2Gas Hydrates in the Qilian Mountain Permafrost, Qinghai, Northwest China显示文摘Qilian Mountain permafrost, with area about 10×104 km2, locates in the north of Qinghai-Tibet plateau. It equips with perfect conditions and has great prospecting potential for gas hydrate. The Scientific Drilling Project of Gas Hydrate in Qilian Mountain permafrost, which locates in Juhugeng of Muri Coalfield, Tianjun County, Qinghai Province, has been implemented by China Geological Survey in 2008–2009. Four scientific drilling wells have been completed with a total footage of 2059.13 m. Samples of gas hydrate are collected separately from holes DK-1, DK-2 and DK-3. Gas hydrate is hosted under permafrost zone in the 133–396 m interval. The sample is white crystal and easily burning. Anomaly low temperature has been identified by the infrared camera. The gas hydrate-bearing cores strongly bubble in the water. Gas-bubble and water-drop are emitted from the hydrate-bearing cores and then characteristic of honeycombed structure is left. The typical spectrum curve of gas hydrate is detected using Raman spectrometry. Furthermore, the logging profile also indicates high electrical resistivity and sonic velocity. 4Genome Editing Enables Next-Generation Hybrid Seed Production Technology显示文摘The next-generation hybrid seed technology enables the successful production of sortable hybrid seeds from genic male sterile(GMS)lines and maintainers;however,it requires multiple laborious and complicated steps.Here,we designed a simple next-generation hybrid seed production strategy that takes advantage of the CRISPR/Cas9 technology to create a Manipulated GMS Maintainer(MGM)system via a single transformation.Under this schema,the maize male fertility gene ZmMS26 was nullified by removal of its fifth exon using the CRISPR/Cas9 system on a vector,and a second vector carrying a functional ZmMS26 cDNA was co-transformed to restore fertility.The second vector also contains a male gametophyte inactivation gene(ZmAA1)encoding maizeα-amylase driven by the pollen-specific promoter PG47 and an endosperm fluorescent marker(DsRED)driven by the barley endosperm aleurone-specific promoter Ltp2.The derived single-copy hemizygous MGM lines bore a mutated MS26 gene,leading to complete male sterility but normal vegetative growth and grain yield.The MGM system could prevent genetic transmission of the MGM elements via male gametophytes,providing an efficient method for sorting maintainer seeds labeled by DsRED.This strategy can be extended to any GMS gene and to hybrid crops other than maize.Xiantao Qi Congsheng Zhang Jinjie Zhu Changlin Liu Changling Huang Xinhai Li Chuanxiao Xie 2020Molecular Plant2020,13,9:18
5Undrained shear strength evaluation for hydrate-bearing sediment overlying strata in the Shenhu area, northern South China Sea显示文摘The undrained shear strength of shallow strata is a critical parameter for safety design in deep-water operations.In situ piezocone penetration tests(CPTU) and laboratory experiments are performed at Site W18-19 in the Shenhu area, northern South China Sea, where China's first marine hydrate exploitation operation is due to be located. The validation of the undrained shear strength prediction model based on CPTU parameters. Different laboratory tests, including pocket penetrometer, torvane, miniature vane and unconsolidated undrained triaxial tests, are employed to solve empirical cone coefficients by statistical and mathematical methods. Finally, an optimized model is proposed to describe the longitudinal distribution of undrained shear strength in calcareous clay strata in the Shenhu area. Research results reveal that average empirical cone coefficients based on total cone resistance, effective resistance, and excess-pore pressure are 13.8, 4.2 and 14.4, respectively. The undrained shear strength prediction model shows a good fit with the laboratory results only within specific intervals based on their compaction degree and gas-bearing conditions. The optimized prediction model in piecewise function format can be used to describe the longitudinal distribution of the undrained shear strength for calcareous clay within all depth intervals from the mud-line to the upper boundary of hydrate-bearing sediments(HBS). The optimized prediction result indicates that the effective cone resistance model is suitable for very soft to firm calcareous clays,the excess-pore pressure model can depict the undrained shear strength for firm to very stiff but gas-free clays,while the total cone resistance model is advantageous for evaluating the undrained shear strength for very stiff and gassy clays. The optimized model in piecewise function format can considerably improve the adaptability of empirical models for calcareous clay in the Shenhu area. 6Synthetical Efficiency-based Optimization for the Power Distribution of Power-split Hybrid Electric Vehicles显示文摘Now the optimization strategies for power distribution are researched widely,and most of them are aiming to the optimal fuel economy and the driving cycle must be preknown.Thus if the actual driving condition deviates from the scheduled driving cycle,the effect of optimal results will be declined greatly.Therefore,the instantaneous optimization strategy carried out on-line is studied in this paper.The power split path and the transmission efficiency are analyzed based on a special power-split scheme and the efficiency models