1Antibiotic resistance and cagA gene correlation:A looming crisis of Helicobacter pylori显示文摘AIM:To determine antibiotic resistance of Helicobacter pylori(H.pylori) in Pakistan and its correlation with host and pathogen associated factors.METHODS:A total of 178 strains of H.pylori were isolated from gastric biopsies of dyspeptic patients.Susceptibility patterns against first and second-line antibiotics were determined and trends of resistance were analyzed in relation to the sampling period,gastric conditions and cagA gene carriage.The effect of cagA gene on the acquisition of resistance was investigated by mutant selection assay.RESULTS:The observations showed that monoresistant strains were prevalent with rates of 89% for metronidazole,36% for clarithromycin,37% for amoxicillin,18.5% for ofloxacin and 12% for tetracycline.Furthermore,clarithromycin resistance was on the rise from 2005 to 2008(32% vs 38%,P = 0.004) and it is significantly observed in non ulcerative dyspeptic patients compared to gastritis,gastric ulcer and duodenal ulcer cases(53% vs 20%,18% and 19%,P = 0.000).On the contrary,metronidazole and ofloxacin resistance were more common in gastritis and gastric ulcer cases.Distribution analysis and frequencies of resistant mutants in vitro correlated with the absence of cagA gene with metronidazole and ofloxacin resistance.CONCLUSION:The study confirms the alarming levels of antibiotic resistance associated with the degree of gastric inflammation and cagA gene carriage in H.pylori strains.Adnan Khan Amber Farooqui Hamid Manzoor Syed Shakeel Akhtar Muhammad Saeed Quraishy Shahana Urooj Kazmi 2012World Journal of Gastroenterology2012,18,18:15
2PBS3 and EPS1 Complete Salicylic Acid Biosynthesis from Isochorismate in Arabidopsis显示文摘Salicylic acid(SA)is an important phytohormone mediating both local and systemic defense responses in plants.Despite over half a century of research,how plants biosynthesize SA remains unresolved.In Arabidop-sis,a major part of SA is derived from isochorismate,a key intermediate produced by the isochorismate syn-thase,which is reminiscent of SA biosynthesis in bacteria.Whereas bacteria employ an isochorismate pyru-vate lyase(IPL)that catalyzes the turnover of isochorismate to pyruvate and SA,plants do not contain an IPL ortholog and generate SA from isochorismate through an unknown mechanism.Combining genetic and biochemical approaches,we delineated the SA biosynthetic pathway downstream of isochorismate in Ara-bidopsis.We found that PBS3,a GH3 acyl adenylase-family enzyme important for SA accumulation,catalyzes ATP-and Mg2+-dependent conjugation of L-glutamate primarily to the 8-carboxyl of isochorismate and yields the key SA biosynthetic intermediate,isochorismoyl-glutamate A.Moreover,we discovered that EPS1,a BAHD acyltransferase-family protein with a previously implicated role in SA accumulation upon pathogen attack,harbors a noncanonical active site and an unprecedented isochorismoyl-glutamate A pyruvoyl-glutamate lyase activity that produces SA from the isochorismoyl-glutamate A substrate.Together,PBS3 and EPS1 form a two-step metabolic pathway to produce SA from isochorismate in Arabidopsis,which is distinct from how SA is biosynthesized in bacteria.This study closes a major knowledge gap in plant SA meta-bolism and would help develop new strategies for engineering disease resistance in crop plants.Michael P.Torrens-Spence Anastassia Bobokalonova Valentina Carballo Christopher MGlinkerman Toma(s)Pluskal Amber Shen Jing-Ke Weng 2019Molecular Plant2019,12,12:10

6Interprofessional, multiple step simulation course improves pediatric resident and nursing staff management of pediatric patients with diabetic ketoacidosis显示文摘AIM To investigate the use of a multidisciplinary, longitudinal simulation to educate pediatric residents and nurses on management of pediatric diabetic ketoacidosis.METHODS A multidisciplinary, multiple step simulation course was developed by faculty and staff using a modified Delphi method from the Pediatric Simulation Center and pediatric endocrinology department. Effectiveness of the simulation for the residents was measured with a pre- and post-test and a reference group not exposed to simulation. A follow up post-test was completed 3-6 mo after the simulation. Nurses completed a survey regarding the education activity. RESULTS Pediatric and medicine-pediatric residents(n = 20) and pediatric nurses(n = 25) completed the simulation course. Graduating residents(n = 16) were used as reference group. Pretest results were similar in the control and intervention group(74% ± 10% vs 76% ± 15%, P = 0.658). After completing the intervention, participants improved in the immediate post-test in comparison to themselves and the control group(84% ± 12% post study; P < 0.05). The 3-6 mo follow up post-test results demonstrated knowledge decay when compared to their immediate post-test results(78% ± 14%, P = 0.761). 