2Sequential use of transarterial chemoembolization and percutaneous cryosurgery for hepatocellular carcinoma显示文摘AIM: To evaluate the efficacy of sequential use of transarterial chemoembolization (TACE) and percutaneous cryosurgery for unresectable hepatocellular carcinoma (HCC). METHODS: Four hundred and twenty patients were enrolled in this study. The patients, who were considered to have unresectable tumors due to their location or size or comorbidity, were divided into sequential TACE-cryosurgery (sequential) group (n = 290) and cryosurgery alone (cryoalone) group (n = 130). Patients in the sequential group tended to have larger tumors and a greater number of tumors than those in the cryo-alone group. Tumors larger than 10 cm in diameter were only seen in the sequential group. TACE was performed with the routine technique and percutaneous cryosurgery was conducted under the guidance of ultrasound 2-4 wk after TACE. RESULTS: During a mean follow-up period of 42 ± 17 mo (range, 24-70 mo), the local recurrence rateat the ablated area was 17% for all patients, 11% and 23% for patients in sequential group and cryoalone groups, respectively (P = 0.001). The overall 1-, 2-, 3-, 4and 5-year survival rate was 72%, 57%, 47%, 39% and 31%, respectively. The 1and 2-year survival rates (71% and 61%) in sequential group were similar to those (73% and 54%) in cryo-alone group (P = 0.69 and 0.147), while the 4and 5-year survival rates were 49% and 39% in sequential group, higher than those (29% and 23%) in cryo-alone group (P = 0.001). Eighteen patients with large HCC (> 5 cm in diameter) survived for more than 5 years after sequential TACE while no patient with large HCC (> 5 cm in diameter) survived more than 5 years after cryosurgery. The overall complication rate was 24%, and the complication rates were 21% and 26% for the sequential and cryo-alone groups, respectively (P = 0.06). The incidence of hepatic bleeding was higher in cryo-alone group than in sequential group (P = 0.02). Liver crack only occurred in two patients of the cryoalone group. CONCLUSION: Pre-cryosurgical TACE can increase the cryoablation efficacy and decrease its adverse effects, especially bleeding. 5High levels of circulating GM-CSF^(+) CD4^(+) T cells are predictive of poor outcomes in sepsis patients: a prospective cohort study显示文摘Granulocyte colony-stimulating factor(GM-CSF),produced by CD4^(+)T cells,has recently been implicated in the pathogenesis of inflammatory diseases,such as multiple sclerosis and juvenile arthritis.However,the role of GM-CSF-producing CD4^(+)T cells in sepsis remains unknown.This study reports peripheral changes in GM-CSF-producing CD4^(+)T cells in septic patients and the possible underlying mechanism by which GM-CSF influences the outcome of sepsis.Forty-three septic patients,20 SIRS patients,and 20 healthy controls were enrolled in this study and followed for 28 days to assess mortality.We measured the peripheral frequency of GM-CSF^(+)CD4^(+)T cells and recorded their associated relationship with disease progression.Our data demonstrated that peripheral GM-CSF-producing CD4^(+)T cells were significantly higher in septic patients than in both SIRS patients and healthy controls.These cells exhibit a memory phenotype and impaired IFN-γ-secreting capacity in sepsis patients.Using a receiver operating curve analysis with 8.01%as a cut-off point,the percentage of GM-CSF^(+)CD4^(+)T cells could predict the outcome of septic patients.Combined with the increase in GM-CSF-producing CD4^(+)T cells,inflammatory cytokines IL-1βand IL-6 were also upregulated.Using an in vitro neutrophil model,we found that GM-CSF inhibited C3aR expression,while inducing IL-8 production.Furthermore,this effect was transferrable in plasma from sepsis patients and was attenuated by inhibition of GM-CSF using an anti-GM-CSF antibody.These results indicate that GM-CSF-producing CD4^(+)T cells may serve as a marker of sepsis severity.Thus,targeting GM-CSF overproduction may benefit sepsis patients.Huihuang Huang Siyu Wang Tianjun Jiang Rong Fan Zheng Zhang Jinsong Mu Ke Li Yonggang Wang Lei Jin Fang Lin Jie Xia Lijian Sun Biao Xu Chengcheng Ji Jing Chen Juan Chang Bo Tu Bing Song Chao Zhang Fu-Sheng Wang Ruonan Xu 2019Cellular & Molecular Immunology2019,16,6:6