of the power transmitting components are established.The synthetical efficiency optimization model is established for enhancing the transmission efficiency and the fuel economy.The identification of the synthetical efficiency as the optimization objective and the constrain group are discussed emphatically.The optimization is calculated by the adaptive simulated annealing(ASA)algorithm and realized on-line by the radial basis function(RBF)-based similar models.The optimization for power distribution of the hybrid vehicle in an actual driving condition is carried out and the road test results are presented.The test results indicate that the synthetical efficiency optimization method can enhance the transmission efficiency and the fuel economy of the power-split hybrid electric vehicle(HEV)observably.Compared to the rules-based strategy the optimization strategy is optimal and achieves the approximate global optimization solution for the power distribution.The synthetical efficiency optimization solved by ASA algorithm can give attentions to both optimization quality and calculation efficiency,thus it has good application foreground for the power distribution of power-split HEV.WANG Weida HAN Lijin XIANG Changle MA Yue LIU Hui 2014Chinese Journal of Mechanical Engineering2014,27,1:12
7Experimental studies on the P-T stability conditions and influencing factors of gas hydrate in different systems显示文摘The P-T stability conditions of gas hydrate in different systems (i.e., solution, silica sand, and marine sediment) were studied using multi-step decomposition method with our experimental equipment. The effects of different ions with various concentrations and sediment grains on the P-T stability conditions of gas hydrate were investigated. The results show that different ions have different influences on the phase equilibrium of gas hydrate. However, the influence of ions is in a similar trend: the larger the concentration, the bigger the P-T curve shifts to the left. For the silica sand, the influence of pore capillarity of coarse particles (> 460 μm) can be negligible. The P-T curve measured in coarse silica is in agreement with that in pure water. However, the influence of pore capillarity of fine particles (< 35 μm) is significant. The maximum reduction value of temperature is 1.5 K for methane hydrate under stable state. The sediment from the South China Sea significantly affects the P-T stability conditions of methane hydrate, with an average reduction value of 1.9 K within the experimental conditions. This is mainly the result of both the pore water salinity and the pore capillarity of sediment. 10GW5-Like, a homolog of GW5, negatively regulates grain width, weight and salt resistance in rice显示文摘Grain size is an important determinant of yield potential in crops. We previously demonstrated that natural mutations in the regulatory sequences of qSW5/GW5 confer grain width diversity in rice. However, the biological function of a GW5 homolog, named GW5-Like(GW5 L), remains unknown. In this study, we report on GW5 L knockout mutants in Kitaake, a japonica cultivar(cv.)considered to have a weak gw5 variant allele that confers shorter and wider grains. GW5 L is evenly expressed in various tissues, and its protein product is localized to the plasma membrane. Biochemical assays verified that GW5 L functions in a similar fashion to GW5. It positively regulates brassinosteroid(BR) signaling through repression of the phosphorylation activity of GSK2. Genetic data show that GW5 L overexpression in either Kitaake or a GW5 knockout line, Kasaorf3(indica cv. Kasalath background), causes more slender, longer grains relative to the wild-type. We also show that GW5 L could confer salt stress resistance through an association with calmodulin protein OsCa M1-1. 11Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. 12Study of the Surface Morphology of Gas Hydrate显示文摘Study of the surface morphology of gas hydrate is of great importance in understanding its physical properties and occurrence.In order to investigate the surface morphology of different types(sI and sII)and occurrences(pore-filling and fracture-filling)of gas hydrate,both lab-synthesized and drilled-gas hydrate samples were measured using cryo-scanning electron microscopy(cryo-SEM).Results showed that the surface of s I hydrate was relatively smooth,and spongy nano-pores(200–400 nm)gradually occurred at the surface during continuous observation.The surface of sII hydrate was more compact,showing a tier-like structure.Hydrate occurred in quartz sand and usually filled the pores of the sediments and both hydrate and sediments were cemented with each other.SEM observation of the gas hydrates collected from the South China Sea showed that the surface morphology and contact relation with sediments varied with hydrate occurrence.For instance,hydrates dispersed in sediments mainly filled the pores of the sediments.The existence of microorganism shells,such as foraminifera,was beneficial to the formation of gas hydrate.When hydrate occurred as a massive or vein structure,it was easily distinguished from the surrounding sediments.The surface of hydrate with massive or vein structure showed two distinct characters:one was dense and smooth,the other is porous(several to tens of micrometers in diameter).The occurrence of different hydrate morphologies was probably caused by the supplement rates of methane gas.SUN Jianye LI Chengfeng HAO Xiluo LIU Changling CHEN Qiang WANG Daigang 2020Journal of Ocean University of China2020,19,2:9
