| 1 | Interleukin-12 as a Genetic Adjuvant Enhances Hepatitis C Virus NS3 DNA Vaccine Immunogenicity显示文摘Hepatitis C virus (HCV) chronic infection is a worldwide health problem, and numerous efforts have been invested to develop novel vaccines. An efficient vaccine requires broad immune response induction against viral proteins. To achieve this goal, we constructed a DNA vaccine expressing nonstructural 3 (NS3) gene (pcDNA3.1-HCV-NS3) and assessed the immune response in C57BL/6 mice. In this study, the NS3 gene was amplified with a nested-reverse transcriptase-polymerase chain reaction (RT-PCR) method using sera of HCV-infected patients with genotype 1a. The resulting NS3 gene was subcloned into a pcDNA3.1 eukaryotic expression vector, and gene expression was detected by western blot. The resultant DNA vaccine was co-administered with interleukin-12 (IL-12) as an adjuvant to female C57BL/6 mice. After the final immunizations, lymphocyte proliferation, cytotoxicity, and cytokine levels were assessed to measure immune responses. Our data suggest that co-administration of HCV NS3 DNA vaccine with IL-12 induces production of significant levels of both IL-4 and interferon (IFN)-γ (p<0.05). Cytotoxicity and lymphocyte proliferation responses of vaccinated mice were significantly increased compared to control (p<0.05). | 2 | Assessing the performance of decision tree and neural network models in mapping soil properties显示文摘To build any spatial soil database, a set of environmental data including digital elevation model(DEM) and satellite images beside geomorphic landscape description are essentials. Such a database, integrates field observations and laboratory analyses data with the results obtained from qualitative and quantitative models. So far, various techniques have been developed for soil data processing. The performance of Artificial Neural Network(ANN) and Decision Tree(DT) models was compared to map out some soil attributes in Alborz Province, Iran. Terrain attributes derived from a DEM along with Landsat 8 ETM+, geomorphology map, and the routine laboratory analyses of the studied area were used as input data. The relationships between soil properties(including sand, silt, clay, electrical conductivity, organic carbon, and carbonates) and the environmental variables were assessed using the Pearson Correlation Coefficient and Principle Components Analysis. Slope, elevation, geomforms, carbonate index, stream network, wetness index, and the band’s number 2, 3, 4, and 5 were the most significantly correlated variables. ANN and DT did not show the same accuracy in predicting all parameters. The DT model showed higher performances in estimating sand(R^2=0.73), silt(R^2=0.70), clay(R^2=0.72), organic carbon(R^2=0.71), and carbonates(R^2=0.70). While the ANN model only showed higher performance in predicting soil electrical conductivity(R^2=0.95). The results showed that determination the best model to use, is dependent upon the relation between the considered soil properties with the environmental variables. However, the DT model showed more reasonable results than the ANN model in this study. The results showed that before using a certain model to predict variability of all soil parameters, it would be better to evaluate the efficiency of all possible models for choosing the best fitted model for each property. | 4 | Comparing the deactivation behaviour of Co/CNT and Co/γ-Al_2O_3 nano catalysts in Fischer-Tropsch synthesis显示文摘An extensive study of Fischer-Tropsch (FT) synthesis on cobalt nano particles supported on γ-alumina and carbon nanotubes (CNTs) catalysts is reported.20 wt% of cobalt is loaded on the supports by impregnation method.The deactivation of the two catalysts was studied at 220 C,2 MPa and 2.7 L/h feed flow rate using a fixed bed micro-reactor.The calcined fresh and used catalysts were characterized extensively and different sources of catalyst deactivation were identified.Formation of cobalt-support mixed oxides in the form of xCoO yAl2O3 and cobalt aluminates formation were the main sources of the Co/γ-Al2O3 catalyst deactivation.However sintering and cluster growth of cobalt nano particles are the main sources of the Co/CNTs catalyst deactivation.In the case of the Co/γ-Al2O3 catalyst,after 720 h on stream of continuous FT synthesis the average cobalt nano particles diameter increased from 15.9 to 18.4 nm,whereas,under the same reaction conditions the average cobalt nano particles diameter of the Co/CNTs increased from 11.2 to 17.8 nm.Although,the initial FT activity of the Co/CNTs was 26% higher than that of the Co/γ-Al2O3,the FT activity over the Co/CNTs after 720 h on stream decreased by 49% and that over the Co/γ-Al2O3 by 32%.For the Co/γ-Al2O3 catalyst 6.7% of total activity loss and for the Co/CNTs catalyst 11.6% of total activity loss cannot be recovered after regeneration of the catalyst at the same conditions of the first regeneration step.It is concluded that using CNTs as cobalt catalyst support is beneficial in carbon utilization as compared to γ-Al2O3 support,but the Co/CNTs catalyst is more susceptible for deactivation. | Ahmad Tavasoli Saba Karimi Somayeh Taghavi Zahra Zolfaghari Hamideh Amirfirouzkouhi | 2012 | Journal of Natural Gas Chemistry2012,21,5: | 4 |
