| 2 | Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis显示文摘Objective PERK/eI F2α/CHOP is a major signaling pathway mediating endoplasmic reticulum(ER) stress related with atherosclerosis.Oxidized LDL(ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis.The present study was conducted to explore the regulatory effect of ox-LDL on PERK/e IF2α/CHOP signaling pathway in vascular endothelial cells.Methods The effects of ox-LDL on PERK and p-e IF2α protein expression of primary human umbilical vein endothelial cells(HUVECs) were investigated by Western blot analysis.PERK gene silencing and selective eI F2α phosphatase inhibitor,salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis,caspase-3 activity,and CHOP mR NA level.Results Ox-LDL treatment significantly increased the expression of PERK,PERK-mediated inactivation of e IF2α phosphorylation,and the expression of CHOP,as well as the caspase-3 activity and apoptosis.The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral sh RNA or by selective eI F2α phosphatase inhibitor,salubrinal.Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eI F2α/CHOP ER-stress pathway.It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis. | TAO Yong Kang YU Pu Lin BAI Yong Ping YAN Sheng Tao ZHAO Shui Ping ZHANG Guo Qiang | 2016 | Biomedical and Environmental Sciences2016,29,12: | 21 |

| 5 | Genotypic Characterization of Methicillin-resistant Staphylococcus aureus Isolated from Pigs and Retail Foods in China显示文摘Objective To investigate the genotypic diversity of Methicillin-resistant Staphylococcus aureus(MRSA) isolated from pigs and retail foods from different geographical areas in China and further to study the routes and rates of transmission of this pathogen from animals to food. Methods Seventy-one MRSA isolates were obtained from pigs and retail foods and then characterized by multi-locus sequencing typing(MLST), spa typing, multiple-locus variable number of tandem repeat analysis(MLVA), pulsed-field gel electrophoresis(PFGE), and antimicrobial susceptibility testing. Results All isolated MRSA exhibited multi-drug resistance(MDR). Greater diversity was found in food-associated MRSA(7 STs, 8 spa types, and 10 MLVA patterns) compared to pig-associated MRSA(3 STs, 1 spa type, and 6 MLVA patterns). PFGE patterns were more diverse for pig-associated MRSA than those of food-associated isolates(40 vs. 11 pulse types). Among the pig-associated isolates, CC9-ST9-t899-MC2236 was the most prevalent clone(96.4%), and CC9-ST9-t437-MC621(20.0%) was the predominant clone among the food-associated isolates. The CC9-ST9 isolates showed significantly higher antimicrobial resistance than other clones. Interestingly, CC398-ST398-t034 clone was identified from both pig-and food-associated isolates. Of note, some community-and hospital-associated MRSA strains(t030, t172, t1244, and t4549) were also identified as food-associated isolates. Conclusion CC9-ST9-t899-MC2236-MDR was the most predominant clone in pigs, but significant genetic diversity was observed in food-associated MRSA. | 8 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 9 | Behavioral Abnormality along with NMDAR-related CREB Suppression in Rat Hippocampus after Shortwave Exposure显示文摘Objective To estimate the detrimental effects of shortwave exposure on rat hippocampal structure and function and explore the underlying mechanisms. Methods One hundred Wistar rats were randomly divided into four groups(25 rats per group) and exposed to 27 MHz continuous shortwave at a power density of 5, 10, or 30 m W/cm^2 for 6 min once only or underwent sham exposure for the control. The spatial learning and memory, electroencephalogram(EEG), hippocampal structure and Nissl bodies were analysed. Furthermore, the expressions of N-methyl-D-aspartate receptor(NMDAR) subunits(NR1, NR2 A, and NR2 B), c AMP responsive element-binding protein(CREB) and phosphorylated CREB(p-CREB) in hippocampal tissue were analysed on 1, 7, and 14 days after exposure. Results The rats in the 10 and 30 m W/cm^2 groups had poor learning and memory, disrupted EEG oscillations, and injured hippocampal structures, including hippocampal neurons degeneration, mitochondria cavitation and blood capillaries swelling. The Nissl body content was also reduced in the exposure groups. Moreover, the hippocampal tissue in the 30 m W/cm^2 group had increased expressions of NR2 A and NR2 B and decreased levels of CREB and p-CREB. Conclusion Shortwave exposure(27 MHz, with an average power density of 10 and 30 m W/cm^2) impaired rats' spatial learning and memory and caused a series of dose-dependent pathophysiological changes. | 10 | Sedimentary Evolution and