Endovascular stent-induced alterations in host artery mechanical environments and their roles in stent restenosis and late thrombosis显示文摘Cardiovascular stent restenosis remains a major challenge in interventional treatment of cardiovascular occlusive disease.Although the changes in arterial mechanical environment due to stent implantation are the main causes of the initiation of restenosis and thrombosis, the mechanisms that cause this initiation are still not fully understood. In this article,we reviewed the studies on the issue of stent-induced alterations in arterial mechanical environment and discussed their roles in stent restenosis and late thrombosis from three aspects:(i)the interaction of the stent with host blood vessel, involve the response of vascular wall, the mechanism of mechanical signal transmission, the process of re-endothelialization and late thrombosis;(ii)the changes of hemodynamics in the lumen of the vascular segment and(iii)the changes of mechanical microenvironment within the vascular segment wall due to stent implantation. This review has summarized and analyzed current work in order to better solve the two main problems after stent implantation, namely in stent restenosis and late thrombosis, meanwhile propose the deficiencies of current work for future reference. | Jinxuan Wang Xuepu Jin Yuhua Huang Xiaolin Ran Desha Luo Dongchuan Yang Dongyu Jia Kang Zhang Jianhua Tong Xiaoyan Deng Guixue Wang | 2018 | Regenerative Biomaterials2018,5,3: | 13
Effect of intraplaque angiogenesis to atherosclerotic rupture-prone plaque induced by high shear stress in rabbit model显示文摘Atherosclerotic prone-rupture plaque is mainly localized in the region of the entrance to the stenosiswith high shear stress and the reasons are largely unknown. Our hypothesis is that such a distributionof cells in atherosclerotic plaque may depend on the angiogenesis. Silastic collars inducedregions of high shear stress (20.6865.27 dynes/cm2) in the upstream flow and low shear stress(12.2561.28 dynes/cm2) in the downstream flow in carotid arteries. Compared with the low shearstress region, plaques in the high shear stress region showed more intraplaque haemorrhaging, less collagen and higher apoptotic rates of vascular smooth muscle cells;endothelial cells (ECs) inthe high shear stress region were characterized with integrity and high endothelial nitric oxidesynthase (eNOS) expression (1570.36345.5% vs 172.9649.9%). The number of intraplaque microvesselsis very high in the high shear stress region (1561.8 n/mm2 vs 3.560.4 n/mm2), and themicrovessels in the plaque show ECs were abnormal, with membrane blebs, intracytoplasmic vacuolesand leukocyte infiltration. Our current study reveals that the integrity of the endothelium andthe vulnerability of atherosclerotic plaques are simultaneously localized in high shear stress regions, and we provide evidence for the first time that microvessels in the intraplaque maybe responsiblefor rupture-prone plaque formation in the high shear stress region. | Juhui Qiu Daoxi Lei Jianjun Hu Tieying Yin Kang Zhang Donghong Yu Guixue Wang | 2017 | Regenerative Biomaterials2017,4,4: | 12
Initial corrosion behavior and mechanism of 7B04 aluminum alloy under acid immersion and salt spray environments显示文摘The initial corrosion behavior and mechanism of 7B04 aluminum alloy under acid immersion and salt spray environments(pH=3.5)are studied by Scanning Electron Microscope(SEM),optical microscope,Fourier Transform Infrared Spectroscopy(FT-IR),X-Ray Diffraction(XRD),potentiodynamic polarization,Electrochemical Impedance Spectroscopy(EIS),and Scanning Kelvin Probe(SKP).The results show that pitting corrosion occurs at the initial corrosion stage, and the potential difference between the second phase particles is the main cause of pitting.Pitting pits on different locations gradually expand and coalesce with the proceeding of corrosion. The main components of corrosion products are Al_(2)O_(3),Al(OH)_(3),and AlCl_(3),and the generation rate of the corrosion product layer under the salt spray environment is larger than that under acid immersion environment.Under both environments, the Volta potential distribution first disperses and then concentrates, while the charge transfer resistance first decreases and then increases with the corrosion time. The Volta potential gradually shifts in a positive direction, indicating that corrosion products have an inhibitory effect on corrosion. After the same corrosion time, the corrosion product layer resistance and the expectation of the Volta potential of the salt spray sample are higher than those of the immersion sample.Comparatively, the corrosion current density for the salt spray sample is significantly lower than the immersion sample, which indicates that the thicker the corrosion product layer, the stronger the inhibition of corrosion reaction. | Yangguang ZHANG Yueliang CHEN Yong ZHANG Guixue BIAN Chenguang WANG Andong WANG | 2022 | Chinese Journal of Aeronautics2022,35,1: | 8