13Factors affecting genomic selection revealed by empirical evidence in maize显示文摘Genomic selection(GS) as a promising molecular breeding strategy has been widely implemented and evaluated for plant breeding, because it has remarkable superiority in enhancing genetic gain, reducing breeding time and expenditure, and accelerating the breeding process. In this study the factors affecting prediction accuracy(rMG) in GS were evaluated systematically, using six agronomic traits(plant height, ear height, ear length, ear diameter,grain yield per plant and hundred-kernel weight) evaluated in one natural and two biparental populations. The factors examined included marker density, population size, heritability,statistical model, population relationships and the ratio of population size between the training and testing sets, the last being revealed by resampling individuals in different proportions from a population. Prediction accuracy continuously increased as marker density and population size increased and was positively correlated with heritability; rMGshowed a slight gain when the training set increased to three times as large as the testing set. Low predictive performance between unrelated populations could be attributed to different allele frequencies, and predictive ability and prediction accuracy could be improved by including more related lines in the training population. Among the seven statistical models examined, including ridge regression best linear unbiased prediction(RR-BLUP), genomic BLUP(GBLUP), Bayes A, Bayes B, Bayes C, Bayesian least absolute shrinkage and selection operator(Bayesian LASSO), and reproducing kernel Hilbert space(RKHS), the RKHS and additive-dominance model(Add + Dom model) showed credible ability for capturing non-additive effects, particularly for complex traits with low heritability. 14QTL analysis of ear leaf traits in maize(Zea mays L.)under different planting densities显示文摘Modem maize varieties have become more productive than ever, owing largely to increased tolerance of high plant density. However, the genetics of ear leaf traits under different densities remains poorly understood. In this study, Zhongdan 909 recombinant inbred lines(RILs) derived from a cross between Z58 and HD568 were genotyped for 3072 single-nucleotide polymorphisms(SNPs), and phenotyped for leaf length(LL), leaf width(LW), and leaf angle(LA) of the uppermost ear leaf under three planting densities(52,500,67,500, and 82,500 plants ha-1, respectively). A genetic map was then constructed using1358 high-quality SNPs. The total length of the linkage map was 1985.2 cM and the average interval between adjacent markers 1.46 cM. With increasing density, LL and LW decreased from 63.68 to 63.02 cm and from 8.56 to 8.21 cm, respectively, while LA increased from19.42° to 19.66°. All three traits had high heritabilities, of 0.75, 0.78, and 0.84, respectively.Using inclusive composite interval mapping, 23, 25, and 17 quantitative trait loci(QTL) were detected for LL, LW, and LA, respectively. Of these, 35 were simultaneously detected under two or three plant densities, while 30 were detected under only one. 15Influence of foraminifera on formation and occurrence characteristics of natural gas hydrates in fine-grained sediments from Shenhu area, South China Sea显示文摘Marine gas hydrates accumulate primarily in coarse-grained, high-permeability layers; however, highly saturated natural gas hydrates have been discovered in the fine-grained sediments of Shenhu area, South China Sea(SCS). This may be explained by key factors, such as the great abundance of foraminifera shells. In this paper, by analyzing the SCS foraminifera structure and performing hydrate formation experiments in the foraminifera shells, the contribution of foraminifera to hydrate accumulation in the SCS was investigated from a microscopic point of view. Simulations of hydrate formation were carried out in both pure SCS foraminifera shells and the host sediments. Pore structures in typical foraminifera were studied by use of micro-focus X-ray computed tomography(CT) and scanning electron microscopy(SEM). Hydrate growth and occurrence characteristics in the foraminifera shells were observed in-situ. The results showed that the presence of foraminifera significantly enhanced the effective porosity of the SCS sediments. Moreover, while the hydrates grew preferentially in the chambers of the coarse-grained foraminifera by adhering to the inner walls of the foraminifera shells, no apparent hydrate accumulation was observed in the fine-grained or argillaceous matrix. 16Synchrotron radiation X-ray powder diffraction techniques applied in hydrogen storage materials--A review显示文摘Synchrotron radiation is an advanced collimated light source with high intensity. It has particular advantages in structural characterization of materials on the atomic or molecular scale. Synchrotron radiation X-ray powder diffraction(SR-XRPD) has been successfully exploited to various areas of hydrogen storage materials. In the paper, we will give a brief introduction on hydrogen storage materials, X-ray powder diffraction(XRPD), and synchrotron radiation light source. The applications of ex situ and in situ time-resolved SR-XRPD in hydrogen storage materials, are reviewed in detail. 