7Single vs dual(en bloc) kidney transplants from donors ≤ 5 years of age: A single center experience显示文摘AIM: To compare outcomes between single and dual en bloc(EB) kidney transplants(KT) from small pediatric donors. METHODS: Monocentric nonprospective review of KTs from pediatric donors ≤ 5 years of age. Dual EB KT was defined as keeping both donor kidneys attached tothe inferior vena cava and aorta, which were then used as venous and arterial conduits for the subsequent transplant into a single recipient. Donor age was less useful than either donor weight or kidney size in decision-making for kidney utilization as kidneys from donors < 8 kg or kidneys < 6 cm in length were not transplanted. Post-transplant management strategies were standardized in all patients.RESULTS: From 2002-2015, 59 KTs were performed including 34 dual EB and 25 single KTs. Mean age of donors(17 mo vs 38 mo, P < 0.001), mean weight(11.0 kg vs 17.4 kg, P = 0.046) and male donors(50% vs 84%, P = 0.01) were lower in the dual EB compared to the single KT group, respectively. Mean cold ischemia time(21 h), kidney donor profile index(KDPI; 73% vs 62%) and levels of serum creatinine(SCr, 0.37 mg/d L vs 0.49 mg/d L, all P = NS) were comparable in the dual EB and single KT groups, respectively. Actuarial graft and patient survival rates at 5-years follow-up were comparable. There was one case of thrombosis resulting in graft loss in each group. Delayed graft function incidence(12% dual EB vs 20% single KT, P = NS) was slightly lower in dual EB KT recipients. Initial duration of hospital stay(mean 5.4 d vs 5.6 d) and the one-year incidences of acute rejection(6% vs 16%), operative complications(3% vs 4%), and major infection were comparable in the dual EB and single KT groups, respectively(all P = NS). Mean 12 mo SCr and abbreviated MDRD levels were 1.17 mg/d L vs 1.35 mg/d L and 72.5 m L/min per 1.73 m^2 vs 60.5 m L/min per 1.73 m^2(both P = NS) in the dual EB and single KT groups, respectively. 8Temporal changes in inflammatory mitochondria-enriched microRNAs following traumatic brain injury and effects of miR-146a nanoparticle delivery显示文摘MicroRNAs(miRNAs)are small non-coding RNA molecules that regulate post-transcriptional gene expression and contribute to all aspects of cellular function.We previously reported that the activities of several mitochondria-enriched miRNAs regulating inflammation(i.e.,miR-142-3p,miR-142-5p,and miR-146a)are altered in the hippocampus at 3–12 hours following a severe traumatic brain injury.In the present study,we investigated the temporal expression profile of these inflammatory miRNAs in mitochondria and cytosol fractions at more chronic post-injury times following severe controlled cortical impact injury in rats.In addition,several inflammatory genes were analyzed in the cytosol fractions.The analysis showed that while elevated levels were observed in cytoplasm,the mitochondria-enriched miRNAs,miR-142-3p and miR-142-5p continued to be significantly reduced in mitochondria from injured hippocampi for at least 3 days and returned to near normal levels at 7 days post-injury.Although not statistically significant,miR-146a also remained at reduced levels for up to 3 days following controlled cortical impact injury,and recovered by 7 days.In contrast,miRNAs that are not enriched in mitochondria,including miR-124a,miR-150,miR-19b,miR-155,and miR-223 were either increased or demonstrated no change in their levels in mitochondrial fractions for 7 days.The one exception was that miR-223 levels were reduced in mitochondria at 1 day following injury.No major alterations were observed in sham operated animals.This temporal pattern was unique to mitochondria-enriched miRNAs and correlated with injury-induced changes in mitochondrial bioenergetics as well as expression levels of several inflammatory markers.These observations suggested a potential compartmental re-distribution of the mitochondria-enriched inflammatory miRNAs and may reflect an intracellular mechanism by which specific miRNAs regulate injury-induced inflammatory signaling.To test this,we utilized a novel peptide-based nanoparticle strategy for in vitro and in vivo delivery of