Distinctive effects of CD34- and CD133-specific antibody-coated stents on re-endothelialization and in-stent restenosis at the early phase of vascular injury显示文摘It is not clear what effects of CD34-and CD133-specific antibody-coated stents have on reendothelialization and in-stent restenosis(ISR)at the early phase of vascular injury. This study aims at determining the capabilities of different coatings on stents(e.g.gelatin,anti-CD133 and anti-CD34 antibodies)to promote adhesion and proliferation of endothelial progenitor cells(EPCs).The in vitro study revealed that the adhesion force enabled the EPCs coated on glass slides to withstand flow-induced shear stress,so that allowing for the growth of the cells on the slides for 48 h.The in vivo experiment using a rabbit model in which the coated stents with different substrates were implanted showed that anti-CD34 and anti-CD133 antibody-coated stents markedly reduced the intima area and restenosis than bare mental stents(BMS)and gelatin-coated stents.Compared with the anti-CD34 antibody-coated stents, the time of cells adhesion was longer and earlier present in the anti-CD133 antibody-coated stents and anti-CD133 antibody-coated stents have superiority in re-endothelialization and inhibition of ISR.In conclusion, this study demonstrated that anti-CD133 antibody as a stent coating for capturing EPCs is better than anti-CD34 antibody in promoting endothelialization and reducing ISR. | Xue Wu Tieying Yin Jie Tian Chaojun Tang Junli Huang Yinping Zhao Xiaojuan Zhang Xiaoyan Deng Yubo Fan Donghong Yu Guixue Wang | 2015 | Regenerative Biomaterials2015,2,2: | 7

Uptake of oxidative stress-mediated extracellular vesicles by vascular endothelial cells under low magnitude shear stress显示文摘Extracellular vesicles(EVs)are increasingly used as delivery vehicles for drugs and bioactive molecules, which usually require intravascular administration. The endothelial cells covering the inner surface of blood vessels are susceptible to the shear stress of blood flow.Few studies demonstrate the interplay of red blood cell-derived EVs(RBCEVs)and endothelial cells. Thus, the phagocytosis of EVs by vascular endothelial cells during blood flow needs to be elucidated. In this study, red blood cell-derived extracellular vesicles(RBCEVs)were constructed to investigate endothelial cell phagocytosis in vitro and animal models. Results showed that low magnitude shear stress including low shear stress(LSS)and oscillatory shear stress(OSS)could promote the uptake of RBCEVs by endothelial cells in vitro. In addition,in zebrafish and mouse models,RBCEVs tend to be internalized by endothelial cells under LSS or OSS.Moreover,RBCEVs are easily engulfed by endothelial cells in atherosclerotic plaques exposed to LSS or OSS.In terms of mechanism, oxidative stress induced by LSS is part of the reason for the increased uptake of endothelial cells.Overall, this study shows that vascular endothelial cells can easily engulf EVs in areas of low magnitude shear stress, which will provide a theoretical basis for the development and utilization of EVs-based nano-drug delivery systems in vivo. | Xian Qin Kun Zhang Juhui Qiu Nan Wang Kai Qu Yuliang Cui Junli Huang Li Luo Yuan Zhong Tian Tian Wei Wu Yi Wang Guixue Wang | 2022 | Bioactive Materials2022,7,3: | 3
