1Preventive effect of glutamine on intestinal barrier dysfunction induced by severe trauma显示文摘AIM: To investigate the mechanism underlying intestinalbarrier function damage after severe trauma and thetherapeutic effect of glutamine.METHODS: Burned patients, and animal models of severstrauma replicated by hemorrhagic shock combined withendotoxin infusion and burn injury, were included in a serialexperiment. Effects of oral glutamine on intestinal barrierfunction were observed in scalded rets. Parametersmeasured in these experiments were as follows: plasmalevels of diamine oxidase (DAO), tumor necrosis factor(TNFα), endotoxin (LPS), and lactate as well as D-lactateby biochemical methods, lactose/mannitol (L/M) ratio inurine by SP-3400, and pathological examination of intestinalmucosa under light microscopy.RESULTS: Plasma DAO activity was significantly increasedafter injury. There was a negative correlation betweenplasma DAO and intestinal mucosal DAO or pHi ( r= -0.93,plasma 0.80 ± 0.93,2.83 ± 1.71, 1.14 ± 0.64,2.36 ± 2.06 and2.49± 1.67 vs intestinal 0.52± 0.12,0.34 ± 0.03,0.45 ± 0.18,0.37± 0.26 and 0.41 ± 0.07; r = - 0.533, plasma 0.87 ± 0.75,1.89± 1.13, 1.21 ± 0.23,3.03 ± 2.61 and 4.70 ± 1.22 Vs pHi7.03± 0.05,7.05 ± 0.06,7.14 ± 0.096,7.20 ± 0.08 and 7.05 ±0.07; P < 0.01-0.05). Positive correlations were foundbetween DAO activity and plasma TNFα, LPS, lactate, L/Mand D-lactate ( r = 0.817, 0.842, 0.872, and 0.951; plasmaDAO 0.87 ± 0.75,1.89 ± 1.13, 1.21 ± 0.23,3.03 ± 2.61 and 4.70± 1.22 vs TNF 0.08 ± 0.02,0.03 ± 0.25,0.17 ± 0.09,0.34 ± 0.15and 0.33 ± 0.18; vs LPS 0.14 ± 0.03,0.16 ± 0.04,0.21 ± 0.02,0.18± 0.16 and 0.37 ± 0.10; vs lactate 9.03 ± 2.19, 18.30 ±2.56,9.81 ± 2.83,12.01 ± 6.83, 12.01 ± 6.84 and 43.61 ± 11.27;vs L/M 0.03 ± 0.01,0.41 ± 0.27,0.62 ± 0.20, 1.70 ± 0.60; r =0.774, plasma DAO 1.25 ± 0.41,2.17 ± 0.71,2.29 ± 0.87, 1.23± 0.55 and 1.11 ± 0.47 vs D-lactate 8.37 ± 2.48, 18.25 ± 6.18,13.96 ± 4.94, 8.93 ± 3.00 and 12.39 ± 4.94; all P < 0.01),repestively. Damage of intestinal mucosa was found bypathological examination. Intestinal barrier function wasimproved to a certain extent by oral glutamine in scaldedrats.CONCLUSION: Intestinal barrier function was damaged in theearly stage after trauma. 2The effects of anisodamine and dobutamine on gut mucosal blood flow during gut ischemia/reperfusion显示文摘AIM:To detemine if anisodamine is able to augment mucsal perfusion during gut I/R ischemia-reperfusion METHODS:A jejunal sac was formend in Sperague Dawley rat.A Laser Doppler probe and a tonometer were inserted into the sac thich was filled with saline.The superior mesenteric artery was occluded(SMAO)for 60 minutes followed by 90 minutes of reperfusion.At the end of 60 minutes of SMAO.either 0.2mg/kg of anisodmine or dobutamine was injected into the jejunal sac.Lase Doppler mucosal blood flow and regional PCO2(PrCO2) measurements were made .RESULTS:Mucosal blood flow was significantly increased at 30,60 and 90 minutes of reperfusion(R30,R60,R90)when intraluminal anisodamine or dobutamine was present compared to intraluminal saline only(44±3.3%or 48±4.1%vs 37±2.6%at R30,57±5.0%,57±5.0%or56±4.7%vs45±2.7%at R60,64±3.3%or56±4.2%.vs48±3.4%at R90,respectively P<0.05),Blood flow changes were also reflected by lowering of jejunal PrCO2 measurements after imtraluminal anisodamine or dobutamine compared with that of the saline controls(41±3.1mmHg or 44±3.0mmHg vs49±3.7mmHg at R30,38±3.7mmHg or 40±2.1mmHg vs47±3.8mmHg at R60,34±2.1mmHg or 39±3.0mmHg vs46±3.4mmHg at R90,respectively,P<0.05),Most interesting finding was that there were significantly higher mucosal blood flow and lower jejunal PrCO2 in