1Comparative genomic and transcriptomic analyses uncover the molecular basis of high nitrogen-use efficiency in the wheat cultivar Kenong 9204显示文摘Studying the regulatory mechanisms that drive nitrogen-use efficiency(NUE)in crops is important for sustainable agriculture and environmental protection.In this study,we generated a high-quality genome assembly for the high-NUE wheat cultivar Kenong 9204 and systematically analyzed genes related to nitrogen uptake and metabolism.By comparative analyses,we found that the high-affinity nitrate transporter gene family had expanded in Triticeae.Further studies showed that subsequent functional differentiation endowed the expanded family members with saline inducibility,providing a genetic basis for improving the adaptability of wheat to nitrogen deficiency in various habitats.To explore the genetic and molecular mechanisms of high NUE,we compared genomic and transcriptomic data from the high-NUE cultivar Kenong 9204(KN9204)and the low-NUE cultivar Jing 411 and quantified their nitrogen accumulation under high-and low-nitrogen conditions.Compared with Jing 411,KN9204 absorbed significantly more nitrogen at the reproductive stage after shooting and accumulated it in the shoots and seeds.Transcriptome data analysis revealed that nitrogen deficiency clearly suppressed the expression of genes related to cell division in the young spike of Jing 411,whereas this suppression of gene expression was much lower in KN9204.In addition,KN9204 maintained relatively high expression of NPF genes for a longer time than Jing 411 during seed maturity.Physiological and transcriptome data revealed that KN9204 was more tolerant of nitrogen deficiency than Jing 411,especially at the reproductive stage.The high NUE of KN9204 is an integrated effect controlled at different levels.Taken together,our data provide new insights into the molecular mechanisms of NUE and important gene resources for improving wheat cultivars with a higher NUE trait.Xiaoli Shi Fa Cui Xinyin Han Yilin He Long Zhao Na Zhang Hao Zhang Haidong Zhu Zhexin Liu Bin Ma Shusong Zheng Wei Zhang Jiajia Liu Xiaoli Fan Yaoqi Si Shuiquan Tian Jianqing Niu Huilan Wu Xuemei Liu Zhuo Chen Deyuan Meng Xiaoyan Wang Liqiang Song Lijing Sun Jie Han Hui Zhao Jun Ji Zhiguo Wang Xiaoyu He Ruilin Li Xuebin Chi Chengzhi Liang Beifang Niu Jun Xiao Junming Li Hong-Qing Ling 2022Molecular Plant2022,15,9:6

3Transmission of the Chromosome 1R in Winter Wheat Germplasm Aimengniu and Its Derivatives Revealed by Molecular Markers显示文摘In order to clarify the transmission of the rye chromosome 1R in winter wheat germplasm Aimengniu and its derivatives, 17 derivatives and 7 types of Aimengniu were examined through molecular-marker technology. The results showed that the chromosome arm 1RS of Neuzucht was transmitted to 5 of the 7 types of Aimengniu, i.e., Aimengniu II and Aimengniu IV-Aimengniu VII, no segment of 1RS was identified in Aimengniu I or Aimengniu III. As for the 17 derivatives, the 1RS chromosome arm of Aimengniu was transmitted to 11 derivatives, part segments of 1RS were found in 1 derivative, while no segment was found in the remaining 5 ones. 5Optimizing Heavy-duty Anticorrosive Performances of Coating Films Formed by Acrylate-vinylidene Chloride Copolymer Latexes through Twice-painting Technique显示文摘Twice-painting technique was adopted to prepare heavy-duty anticorrosive coating films formed by aqueous latexes of copolymers of vinylidene chloride(VDC) with an acrylate, namely methyl acrylate(MA), ethyl acrylate(EA), butyl acrylate(BA) or 2-ethylhexyl acrylate(EHA). Harsh salt-spray corrosion tests demonstrated that the optimized twicepainting technique was that the acidic latex solution was adjusted to p H 5-6 for the first painting, while it was utilized directly for the second painting. The test of 600 h of harsh salt-spray corrosion showed that MA-VDC85 coating could protect the steel excellently, whereas the other acrylate-VDC coatings with 75%-90% VDC content could not protect the steel so effectively. Further corrosion test showed that(1) MA-VDC85 coating protected steel from loss of metallic luster for at least 1000 h of salt-spray corrosion;(2) adhesion of MA-VDC85 coating to steel was excellent for at least 800 h of saltspray corrosion, but became very poor after 1000 h. 6Genetic dissection of wheat uppermost-internode diameter and its association with agronomic traits in five recombinant inbred line populations at various field environments显示文摘Uppermost-internode diameter(UID)is a key morphological trait associated with spike development and yield potential in wheat.Our understanding of its genetic basis remains largely unknown.Here,quantitative trait loci(QTLs)for UID with high-density genetic maps were identified in five wheat recombinant inbred line(RIL)populations.In total,25 QTLs for UID were detected in five RIL populations,and they were located on chromosomes 1A,1D(3 QTL),2B(2),2D(3),3B,3D,4A,4B(3),4D,5A(5),5B(2),6B,and 7D.Of them,five major and stable QTLs(QUid.sau-2CN-1D.1,QUid.sau-2SY-1D,QUid.sau-QZ-2D,QUid.sau-SC-3D,and QUid.sau-AS-4 B)were identified from each of the five RIL populations in multiple environments.QUid.sau-2CN-1D.1,QUid.sau-2SY-1D and QUid.sau-SC-3D are novel QTLs.Kompetitive Allele Specific