results show that the extreme rainfall occurred in an environment characteristic of warm-sector heavy rainfall,with abundant warm moist air transported from the ocean by an abnormally northward-displaced western Pacific subtropical high and Typhoon In-Fa(2021).However,rather than through back building and echo training of convective cells often found in warm-sector heavy rainfall events,this extreme hourly rainfall event was caused by a single,quasi-stationary storm in Zhengzhou.Scale separation analysis reveals that the extreme-rainproducing storm was supported and maintained by the dynamic lifting of low-level converging flows from the north,south,and east of the storm.The low-level northerly flow originated from a mesoscale barrier jet on the eastern slope of the Taihang Mountain due to terrain blocking of large-scale easterly flows,which reached an overall balance with the southerly winds in association with a low-level meso-β-scale vortex located to the west of Zhengzhou.The large-scale easterly inflows that fed the deep convection via transport of thermodynamically unstable air into the storm prevented the eastward propagation of the weak,shallow cold pool.As a result,the convective storm was nearly stationary over Zhengzhou,resulting in record-breaking hourly precipitation.Peng WEI Xin XU Ming XUE Chenyue ZHANG Yuan WANG Kun ZHAO Ang ZHOU Shushi ZHANG Kefeng ZHU 2023Advances in Atmospheric Sciences2023,40,3:9

10A cylindrical rod ultrasonic motor with 1 mm diameter and its application in endo-scopic OCT显示文摘A tiny ultrasonic motor was constructed by using a piezoelectric ceramic cylindrical rod as a vibration generator of the stator. The fabricated tiny motor has a di- ameter of 1 mm, a length of 5 mm and a weight of 36 mg. The resonant frequency is 32 kHz with one fixed boundary or 83 kHz with two free boundaries. Four silver electrodes were deposited on the cylindrical surface. The spacing be- tween two adjacent electrodes was carefully designed. A method was developed for the polarization of the stator. By using the bending vibration mode of the stator, the motor shows a performance with a speed of 1800 rpm and a stalled torque of 4 μ Nm, and can bear a high driving voltage up to 350 Vpp. The motor was used to drive an optical prism for circular scanning in an endoscopic OCT system. 11Preconditioned deconvolution method for high-resolution ghost imaging显示文摘Ghost imaging(GI)can nonlocally image objects by exploiting the fluctuation characteristics of light fields,where the spatial resolution is determined by the normalized second-order correlation function g^(2) .However,the spatial shift-invariant property of g^(2) is distorted when the number of samples is limited,which hinders the deconvolution methods from improving the spatial resolution of GI.In this paper,based on prior imaging systems,we propose a preconditioned deconvolution method to improve the imaging resolution of GI by refining the mutual coherence of a sampling matrix in GI.Our theoretical analysis shows that the preconditioned deconvolution method actually extends the deconvolution technique to GI and regresses into the classical deconvolution technique for the conventional imaging system.The imaging resolution of GI after preconditioning is restricted to the detection noise.Both simulation and experimental results show that the spatial resolution of the reconstructed image is obviously enhanced by using the preconditioned deconvolution method.In the experiment,1.4-fold resolution enhancement over Rayleigh criterion is achieved via the preconditioned deconvolution.Our results extend the deconvolution technique that is only applicable to spatial shift-invariant imaging systems to all linear imaging systems,and will promote their applications in biological imaging and remote sensing for high-resolution imaging demands.ZHISHEN TONG ZHENTAO LIU CHENYU HU JIAN WANG SHENSHENG HAN 2021Photonics Research2021,9,6:5
