| 3 | Three-dimensional morphometric analysis for hepatectomy of centrally located hepatocellular carcinoma:A pilot study显示文摘AIM: To describe a three-dimensional model(3DM) to accurately reconstruct anatomic relationships of centrally located hepatocellular carcinomas(HCCs).METHODS: From March 2013 to July 2014, reconstructions and visual simulations of centrally located HCCs were performed in 39 patients using a 3D subject-based computed tomography(CT) model with customdeveloped software. CT images were used for the 3D reconstruction of Couinaud's pedicles and hepatic veins, and the calculation of corresponding tumor territories and hepatic segments was performed using Yorktal DMIT software. The respective volume, surgical margin, and simulated virtual resection of tumors were also estimated by this model preoperatively. All patients were treated surgically and the results were retrospectively assessed. Clinical characteristics, imaging data, procedure variables, pathologic features, and postoperative data were recorded and compared to determine the reliability of the model.RESULTS: 3D reconstruction allowed stereoscopic identification of the spatial relationships between physiologic and pathologic structures, and offered quantifiable liver resection proposals based on individualized liver anatomy. The predicted values were consistent with the actual values for tumor mass volume(82.4 ± 109.1 m L vs 84.1 ± 108.9 m L, P = 0.910), surgical margin(10.1 ± 6.2 mm vs 9.1 ± 5.9 mm, P = 0.488), and maximum tumor diameter(4.61 ± 2.16 cm vs 4.53 ± 2.14 cm, P = 0.871). In addition,the number and extent of portal venous ramifications, as well as their relation to hepatic veins, were visualized. Preoperative planning based on simulated resection facilitated complete resection of large tumors located in the confluence of major vessels. | 5 | Schisandra sphenanthera extract(Wuzhi Tablet)protects against chronic-binge and acute alcohol-induced liver injury by regulating the NRF2-ARE pathway in mice显示文摘Alcohol abuse leads to alcoholic liver disease and no effective therapy is currently available.Wuzhi Tablet(WZ), a preparation of extract from Schisandra sphenanthera that is a traditional hepatoprotective herb, exerted a significant protective effect against acetaminophen-induced liver injury in our recent studies, but whether WZ can alleviate alcohol-induced toxicity remains unclear. This study aimed to investigate the contribution of WZ to alcohol-induced liver injury by using chronic-binge and acute models of alcohol feeding. The activities of ALT and AST in serum were assessed as well as the level of GSH and the activity of SOD in the liver. The expression of CYP2E1 and proteins in the NRF2-ARE signaling pathway including NRF2, GCLC, GCLM, HO-1 were measured, and the effect of WZ on NRF2 transcriptional activity was determined. We found that both models resulted in liver steatosis accompanied by increased transaminase activities, but that liver injury was significantly attenuated by WZ. WZ administration also inhibited CYP2E1 expression induced by alcohol, and elevated the level of GSH and the activity of SOD in the liver. Moreover, the NRF2-ARE signaling pathway was activated by WZ andthe target genes were all upregulated. Furthermore, WZ significantly activated NRF2 transcriptional activity. | 7 | Evaluation of the simulation capability of the Wavewatch Ⅲ model for Pacific Ocean wave显示文摘Wave climate analysis and other applications for the Pacific Ocean require a reliable wave hindcast. Five source and sink term packages in the Wavewatch III model(v3.14 and v4.18) are compared and assessed in this study through comprehensive observations, including altimeter significant wave height, advanced synthetic aperture radar swell, and buoy wave parameters and spectrum. In addition to the evaluation of typically used integral parameters, the spectra partitioning method contributes to the detailed wave system and wave maturity validation. The modified performance evaluation method(PS) effectively reduces attribute numbers and facilitates the overall assessment. To avoid possible misleading results in the root mean square error-based validations, another indicator called HH(indicating the two authors) is also calculated to guarantee the consistency of the results. The widely used Tolman and Chalikov(TC) package is still generally efficient in determining the integral properties of wave spectra but is physically deficient in explaining the dissipation processes. The ST4 package performs well in overall wave parameters and significantly improves the accuracy of wave systems in the open ocean. Meanwhile, the newly published ST6 package is slightly better in determining swell energy variations. The two packages(ACC350 and BJA) obtained from Wavewatch III v3.14 exhibit large scatters at different sea states. The three most ideal packages are further examined in terms of reproducing waveinduced momentum flux from the perspective of transport. Stokes transport analysis indicates that ST4 is the closest to the NDBC-buoy-spectrum-based transport values, and TC and ST6 tend to overestimate and underestimate the transport magnitude, respectively, in swell mixed areas. This difference must be considered,particularly in air–wave–current coupling research and upper ocean analysis. | 8 | Effect of artesunate supplementation on bacterial translocation and dysbiosis of gut microbiota in rats with liver cirrhosis显示文摘AIM: To evaluate the effect of artesunate(AS) supplementation on bacterial translocation(BT) and gut microbiota in a rat