| 1 | Experimental and numerical study of the fragmentation of expanding warhead casings by using different numerical codes and solution techniques显示文摘There has been increasing interest in numerical simulations of fragmentation of expanding warheads in 3D.Accordingly there is a pressure on developers of leading commercial codes,such as LS-DYNA.AUTODYN and IMPETUS Afea.to implement the reliable fracture models and the efficient solution techniques.The applicability of the Johnson—Cook strength and fracture model is evaluated by comparing the fracture behaviour of an expanding steel casing of a warhead with experiments.The numerical codes and different numerical solution techniques,such as Eulerian,Lagrangian.Smooth particle hydrodynamics(SPH).and the corpuscular models recently implemented in IMPETUS Afea are compared.For the same solution techniques and material models we find that the codes give similar results.The SPH technique and the corpuscular technique are superior to the Eulerian technique and the Lagrangian technique(with erosion) when it is applied to materials that have fluid like behaviour such as the explosive and the tracer.The Eulerian technique gives much larger calculation time and both the Lagrangian and Eulerian techniques seem to give less agreement with our measurements.To more correctly simulate the fracture behaviours of the expanding steel casing,we applied that ductility decreases with strain rate.The phenomena may be explained by the realization of adiabatic shear bands.An implemented node splitting algorithm in IMPETUS Afea seems very promising. | John F.MOXNES Anne K.PRYTZ Фyvind FRФYLAND Siri KLOKKEHAUG Stian SKRIUDALEN Eva FRIIS Jan A.TELAND Cato DФRUM Gard ФDEGARDSTUEN | 2014 | Defence Technology(防务技术)2014,10,2: | 12 |

| 4 | Heparin resistance in severe thermal injury:a prospective cohort study显示文摘Background:Low molecular-weight heparin(LMWH)is routinely administered to burn patients for thromboprophylaxis.Some studies have reported heparin resistance,yet the mechanism(s)and prevalence have not been systematically studied.We hypothesized that nucleosomes,composed of histone structures with associated DNA released from injured tissue and activated immune cells in the form of neutrophil extracellular traps(NETs or NETosis),neutralize LMWH resulting in suboptimal anticoagulation,assessed by reduction in anti-factor Xa activity.Methods:Blood was sampled from>15%total body surface area(TBSA)burn patients receiving LMWH on days 5,10 and 14.Peak anti-factor Xa(AFXa)activity,anti-thrombin(ATIII)activity,cellfree DNA(cfDNA)levels and nucleosome levels were measured.Mixed effects regression was adjusted for multiple confounders,including injury severity and ATIII activity,and was used to test the association between nucleosomes and AFXa.Results:A total of 30 patients with severe burns were included.Mean TBSA 43%(SD 17).Twentythree(77%)patients were affected by heparin resistance(defined by AFXa activity<0.2 IU/mL).Mean peak AFXa activity across samples was 0.18 IU/mL(SD 0.11).Mean ATIII was 81.9%activity(SD 20.4).Samples taken at higher LWMH doses were found to have significantly increased AFXa activity,though the effect was not observed at all doses,at 8000 IU no samples were heparin resistant.Nucleosome levels were negatively correlated with AFXa(r=−0.29,p=0.050)consistent with the hypothesis.The final model,with peak AFXa as the response variable,was adjusted for nucleosome levels(p=0.0453),ATIII activity(p=0.0053),LMWH dose pre-sample(p=0.0049),drug given(enoxaparin or tinzaparin)(p=0.03),and other confounders including severity of injury,age,gender,time point of sample.Conclusions:Heparin resistance is a prevalent issue in severe burns.Nucleosome levels were increased post-burn,and showed an inverse association with AFXa consistent with the hypothesis that they may interfere with the anticoagulant effect of heparin in vivo and contribute to heparin resistance.Accurate monitoring of AFXa activity with appropriate therapy escalation plans are recommended with dose adjustment following severe burn injury. | Liam D Cato Benjamin Bailiff Joshua Price Christos Ermogeneous Jon Hazeldine William Lester Gillian Lowe Christopher Wearn Jonathan RB Bishop Janet M Lord Naiem Moiemen Paul Harrison | 2021 | Burns & Trauma2021,9,1: | 3 |
| 5 | Nanofiber–microsphere (nano-micro) matrices for bone regenerative engineering: a convergence approach toward matrix design显示文摘Bone is an essential organ for health and quality of life.Due to current shortfalls in therapy for bone tissue engineering,scientists have sought the application of synthetic materials as bone graft substitutes.As a composite organic/inorganic material with significant extra cellular matrix(ECM),one way to improve bone graft substitutes may be to engineer a synthetic matrix that is influenced by the physical appearance of natural ECM networks.In this work,the authors evaluate composite,hybrid scaffolds for bone tissue engineering based on composite ceramic/polymer microsphere scaffolds with synthetic ECM-mimetic networks in their pore spaces.Using thermally induced phase separation,nanoscale fibers were deposited in the pore spaces of structurally sound microsphere-based scaffold with a density proportionate to the initial polymer concentration.Porosimetry and mechanical testing indicated no significant changes in overall pore characteristics or mechanical integrity as a result of the fiber deposition process.These scaffolds displayed adequate mechanical integrity on the scale of human trabecular bone and supported the adhesion and proliferation of cultured mouse calvarial osteoblasts.Drawing from natural cues,these scaffolds may represent a new avenue forward for advanced bone tissue engineering scaffolds. | Clarke Nelson Yusuf Khan Cato T.Laurencin | 2014 | Regenerative Biomaterials2014,1,1: | 2 |

| 14 | 基于G-蛋白偶联受体激酶2抑制剂治疗心力衰竭的合理药物设计(英文)显示文摘G protein-coupled receptors(GPCRs)convert extracellular stimuli in the form of hormones,odorants and light into profound changes in cell homeostasis.Their timely desensitization is critical for cells to rapidly respond to changes in their environment and to avoid damage from sustained signaling.Seven GPCR kinases(GRKs)phosphorylate and regulate the activity of most of the^800 GPCRs in the human genome.Although GRKs normally play an adaptive role,in conditions such as chronic heart failure they are overexpressed and linked to disease progression.GRK2 and GRK5 have thus become important targets for the treatment of heart failure and pathological cardiac hypertrophy,respectively.Our lab has determined atomic structures representing all three vertebrate GRK subfamilies,and is now in the midst of a campaign to develop selective inhibitors of these enzymes using structure-based rational design.We have identified the FDA approved drug paroxetine as a selective GRK2 inhibitor,determined the crystal structure of the GRK2·paroxetine complex and,in collaboration with the Koch lab,showed that the drug improves contractility in myocytes and,most impressively,recovery in postmyocardial infarcted mice.Since then,we have identified additional chemical scaffolds that exhibit even higher potency and/or selectivity for GRK5.Using a'hybrid'inhibitor design approach we have generated GRK selective chemical probes that exhibit improved potency and stability and are able to increase inotropy and dampen the hypertrophic response in cardiomyocytes and small animal models.Structural analysis has revealed the molecular basis for selectivity and potency in many of these compounds,allowing for the design of future generations of GRK chemical probes. | John J TESMER Helen V WALDSCHMIDT Marie C CATO Renee BOULEY Osvaldo CRUZ-RODRIGUEZ Scott D LARSEN | 2017 | 中国药理学与毒理学杂志2017,31,10: | 1 |