anisodamine group than those in dobutamine group at 90 minutes of reperfusion(64±3.3%vs56±4.2%for blood flow or 34±2.1mmHg vs39±3.0mmHg for PrCOs,respectively,P<0.05).suggesting that anisodamine had a more lasting effect on mucosal perfusion than dobutamine.CONCLUSION.Intraluminal anisodamine and dobutamine can augment mucosal blood flow during gut I/R and alleviate mucosal acidosis.The results provided benifical effects on the treatment of splanchnic hypoperfusion following traumatic or burn shock.Sen Hu Zhi-Yong Sheng,Burns Institute,304th Hospital of PLA,Beijing 100037,China 2002World Journal of Gastroenterology2002,8,3:23
3Effects of hydrogen peroxide on mitochondrial gene expression of intestinal epithelial cells显示文摘AIM: To study the effects of hydrogen peroxide on mitochondrial gene expression of intestinal epithelial cells in in vitro model of hydrogen peroxide-stimulated SW-480 cells.METHODS: RNA of hydrogen peroxide-induced SW-480 cells was isolated, and reverse-transcriptional polymerase chain reaction was performed to study gene expression of ATPase subunit 6, ATPase subunit 8, cytochrome c oxidase subunit Ⅰ (COⅠ), cytochrome coxidase subuit Ⅱ (COⅡ) and cytochrome c oxidase subunit Ⅲ (COⅢ). Mitochondria were isolated and activities of mitochondrial cytochrome c oxidase and ATPase were also measured simultaneously.RESULTS: Hydrogen peroxide led to differential expression of mitochondrial genes with some genes up-regulated or down-regulated in a dose dependent manner. Differences were very obvious in expressions of mitochondrial genes of cells treated with hydrogen peroxide in a concentration of 400 μmol/L or 4 mmol/L. In general, differential expression of mitochondrial genes was characterized by up-regulation of mitochondrial genes in the concentration of 400 μmol/L and down-regulation in the concentration of 4 mmol/L. 4Rapid mitogen-activated protein kinase by basic fibroblast growth factor in rat intestine after ischemia/reperfusion injury显示文摘AIM: Previous studies showed that exogenous basic fibroblast growth factor (bFGF or FGF-2) could improve physiological dysfunction after intestinal ischemia/reperfusion (I/R) injury. However, the mechanisms of this protective effect of bFGF are still unclear. The present study was to detect the effect of bFGF on the activities of mitogen-activated protein kinase (MAPK) signaling pathway in rat intestine after I/R injury, and to investigate the protective mechanisms of bFGF on intestinal ischemiain jury.METHODS: Rat intestinal I/R injury was produced by clamping the superior mesenteric artery (SMA) for 45 minutes and followed by reperfusion for 48 hours. Seventy-eight Wistar rats were used and divided randomly into sham-operated group (A), normal saline control group (B),bFGF antibody pre-treated group (C), and bFGF treated group (D). In group A, SMA was separated without occlusion. In groups B, C and D, SMA was separated and occluded for 45 minutes, then, released for reperfusion for 48 hours. After the animals were sacrificed, blood and tissue samples were taken from the intestine 45 minutes after ischemia in group A and 2, 6, 24, and 48 hours after reperfusion in the other groups. Phosphorylated forms of p42/p44 MAPK, p38 MAPK and stress activated protein kinase/C-Jun N-terminal kinase (SAPK/JNK) were measured by immunohistochemistry. Plasma levels of D-lactate were examined and histological changes were observed under the light microscope.RESULTS: Intestinal I/R injury induced the expression of p42/p44 MAPK, p38 MAPK, and SAPK/JNK pathways and exogenous bFGF stimulated the early activation of p42/p44 MAPK and p38 MAPK pathways. The expression of