PCR(KASP)markers tightly linked to them were further investigated for developing near-isogenic lines(NILs)carrying the major loci.Furthermore,candidate genes at these intervals harboring major and stable QTLs were predicted,and they were associated with plant development and water transportation in most cases.Comparison of physical locations of the identified QTL on the‘Chinese Spring’reference genome showed that several QTLs including two major ones,QUid.sau-2CN-1D.1 and QUid.sau-2 SY-1 D,are likely allelic confirming their validity and effectiveness.The significant relationships detected between UID and other agronomic traits and a proper UID were discussed.Collectively,our results dissected the underlying genetic basis for UID in wheat and laid a foundation for further fine mapping and map-based cloning of these QTLs.LIU Hang TANG Hua-ping LUO Wei MU Yang JIANG Qian-tao LIU Ya-xi CHEN Guo-yue WANG Ji-rui ZHENG Zhi QI Peng-fei JIANG Yun-feng CUI Fa SONG Yin-ming YAN Gui-jun WEI Yu-ming LAN Xiu-jin ZHENG You-liang MA Jian 2021Journal of Integrative Agriculture2021,20,11:1

18Intramolecular hydrogen bonds in a single macromolecule:Strength in high vacuum versus liquid environments显示文摘As a weak non-covalent interaction,hydrogen bond(H-bond)is highly susceptible to the environmental interference.However,the direct quantification of a single H-bond under an interference-free condition is still a challenge.Herein,the intramolecular H-bond in a model system,poly(N-isopropylacrylamide),is studied in high vacuum by single-molecule atomic force microscopy and steered molecular dynamics simulations,which allows the precise quantification of H-bond strength in an interference-free state.Control experiments show that the H-bond is significantly weakened in nonpolar solvent,even if the dielectric constant is very close to vacuum.If a polar solvent is used as the environment,the H-bond will be further weaker or even broken.These results imply that for experiments in any liquid environment,the H-bond strength(△G)will be only -50% or even less of that measured in vacuum.Further analysis shows that in liquid environments,AG decays in a quasi-linear way with the increase of the dielectric constant(ε).For H-bond studies in future,the result measured in vacuum can be set as the standard value,namely,the inherent strength.This approach will provide fundamental insights into the H-bond participated nano-structures and materials in different environments.Wanhao Cai Duo Xu Fa Zhang Junhao Wei Song Lu Linmao Qian Zhongyuan Lu Shuxun Cui 2022Nano Research2022,15,2:0
19SYSTHESIS OF DOUBLE-HYDROPHILIC CORE-SHELL TYPE MULTIARM STAR COPOLYMER POLYETHYLENIMINE-block-POLY(2-HYDROXYETHYL METHACRYLATE)显示文摘Multiarm star block copolymers hyperbranched polyethylenimine-b-poly(2-hydroxyethyl methacrylate) (HPEI-b-PHEMA) with average 28 PHEMA arms have been prepared by atom transfer radical polymerization (ATRP) of HEMA in a mixed solvent of methanol and water using a core-first strategy. The hyperbranched macroinitiator employed was prepared on the basis of well-defined hyperbranched polyethylenimine with Mw/Mn of 1.04 by amidation with 2-bromo-isobutyryl bromide. The polymerization condition was optimized to prepare star copolymers with narrow dispersity, and the variables included the volume ratio of methanol to water, the molar ratio of initiating site to CuCl and the molar ratio of [CuCl]:[CuBr2]. Under the optimized polymerization condition, the lowest Mw/Mn value of the obtained star copolymers was around 1.3. Kinetic analysis showed that an induction period existed in the polymerization of HEMA. After this induction period, a linear dependence of ln([M]0/[M]t) on time was observed. The obtained HPEI-b-PHEMA could adsorb hydrophilic molecules. 20Mifepristone in Combination with Misoprostol vs. Low Dose Mifepristone Alone in Emergency Contraception: a Multi-center Double-blind Randomized Clinical Trial显示文摘To compare the effectiveness and side effects of various low dose of Mifepristone in combination with Misoprostol and low doses Mifepristone alone in emergency contraception Materials & Methods This is a multi center double blind randomized controlled clinical trial. A total of 899 healthy women were allocated into this study and were randomly divided into 3 groups. They were orally administrated with different emergency contraceptives with 120 h after unprotected intercourse. Group I (n=300) was given 25 mg Mifepristone plus 0.2 mg Misoprostol after 24 h. Group II (n=299) was given 10 mg Mifepristone plus 0.2 mg Misoprostol after 24 h. Group III (n=300) was administrated with 10 mg Mifepristone alone. The effective rates in different groups were calculated with Dixon method. Results Altogether 11 pregnancies occurred, among which 2 cases were in Group I, 2 cases in Group II, and 7 cases were in Group III. After correction with method failure, there was only one case in Group I, 0 case in Group II, and 5 cases in Group III. The contraceptive effectiveness in these groups was 95.5%, 100% and 76.9% respectively. The pregnancy rate was significantly lower in Group I and Group II than that of Group III (P<0.01). The side effects were slight and tolerable, and there was no significant difference between different groups (P>0.05).