7Substantial transition to clean household energy mix in rural China显示文摘The household energy mix has significant impacts on human health and climate,as it contributes greatly to many health-and climate-relevant air pollutants.Compared to the well-established urban energy statistical system,the rural household energy statistical system is incomplete and is often associated with high biases.Via a nationwide investigation,this study revealed high contributions to energy supply from coal and biomass fuels in the rural household energy sector,while electricity comprised∼20%.Stacking(the use of multiple sources of energy)is significant,and the average number of energy types was 2.8 per household.Compared to 2012,the consumption of biomass and coals in 2017 decreased by 45%and 12%,respectively,while the gas consumption amount increased by 204%.Increased gas and decreased coal consumptions were mainly in cooking,while decreased biomass was in both cooking(41%)and heating(59%).The time-sharing fraction of electricity and gases(E&G)for daily cooking grew,reaching 69%in 2017,but for space heating,traditional solid fuels were still dominant,with the national average shared fraction of E&G being only 20%.The non-uniform spatial distribution and the non-linear increase in the fraction of E&G indicated challenges to achieving universal access to modern cooking energy by 2030,particularly in less-developed rural and mountainous areas.In some non-typical heating zones,the increased share of E&G for heating was significant and largely driven by income growth,but in typical heating zones,the time-sharing fraction was<5%and was not significantly increased,except in areas with policy intervention.The intervention policy not only led to dramatic increases in the clean energy fraction for heating but also accelerated the clean cooking transition.Higher income,higher education,younger age,less energy/stove stacking and smaller family size positively impacted the clean energy transition.Guofeng Shen Rui Xiong Yanlin Tian Zhihan Luo Bahabaike Jiangtulu Wenjun Meng Wei Du Jing Meng Yuanchen Chen Bing Xue Bin Wang Yonghong Duan Jia Duo Fenggui Fan Lei Huang Tianzhen Ju Fenggui Liu Shunxin Li Xianli Liu Yungui Li Mu Wang Ying Nan Bo Pan Yanfang Pan Lizhi Wang Eddy Zeng Chao Zhan Yilin Chen Huizhong Shen Hefa Cheng Shu Tao 2022National Science Review2022,9,7:4

10Ginsenoside Rb1 relieves glucose fluctuationinduced oxidative stress and apoptosis in Schwann cells显示文摘Cultured Schwann cells were treated with 5.6 mM and 50 mM glucose alternating every 8 hours to simulate intermittent high glucose. The present study analyzed the neuroprotective effects of 1, 10 and 100 μM ginsenoside Rb1 on oxidative damage and apoptosis in Schwann cells induced by intermittent high glucose. Flow cytometry demonstrated that ginsenoside Rb1 reduced intermittent high glucose-mediated reactive oxygen species production. Enzyme linked immunosorbent assay showed that 8-hydroxy-2-deoxy guanosine levels in Schwann cells decreased following ginsenoside Rb1 treatment. Quantitative real-time reverse transcription-PCR and western blot assay results revealed that ginsenoside Rb1 inhibited intermittent high glucose-upregulated Bax expression, but antagonized intermittent high glucose-downregulated Bcl-2 expression in Schwann cells. These effects were most pronounced with 100 μM ginsenoside Rb1. 