| 1 | 血液甲胎蛋白mRNA对肝癌术后复发转移的影响:3年回顾性研究显示文摘目的 探讨血液肝癌细胞相关甲胎蛋白(AFP) mRNA阳性与肝癌患者术后肿瘤复发或转移的关系.方法 应用巢式RT-PCR检测62例原发性肝癌患者术前1天周围静脉血液中AFPmRNA,术后随访3年.结果 24例术前血液检测AFPmRNA阳性,38例患者术前AFPmRNA呈阴性.术前血AFPmRNA阳性的患者术后1、2、3年肿瘤复发率分别为58.3% (14/24)、75.0%(18/24)、100.0% (24/24);而术前血AFPmRNA阴性的患者术后1、2、3年肿瘤复发率则分别为23.6% (10/38)、42.1%(16/38)、68.4%(26/38),两者比较复发率差异具有统计学意义(P<0.01).术前血AFPmRNA阳性的患者术后3年有8.3% (2/24)发生肺转移;而阴性患者术后3年内无一发生肺或其他远处转移(0.0%,0/38;P<0.01).结论 术前血液AFPmRNA阳性的肝癌患者较AFPmRNA阴性的肝癌患者术后更容易产生复发或远处转移. | 刘扬 王跃如 王龙 周波 宋振顺 Butterfield Lisa H. 张柏和 吴孟超 | 2014 | 中华肝胆外科杂志2014,20,10: | 6 |
| 2 | AFP特异性CD8^+ T/CD4^+ T肝癌疫苗体外杀伤肝癌细胞的实验研究显示文摘目的甲胎蛋白作为一种肿瘤特异性抗原,是临床上诊断肝癌的重要指标。本研究中,检测AFP特异性T细胞的体外抗肿瘤免疫活性。方法采用经过AFP抗原决定簇肽刺激或慢病毒转染的的树突状细胞(dendriticcells,DC),活化AFP特异性的CD4+T细胞和CD8+T细胞,并在体外检测这群T细胞对人肝癌细胞HepG2的杀伤活性。结果本研究结果显示,AFP特异性的CD4+T细胞和CD8+T细胞体外能有效杀伤HepG2细胞,上调培养体系中IL-2、IFN-γ、TNF-α、穿孔素、颗粒酶的水平,对于T细胞活化的负调控因子IL-10,具有显著的抑制作用。结论经DC活化后的AFP特异性T细胞具有显著的体外抗肿瘤免疫活性,有望成为肝细胞癌的免疫治疗新方法。 | 刘扬 宋振顺 周波 傅晓辉 吴孟超 Butterfield LH | 2011 | 同济大学学报(医学版)2011,32,1: | 6 |
| 3 | Intranasal rapamycin ameliorates Alzheimerlike cognitive decline in a mouse model of Down syndrome显示文摘Background:Down syndrome(DS)individuals,by the age of 40s,are at increased risk to develop Alzheimer-like dementia,with deposition in brain of senile plaques and neurofibrillary tangles.Our laboratory recently demonstrated the disturbance of PI3K/AKT/mTOR axis in DS brain,prior and after the development of Alzheimer Disease(AD).The aberrant modulation of the mTOR signalling in DS and AD age-related cognitive decline affects crucial neuronal pathways,including insulin signaling and autophagy,involved in pathology onset and progression.Within this context,the therapeutic use of mTOR-inhibitors may prevent/attenuate the neurodegenerative phenomena.By our work we aimed to rescue mTOR signalling in DS mice by a novel rapamycin intranasal administration protocol(InRapa)that maximizes brain delivery and reduce systemic side effects.Methods:Ts65Dn mice were administered with InRapa for 12 weeks,starting at 6 months of age demonstrating,at the end of the treatment by radial arms maze and novel object recognition testing,rescued cognition.Results:The analysis of mTOR signalling,after InRapa,demonstrated in Ts65Dn mice hippocampus the inhibition of mTOR(reduced to physiological levels),which led,through the rescue of autophagy and insulin signalling,to reduced APP levels,APP processing and APP metabolites production,as well as,to reduced tau hyperphosphorylation.In addition,a reduction of oxidative stress markers was also observed.Discussion:These findings demonstrate that chronic InRapa administration is able to exert a neuroprotective effect on Ts65Dn hippocampus by reducing AD pathological hallmarks and by restoring protein homeostasis,thus ultimately resulting in improved cognition.Results are discussed in term of a potential novel targeted therapeutic approach to reduce cognitive decline and AD-like neuropathology in DS individuals. | Antonella Tramutola Chiara Lanzillotta Eugenio Barone Andrea Arena Ilaria Zuliani Luciana Mosca Carla Blarzino D.Allan Butterfield Marzia Perluigi Fabio Di Domenico | 2018 | Translational Neurodegeneration2018,7,1: | 3 |

| 7 | The Relationship between Shell Morphology and Crevice Size Affecting Retreat Selection of the Keeled Box Turtle(Cuora mouhotii)显示文摘Rock crevices are important retreat sites for many reptiles.Unlike lizards and snakes,crevicedwelling turtles have rigid shells which severely limit the size and shape of crevices they are able to use.Few studies have addressed the relationship between turtle's shell morphology,behavior,and crevice size.Here,we investigate the relationship between morphology and crevice size selection in the Keeled box turtle(Cuora mouhotii).We radio tracked turtles in the field to document crevice dimensions and turtle behavior associated with crevices,and we conducted a controlled experiment in the lab to understand how a turtle's morphology influences its decision to select crevices with different dimensions.Both laboratory and field experiments demonstrate that turtles select deep and moderately wide crevices that are low enough to barely accommodate the carapace.Crevices used by turtles tend to be about 2 times as wide,1.2 times as high,and 3 times longer than the turtle's shell.Moreover,turtles tended to prefer crevices with horizontal openings in the field and position themselves head first in the crevice.We conclude that morphology,especially carapace height,influences the Keeled box turtles' decision to select specific sizes of crevice.We argue that the tendency for turtles to select crevices that come into contact with their carapace contributes to successful defense against predators,as it would be more difficult for a predator to remove a turtle when the crevice is nearly touching the turtles shell. | Fanrong XIAO Zhen HONG Jichao WANG Taggert BUTTERFIELD Haitao SHI | 2020 | Asian Herpetological Research2020,11,4: | 2 |
