| 2 | Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. | 6 | Study on the Estimation of Groundwater Withdrawals Based on Groundwater Flow Modeling and Its Application in the North China Plain显示文摘The amount of water withdrawn by wells is one of the quantitative variables that can be applied to estimate groundwater resources and further evaluate the human influence on groundwater systems. The accuracy for the calculation of the amount of water withdrawal significantly influences the regional groundwater resource evaluation and management. However, the decentralized groundwater pumping, inefficient management, measurement errors and uncertainties have resulted in considerable errors in the groundwater withdrawal estimation. In this study, to improve the estimation of the groundwater withdrawal, an innovative approach was proposed using an inversion method based on a regional groundwater flow numerical model, and this method was then applied in the North China Plain. The principle of the method was matching the simulated water levels with the observation ones by adjusting the amount of groundwater withdrawal. In addition, uncertainty analysis of hydraulic conductivity and specific yield for the estimation of the groundwater withdrawal was conducted. By using the proposed inversion method, the estimated annual average groundwater withdrawal was approximately 24.92×109 m3 in the North China Plain from 2002 to 2008. The inversion method also significantly improved the simulation results for both hydrograph and the flow field. | 7 | Preparation and characterization of Fe_(3)O_(4)/Au composite particles显示文摘Using Fe3O4 nano-particles as seeds, a new type of Fe3O4/Au composite particles with core/shell structure and diameter of about 170 nm was prepared by reduction of Au3+ with hydroxylamine in an aqueous solution. Particle size analyzer and transmission electron micro-scope were used to analyze the size distribution and microstructure of the particles in different conditions. The result showed that the magnetically responsive property and suspension stability of Fe3O4 seeds as well as reduction conditions of Au3+ to Au0 are the main factors which are crucial for obtaining a colloid of the Fe3O4/Au composite particles with uniform particle dispersion, excellent stability, homogeneity in particle sizes, and effective response to an external magnet in aqueous suspension solutions. UV-Vis analysis revealed that there is a characteristic peak of Fe3O4/Au fluid. | 8 | Applicability of Artificial Recharge of Groundwater in the Yongding River Alluvial Fan in Beijing through Numerical Simulation显示文摘A groundwater transient flow model was developed to evaluate the applicability and effectiveness of artificial recharge scenarios in the middle-upper part of the Yongding River alluvial fan in Beijing. These scenarios were designed by taking into account different types of recharge facilities and their infiltration rate with the Middle Route Project for South-to-North Water Transfer(MRP) as the recharge water source. The simulation results suggest that:(1) the maximum amount of artificial recharge water, for scenario I, would be 127.42×106 m3 with surface infiltration facilities; and would be243.48×106 m3 for scenario II with surface infiltration and recharge wells under the constraint of the upper limit of groundwater;(2) with preferred pattern of recharge facilities, groundwater levels in both optimized recharge scenarios would not exceed the upper limit within the given recharge period; and(3implementation of the recharge scenarios would efficiently increase the aquifer replenishment and the groundwater budget will change from-54.11×106 to 70.89×104 and 183.36×104 m3, respectively. In addition, under these two scenarios groundwater level would rise up to 30 and 34 m, respectively, without increasing the amount of evaporation. | 9 | Microbial population dynamics and changes in main nutrients during the acidification process of pig manures显示文摘This study evaluated the impact of pig manure acidification on anaerobic treatment and composition of the fecal microbial community.According to the different chemical oxygen demand (COD) in the anaerobic treatment processes,pig manure was diluted 4 times (×4),16 times (×16),or 64 times (×64) and subjected to acidification.During the acidification process,pH,soluble chemical oxygen demand (SCOD),volatile fatty acids (VFAs),nitrogen (N),phosphorus (P) and potassium (K) were determined along with microbial population dynamics.The pH of the three dilutions first declined,and then slowly increased.The total VFAs of ×4 and ×16 dilutions peaked on day 15 and 20,respectively.The content of acetic acid,propanoic acid,butanoic acid and valeric acid