12Identification of Ebola virus microRNAs and their putative pathological function显示文摘Ebola virus(EBOV),a member of the filovirus family,is an enveloped negative-sense RNA virus that causes lethal infections in humans and primates.Recently,more than 1000 people have been killed by the Ebola virus disease in Africa,yet no specific treatment or diagnostic tests for EBOV are available.In this study,we identified two putative viral microRNA precursors(pre-miRNAs)and three putative mature microRNAs(miRNAs)derived from the EBOV genome.The production of the EBOV miRNAs was further validated in HEK293T cells transfected with a pcDNA6.2-GW/EmGFP-EBOV-pre-miRNA plasmid,indicating that EBOV miRNAs can be produced through the cellular miRNA processing machinery.We also predicted the potential target genes of these EBOV miRNAs and their possible biological functions.Overall,this study reports for the first time that EBOV may produce miRNAs,which could serve as non-invasive biomarkers for the diagnosis and prognosis of EBOV infection and as therapeutic targets for Ebola viral infection treatment.LIANG HongWei ZHOU Zhen ZHANG SuYang ZEN Ke CHEN Xi ZHANG ChenYu 2014Science China(Life Sciences)2014,57,10:5
13Global and regional trends in incidence and mortality of female breast cancer and associated factors at national level in 2000 to 2019显示文摘Background: Female breast cancer (FBC) has become the most prevalent malignancy worldwide. We aimed to evaluate the global and regional burden in epidemiological trends and factors associated with the incidence and mortality of FBC.Methods: FBC incidence and mortality in 60 selected countries by cancer registry data integrity in 2020 were estimated from the GLOBOCAN database, and their association with the human development index (HDI) was further evaluated. Trends of age-standardized rates of incidence and mortality in 60 countries from 2000 through 2019 were evaluated by joinpoint regression analysis using data of Global Burden of Disease 2019. The association between potential behavioral, metabolic, and socioeconomic risk factor exposure at the nation level retrieved from the World Bank and Global Health Observatory and the incidence and mortality of FBC were evaluated by multivariate linear regression.Results: FBC incidence and mortality varied greatly in the 60 included countries. Higher incidence and mortality rates were typically observed in countries with higher HDIs and vice versa. During 2000 to 2019, significantly increasing trends in incidence and mortality were observed in 26 (average annual percent changes [AAPCs], 0.35-2.96) and nine countries (AAPC, 0.30-1.65), respectively, while significantly decreasing trends in both incidence and mortality were observed in 22 countries, most of which were high-HDI countries. Among the population aged ≥40 years, there were 26 and 11 countries showing significantly increased trends in incidence and mortality, respectively. Ecological analysis showed that countries with higher prevalence rates of high cholesterol and higher health expenditures were more likely to have higher FBC incidence, and countries with higher rates of obesity and poorer universal health coverage were more likely to have higher FBC mortality.Conclusions: Despite decreased or stabilized FBC incidence and mortality rates were observed in some countries with high HDI over the past decades, disease burden became even severer in developing countries, especially for the population aged ≥40 years. 15Bistratal Au@Bi2S3 nanobones for excellent NIR-triggered/multimodal imaging-guided synergistic therapy for liver cancer显示文摘To fabricate a highly biocompatible nanoplatform enabling synergistic therapy and real-time imaging,novel Au@Bi2S3 core shell nanobones(NBs)(Au@Bi2S3 NBs)with Au nanorods as cores were synthesized.The combination of Au nanorods with Bi2S3 film made the Au@Bi2S3 NBs exhibit ultrahigh photothermal(PT)conversion efficiency,remarkable photoacoustic(PA)imaging and high computed tomography(CT)performance;these Au@Bi2S3 NBs thus are a promising nanotheranostic agent for PT/PA/CT imaging.Subsequently,poly(N-vinylpyrrolidone)-modified Au@Bi2S3 NBs(Au@Bi2S3-PVP NBs)were successfully loaded with the anticancer drug doxorubicin(DOX),and a satisfactory pH sensitive release profile was achieved,thus revealing the great potential of Au@Bi2S3-PVP NBs in chemotherapy as a drug carrier to deliver DOX into cancer cells.Both in vitro and in vivo investigations demonstrated that the Au@Bi2S3-PVP NBs possessed multiple desired features for cancer therapy,including extremely low toxicity,good biocompatibility,high drug loading ability,precise tumor targeting and effective accumulation.Highly efficient ablation of the human liver cancer cell HepG2 was achieved through Au@Bi2S3-PVP NB-mediated photothermal therapy(PTT).As both a contrast enhancement probe and therapeutic agent,Au@Bi2S3-PVP NBs provided outstanding NIR-triggered multi-modal PT/PA/CT imaging-guided PTT and effectively inhibited the growth of HepG2 liver cancer cells via synergistic chemo/PT therapy.Ruizhuo Ouyang Penghui Cao Pengpeng Jia Hui Wang Tianyu Zong Chenyu Dai Jie Yuan Yuhao Li Dong Sun Ning Guo Yuqing Miao Shuang Zhou 2021Bioactive Materials2021,6,2:5