17Conversion of a normal maize hybrid into a waxy version using in vivo CRISPR/Cas9 targeted mutation activity显示文摘Waxy maize is a specialty maize that produces mainly amylopectin starch with special food or industrial values. The objective of this study was to overcome the limitations of wx mutant allele acquisition and breeding efficiency by conversion of parental lines from normal to waxy maize. The intended mutation activity was achieved by in vivo CRISPR/Cas9 machinery involving desired-target mutation of the Wx locus in the ZC01 background,abbreviated as ZC01-DTM^(wx). Triple selection was applied to segregants to obtain high genome background recovery with transgene-free wx mutations. The targeted mutation was identified, yielding six types of mutations among progeny crossed with ZC01-DTM^(wx).The amylopectin contents of the endosperm starch in mutant lines and hybrids averaged94.9%, while those of the wild-type controls were significantly(P < 0.01) lower, with an average of 76.9%. Double selection in transgene-free lines was applied using the Bar strip test and Cas9 PCR screening. The genome background recovery ratios of the lines were determined using genome-wide SNP data. That of lines used as male parents was as high as98.19% and that of lines used as female parents was as high as 86.78%. Conversion hybrids and both parental lines showed agronomic performance similar to that of their wild-type counterparts. 18Investigation of Transformation for Ultrahigh Strength Steel Aermet 100显示文摘The heat-stagnation curves of two quenched and cryogenically treated ultrahigh strength AF1410 and Aermet 100 steels during heating and cooling in furnace have been measured. The results showed that the curve displayed an abnormal terrace in the course of heating, and within the temperature range corresponding to the terrace, the formation and growth of both cementite Fe3C and secondary hardening phase M2C in the steel make the carbon content in matrix ob- viously vary. It is in the region of terrace that the optimum combination of tensile strength and fracture toughness is obtained because of secondary hardening. Based on the measured heat-stagnation curve, several transformation points have been determined and the correctness was also verified by X-ray diffraction (XRD) and transmission electron microscopy (TEM). 19Enhancing Genetic Gain through Genomic Selection: From Livestock to Plants显示文摘Although long-term genetic gain has been achieved through increasing use of modern breeding methods and technologies,the rate of genetic gain needs to be accelerated to meet humanity’s demand for agricultural products.In this regard,genomic selection(GS)has been considered most promising for genetic improvement of the complex traits controlled by many genes each with minor effects.Livestock scientists pioneered GS application largely due to livestock’s significantly higher individual values and the greater reduction in generation interval that can be achieved in GS.Large-scale application of GS in plants can be achieved by refining field management to improve heritability estimation and prediction accuracy and developing optimum GS models with the consideration of genotype-by-environment interaction and non-additive effects,along with significant cost reduction.Moreover,it would be more effective to integrate GS with other breeding tools and platforms for accelerating the breeding process and thereby further enhancing genetic gain.In addition,establishing an open-source breeding network and developing transdisciplinary approaches would be essential in enhancing breeding efficiency for small-and medium-sized enterprises and agricultural research systems in developing countries.New strategies centered on GS for enhancing genetic gain need to be developed.Yunbi Xu Xiaogang Liu Junjie Fu Hongwu Wang Jiankang Wang Changling Huang Boddupalli MPrasanna Michael SOlsen Guoying Wang Aimin Zhang 2020Plant Communications2020,1,1:4
20Creation of aromatic maize by CRISPR/Cas显示文摘Aroma is an important quality parameter for breeding in rice(Oryza sativa).For example,the aromatic rice varieties basmati and jasmine rice,with a popcorn-like scent,are popular worldwide and routinely command a price premium.2-acetyl-1-pyrroline(2AP)is a key flavor compound among over 200 volatiles identified in fragrant rice.A naturally fragrant germplasm exists in multiple plant species besides rice,which all exhibit lower activity of BETAINE ALDEHYDE DEHYDROGENASE 2(BADH2).However,no equivalent aromatic germplasm has been described in maize(Zea mays).Here,we characterized the two maize BADH2 homologs,ZmBADH2a and ZmBADH2b.We generated zmbadh2a and zmbadh2b single mutants and the zmbadh2a-zmbadh2b double mutant by CRISPR/Cas in four inbred lines.A popcorn-like scent was only noticeable in seeds from the double mutant,but not from either single mutant or in wild type.In agreement,we only detected 2AP in fresh kernels and dried mature seeds from the double mutant,which accumulated between 0.028 and 0.723 mg/kg 2AP.These results suggest that ZmBADH2a and ZmBADH2b redundantly participate in 2AP biosynthesis in maize,and represent the creation of the world's first aromatic maize by simultaneous genome editing of the two BADH2 genes.Yanxiao Wang Xiaoqin Liu Xiuxiu Zheng Wenxia Wang Xunqing Yin Haifeng Liu Changle Ma Xiaomu Niu Jian-Kang Zhu Fei Wang 2021Journal of Integrative Plant Biology2021,63,9:3