model of liver cirrhosis. METHODS: Fifty-four male Sprague-Dawley rats were randomly divided into a normal control group(N), a liver cirrhosis group(M) and a liver cirrhosis group intervened with AS(MA). Each group was sampled at 4, 6 and 8 wk. Liver cirrhosis was induced by injection of carbon tetrachloride(CCl4), intragastric administration of 10% ethanol, and feeding a high fat diet. Rats in the MA group were intragastrically administered with AS(25 mg/kg body weight, once daily). Injuries of the liver and intestinal mucosa were assessed by hematoxylineosin or Masson's trichrome staining. Liver index was calculated as a ratio of the organ weight(g) to body weight(g). The gut microbiota was examined by automated ribosomal intergenic-spacer analysis of fecal DNA. BT was assessed by standard microbiological techniques in the blood, mesenteric lymph nodes(MLNs), liver, spleen, and kidney. RESULTS: Compared to group N, the body weight was reduced significantly in groups M and MA due to the development of liver cirrhosis over the period of 8 wk. The body weight was higher in group MA than in group M. The liver indices were significantly elevated at 4, 6 and 8 wk in groups M and MA compared to group N. AS supplementation partially decreased the liver indices in group MA. Marked histopathologic changes in the liver and small intestinal mucosa in group M were observed, which were alleviated in group MA. Levels of pro-inflammatory interleukin-6 and tumor necrosis factor-α were significantly elevated at 8 wk in ileal homogenates in group M compared to group N, which were decreased after AS supplementation in group MA. The dysbiosis of gut microbiota indicated by the mean diversity(Shannon index) and mean similarity(Sorenson index) was severe as the liver cirrhosis developed, and AS supplementation had an apparent intervention effect on the dysbiosis of gut microbiota at 4 wk. The occurrence of BT was increased in the liver of group M compared to that of group N. AS supplementation reduced BT in group MA at 8 wk. BT also occurred in the MLNs, spleen, and kidney, which was reduced by AS supplementation. BT was not detected in the blood in any group.CONCLUSION: Dysbiosis of gut microbiota, injury of intestinal mucosal barrier and BT occurred as liver cirrhosis progressed, which might enhance inflammation and aggravate liver injury. | 14 | OpenKG Chain:A Blockchain Infrastructure for Open Knowledge Graphs显示文摘The early concept of knowledge graph originates from the idea of the semantic Web,which aims at using structured graphs to model the knowledge of the world and record the relationships that exist between things.Currently publishing knowledge bases as open data on the Web has gained significant attention.In China,Chinese Information Processing Society of China(CIPS)launched the OpenKG in 2015 to foster the development of Chinese Open Knowledge Graphs.Unlike existing open knowledge-based programs,OpenKG chain is envisioned as a blockchain-based open knowledge infrastructure.This article introduces the first attempt at the implementation of sharing knowledge graphs on OpenKG chain,a blockchain-based trust network.We have completed the test of the underlying blockchain platform,and the on-chain test of OpenKG’s data set and tool set sharing as well as fine-grained knowledge crowdsourcing at the triple level.We have also proposed novel definitions:K-Point and OpenKG Token,which can be considered to be a measurement of knowledge value and user value.1,033 knowledge contributors have been involved in two months of testing on the blockchain,and the cumulative number of on-chain recordings triggered by real knowledge consumers has reached 550,000 with an average daily peak value of more than 10,000.For the first time,we have tested and realized on-chain sharing of knowledge at entity/triple granularity level.At present,all operations on the data sets and tool sets at OpenKG.CN,as well as the triplets at OpenBase,are recorded on the chain,and corresponding value will also be generated and assigned in a trusted mode.Via this effort,OpenKG chain looks forward to providing a more credible and traceable knowledge-sharing platform for the knowledge graph community. | Huajun Chen Ning Hu Guilin Qi Haofen Wang Zhen Bi Jie Li Fan Yang | 2021 | Data Intelligence2021,3,2: | 7 |
| 15 | Climate and extrema of ocean waves in the East China Sea显示文摘Wave climate plays an important role in the air-sea interaction over marginal seas. Extreme wave height provides fundamental information for various ocean engineering practices, such as hazard mitigation, coastal structure design, and risk assessment. In this paper, we implement a third generation wave model and conduct a high-resolution wave hindcast over the East China Sea to reconstruct a 15-year wave field from 1988 to 2002 for derivation of monthly mean wave parameters and analysis of extreme wave conditions. The numerical results of the wave field are validated through comparison with satellite altimetry measurements, low-resolution reanalysis, and the ocean wave buoy record. The monthly averaged wave height and wave period show seasonal variation and refined spatial patterns of surface waves in the East China Sea. The climatological significant wave height and mean wave period decrease from the open ocean in the southeast toward the continental area in the northwest, with the pattern generally following the bathymetry. Extreme analysis on the significant wave height at the buoy station indicates the hindcast data underestimate the extreme values relative to the observations.