| 5 | Enhancement of bimetallic Fe-Mn/CNTs nano catalyst activity and product selectivity using microemulsion technique显示文摘Bimetallic Fe-Mn nano catalysts supported on carbon nanotubes(CNTs) were prepared using microemulsion technique with water-to-surfactant ratios of 0.4-1.6. The nano catalysts were extensively characterized by different methods and their activity and selectivity in Fischer-Tropsch synthesis(FTS) have been assessed in a fixed-bed microreactor. The physicochemical properties and performance of the nanocatalysts were compared with the catalyst prepared by impregnation method. Very narrow particle size distribution has been produced by the microemulsion technique at relatively high loading of active metal. TEM images showed that small metal nano particles in the range of 3–7 nm were not only confined inside the CNTs but also located on the outer surface of the CNTs. Using microemulsion technique with water to surfactant ratio of0.4 decreased the average iron particle sizes to 5.1 nm. The reduction percentage and dispersion percentage were almost doubled. Activity and selectivity were found to be dependent on the catalyst preparation method and average iron particle size. CO conversion and FTS rate increased from 49.1% to 71.0% and 0.144 to 0.289 gHC/(gcat h), respectively. While the WGS rate decreased from 0.097 to 0.056 gCO2/(gcat h). | 8 | Soil Atterberg Limits and Consistency Indices as Influenced by Land Use and Slope Position in Western Iran显示文摘Atterberg limits and consistency indices are used for classifications of cohesive(fine-grained) soils in relation with compaction and tillage practices. They also provide information for interpreting several soil mechanical and physical properties such as shear strength, compressibility, shrinkage and swelling potentials. Although, several studies have been conducted regarding the land use effects on various soil mechanical properties, little is known about the effects of land use and slope positions on Atterberg limits and consistency indices. This study was conducted to investigate the effects of land use and slope position on selected soil physical and chemical properties, Atterberg limits and consistency indices in hilly region of western Iran. Three land uses including dryland farming, irrigated farming and pasture and four slope positions(i.e., shoulder, backslope, footslope, and toeslope) were used for soil samplings. One hundred eleven soil samples were collected from the surface soil(0-10 cm). Selected physical and chemical properties, liquid limit(LL), plastic limit(PL) and shrinkage limit(SL) were measured using the standard methods; and consistency indices including plastic index(PI), friability index(FI), shrinkage index(SI) and soil activity(A=PI/clay) were calculated. The results showed that irrigated farming significantly increased organic matter content(OM) and OM/clay ratio, and decreased bulk density(ρb) and relative bulk density(ρb-rel) as a result of higher biomass production and plant residues added to the soil compared to other land uses. Except for sand content, OM, ρb, cation exchange capacity(CEC) and calcium carbonate equivalent(CCE), slope position significantly affected soil physical and chemical properties. The highest values of silt, OM/clay and CEC/clay were found in the toeslope position, predominantly induced by soil redistribution within the landscape. The use of complexed(COC)- noncomplexed organic carbon(NCOC) concept indicated that majority of the studied soils were located below the saturation line and the OM in the soils was mainly in the COC form. The LL, PI, FI and A showed significant differences among the land uses; the highest values belonged to the irrigated farming due to high biomass production and plant residues returned to the soils. Furthermore, slope position significantly affected the Atterberg limits and consistency indices except for SL. The highest values of LL, PI, SI and A were observed in the toeslope position probably because of higher OM and CEC/clay due to greater amount of expandable phyllosilicate clays. | 9 | Investigation on removal pathways of Di 2-ethyl hexyl phthalate from synthetic municipal wastewater using a submerged membrane bioreactor显示文摘Highly hydrophobic Di 2-ethyl hexyl phthalate(DEHP) is one of the most prevalent plasticizers in wastewaters. Since its half-life in biological treatment is around 25 days, it can be used as an efficiency indicator of wastewater treatment plant for the removal of hydrophobic emerging contaminants. In this study, the performance of submerged membrane bioreactor was monitored to understand the effect of DEHP on the growth of aerobic microorganisms. The data showed that the chemical oxygen demand(COD)and ammonia concentration were detected below 10 and 1.0 mg/L, respectively for operating conditions of hydraulic retention time(HRT) = 4 and 6 hr, sludge retention time(SRT) = 140 day and sludge concentration between 11.5 and 15.8 g volatile solid(VS)/L. The removal efficiency of DEHP under these conditions was higher and ranged between 91% and 98%. Results also showed that the removal efficiency of DEHP in biological treatment depended on the concentration of sludge, as adsorption is the main mechanism of its removal. For the submerged membrane bioreactor, the pore size is the pivotal factor for DEHP removal, since it determines the amount of soluble microbial products coming out of the process. Highly assimilated microorganisms increase the biodegradation rate, as 74% of inlet DEHP was biodegraded; however, the concentration of DEHP inside sludge was beyond the discharge limit.