Transgressions of the Western Subei Basin in Eastern China since the Late Pliocene显示文摘Sedimentary sequences in the Subei Basin are complex and have been affected by interactions between the ocean and rivers since the Late Pliocene, including the Yellow River, Huaihe River, and the Yangtze River. This sedimentary evolution, in particular the timing of Pleistocene transgressions, has long been a matter of controversy owing to the lack of precise chronological evidence. The aim of this study is to explore the evolution of the sedimentary environment throughout the past 3.00 Ma in this region on the basis of a comprehensive analysis of particle size and foraminifera and ostracods collected in the TZK9 core from the Subei Basin combined with geochronological studies of magnetostratigraphy, AMS^(14)C and optically stimulated luminescence(OSL). The results show that fluvial facies in the sedimentary environment from 3.00 to 1.01 Ma. There were fluvial facies and reflects six sea-level high stands from 1.01 to 0.25 Ma. The study area was affected by four large-scale transgressions since 0.25 Ma. The four marine sedimentary layers known as DU7(buried at 48–52 m), DU5(buried at 35–41 m), DU3(buried at 16–23 m), and DU1(buried at 2–4 m) are recorded in the MIS7(210–250 ka), MIS5, MIS3, and Holocene, respectively. The magnitude of the DU5 transgression was identical to that of the DU3 transgression, both were larger than the DU7 transgression, and the DU1 transgression was the weakest. The variation of transgression strength reflects the influence of global changes in sea level, tectonic subsidence, shell ridges, and sand dams. In the TZK9 core, we found evidence of seven sea-level high stands from the Early–Middle Pleistocene, and the first one caused by regional rapid subsidence and could be traced back to 0.83-0.84 Ma. The sea-level high stands and the age of the first one recorded above was different from other cores in eastern China, this was caused by the lack of absolute age control and the differences in paleotopography during this period. This study reconstructs sedimentary evolution, determines the transgression and its age, establishes the chronology since the Late Pliocene, and provides a scientific framework for further paleoenvironmental and tectonic studies. | 11 | Epidemiology of urticaria in China:a population-based study显示文摘Background:Urticaria is a common skin disease characterized by episodes of wheals,and it has a negative effect on patients’quality of life.Large-scale population-based epidemiological studies of urticaria are scarce in China.The aim of this survey was to determine the prevalence,clinical forms,and risk factors of urticaria in the Chinese population.Methods:This survey was conducted in 35 cities from 31 provinces,autonomous regions,and municipalities of China.Two to three communities in each city were selected in this investigation.Participants completed questionnaires and received dermatological examinations.We analyzed the prevalence,clinical forms,and risk factors of urticaria.Results:In total,44,875 questionnaires were distributed and 41,041 valid questionnaires were collected(17,563 male and 23,478 female participants).The lifetime prevalence of urticaria was 7.30%,with 8.26%in female and 6.34%in male individuals(P<0.05).The point prevalence of urticaria was 0.75%,with 0.79%in female and 0.71%in male individuals(P<0.05).Concomitant angioedema was found in 6.16%of patients.Adults had a higher prevalence of urticaria than adolescents and children.Living in urban areas,exposure to pollutants,an anxious or depressed psychological status,a personal and family history of allergy,thyroid diseases,and Helicobacter pylori infection were associated with a higher prevalence of urticaria.Smoking was correlated with a reduced risk of urticaria.Conclusion:This study demonstrated that the lifetime prevalence of urticaria was 7.30%and the point prevalence was 0.75%in the Chinese population;women had a higher prevalence of urticaria than men.Various factors were correlated with urticaria. | Jiaqing Li Dandan Mao Shuoshuo Liu Ping Liu Jing Tian Chenhong Xue Xiaojing Liu Ruiqun Qi Bingxue Bai Jianjun Nie Siqi Ye Yu Wang Yuye Li Qing Sun Juan Tao Shuping Guo Hong Fang Jianqin Wang Qiri Mu Quanzhong Liu Yan Ding Jianzhong Zhang | 2022 | Chinese Medical Journal2022,,11: | 6 |

