1Multi-spectroscopic characterization of bovine serum albumin upon interaction with atomoxetine显示文摘The quenching interaction of atomoxetine(ATX) with bovine serum albumin(BSA) was studied in vitro under optimal physiological condition(pH=7.4) by multi-spectroscopic techniques. The mechanism of ATX-BSA system was a dynamic quenching process and was confirmed by the fluorescence spectra and lifetime measurements. The number of binding sites, binding constants and other binding characteristics were computed. Thermodynamic parameters ΔH^0 and ΔS^0 indicated that intermolecular hydrophobic forces predominantly stabilized the drug-protein system. The average binding distance between BSA and ATX was studied by F?rsters theory. UV-absorption, Fourier transform infrared spectroscopy(FT-IR), circular dichroism(CD), synchronous spectra and three-dimensional(3D) fluorescence spectral results revealed the changes in micro-environment of secondary structure of protein upon the interaction with ATX. 2Endoscopic submucosal dissection of gastric tumors: A systematic review and meta-analysis显示文摘AIM: To systematically review the medical literature in order to evaluate the safety and efficacy of gastric endoscopic submucosal dissection(ESD).METHODS: We performed a comprehensive literature search of MEDLINE, Ovid, CINAHL, and Cochrane for studies reporting on the clinical efficacy and safety profile of gastric ESD.RESULTS: Twenty-nine thousand five hundred and six tumors in 27155 patients(31% female) who underwent gastric ESD between 1999 and 2014 were included in this study. R0 resection rate was 90%(95%CI: 87%-92%) with significant between-study heterogeneity(P < 0.001) which was partly explained by difference in region(P = 0.02) and sample size(P = 0.04). Endoscopic en bloc and curative resection rates were 94%(95%CI: 93%-96%) and 86%(95%CI: 83%-89%) respectively. The rate of immediate and delayed perforation rates were 2.7%(95%CI: 2.1%-3.3%) and 0.39%(95%CI: 0.06%-2.4%) respectively while rates of immediate and delayed major bleeding were 2.9%(95%CI: 1.3-6.6) and 3.6%(95%CI: 3.1%-4.3%). After an average follow-up of about 30 mo post-operative, the rate of tumor recurrence was 0.02%(95%CI: 0.001-1.4) among those with R0 resection and 7.7%(95%CI: 3.6%-16%) among those without R0 resection. 3Species diversity, regeneration and dominance as influenced by canopy gaps and their characteristics in tropical evergreen forests of Western Ghats, India显示文摘Canopy gaps play a significant role in maintaining structure and composition of tropical forests. This study was carried out in tropical evergreen forests of central Western Ghats in India to understand the influence of canopy gap size and the relationship of gap regime attributes to diversity measures and regeneration. The average gap size in the study area was found to be 396 m2 and around half of gaps were 4–8 years old. Gaps created by natural single tree fall were smaller in size but significantly higher in number. Diversity and regeneration of woody species were compared with canopy gaps and intact vegetation. Species richness and diversity was higher in gaps than in intact vegetation. Macaranga peltata, a shade intolerant species dominated gaps while intact vegetation was dominated by shade tolerant Kingiodendron pinnatum.Gap size significantly influenced species diversity and regeneration. Gap area and age were significantly and negatively correlated with diversity measures but positively correlated with regeneration. Among all the attributes of gaps, regeneration was significantly positively correlated with light intensity. Gaps maintained species diversity and favored regeneration of woody species. 4COVID-19: An overview and a clinical update显示文摘The outbreak of coronavirus disease-2019(COVID-19,previously known as 2019 nCoV)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection in Wuhan City,China,has spread rapidly around the world.Most patients from the first cluster had an epidemiological connection to the Wuhan's Huanan Seafood Wholesale Market.Available evidence has shown that SARSCoV-2 can be easily transmitted from person to person through close contact and respiratory droplets,posing a substantial challenge to public health.At present,the research on SARS-CoV-2 is still in the primary stages.However,dexamethasone and remdesivir are appeared