18Squalene epoxidase promotes colorectal cancer cell proliferation through accumulating calcitriol and activating CYP24A1-mediatedMAPK signaling显示文摘Background:Colorectal cancer(CRC)is one of the most malignant tumorswith high incidence,yet its molecular mechanism is not fully understood,hindering the development of targeted therapy.Metabolic abnormalities are a hallmark of cancer.Targeting dysregulated metabolic features has become an important direction for modern anticancer therapy.In this study,we aimed to identify a new metabolic enzyme that promotes proliferation of CRC and to examine the related molecular mechanisms.Methods:We performed RNA sequencing and tissue microarray analyses of human CRC samples to identify new genes involved in CRC.Squalene epoxidase(SQLE)was identified to be highly upregulated in CRC patients.The regulatory function of SQLE in CRC progression and the therapeutic effect of SQLE inhibitors were determined by measuring CRC cell viability,colony and organoid formation,intracellular cholesterol concentration and xenograft tumor growth.Themolecularmechanism of SQLE functionwas explored by combining transcriptome and untargeted metabolomics analysis.Western blotting and realtime PCR were used to assess MAPK signaling activation by SQLE.Results:SQLE-related control of cholesterol biosynthesis was highly upregulated in CRC patients and associated with poor prognosis.SQLE promoted CRC growth in vitro and in vivo.Inhibition of SQLE reduced the levels of calcitriol(active form of vitamin D3)and CYP24A1,followed by an increase in intracellular Ca2+concentration.Subsequently,MAPK signaling was suppressed,resulting in the inhibition of CRC cell growth.Consistently,terbinafine,an SQLE inhibitor,suppressed CRC cell proliferation and organoid and xenograft tumor growth.Conclusions:Our findings demonstrate that SQLE promotes CRC through the accumulation of calcitriol and stimulation of CYP24A1-mediated MAPK signaling,highlighting SQLE as a potential therapeutic target for CRC treatment.Luwei He Huaguang Li Chenyu Pan Yutong Hua Jiayin Peng Zhaocai Zhou Yun Zhao Moubin Lin 2021Cancer Communications2021,41,8:4
19Memristor bridge-based low pass filter for image processing显示文摘This paper highlights the memristor bridge-based lowpass filter (LPF) and improved image processing algorithms along with a novel adaptive Gaussian filter for denoising image and a new Gaussian pyramid for scale invariant feature transform (SIFT). First, a novel kind of LPF based on the memristor bridge is designed, whose cut-off frequency and other traits are demonstrated to change with different time and memristance. In light of the changeable parameter of the memristor bridge-based LPF, a new adaptive Gaussian filter and an improved SIFT algorithm are presented. 20Analysis of Malware Application Based on Massive Network Traffic显示文摘Security and privacy issues are magnified by velocity, volume, and variety of big data. User's privacy is an even more sensitive topic attracting most people's attention. While XcodeGhost, a malware of i OS emerging in late 2015, leads to the privacy-leakage of a large number of users, only a few studies have examined XcodeGhost based on its source code. In this paper we describe observations by monitoring the network activities for more than 2.59 million i Phone users in a provincial area across 232 days. Our analysis reveals a number of interesting points. For example, we propose a decay model for the prevalence rate of Xcode Ghost and we find that the ratio of the infected devices is more than 60%; that a lot of popular applications, such as Wechat, railway 12306, didi taxi, Youku video are also infected; and that the duration as well as the traffic volume of most Xcode Ghost-related HTTP-requests is similar with usual HTTP-request which makes it difficult to be found. Besides, we propose a heuristic model based on fingerprint and its web-knowledge to identify the infected applications.