The vertical pressure on the base of the retaining wall, the horizontal displacement of the wall and the horizontal soil pressure acting on the wall were analyzed. | 14 | Proteomic analysis of differentially expressed proteins involving in liver metastasis of human colorectal carcinoma显示文摘BACKGROUND:Early diagnosis of liver metastasis of colorectal carcinoma is very important for the appropriate treatment of such patients.However,there has been no effective approach available for clinical application.The present study aimed to investigate the differential expression of proteins in patients with liver metastasis of colorectal carcinomas using proteomic analysis and evaluate its potentiality in clinical diagnosis. METHODS:Fluorescence two-dimensional differential in-gel electrophoresis(2-D DIGE)was used to analyze and compare the protein expression between normal mucosa,the primary focus,and liver metastases.Proteomic analysis was made to identify the differentially expressed proteins.Immunohistological staining was used to confirm the expression of differentially expressed proteins in colorectal carcinomas and areas of liver metastasis. RESULTS:A 1.5-fold difference was found with 46 differentially expressed proteins.In 20 differentially expressed proteins,3 were down-regulated and 17 up-regulated in liver metastases. Proteomic analysis showed that the S-adenosylmethionine transgelin variant was down-regulated in liver metastasis tissues.Zinc finger protein 64 homolog(Zfp64),guanine nucleotide exchange factor 4(GEF4),human arginase,glutathione S-transferases(GSTs)A3,and tumor necrosis factorα(TNF-α)-induced protein 9 were up-regulated in liver metastasis tissues.Immunohistochemical staining confirmed that human arginase expression was higher in liver metastases than in the primary focus. CONCLUSIONS:There was a significant difference in protein expression between the primary focus of colorectal carcinoma and liver metastases.The differentially regulated proteins were closely related to liver metastasis of colorectal carcinoma. | 16 | Effect of acupuncture plus conventional treatment on brain activity in ischemic stroke patients: a regional homogeneity analysis显示文摘OBJECTIVE: To determine differences in cerebral activity evoked by acupuncture and conventional stroke treatment, and identify the treatment targets.METHODS: In total, 21 patients were randomly divided into two groups. Group A(11 patients) received both acupuncture and conventional treatment, while group B(10 patients) received conventional treatment only. Resting-state functional magnetic resonance imaging(f MRI) was performed on each participant before and after treatment. Regional homogeneity analysis was performed to investigate the potential mechanism of acupuncture treatment by comparing differences in cerebral activity between treatments.RESULTS: Group A showed higher Re Ho in the frontal lobe(BA6, BA46), supra-marginal gyrus(BA40),middle temporal gyrus(BA21), cerebellum, and insula. Group B showed higher Re Ho in the frontal lobe(BA6) and parietal lobe(BA3, BA7).CONCLUSION: Acupuncture and conventional treatment triggered relatively different clinical efficacy and brain responses. Acupuncture treatment more significantly improved the symptoms of stroke patients. | 17 | CD146 as a promising therapeutic target for retinal and choroidal neovascularization diseases显示文摘Blood vessel dysfunction causes several retinal diseases,including diabetic retinopathy,familial exudative vitreoretinopathy,macular degeneration and choroidal neovascularization in pathological myopia.Vascular endothelial growth factor(VEGF)-neutralizing proteins provide benefits in most of those diseases,yet unsolved haemorrhage and frequent intraocular injections still bothered patients.Here,we identified endothelial CD146 as a new target for retinal diseases.CD146 expression was activated in two ocular pathological angiogenesis models,a laser-induced choroid neovascularization model and an oxygeninduced retinopathy model.The absence of CD146 impaired hypoxia-induced cell migration and angiogenesis both in cell lines and animal model.Preventive or therapeutic treatment with anti-CD146 antibody AA98 significantly inhibited hypoxia-induced aberrant retinal angiogenesis in two retinal disease models.Mechanistically,under hypoxia condition,CD146 was involved in the activation of NFκB,Erk and Akt signalling pathways,which are partially independent of VEGF.Consistently,anti-CD146therapy combined with anti-VEGF therapy showed enhanced impairment effect of hypoxia-induced angiogenesis in vitro and in vivo.Given the critical role of abnormal angiogenesis in retinal and choroidal diseases,our results provide novel insights into combinatorial therapy for neovascular fundus diseases. | Bai Xue Ping Wang Wenzhen Yu Jing Feng Jie Li Rulian Zhao Zhenglin Yang Xiyun Yan Hongxia Duan | 2022 | Science China(Life Sciences)2022,65,6: | 3 |

| 19 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin.