Photocrosslinking silver nanoparticles–aloe vera–silk fibroin composite hydrogel for treatment of full-thickness cutaneous wounds显示文摘Damage to the skin causes physiological and functional issues. The most effective treatment approach is the use of wound dressings.Silk fibroin(SF)is a promising candidate biomaterial for regulating wound healing;however, its antibacterial properties and biological activity must be further improved. In this study,a photocrosslinking hydrogel was developed to treat full-thickness cutaneous wounds. The composite hydrogel(Ag-AV-SF hydrogel)was prepared by introducing the silver nanoparticles(AgNPs)and aloe vera(AV)as the modifiers. In vitro study exhibited great antibacterial ability, biocompatibility and cell-proliferation and-migration-promoting capacities. It also showed the pH-response releasing properties which release more AgNPs in a simulated chronic infection environment. The healing effect evaluation in vivo showed the healing-promoting ability of the Ag-AV-SF hydrogel was stronger than the single-modifiers groups, and the healing rate of it reached 97.02%on Day 21,higher than the commercial wound dressing, silver sulfadiazine(SS)cream on sale.Additionally, the histological and protein expression results showed that the Ag-AV-SF hydrogel has a greater effect on the pro-healing regenerative phenotype with M2 macrophages at the early stage, reconstructing the blood vessels networks and inhibiting the formation of scars. In summary, the Ag-AV-SF hydrogel developed in this study had good physical properties, overwhelming antibacterial properties, satisfactory biocompatibility and significantly promoting effect on cell proliferation, migration and wound healing.Overall, our results suggest that the Ag-AV-SF hydrogel we developed has great potential for improving the wound healing in clinical treatment. | Yangkun Liu JinChuan Fan MingQi Lv Kepeng She Jiale Sun Qingqing Lu Changhao Han SongTao Ding Shuang Zhao GuiXue Wang YuChan Zhang GuangChao Zang | 2021 | Regenerative Biomaterials2021,8,6: | 2

trans-2-Enoyl-CoA Reductase Tecr-Driven Lipid Metabolism in Endothelial Cells Protects against Transcytosis to Maintain Blood-Brain Barrier Homeostasis显示文摘The transport and metabolism of lipids in cerebrovascular endothelial cells(ECs)have been hypothesized to regulate blood-brain barrier(BBB)maturation and homeostasis.Long-chain polyunsaturated fatty acids(LCPUFAs)as the important lipids components of cell membranes are essential for the development and function of BBB,but the direct links of lipid metabolism and ECs barrier function remain to be established. Here,we comprehensively characterize the transcriptomic phenotype of developmental cerebrovascular ECs in single-cell resolution and firstly find that trans-2-enoyl-CoA reductase(Tecr),a verylong-chain fatty acid synthesis,is highly expressed during barriergenesis and decreased after BBB maturation.EC-specific knockout of Tecr compromises angiogenesis due to delayed vascular sprouting.Importantly,EC-specific deletion of Tecr loss restrictive quality of vascular permeability from neonatal stages to adulthood, with high levels of transcytosis, but maintains the vascular tight junctions. Moreover, lipidomic analysis shows that the expression of Tecr in ECs is associated with the containing of omega-3 fatty acids, which directly suppresses caveolae vesicles formation. These results reveal a protective role for Tecr in BBB integrity and suggest that Tecr as a novel therapeutic target in the central nervous system(CNS)diseases associated with BBB dysfunction. | Jinxuan Wang Jianxiong Xu Guangchao Zang Tao Zhang Qi Wu Hongping Zhang Yidan Chen Yi Wang Weixi Qin Shuang Zhao Erdai Qin Juhui Qiu Xiaojuan Zhang Lin Wen Yeqi Wang Guixue Wang | 2022 | Research2022,,3: | 1
Remodeling tumor immunosuppressive microenvironment via a novel bioactive nanovaccines potentiates the efficacy of cancer immunotherapy显示文摘The clinical outcomes of cancer nanovaccine have been largely impeded owing to the low antigen-specific T cell response rate and acquired resistance caused by the immunosuppressive tumor microenvironment(TME).Here,we reported a tumor acidity-responsive nanovaccine to remodel the immunosuppressive TME and expand the recruitment of tumor infiltrating lymphocytes(TILs)using hybrid micelles(HM),which encapsulated colony stimulating factor 1 receptor(CSF1-R)inhibitor BLZ-945 and indoleamine 2,3-dioxygenase(IDO)inhibitor NLG-919 in its core and displayed a model antigen ovalbumin(OVA)on its surface(denoted as BN@HM-OVA).The bioactive nanovaccine is coated with a polyethylene glycol(PEG)shell for extending nanoparticle circulation. The shell can be shed in response to the weakly acidic tumor microenvironment. The decrease in size and the increase in positive charge may cause the deep tumor penetration of drugs. We demonstrated that the bioactive nanovaccine dramatically enhance antigen presentation by dendritic cells(DCs)and drugs transportation into M1-like tumor-associated macrophages(TAMs)and tumor cells via size reduction and increasing positive charge caused by the weakly acidic