to be promising medical therapies.Still,there is no definite specific treatment,and the mainstay of treatment is still focused on supportive therapies.Currently,over 150 vaccines are under investigation.It is necessary to understand the nature of the virus and its clinical characteristics in order to find effectively manage the disease.The knowledge about this virus is rapidly evolving,and clinicians must update themselves regularly.The present review comprehensively summarizes the epidemiology,pathogenesis,clinical characteristics,and management of COVID-19 based on the current evidence.Arunkumar Krishnan James P Hamilton Saleh A Alqahtani Tinsay A Woreta 2021World Journal of Clinical Cases2021,9,1:5
5Impact of poly chlorinated biphenyl (Aroclor 1254) and vitamin C on antioxidant system of rat ventral prostate显示文摘Aim:To evaluate the effect of polychlorinated biphenyls (PCB) and vitamin C on ventral prostatic antioxidant system in adult male rats.Methods:A group of 20 adult male rats were administered ip Aroclor 1254 in corn oil at a dose of 2 mg.kg^-1·day^-1 for 30 days.Ten control rats were administered only the vehicle.After 30 days the treated rats were divided at random into 2 sub-groups of 10 animals each.One sub-group received vitamin C at a dose of 500 mg.kg^-1·day^-1 for 10 days.The other group was maintained as Aroclor 1254 control.Twenty-four hours after the last treatment the rats were killed by decapitation.Ventral prostatic homogenate was prepared and used for the estimation of enzymatic antioxidants,including superoxide dismutase (SOD),catalase (CAT) and glutathione-S-transferase (GST) and hydrogen peroxide (H2O2),lipid peroxidation (LPO) and prostatic acid phosphatase.The serum levels of total T3,total T4,TSH,testosterone and estradiol were also assayed.Results:The body weight and ventral prostatic weight were reduced in PCB treated rats.The activities of SOD,CAT,GST and acid phosphatase were decreased while the levels of H2O2 and lipid peroxidation were increased in the ventral prostate of PCB treated rats.Administration of vitamin C restored these parameters.Serum levels of thyroid hormones,estradiol and testosteron ewere decreased in PCB treated animals.Administration of vitamin C restored the thyroid hormone levels.Conclusion:PCB induces oxidative stress and decreases the antioxidant enzymes in the ventral prostate of adult male rats;the effects could be reversed by the administration of vitamin C.(Asian J Androl 2004 Mar;6:19-22)M. 6Interleukin-6 mediates neutrophil mobilization from bone marrow in pulmonary hypertension显示文摘Myeloid cells,such as neutrophils,are produced in the bone marrow in high quantities and are important in the pathogenesis of vascular diseases such as pulmonary hypertension(PH).Although neutrophil recruitment into sites of inflammation has been well studied,the mechanisms of neutrophil egress from the bone marrow are not well understood.Using computational flow cytometry,we observed increased neutrophils in the lungs of patients and mice with PH.Moreover,we found elevated levels of IL-6 in the blood and lungs of patients and mice with PH.We observed that transgenic mice overexpressing Il-6 in the lungs displayed elevated neutrophil egress from the bone marrow and exaggerated neutrophil recruitment to the lungs,resulting in exacerbated pulmonary vascular remodeling,and dysfunctional hemodynamics.Mechanistically,we found that IL-6-induced neutrophil egress from the bone marrow was dependent on interferon regulatory factor 4(IRF-4)-mediated CX3CR1 expression in neutrophils.Consequently,Cx3cr1 genetic deficiency in hematopoietic cells in Il-6-transgenic mice significantly reduced neutrophil egress from bone marrow and decreased neutrophil counts in the lungs,thus ameliorating pulmonary remodeling and hemodynamics.In summary,these findings define a novel mechanism of IL-6-induced neutrophil egress from the bone marrow and reveal a new therapeutic target to curtail neutrophil-mediated inflammation in pulmonary vascular disease.Jonathan Florentin Jingsi Zhao Yi-Yin Tai Sathish Babu Vasamsetti Scott P.O’Neil Rahul Kumar Anagha Arunkumar Annie Watson John Sembrat Grant C.Bullock Linda Sanders Biruk Kassa Mauricio Rojas Brian B.Graham Stephen Y.Chan Partha Dutta 2021Cellular & Molecular Immunology2021,18,2:3