of the × 4 dilution were 23.6,11.4,8.8 and 0.6 g/L respectively,and that of the ×16 dilution was 5.6,2.3,0.9 and 0.2 g/L respectively.Only acetic acid was detected in the ×64 dilution,and its level peaked on day 10.The results showed that the liquid pig manure was more suitable to enter the anaerobic methanogenic bioreactors after two weeks of acidification.During the acidification process,total P concentration increased during the first ten days,then dropped sharply,and rose again to a relatively high final concentration,while total N concentration rose initially and then declined.Based on the analysis of denaturing gradient gel electrophoresis (DGGE) and 16S rRNA gene clone library,we concluded that the acidification process reduced the number of pathogenic bacteria species in pig manure. | Dongdong Zhang Xufeng Yuan Peng Guo Yali Suo Xiaofen Wang Weidong Wang Zongjun Cui | 2011 | Journal of Environmental Sciences2011,23,3: | 4 |

| 11 | Microstructures and Tensile Properties of Hot-extruded Mg-6Zn-xCe(x=0,0.6,1.0,2.0) Alloys显示文摘Mg-6Zn-x Ce(x = 0, 0.6, 1.0, 2.0) alloy ingots with diameter of 50 mm were extruded into bars with diameter of 12 mm at 300 ℃. The microstructures were analyzed by X-ray diffraction, optical microscopy, scanning electron microscopy and transmission electron microscopy, and mechanical properties were tested at room temperature. The results showed that major intermetallic composition in as-cast Mg-6Zn and Mg-6Zn-0.6Ce alloys was Mg_4Zn_7 phase, during extrusion Mg_4Zn_7 phase was dissolved into matrix and then precipitated as MgZn_2. In as-cast and as-extruded Mg-6Zn-1Ce and Mg-6Zn-2Ce alloys the major intermetallic composition was T phase. The microstructure of as-extruded alloy was refined due to complete dynamic recrystallization, the average grain size decreased with increasing Ce content, which were 12.1, 11.7, 11.0 and 10.0 mm, respectively. High density MgZn_2 precipitated in Mg-6Zn and Mg-6Zn-0.6Ce alloys. The broken T phase particles were distributed linearly along extrusion direction. Mg-6Zn-0.6Ce alloy exhibited a high yield strength of 226.3 MPa that was about 24 MPa higher than Mg-6Zn alloy. | 12 | Dark current mechanism of unpassivated mid wavelength type II InAs/GaSb superlattice infrared photodetector显示文摘We investigate the dark current mechanism for an unpassivated mid wavelength(MW) type II InAs/GaSb superlattice infrared photodetector by doing the variablearea diode tests. The bulk resistance-area product and the resistivity due to the surface current are determined to be17.72 X cm2 and 704.23 X cm at 77 K, respectively. It is found that for all the mesa sizes used, the dark current is dominated or predominated by the surface component, and with scaling back the mesa size, the surface current increases while the bulk component decreases. The activation energy is determined to be 145 meV for the temperature range around 140–280 K, while it is 6 meV when temperature is below 100 K. | 13 | Development and Application of a Regional Land Subsidence Model for the Plain of Tianjin显示文摘In this paper, a coupled numerical groundwater and land subsidence model was developed for the Tianjin Plain. The model was employed to investigate the groundwater resources and their changes over the last decade, and to further predict the changing patterns of the groundwater level and associated land subsidence in future. First, according to the regional hydrogeology, the simulation area was defined with an area of 10.6×103 km2, which was divided into six aquifer units. A coupled groundwater and land subsidence numerical model was built by using Modflow2005 and the land subsidence simulation module SUB(subsidence and aquifer-system compaction), in which the groundwater flow was modeled as three-dimensional unsteady flow and the land subsidence simulation was based on one-dimensional consolidation theory. The model was then calibrated by using the groundwater level contour lines, hydrographs, and land subsidence hydrographs over the period of 1998–2008. In addition, groundwater balance analysis of the simulation period indicated that under multi-year groundwater withdrawal condition the cross-flow recharge, compression release, and lateral boundary inflow contributed 44.43%, 32.14%, and 21.88% to the deep aquifer recharge, respectively. Finally, the model was applied to predict the changing patterns of the groundwater levels and the associated variations in land subsidence under the control of groundwater exploitation after implementation of the south-to-north water diversion project. | 14 | Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors. | Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang | 2022 | Cell Research2022,32,11: | 2 |
