| 1 | Development and characterization of an atorvastatin solid dispersion formulation using skimmed milk for improved oral bioavailability显示文摘Atorvastatin has low aqueous solubility resulting in low oral bioavailability(12%)and thus presents a challenge in formulating a suitable dosage form.To improve the aqueous solubility,a solid dispersion formulation of atorvasta tin was prepared by lyophilization utilising skimmed milk as a carrier.Six different formulations were prepared with varying ratios of drug and carrier and the corresponding physical mixtures were also prepared.The formation of a solid dispersion formulation was confirmed by differential scanning ca lorimetry and X-ray diffraction studies.The optimum drug-to-carrier ratio of 1:9 en hanced solubility nearly 33-fold as compared to pure drug.In vitro drug release studies exhibited a cumulative release of 83.69% as compared to 22.7% for the pure drug.Additionally,scanning electron microscopy studies suggested the conversion of crystalline atorvastatin to an amorphous form.In a Triton-induced hyperlipidemia model,a 3-fold increase in the lipid lowering potential was obtained with the reformulated drug as compared to pure drug.These results suggest that solid dispersion of atorvastatin using skimmed milk as carrier is a promising approach for oral delivery of atorvastatin. | Ankush Choudhary Avtar C.Rana Geeta Aggarwal Virender Kumar Foziyah Zakir | 2012 | Acta Pharmaceutica Sinica B2012，2，4： | 10 |
| 2 | Experimental investigation and optimization of weld bead characteristics during submerged arc welding of AISI 1023 steel显示文摘Submerged arc welding(SAW), owing to its high deposition rate and high welding quality, is widely used in the fabrication of pressure vessel, marine vessel, pipelines and offshore structures. However, selection of an optimum combination of welding parameters is critical in achieving high weld quality and productivity. In this work, initially, the SAW experiments were performed using fractional factorial design to analyze the effect of direct and indirect input parameters, namely, welding voltage, wire feed rate,welding speed, nozzle to plate distance, flux condition, and plate thickness on weld bead geometrical responses viz. bead width, reinforcement, and penetration. The bead on plate technique was used to deposit weld metal on AISI 1023 steel plates. The effect of SAW input parameters on response variables were analyzed using main and interaction effects. The linear regression was used to develop the mathematical models for the response variable. Then, the multi-objective optimization of input parameters was carried out using desirability approach, genetic algorithm and Jaya algorithm. | 3 | Hepatoprotective and antioxidant activity of Bombax ceiba flowers against carbon tetrachloride-induced hepatotoxicity in rats显示文摘Aim:The flowers of Bombax ceiba are traditionally used as home remedy in the treatment of jaundice and spleen enlargement.The present work investigated the effect of aqueous extract of flowers of Bombax ceiba(BCAE)on experimentally induced hepatotoxicity in rats to substantiate its traditional use as hepatoprotective agent.Methods:Hepatotoxicity was induced in rats by carbon tetrachloride(CCl_(4))treatment;at the same time vehicle or BCAE(250 or 500 mg/kg)or silymarin(25 mg/kg)were administered daily orally for seven days.Hepatotoxicity was assessed by estimating the activities of marker enzymes and by histological studies.The antioxidant effect of BCAE was assessed by measuring amount of antioxidant phytochemicals(total phenolics and flavonoids),and DPPH free radical scavenging assay of the extract.Results:BCAE treatment significantly prevented the CCl_(4)-induced elevations in levels of glutamate oxaloacatate transaminase,glutamic pyruvic transaminase,alkaline phosphatase,bilirubin,and triglycerides,and decreased the total protein levels.Treatment with BCAE attenuated the CCl_(4)-induced cytotoxic damage to liver.BCAE exhibited presence of antioxidant phytochemicals and showed scavanging action on DPPH radicals.The hepatoprotective effect of BCAE was comparable to that of the standard antioxidant hepatoprotective agent,silymarin.These findings indicated that BCAE showed hepatoprotective effect against CCl_(4)-induced hepatotoxicity and exhibited in vitro antioxidant effects.Conclusion:Bombax ceiba flowers exhibited hepatoprotective effect which may be attributed to antioxidant potential.This study also validated their traditional medicinal use in liver disorders. | Manish M.Wanjari Rachna Gangoria Yadu Nandan Dey Sudesh N.Gaidhani Narendra K.Pandey Ankush D.Jadhav | 2016 | Hepatoma Research2016，2，1： | 3 |
| 4 | Superior safety of direct oral anticoagulants compared to Warfarin in patients with atrial fibrillation and underlying cancer: a national veterans affairs database study显示文摘Background Studies evaluating safety of warfarin and direct oral anticoagulants(DOACs) for prevention of stroke in patients with atrial fibrillation(AF) are lacking. Methods & Results All patients(n = 196,521) receiving care at veteran’s affairs with active cancer and AF from 2010–2015 were included. One-year mortality was significantly higher in unadjusted analysis with warfarin(44.9%) compared to dabigatran(25%, P < 0.001), rivaroxaban(24.4%, P < 0.001) and apixaban(30%, P < 0.001) and after adjusting for age, sex and type of cancer mortality(OR = 2.66, 95% CI: 2.52–2.82, P < 0.001). Risk of ischemic stroke(13.5% vs. 11.1%, 12.0%, 14.0%) was similar, however risk of hemorrhagic stroke was significantly higher among patients receiving warfarin(1.2%) compared to patients receiving dabigatran(0.5%), rivaroxaban(0.7%) and apixaban(0.8%) respectively, P = 0.04. Conclusions We demonstrated the superior safety profile of DOACs compared to warfarin among patients with underlying cancer and AF.