11Evaluation of the in vitro antimicrobial properties of ultraviolet A/riboflavin mediated crosslinking on Candida albicans and Fusarium solani显示文摘AIM:To evaluate the antimicrobial properties of ultraviolet A(UVA)(365 nm)/riboflavin against Candida albicans and Fusarium solani.METHODS:Two fungus isolates were cultured in vitro and prepared with 10-fold serial PBS dilutions of cell concentration.For each dilution of fungus suspension,the concentration(colony-forming units/mL,CFU/mL)and the inactivation ratio of fungal cells were evaluated under 4 conditions:no treatment(control),UVA(365 nm)/riboflavin,riboflavin,and UVA(365 nm).RESULTS:The cell concentration decreased in UVA(365 nm)/riboflavin group for Candida albicans at each dilution and Fusarium solani at dilutions of 104,103,102CFU/mL,when compared with that in control,riboflavin,and UVA(365 nm)groups(P<0.01).No difference of cell concentration was detected amongst the culture of control,riboflavin,and UVA(365 nm)groups for the two fungus.There is a negative correlation between suspension concentration(log-transformed)and the inactivation ratio in UVA(365 nm)/riboflavin group for Candida albicans and Fusarium solani(P<0.01).CONCLUSION:According to the standard protocol of corneal collagen cross-linking,UVA(365 nm)/riboflavin combination treatment is found to moderately inactivate the viability of Candida albicans and Fusarium solani in vitro.The inactivation ratio was found to increase withthe decrease of cell concentration under UVA(365 nm)/riboflavin condition.Bing Sun Zhi-Wei Li Hai-Qun Yu Xiang-Chen Tao Yong Zhang Guo-Ying Mu 2014International Journal of Ophthalmology(English edition)2014,7,2:2
12Processes and mechanisms of multi-collapse of loess roads in seasonally frozen ground regions: A review显示文摘Usually, the collapsible loess widely distributed across the world can serve as a type of foundation soil that meets its strength requirement after dense compaction and elimination of collapsibility. However, many problems such as cracks and differential settlement still occur in loess roads in the seasonally frozen ground regions after several years of operation. Many studies have demonstrated that these secondary or multiple collapses primarily result from the repeated freezing-thawing, wetting-drying, and salinization-desalinization cycles. Therefore, we conducted a research program to(1) monitor the in-situ ground temperatures and water content in certain loess roads to understand their changes,(2) study the effects of freezing-thawing, wetting-drying, and salinization-desalinization cycles on geotechnical properties and micro-fabrics of compacted loess in the laboratory, and(3) develop mitigative measures and examine their engineered effectiveness, i.e., their thermal insulating and water-proofing effects in field and laboratory tests. Our results and advances are reviewed and some further research needs are proposed. These findings more clearly explain the processes and mechanisms of secondary and multiple collapse of loess roads. 13Nr2e1 Downregulation Is Involved in Excess Retinoic Acid-induced Developmental Abnormality in the Mouse Brain显示文摘Objective This study aimed to investigate the expression pattern and function of Nuclear receptor subfamily 2 group E member 1(Nr2e1) in retinoic acid(RA)-induced brain abnormality. Methods The mouse model of brain abnormality was established by administering 28 mg/kg RA, and neural stem cells(NSCs) were isolated from the mouse embryo and cultured in vitro. Nr2e1 expression was detected by whole mount in situ hybridization, RT-PCR, and Western blotting. Nr2e1 function was determined by transducing Nr2e1 shRNA into NSCs, and the effect on the sonic hedgehog(Shh) signaling pathway was assessed in the cells. In addition, the regulation of Nr2e1 expression by RA was also determined in vitro. Results Nr2e1 expression was significantly downregulated in the brain and NSCs of RA-treated mouse embryos, and knockdown of Nr2e1 affected the proliferation of NSCs in vitro. In addition, a similar expression pattern of Nr2e1 and RA receptor(RAR) α was observed after treatment of NSCs with different concentrations of RA. 14Human activity during the late Pleistocene in the Lop Nur region, northwest China: Evidence from a buried stone artifact显示文摘The Lop Nur region, in the east part of Tarim Basin, was an important transportation junction between west and east,north and south Eurasia. However, previous studies on prehistoric human activity have concentrated mostly on the Bronze Age,whereas that during the Stone Age remains largely unresearched. Here, we present a new direct evidence of human activity in the late Pleistocene, recorded