7Science behind cisplatin-induced nephrotoxicity in humans:A clinical study显示文摘Objective:To investigate the relationship between serum electrolyte changes and cisplatin induced nephrotoxicity.Methods:We collected data from 18 patients undergoing cisplatin chemotherapy including serum electrolytes,creatinine,blood urea nitrogen(BUN) and urine potassium,sodium and pH levels before and after the cisplatin chemotherapy.All the patients had cancer and were treated with 40-50 mg/day cisplatin.Renal injury was assessed by measuring serum electrolytes,creatinine,BUN levels and urine potassium,sodium and pH levels. Results:The five cycles of cisplatin based chemotherapy resulted in hypomagnesia(P=0.029), hypocalcaemia(P=0.001~*),hypophosphatemia(P=0.003*),hypokalemia(P=0.001~*)and increased serum creatinine(P=0.001~*)and BUN(P=0.292~*)levels.In urine analysis,decrease in potassium(P=0.024~*)was found,except potassium there was no significant changes in sodium and urine pH. 10Abnormal liver chemistries as a predictor of COVID-19 severity and clinical outcomes in hospitalized patients显示文摘BACKGROUND Abnormal liver chemistries are common findings in patients with Coronavirus Disease 2019(COVID-19).However,the association of these abnormalities with the severity of COVID-19 and clinical outcomes is poorly understood AIM We aimed to assess the prevalence of elevated liver chemistries in hospitalized patients with COVID-19 and compare the serum liver chemistries to predict the severity and in-hospital mortality.METHODS This retrospective,observational study included 3380 patients with COVID-19 who were hospitalized in the Johns Hopkins Health System(Baltimore,MD,United States).Demographic data,clinical characteristics,laboratory findings,treatment measures,and outcome data were collected.Cox regression modeling was used to explore variables associated with abnormal liver chemistries on admission with disease severity and prognosis RESULTS A total of 2698(70.4%)had abnormal alanine aminotransferase(ALT)at the time of admission.Other more prevalent abnormal liver chemistries were aspartate aminotransferase(AST)(44.4%),alkaline phosphatase(ALP)(16.1%),and total bilirubin(T-Bil)(5.9%).Factors associated with liver injury were older age,Asian ethnicity,other race,being overweight,and obesity.Higher ALT,AST,T-Bil,and ALP levels were more commonly associated with disease severity.Multivariable adjusted Cox regression analysis revealed that abnormal AST and T-Bil were associated with the highest mortality risk than other liver injury indicators during hospitalization.Abnormal AST,T-Bil,and ALP were associated with a need for vasopressor drugs,whereas higher levels of AST,T-Bil,and a decreased albumin levels were associated with mechanical ventilation CONCLUSION Abnormal liver chemistries are common at the time of hospital admission in COVID-19 patients and can be closely related to the patient’s severity and prognosis.Elevated liver chemistries,specifically ALT,AST,ALP,and T-Bil levels,can be used to stratify risk and predict the need for advanced therapies in these patients.Arunkumar Krishnan Laura Prichett Xueting Tao Saleh A Alqahtani James P Hamilton Esteban Mezey Alexandra T Strauss Ahyoung Kim James J Potter Po-Hung Chen Tinsay A Woreta 2022World Journal of Gastroenterology2022,28,5:2
11Alternative splicing enhances transcriptome complexity in desiccating seeds显示文摘Before being dispersed in the environment, mature seeds need to be dehydrated. The survival of seeds after dispersal depends on their low hydration in combination with high desiccation tolerance. These characteristics are established during seed maturation. Some key seed maturation genes have been reported to be regulated by alternative splicing(AS). However, so far AS was described only for single genes and a comprehensive analysis of AS during seed maturation has been lacking. We investigated gene expression and AS during Arabidopsis thaliana seed development at a global level, before and after desiccation. Bioinformatics tools were developed to identify differentially spliced regions within genes. Our data suggest the importance and shows the peculiar features of AS during seed desiccation. We identified AS in 34% of genes that are expressed at both timepoints before and after desiccation. Most of these AStranscript variants had not been found before in other tissues.Among the