1Unraveling a genetic roadmap for improved taste in the domesticated apple显示文摘Although taste is an important aspect of fruit quality, an understanding of its genetic control remains elusive in apple and other fruit crops. In this study, we conducted genomic sequence analysis of 497 Malus accessions and revealed erosion of genetic diversity caused by apple breeding and possible independent domestication events of dessert and cider apples. Signatures of selection for fruit acidity and size, but not for fruit sugar content, were detected during the processes of both domestication and improvement. Furthermore, we found that single mutations in major genes affecting fruit taste, including Ma1, MdTDT, and MdSOT2, dramatically decrease malate, citrate, and sorbitol accumulation, respectively, and correspond to important domestication events. Interestingly, Ma1 was identified to have pleiotropic effects on both organic acid content and sugar:acid ratio, suggesting that it plays a vital role in determining fruit taste. Fruit taste is unlikely to have been negatively affected by linkage drag associated with selection for larger fruit that resulted from the pyramiding of multiple genes with minor effects on fruit size. 2Simultaneous evaluation of two branches of a multifunctional integrated optic chip with an ultra-simple dual-channel configuration显示文摘We propose an ultra-simple dual-channel configuration for simultaneously evaluating two branches of a multifunctional integrated optic chip(MFIOC). In the configuration, the MFIOC is employed as a beam splitter to construct the demodulation interferometer together with a 2 × 2 fiber coupler. Interference happens between polarization modes traveling through different channels of the MFIOC. The cross-couplings of each channel are respectively characterized by the interference peaks which distribute on opposite sides of the central interference peak. Temperature responses of the MFIOC are experimentally measured from-40°C to 80°C. Results show that the proposed configuration can achieve simultaneous dual-channel transient measurements with resolution of-90 d B and dynamic range of 90 d B. 4An R2R3-type myeloblastosis transcription factor MYB103 is involved in phosphorus remobilization显示文摘The members of myeloblastosis transcription factor(MYB TF)family are involved in the regulation of biotic and abiotic stresses in plants.However,the role of MYB TF in phosphorus remobilization remains largely unexplored.In the present study,we show that an R2R3 type MYB transcription factor,MYB103,is involved in phosphorus(P)remobilization.MYB103 was remarkably induced by P deficiency in cabbage(Brassica oleracea var.capitata L.).As cabbage lacks the proper mutant for elucidating the mechanism of MYB103 in P deficiency,another member of the crucifer family,Arabidopsis thaliana was chosen for further study.The transcript of its homologue AtMYB103 was also elevated in response to P deficiency in A.thaliana,while disruption of AtMYB103(myb103)exhibited increased sensitivity to P deficiency,accompanied with decreased tissue biomass and soluble P concentration.Furthermore,AtMYB103 was involved in the P reutilization from cell wall,as less P was released from the cell wall in myb103 than in wildtype,coinciding with the reduction of ethylene production.Taken together,our results uncover an important role of MYB103 in the P remobilization,presumably through ethylene signaling.Fangwei Yu Shenyun Wang Wei Zhang Hong Wang Li Yu Zhangjun Fei Jianbin Li 2020Food Production, Processing and Nutrition2020,2,1:0
5An RNA-RNA crosstalk network involving HMGB1 and RICTOR facilitates hepatocellular carcinoma tumorigenesis by promoting glutamine metabolism and impedes immunotherapy by PD-L1+ exosomes activity显示文摘Hepatocellular carcinoma(HCC)is the global leading cause of cancer-related deaths due to the deficiency of targets for precision therapy.A new modality of epigenetic regulation has emerged involving RNA-RNA crosstalk networks where two or more competing endogenous RNAs(ceRNAs)bind to the same microRNAs.However,the contribution of such mechanisms in HCC has not been well studied.Herein,potential HMGB1-driven RNA-RNA crosstalk networks were evaluated at different HCC stages,identifying the mT0RC2 component RICTOR as a potential HMGB1 ceRNA in HBV^(+)early stage HCC.Indeed,elevated HMGB1 mRNA was found to promote the expressio n of RICTOR mRNA through competitively bin ding with the miR-200 family,especially miR-429.Functio nal assays emplo ying overexpressi on or in terference strategies dem on strated that the HMGB1 and RICTOR 3zuntra nslated regions(UTR)epigenetically promoted the malignant proliferation,self-renewal,and tumorigenesis in HCC cells.Intriguingly,in terference agai nst HMGB1 and RICTOR in HCC cells promoted a stron ger an ti-PD-L1 immuno therapy resp on se,which appeared to associate with the production of PD-L1^(+)exosomes.Mechanistically,the HMGB1-driven RNA-RNA crosstalk network facilitated HCC cell glutamine metabolism via dual mechanisms,activating a positive feedback loop involving mT0RC2-AKT-C-MYC to upregulate glutamine synthetase(GS)expression,and inducing mTORCI signaling to derepress SIRT4 on glutamate dehydrogenase(GDH).Meanwhile,this crosstalk network could impede the efficacy of immunotherapy through mTORCI-P70S6K dependent PD-L1 production and PD-L1^(+)exosomes activity.In conclusion,our study highlights the non-coding regulatory role of HMGB1 with implicatio ns for RNA-based therapeutic targeting together with a predictio n of an ti-PD-L1 immuno therapy in HCC.Yanping Wei Xuewu Tang Yibin Ren Yun Yang Fengliang Song Jingbo Fu Shuowu Liu Miao Yu Jing Chen Suyang Wang Kecheng Zhang Yexiong Tan Zhipeng Han Lixjn Wei Baohua Zhang Zhangjun Cheng Liang Li Hongyang Wang 2022Signal Transduction and Targeted Therapy2022,7,1:0
6A state‑domain robust autonomous integrity monitoring with an extrapolation method for single receiver positioning in the presence of slowly growing fault显示文摘Single receiver positioning has been widely used as a standard and standalone positioning technique for about 25 years.To detect the slowly growing faults caused by satellite and receiver clocks in single receiver positioning,the Autonomous Integrity Monitoring with an Extrapolation method(AIME)was proposed based on the Kalman filter measurement domain.However,AIME was designed with the assumption of there is the same number of visible satellites at each epoch,which limits its application.To address this issue,this paper proposes a state-domain Robust Autonomous Integrity Monitoring with the Extrapolation Method(SRAIME).The slowly growing fault detection statistics is established based on the difference between the estimates of the state propagator and the posterior state estimation in Kalman filtering.Meanwhile,singular value decomposition is adopted to factor the covariance matrix of the difference to increase computational robustness.Besides,the relevant formulas of the proposed method are theoretically derived,and it is proven that the proposed method is suitable for any positioning model based on the Kalman filter.Additionally,the results of two experiments indicate that SRAIME can detect slowly growing faults in single receiver positioning earlier than AIME.Zhangjun Yu Qiuzhao Zhang Shubi Zhang Nanshan Zheng Keqiang Liu 2023Satellite Navigation2023,4,3:0
