| 2 | Bidirectional propagation of lightning leader显示文摘The characteristics of initial leader of triggered lightning flashes are studied by use of a highspeed digital camera system with the rate of 1000 frame per second and simultaneous measurements of current and electric field change with 0.1 μs time resolution. The results show that the altitude-triggered lightning was initiated by bidirectional propagation leader because of 'current interruption' by air gap,whereas the grounded-triggered one was not. The onset of downward-moving positive leader was prior to that of the upward-going negative leader for 843 ms in an altitude-triggered lightning under the positive ambient electric field. | 3 | LncRNA TUG1 promotes cisplatin resistance in esophageal squamous cell carcinoma cells by regulating Nrf2显示文摘Esophageal squamous cell carcinoma(ESCC)is a common malignancy with poor prognosis.The drug resistance compromises the efficacy of chemotherapy for ESCC.Long non-coding RNA taurine upregulated gene 1(TUG1)has been identified as a promoter of cancer progression and chemotherapy resistance in many malignancies.However,the exact role of TUG1 in ESCC chemotherapy resistance remains unclear.In this study,we showed that TUG1 expression in TE-1-derived cisplatin(DDP)-resistant(TE-1/DDP)cells was higher than that in TE-1 cells.Furthermore,TUG1 promoted DDP resistance in TE-1 and TE-1/DDP cells by promoting cell proliferation,suppressing cell apoptosis,and elevating protein expression of the classical multi-drug resistance-related P-gp.In contrast,TUG1 knockdown exerted an opposite effect.Mechanistically,RNA pull-down and RNA immunoprecipitation assays confirmed that TUG1 directly bound to nuclear factor(erythroid-derived 2)-like 2(Nrf2)protein and elevated Nrf2 protein expression.Moreover,Nrf2-neutralizing antibody effectively reversed the TUG1 overexpression-mediated promotion of ESCC cell resistance to DDP.In conclusion,our findings demonstrated that TUG1 promoted ESCC cell resistance to DDP,at least in part,through upregulating Nrf2. | Zhenghua Zhang Ran Xiong Caiwei Li Meiqing Xu Mingfa Guo | 2019 | Acta Biochimica et Biophysica Sinica2019,51,8: | 7 |

| 6 | Preparation of an anti-diethylstilbestrol monoclonal antibody and development of an indirect competitive ELISA to detect diethylstilbestrol in biological samples显示文摘Based on the preparation of an anti-diethylstilbestrol(DES) monoclonal antibody,a simple and convenient indirect competitive enzyme-linked immunosorbent assay(ELISA) method for DES detection has been developed.The monoclonal antibody demonstrated high sensitivity to DES with an IC50 value of 275 pg mL-1 and detection limit(LOD) of 90 pg mL-1.The specificity of the assay was studied by measuring cross-reactivity of the antibody with structurally related compounds of ethinyl estradiol(<7%),estrone(<0.1%),estriol(<0.1%),and diethylstilbestrol benzoate(<0.1%).Chicken,fish,shrimp,urine and bile spiked with different concentration of DES were detected by the developed method,and the recovery rates were greater than 79.5%.Intra-and inter-assay variations were about 6%.This method exhibited high stability with a coefficient of variation less than 10% in buffer and in real samples.The LODs in fish/shrimp,liver,feed and urine spiked with DES were 600,600,4800 and 600 pg mL-1,respectively.These results confirmed that the antibody to DES was successfully produced and could be used to establish ELISA methods for DES detection in food producing animals. | LI WeiHua MENG Meng HE FangYang WAN YuPing XUE HuYin LIU Wei YIN WeiWei XU Jing FENG CaiWei WANG ShanLiang LU Xiao LIU JinTing XI RiMo | 2011 | Chinese Science Bulletin2011,56,8: | 4 |
| 7 | Is longer sevoflurane preconditioning neuroprotective in permanent focal cerebral ischemia?显示文摘Sevoflurane preconditioning has neuroprotective effects in the cerebral ischemia/reperfusion model. However, its influence on permanent cerebral ischemia remains unclear. In the present study, the rats were exposed to sevoflurane for 15, 30, 60, and 120 minutes, followed by induction of perma-nent cerebral ischemia. Results demonstrated that 30-and 60-minute sevoflurane preconditioning significantly reduced the infarct volume at 24 hours after cerebral ischemia, and 60-minute se-voflurane preconditioning additionally reduced the number of TUNEL-and caspase-3-positive cells in the ischemic penumbra. However, 120-minute sevoflurane preconditioning did not show evident neuroprotective effects. Moreover, 60-minute sevoflurane preconditioning significantly attenuated neurological deficits and infarct volume in rats at 4 days after cerebral ischemia. | 10 | RelB intrinsically regulates the development and function of medullary thymic epithelial cells显示文摘Medullary thymic epithelial cells(mTECs) act as one of the major stromal components in the thymus for selection and maturation of both conventional T cells and non-conventional T cells. Extensive efforts have been spent to understand how mTEC development and function are regulated. Although RelB has been well accepted to be a critical transcriptional factor for mTEC development, the underlying mechanisms still remain largely unclear. In this study, by generating thymic epithelial cell specific RelB deficient mice, we found that epithelial intrinsic RelB is required for mTEC homeostatic proliferation. Mechanistically, RelB regulates the expression of genes involved in cell cycle. Functionally, lack of intrinsic RelB in thymic epithelial cells results in dramatically reduced population of mTECs and impaired development of thymic invariant natural killer T(iNKT) cells and intraepithelial lymphocyte precursors(IELPs). | 18 | Fluid