AS genes some seed master regulators could be found. Interestingly, 6% of all expressed transcripts were not transcriptionally regulated during desiccation, but only modified by AS. We propose that AS should be more routinely taken into account in the analysis of transcriptomic data to prevent overlooking potentially important regulators.Arunkumar Srinivasan Jose M. 12Vascular-confined multi-passage discoidal nanoconstructs for the low-dose docetaxel inhibition of triple-negative breast cancer growth显示文摘Taxane efficacy in triple negative breast cancer(TNBC)is limited by insufficient tumor accumulation and severe off-target effects.Nanomedicines offer a unique opportunity to enhance the anti-cancer potency of this drug.Here,1,000 nm×400 nm discoidal polymeric nanoconstructs(DPN)encapsulating docetaxel(DTXL)and the near infrared compound Iipid-Cy5 were engineered.DPN were obtained by filling multiple times cylindrical wells in a poly(vinyl alcohol)template with a polymer mixture comprising poly(lactic-co-glycolic acid)(PLGA)and poly(ethylene glycol)diacrylate(PEG-DA)chains together with therapeutic and imaging agents.The resulting“multi-passage”DPN exhibited higher DTXL loading,Iipid-Cy5 stability,and stiffness as compared to the conventional'single-passage'approach.Confocal microscopy confirmed that DTXL-DPN were not taken up by MDA-MB-231 cells but would rather sit next to the cell membrane and slowly release DTXL thereof.Empty DPN had no toxicity on TNBC cells,whereas DTXL-DPN presented a cytotoxic potential comparable to free DTXL(IC_(50)=2.6 nM±1.0 nM vs.7.0 nM±1.09 nM at 72 h).In orthotopic murine models,DPN accumulated in TNBC more efficiently than free-DTXL.With only 2 mg/kg DTXL,intravenously administered every 2 days for a total of 13 treatments,DTXL-DPN induced tumor regression and were associated to an overall 80%survival rate as opposed to a 30%survival rate for free-DTXL,at 120 days.All untreated mice succumbed before 90 days.Collectively,this data demonstrates that vascular confined multi-passage DPN,biomimicking the behavior of circulating platelets,can efficiently deliver chemotherapeutic molecules to malignant tissues and effectively treat orthotopic TNBC at minimal taxane doses.Alessia Felici Daniele Di Mascolo Miguel Ferreira Simone Lauciello Luca Bono Andrea Armirotti Arunkumar Pitchaimani Anna Lisa Palange Paolo Decuzzi 2022Nano Research2022,15,1:2

17MELD or MELD-Na as a Predictive Model for Mortality Following Transjugular Intrahepatic Portosystemic Shunt Placement显示文摘Background and Aim:The model for end-stage liver disease(MELD)was originally developed to predict survival after transjugular intrahepatic portosystemic shunt(TIPS).The MELD-sodium(MELD-Na)score has replaced MELD for organ allocation for liver transplantation.However,there are limited studies to compare the MELD with MELD-Na to predict mortality after TIPS.Methods:We performed a retrospective chart review of patients who underwent TIPS placement between 2006 and 2016 at our institution.The primary outcome was mortality,and the secondary outcomes sought to assess which variables could provide prognostic information for mortality after TIPS placement.We performed receiver operating characteristic(ROC)curve analysis to assess the performance of MELD and MELD-Na.Results:There were 186 eligible patients in the analysis.The mean pre-TIPS MELD and MELD-Na were 13 and 15,respectively.Overall,mortality after TIPS was 15%at 30 days and 16.7%at 90 days.In a comparison of the areas under the ROCs for MELD and MELD-Na,MELD was superior to MELD-Na for 30-day(0.762 vs.0.709)and 90-day(0.780 vs.0.730)mortality after TIPS.The optimal cutoff score for 30-day mortality was 15(0.676–0.848)for MELD and 17(0.610–0.808)for MELD-Na,whereas the optimal cutoff score for 90-day mortality was 16(95%CI:0.705–0.855)for MELD and 17(95%CI:0.643–0.817)for MELDNa.There were 24 patients with high MELD-Na≥17,but with low MELD<15,and 90-day mortality in this group was 8.3%.Conclusions:Although MELD-Na is a superior prognostic tool to MELD for predicting overall mortality in cirrhotic patients,MELD tended to outperform MELD-Na to predict mortality after TIPS.Arunkumar Krishnan Tinsay A.Woreta Dhananjay Vaidya Yisi Liu James P.Hamilton Kelvin Hong Alia Dadabhai Michelle Ma 2023Journal of Clinical and Translational Hepatology2023,11,1:1