a miR-146a mimic as a potential therapeutic strategy for targeting nuclear factor-kappa B inflammatory modulators in the injured brain.Nanoparticle delivery of miR-146a to BV-2 or SH-SY5Y cells significantly reduced expression of TNF receptor-associated factor 6(TRAF6)and interleukin-1 receptor-associated kinase 1(IRAK1),two important modulators of the nuclear factor-kappa B(NF-κB)pro-inflammatory pathway.Moreover,injections of miR-146a containing nanoparticles into the brain immediately following controlled cortical impact injury significantly reduced hippocampal TNF receptor-associated factor 6 and interleukin-1 receptor-associated kinase 1 levels.Taken together,our studies demonstrate the subcellular alteration of inflammatory miRNAs after traumatic brain injury and establish proof of principle that nanoparticle delivery of miR-146a has therapeutic potential for modulating pro-inflammatory effectors in the injured brain.All of the studies performed were approved by the University of Kentucky Institutional Animal Care and Usage Committee(IACUC protocol#2014-1300)on August 17,2017.Wang-Xia Wang Paresh Prajapati Hemendra J.Vekaria Malinda Spry Amber L.Cloud Patrick G.Sullivan Joe E.Springer 2021Neural Regeneration Research2021,16,3:3

10Creation and judicious application of a wheat resistance gene atlas显示文摘Disease-resistance(R)gene cloning in wheat(Triticum aestivum)has been accelerated by the recent surge of genomic resources,facilitated by advances in sequencing technologies and bioinformatics.However,with the challenges of population growth and climate change,it is vital not only to clone and functionally characterize a few handfuls of R genes,but also to do so at a scale that would facilitate the breeding and deployment of crops that can recognize the wide range of pathogen effectors that threaten agroecosystems.Pathogen populations are continually changing,and breeders must have tools and resources available to rapidly respond to those changes if we are to safeguard our daily bread.To meet this challenge,we propose the creation of a wheat R-gene atlas by an international community of researchers and breeders.The atlas would consist of an online directory from which sources of resistance could be identified and deployed to achieve more durable resistance to the major wheat pathogens,such as wheat rusts,blotch diseases,powdery mildew,and wheat blast.We present a costed proposal detailing how the inter-acting molecular components governing disease resistance could be captured from both the host and the pathogen through biparental mapping,mutational genomics,and whole-genome association genetics.We explore options for the configuration and genotyping of diversity panels of hexaploid and tetraploid wheat,as well as their wild relatives and major pathogens,and discuss how the atlas could inform a dynamic,durable approach to R-gene deployment.Set against the current magnitude of wheat yield losses worldwide,recently estimated at 21%,this endeavor presents one route for bringing R genes from the lab to the field at a considerable speed and quantity.Amber N.Hafeez Sanu Arora Sreya Ghosh David Gilbert Robert L.Bowden Brande B.H.Wulff 2021Molecular Plant2021,14,7:3

12UVRAG基因缺失促进饥饿诱导的肝脏脂肪变性显示文摘[目的]紫外线辐射耐受相关(ultraviolet resistance-associated gene,UVRAG)是一个自噬相关基因,具有多种生物学功能.利用UVRAG基因缺失的小鼠证实了该基因缺失可阻断体内自噬流,并鉴于自噬对脂代谢有一定调控作用,研究了UVRAG在饥饿诱导的肝脏脂肪变性中的作用.[方法与结果]野生型小鼠和UVRAG基因缺失小鼠给予正常饮食或24 h饥饿处理.在正常生理条件下,油红染色显示UVARG基因缺失小鼠和野生型小鼠肝脏无明显脂质积聚;但是在24 h饥饿条件下,该基因缺失可导致肝脏脂质积聚显著增多,且UVRAG基因缺失小鼠肝脏甘油三酯、游离脂肪酸质量比显著升高,而总胆固醇质量比无显著性差异.实时荧光定量聚合酶链式反应(reverse transcription-polymerize chain reaction,RT-PCR)显示,在正常生理条件下的胆固醇调节元件结合蛋白-1(sterol regulatory element-binding protein1,SREBP-1)基因在UVRAG基因缺失小鼠肝脏表达显著下调,但在饥饿条件下,UVARG基因缺失小鼠和野生型小鼠肝脏SREBP-1表达无显著差异;而在实验组和对照组中其他肝脏脂代谢相关基因CD 36,FAS,CPT-1,PPAR和FGF 21的表达无显著性变化.此外,UVARG基因缺失对脂代谢调控分子AMPK活性无显著影响.在饥饿条件下,UVRAG基因缺失可导致血浆甘油三酯和游离脂肪酸浓度上升,血糖浓度下降.[结论]UVARAG基因缺失促进饥饿诱导的肝脏脂肪变性,这可能与肝脏自噬阻断和肝脏摄取血浆游离脂肪酸增多有关.胡晓雯 张沙沙 安琳 AMBER Naz 朱洪新 2018上海大学学报(自然科学版)2018,24,3:2