phosphorylated forms of p42/p44 MAPK was primarily localized in the nuclei of crypt cells and in the cytoplasm and nuclei of villus cells. The positive expression of p38 MAPK was localized mainly in the nuclei of crypt cells, very few in villus cells. The activities of p42/p44 MAPK and p38 MAPK peaked 6 hours after reperfusion in groups B and C,while SAPK/JNK peaked 24 hours after reperfusion. The activities of p42/p44 MAPK and p38 MAPK peaked 2 hours after reperfusion in group D and those of SAPK/JNK were not changed in group B. D-lactate levels and HE staining showed that the intestinal barrier was damaged severely 6 hours after reperfusion, however, histological structures were much improved 48 hours after reperfusion in group D than in the other groups.CONCLUSION: The results indicate that intestinal I/R injury stimulates the activities of MAPK pathways, and that p42/p44 MAPK and p38MAPK activities are necessary for the protective effect of exogenous bFGF on intestinal I/R injury.The protective effect of bFGF on intestinal dysfunction may be mediated by the early activation of p42/p44 MAPK and p38 MAPK signaling pathways.Xiao-Bing Fu Yin-Hui Yang Tong-Zhu Sun Wei Chen Jun-You Li Zhi-Yong Sheng Wound Healing and Cell Biology Laboratory,Burns Institute,304 Hospital,Trauma Center of Postgraduate Medical College,Beijing 100037,China 2003World Journal of Gastroenterology2003,9,6:11
5Distribution of nitric oxide synthase in stomach myenteric plexus of rats显示文摘AIM: To study the distribution of nitric oxide synthase (NOS) in rat stomach myenteric plexus.METHODS: The distribution of NOS in gastric wall was studied in quantity and location by the NADPH-diaphorase (NDP) histochemical staining method and whole mount preparation technique.RESULTS: NOS was distributed in whole stomach wall, most of them were located in myenteric plexus, and distributed in submucosal plexus. The shape of NOS positive neurons was basically similar, most of them being round and oval in shape. But their density, size and staining intensity varied greatly in the different parts of stomach. The density was 62 -± 38 cells/mm2(antrum), 43 ± 32 cells/mm2(body), and 32 ± 28 cells/mm2 (fundus), respectively. The size and staining intensity of NOS positive neurons in the fundus were basically the same, the neurons being large and dark stained, while they were obviously different in antrum. In the body of the stomach, the NOS positive neurons were in an intermediate state from fundus to antrum. 6Activation of phosphorylating-p38 mitogen-activated protein kinase and its relationship with localization of intestinal stem cells in rats after ischemia-reperfusion injury显示文摘AIM: To investigate the expression of phosphorylating p38mitogen-activated protein kinase (MAPK) in rat small intestine after ischemia-reperfusion (I/R) insult and its relationship with the localization of intestinal stem cells.METHODS: Forty-eight Wistar rats were divided randomly into three groups, namely intestinal ischemia-reperfusion group (R), intestinal ischemia group (Ⅰ) and sham-operated control group (C). In group I, the animals were killed 45minutes after superior mesenteric artery (SMA) occlusion,while in group R the rats sustained SMA occlusion for 45 minutes and reperfusion for 2, 6, 12 or 24 hours respectively. In shamoperated control group, SMA was separated, but without occlusion. The activity of plasma diamine oxidase (DAO)was determined. Intestinal tissue samples were also taken for histological analysis and immunohistochemical analysis of MAPK p38 detection and intestinal stem cell localization.RESULTS: The changes in histological structure and plasma DAO levels indicated that the intestinal barrier was damaged after intestinal I/R injury. In group C and I, each crypt contained 5-6 p38 MAPK positive cells, which were mainly located in the lower region of the crypts. This was consistent with the distribution of intestinal stem cells. 