on a grinding stone buried in a lacustrine sediment section of the Lop Nur region. The grain size distribution of the sediment section indicates that the site was probably in the center of a lake with weak hydrodynamic environment. Therefore, the stone artifact can only be carried to here by people instead of river and it was never move as soon as left here. Results of radiocarbon dating, the evident stratigraphic relations between the stone artifact and the sedimentary formation, indicate that the human activity could extend to approximately 13 ka BP. Furthermore, the results of starch-grain and use-wear analyses suggest that ancient humans gathered seeds of Triticeae, roots, and tubers and used the grinding stone to simply process selected plant as plant foodstuffs during this period. 15Interwoven nickel(II)-dimethylglyoxime nanowires in 3D nickel foam for dendrite-free lithium deposition显示文摘Metal skeletons,such as Nickel Foam(NF) has attracted worldwide interests as stable host for lithium metal anode because of its high stability,large specific surface area and high conductivity.However,most metal skeletons have lithophobic surface and uneven current distribution that result in sporadic lithium nucleation and uncontrolled dendrites growth.Herein,we describe a sequential immersing strategy to generate interwoven Nickel(Ⅱ)-dimethylglyoxime(Ni-DMG) nanowires at NF to obtain composite skeleton(NDNF),which can be used as an stable host for Li metal storage.The Ni-DMG has proved effective to realize uniform lithium nucleation and dendrite-free lithium deposition.Combing with the three dimensional(3 D) hierarchical porous structure,the composite host shows a significantly improved coulombic efficiency(CE) than pristine commercial nickel foam.Moreover,the corresponding Li‖Li symmetrical cells can run more than 700 h with low voltage hysteresis 22 mV at 1.0 mA/cm^(2),and Li@NDNF‖LiFePO;full-cell exhibits a high capacity retention of 82.03% at 1.0 C during 630 cycles.These results proved the effectiveness of metal-organic complexes in governing Li metal growth and can be employed as a new strategy for dendrite-free Li metal anode and safe Li metal batteries(LMBs).Tianyi Zhou Yanlu Mu Jianyang Wu Bing Zhong Chengkai Yang Qian Wang Wen Liu Henghui Zhou Peng Jiang 2022Chinese Chemical Letters2022,33,4:1

20Graphene oxide-composited chitosan scaffold contributes to functional recovery of injured spinal cord in rats显示文摘The study illustrates that graphene oxide nanosheets can endow materials with continuous electrical conductivity for up to 4 weeks. Conductive nerve scaffolds can bridge a sciatic nerve injury and guide the growth of neurons;however, whether the scaffolds can be used for the repair of spinal cord nerve injuries remains to be explored. In this study, a conductive graphene oxide composited chitosan scaffold was fabricated by genipin crosslinking and lyophilization. The prepared chitosan-graphene oxide scaffold presented a porous structure with an inner diameter of 18–87 μm, and a conductivity that reached 2.83 mS/cm because of good distribution of the graphene oxide nanosheets, which could be degraded by peroxidase. The chitosan-graphene oxide scaffold was transplanted into a T9 total resected rat spinal cord. The results show that the chitosan-graphene oxide scaffold induces nerve cells to grow into the pores between chitosan molecular chains, inducing angiogenesis in regenerated tissue, and promote neuron migration and neural tissue regeneration in the pores of the scaffold, thereby promoting the repair of damaged nerve tissue. The behavioral and electrophysiological results suggest that the chitosan-graphene oxide scaffold could significantly restore the neurological function of rats. Moreover, the functional recovery of rats treated with chitosangraphene oxide scaffold was better than that treated with chitosan scaffold. The results show that graphene oxide could have a positive role in the recovery of neurological function after spinal cord injury by promoting the degradation of the scaffold, adhesion, and migration of nerve cells to the scaffold.