14Predicting(side) effects for patients with inflammatory bowel disease: The promise of pharmacogenetics显示文摘Inflammatory bowel disease (IBD) is a chronic and heterogeneous intestinal inflammatory disorder. The medical management of IBD aims for long-lasting disease remission to prevent complications and disease progression. Early introduction of immunosuppression forms the mainstay of medical IBD management. Large inter-individual variability in drug responses, in terms of both efficacy and toxicity, leads to high rates of therapeutic failure in the management of IBD. Better patient stratification is needed to maximize patient benefit and minimize the harm caused by adverse events. Pre-treatment pharmacogenetic testing has the potential to optimize drug selection and dose, and to minimize harm caused by adverse drug reactions. In addition, optimizing the use of cheap conventional drugs, and avoiding expensive ineffective drugs, will lead to a significant reduction in costs. Genetic variation in both TPMT and NUDT15, genes involved in thiopurine metabolism, is associated to an increased risk of thiopurine-induced myelosuppression. Moreover, specific HLA haplotypes confer risk to thiopurine-induced pancreatitis and to immunogenicity to tumor necrosis factor-antagonists, respectively. Falling costs and increased availability of genetic tests allow for the incorporation of pre-treatment genetic tests into clinical IBD management guidelines. 15Green Design Studio: A modular-based approach for high-performance building design显示文摘A modular-based Green Design Studio(GDS)platform has been developed in this study for fast and accurate performance analysis for early stage green building design.The GDS platform aims to simplify the design and analysis process by embedding performance parameters into design elements in modules and employing near-real-time model for whole building performance simulation as well as by providing an easy-to-use and intuitive user interface to assist users without extensive knowledge on building physics.The platform consists of building modules as fundamental building blocks,performance predicting models,and a user interface for visualization and interactive design.In the platform,a whole building is composed of modules organized in a hierarchical structure,including spaces,enclosures,service systems,sustainable resource systems and sites.Both physics-based and data-driven models can be used to simulate the building performance and optimize building systems.A simplified physics-based model,the Resistance–Capacitance(RC)model,has been proposed as a generic simulation model for the flows of heat,air,moisture and pollutants,which is significantly faster than conventional simulation tools such as EnergyPlus,and hence more practical for use in real-time design interaction and optimization.A pilot case study is conducted to illustrate the modular-based design approach using a section of an office building.Compared to conventional building performance analysis tools,the GDS platform can provide fast and reliable feedback on performance prediction for early design.The modular approach makes it easier to modify the building design and evaluate the potentials and contributions of various green design features and technologies.Jialei Shen Bess Krietemeyer Amber Bartosh Zhi Gao Jianshun Zhang 2021Building Simulation2021,14,2:2

19Myelin repair and functional recovery mediated by neural cell transplantation in a mouse model of multiple sclerosis显示文摘Cellular therapies are becoming a major focus for the treatment of demyelinating diseases such as multiple sclerosis (MS), therefore it is important to identify the most effective cell types that promote myelin repair. Several components contribute to the relative benefits of specific cell types including the overall efficacy of the cell therapy, the reproducibility of treatment, the mechanisms of action of distinct cell types and the ease of isolation and generation of therapeutic populations. A range of distinct cell populations promote functional recovery in animal models of MS including neural stem cells and mesenchymal stem cells derived from different tissues. Each of these cell populations has advantages and disadvantages and likely works through distinct mechanisms. The relevance of such mechanisms to myelin repair in the adult central nervous system is unclear since the therapeutic cells are generally derived from developing animals. Here we describe the isolation and characterization of a population of neural cells from the adult spinal cord that are characterized by the expression of the cell surface glycoprotein NG2. In functional studies, injection of adult NG2^+ cells into mice with ongoing MOG_35-55-induced experimental autoimmune encephalomyelitis (EAE) enhanced remyelination in the CNS while the number of CD3^+ T cells in areas of spinal cord demyelination was reduced approximately three-fold. In vivostudies indicated that in EAE, NG2^+ cells stimulated endogenous repair while in vitro they responded to signals in areas of induced inflammation by differentiating into oligodendrocytes.