7急性脊髓损伤患者治疗的临床操作指南:关于MRI基线在临床治疗决策与结果预测中作用的几点建议(英文)显示文摘The objective of this guideline is to outline the role of magnetic resonance imaging(MRI) in clinical decision making and outcome prediction in patients with traumatic spinal cord injury(SCI).Methods A systematic review of the literature was conducted to address key questions related to the use of MRI in patients with traumatic SCI.This review focused on longitudinal studies that controlled for baseline neurologic status.A multidisciplinary Guideline Development Group(GDG) used this information,their clinical expertise,and patient input to develop recommendations on the use of MRI for SCI patients.Based on GRADE(Grading of Recommendation,Assessment,Development and Evaluation),a strong recommendation is worded as ' we recommend,' whereas a weaker recommendation is indicated by 'we suggest.' 11Effect of the current density on electrodepositing alpha-lead dioxide coating on aluminum substrate显示文摘The α-PbO2 electrodes are prepared by anodic electrodeposition on A1/conductive coating electrode from alkaline plumbite solutions in order to investigate the effect of the different current densities on the properties of α-PbO2 electrodes. The physic-ochemical properties of the α-PbO2 electrodes are analyzed by using SEM, EDS, XRD, Tafel plot, linear sweep voltammetry (LSV) and A.C. impedance. A compact and uniform layer of lead dioxide was obtained at the current density of 3 mA·cm-2. A further increase in current density results in smaller particles with high porosity. EDS and XRD analyses have shown that the PbO2 deposited in alkaline conditions is highly non stoichiometric, and the PbO impurities are formed on the surface layer besides the α-PbO2. The corrosion resistance of α-PbO2 at the low current density is superior to that of the high current density. It can be attributed to a porous layer of deposited films at high current densities. When used as anodes for oxygen evolution in aqueous Zn2+ 50 g·L-1, H2SO4 150 g·L-1, the Al/conductive coating/α-PbO2 exhibits lower potential compared to Pb electrode. Al/conductive coating/α-PbO2 electrode with the best electrocatalytic activity was obtained at current density of 1 mA·cm-2. 15Role of mitochondrial dysfunction in hydrogen peroxide-induced apoptosis of intestinal epithelial cells显示文摘AIM: To study the role of mitochondrial dysfunction in hydrogen peroxide-induced apoptosis of intestinal epithelial cells.METHODS: Hydrogen peroxide-induced apoptosis of human intestinal epithelial cell line SW-480 was established. Cell apoptosis was determined by Annexin-V and PI doublestained flow cytometry and DNA gel electrophoresis.Morphological changes were examined with light and electron microscopy. For other observations, mitochondrial function,cytochrome c release, mitochondrial translocation and membrane potential were determined simultaneously.RESULTS: Percentage of apoptotic cells induced with 400μ mol/L hydrogen peroxide increased significantly at I h or 3h after stimulation and recovered rapidly. Meanwhile percentage of apoptotic cells induced with 4 mmol/L hydrogen peroxide increased with time. In accordance with these changes, we observed decreased mitochondrial function in 400 μmol/L H2O2-stimualted cells at 1 h or 3 h and in 4 mmol/L H2O2-stimualted cells at times examined.Correspondingly, swelling cristae and vacuole-like mitochondria were noted. 