Charging and Paleo-pressure Evolution in the Ledong Slope Zone of the Yinggehai Basin,South China Sea显示文摘Large numbers of gas reservoirs have been discovered in overpressure basins.Fluid charging has a close relationship with paleo-pressure evolution,affecting the migration of gas reservoirs.To study fluid charging and the related pressure system,we analyzed burial histories and fluid inclusion(PVTx)simulations and conducted basin modeling of the Ledong Slope Zone in the Yinggehai Basin as an example.On the basis of fluid-inclusion assemblages(FIAs),homogenization temperature(Th),final melting temperature(Tm,ice)and Raman spectroscopy in fluid inclusions,there are three stages of fluid charging:during the first and second stage,methane-dominated fluid was charged at 2.2–1.7 Ma and 1.7–0.9 Ma,respectively.In the third stage,CO_(2)-rich hydrothermal fluid was charged since 0.9 Ma.It could be concluded from the well-logging data that the disequilibrium compaction in the Yinggehai Fm.,along with the fluid expansion and clay diagenesis in the Huangliu and Meishan formations,resulted in the overpressure in the Ledong slope zone.The evolution of paleo-pressure was affected by the sedimentation rate of the Yinggehai Fm.,as well as the hydrocarbon generation rate.Additionally,the Ledong Slope Zone is less affected by diapir activity than the nearby diapir area.Based on fluid inclusions,paleo-pressure,basin modeling and geological background,the gas migration history of the Ledong Slope Zone can be divided into four stages:in the first stage,excess pressure was formed around 5 Ma;from 2.2 to 1.7 Ma,there was a reduction in the charging of hydrocarbon fluid and steadily increasing excess pressure;during the 1.7–0.9 Ma period a large amount of hydrocarbon was generated,excess pressure increasing significantly and hydraulic fractures forming in mudstones,With gas reservoirs developing in structural highs;since 0.9 Ma,CO_(2)-rich hydrothermal fluid accumulated in reservoirs adjacent to faults and the pressure coefficient remained stable.The research results are helpful in the study of fluid migration and accumulation mechanisms in overpressure basins. | ZHAO Jing HUANG Zhilong MA Jian WANG Rui YANG Yizhuo FAN Caiwei ZHOU Ying XU Maguang | 2023 | Acta Geologica Sinica(English Edition)2023,97,3: | 0 |
| 19 | A novel Ti cored wire developed for wire-feed arc deposition of TiB/Ti composite coating显示文摘A novel Ti cored wire containing Ti B2,Al60 V40 and Ti6 Al4 V mixed powders was developed for wirefeed arc deposition of Ti B/Ti composite coating,to enhance the hardness and wear resistance of Ti alloy.Results showed that after experiencing several chemical reactions,the wire was melted in the arc zone and turned into nonuniform droplets composed of Ti-Al-V-B melt and undecomposed Ti B2 particles.With the increase of welding current,the detachment time of droplet shortened while the transfer frequency accelerated,accompanied by the improvement in coating surface quality.The spatial distribution of Ti B whiskers in coating was governed by welding current.A uniform distribution could be achieved as welding current was sufficient at the expense of elevated dilution ratio,while increasing wire feeding speed could compensate the dilution loss of Ti B whisker to some extent.The decomposition process of Ti B_(2)particles and the microstructure evolution mechanism of coating was discussed in detail.The optimum coating possessed uniform microstructure,relatively low dilution ratio,and high hardness(639.1 HV_(0.5))as compared with Ti6 Al4 V substrate(326 HV_(0.5)).Indentation morphology analysis verified the excellent performance was ascribed to the load-sharing strengthening of Ti B whiskers.This study provides a high-efficiency fabrication method for the ever-developing titanium matrix composites(TMCs)coating. | Yang Bao Lujun Huang Shan Jiang Rui Zhang Qi An Caiwei Zhang Lin Geng Xinxin Ma | 2021 | Journal of Materials Science & Technology2021,,24: | 0 |
| 20 | Characterization of SARS-COV-2 main protease inhibitory peptides from Ulva prolifera proteins显示文摘The main protease(M^(pro))is essential for the replication of SARS-COV-2 and therefore represents a promising anti-viral target.In this study,we screened M^(pro)inhibitory peptides from Ulva prolifera protein on in-silico proteolysis.Cytotoxicity analysis using the online toxic prediction tool ToxinPred revealed that all the peptides were non-cytotoxic.The hexapeptide(SSGFID)exhibited high M^(pro)inhibitory activity in molecular docking and its IC_(50)value was 139.40±0.82μmol/L in vitro according to fluorescence resonance energy transfer assay(FRET).Quantitative real-time(qRT-)PCR results show that SSGFID could stimulate the expression of mitosis-related factors,including nuclear factor-κB,cyclin D1,and cyclin-dependent kinase 4,to promote the proliferation of mice splenocytes.Stability study revealed that SSGFID showed resistance against pepsin and trypsin but lost D(Asp)after pretreatment at121℃ for 15 min.Besides,SSGFID was mainly transported through the Caco-2 cell monolayer by the peptide transporter PepT1 and passive-mediated transport during the transport study.Unfortunately,the peptide was also degraded by Caco-2 intracellular enzymes,and the transfer rate of intact peptide was4.2%.Furthermore,Lineweaver–Burk plots demonstrated that SSGFID possessed a mixed inhibitory characteristic with M^(pro).Our study indicated the potential of Ulva prolifera as antiviral and immuneenhancing functional food ingredients and nutraceuticals. | Zhiyong LI Yehua WANG Caiwei FU Dongren ZHANG Tuanjie CHE Songdong SHEN | 2023 | Journal of Oceanology and Limnology2023,41,5: | 0 |