16The mRNA expression patterns of tumor necrosis factor-a and TNFR-I in some vital organs after thermal injury显示文摘AIM: To investigate changes of tumor necrosis factor-α (TNFα) and TNFR-Ⅰ expression in vital organs and their significance in the pathogenesis of multiple organ damage associated with endogenous endotoxin following major burns.METHODS: Wistar rats subjected to a 35 % full-thickness scald injury were sacrificed at 12 h, 24 h, 48 h, and 72 h postburn, respectively. Meanwhile, eight rats were taken as normal controls. Tissue samples from liver, spleen, kidney,lung and intestine were collected to assay tissue endotoxin levels and measure TNF-α and TNFR-Ⅰ expression, In addition, blood samples were obtained for the determination of organ function parameters.RESULTS: Endotoxin levels in liver, spleen and lung increased markedly after thermal injury, with the highest level in liver. The gene expression of TNF-α in liver, lung and kidney was up-regulated after thermal injury, while the TNFR-Ⅰ mRNA expression in liver, lung, kidney and intestine was shown decreased throughout the observation period. Thus, the mRNA expression ratio of TNF-α to TNFRⅠ was significantly increased postburn, particularly in pulmonary tissue (67-fold). In addition, the significant correlations between the expression of TNFR-Ⅰ or the expression ratio of TNF-α/TNFR mRNA in liver tissue and serum aspartate aminotransferase levels were noted (P <0.05-0.01). Similar results were also obtained between pulmonary TNF-α mRNA expression and myeloperoxidase activities (P<0.01), whereas there was a highly negative correlation between levels of renal TNFR-Ⅰ mRNA expression and serum creatinine.CONCLUSION: Burn injury could result in the translocation of gut-derived endotoxin that was mainly distributed in the liver, spleen and lung. 18Conceptus elongation in ruminants: roles of progesterone, prostaglandin, interferon tau and cortisol显示文摘The majority of pregnancy loss in ruminants occurs during the first three weeks after conception, particularly during the period of conceptus elongation that occurs prior to pregnancy recognition and implantation. This review integrates established and new information on the biological role of ovarian progesterone(P4), prostaglandins(PGs),interferon tau(IFNT) and cortisol in endometrial function and conceptus elongation. Progesterone is secreted by the ovarian corpus luteum(CL) and is the unequivocal hormone of pregnancy. Prostaglandins(PGs) and cortisol are produced by both the epithelial cells of the endometrium and the trophectoderm of the elongating conceptus. In contrast, IFNT is produced solely by the conceptus trophectoderm and is the maternal recognition of pregnancy signal that inhibits production of luteolytic pulses of PGF2αby the endometrium to maintain the CL and thus production of P4. Available results in sheep support the idea that the individual, interactive, and coordinated actions of P4, PGs, IFNT and cortisol regulate conceptus elongation and implantation by controlling expression of genes in the endometrium and/or trophectoderm. 19利用测井曲线评价碎屑岩岩石物理参数——以埃及尼罗河三角洲El-Wastani气田上中新统为例显示文摘利用埃及尼罗河三角洲El-Wastani气田10口气井的测井数据,对该气田主要储集层上中新统Qawasim组和Abu Madi组进行岩石物理分析,利用测井参数交会图进行岩性分析,并评估泥质含量、孔隙度、含水饱和度、含油气孔隙体积等参数。分析各井中子孔隙度-密度交会图、中子孔隙度-无铀伽马交会图和岩性-饱和度交会图发现:下Abu Madi组主要为砂岩,偶夹页岩;Qawasim组主要为页岩,偶夹砂岩。利用参数交会图及无铀伽马测井结果,得到有效产层各参数的临界值分别为:泥质含量50%,孔隙度10%,含水饱和度70%。分别绘制了产层有效厚度、平均孔隙度、泥质含量、含水饱和度等参数的等值线图,综合分析发现,研究区Qawasim组和Abu Madi组碎屑岩为优质储集层,天然气有利聚集区呈北西—南东向展布,中部尤为有利。Ezz S El-Din Maher A Mesbah Mohamed A Kassab Ibtehal F Mohamed Burns A Cheadle Mostafa A Teama 2013石油勘探与开发2013,40,4:3