TME.Such bioactive nanovaccine could remodel the immunosuppressive TME into an effector T cells favorable environment, leading to tumor growth inhibition in prophylactic and therapeutic E.G7-OVA tumor models.Furthermore, combining the bioactive nanovaccine with simultaneous anti-PD-1 antibody treatment leads to a long-term tumor inhibition, based on the optimal timing and sequence of PD-1 blockade against T cell receptor. This research provides a new strategy for the development of efficient cancer immunotherapy. | Xiaoxue Xie Yi Feng Hanxi Zhang Qingqing Su Ting Song Geng Yang Ningxi Li Xiaodan Wei Tingting Li Xiang Qin Shun Li Chunhui Wu Xiaojuan Zhang Guixue Wang Yiyao Liu Hong Yang | 2022 | Bioactive Materials2022,7,10: | 1
Effect of ultrasound with methylene blue as sound sensitive agent on virus inactivation显示文摘The goal of this paper is to explore a method for virus inactivation based on ultrasonic treatment, and on this basis,to explore the synergistic effect of methylene blue as a sonosensitizer in virus inactivation. The titer of human parainfluenza virus type 3(HPIV3),Autographacaliforinica nuclear polyhedrosis virus carried a green fluorescent reporter gene(AcNPV-GFP)and Enterovirus group D 68(EV-D68)were determined by plaque assays or TCID50 methods after treatment by MB combined with illumination(MB photochemical,MBP)or ultrasonic excitation.Different ultrasonic power and time,MB concentration gradient were set to determine the best antiviral combination. For the enveloped virus HPIV3 and AcNPV-GFP,pure ultrasonic or MBP treatment could reduce viral titer more than 104,and ultrasonic combined with MB could completely inactivate HPIV3 and AcNPV-GFP in 5min while traditional MBP methods could only reduce viral titer about 10X.For the nonenveloped virus EV-D68,pure ultrasonic or MBP treatment only reduced the viral titer about 102,but ultrasonic combined with MB treatment could reduce the viral titer about 103 in 5min and completely inactive EV-D68 in 10min(reduced 105).Compared with MBP method and pure ultrasonic inactivation, ultrasonic combined with MB has better inactivation effect on either enveloped or non-enveloped viruses, and the appropriate combination of parameters is expected to be a new blood transfusion transmitted virus inactivation method. | Nan Lu Yunbo Tian Lan Wei Guangyuan Zhang Chengbing Xie Tao He Yongzhu Xu Ziqiu Hu Guixue Wang | 2023 | Medicine in Novel Technology and Devices2023,,1: | 1
Cathepsin K contributed to disturbed flow-induced atherosclerosis is dependent on integrin-actin cytoskeleton–NF–κB pathway显示文摘Atherosclerosis is a chronic inflammatory disease, occurring preferentially in bifurcation, branching, and bending of blood vessels exposed to disturbed flow. Disturbed flow in atheroprone areas activates elevated proteases, degrading elastin lamellae and collagenous matrix, resulting in endothelial dysfunction and vascular remodeling. As a mediator for extracellular matrix protein degradation, cathepsin K (CTSK) was directly regulated by hemodynamics and contributed to atherosclerosis. The mechanism of CTSK responding to disturbed flow and contributing to disturbed flow-induced atherosclerosis is unclear. In this study, the partial carotid ligation model of mice and in vitro disturbed shear stress model were constructed to explore the contribution and potential mechanism of CTSK in atherosclerosis. Our results indicated that CTSK elevated in the disturbed flow area in vivo and in vitro along with endothelial inflammation and atherogenesis. Additionally, the expression of integrin αvβ3 was upregulated in these atheroprone areas. We found that inhibition of the integrin αvβ3-cytoskeleton pathway could significantly block the activation of NF-κB and the expression of CTSK. Collectively, our findings unraveled that disturbed flow induces increased CTSK expression, and contributes to endothelial inflammation and vascular remodeling, leading to atherogenesis eventually. This study is helpful to provide new enlightenment for the therapy of atherosclerosis. | Fei Fang Tang Feng Jianwei Li Huaiyi Zhang Qin Wang Yidan Chen Guixue Wang Yang Shen Xiaoheng Liu | 2023 | Genes & Diseases2023,10,2: | 1

The Janus-faced role of Piezo1 in cardiovascular health under mechanical stimulation显示文摘In recent years, cardiovascular health problems are becoming more and more serious. At the same time, mechanical stimulation closely relates to cardiovascular health. In this context,Piezo1,which is very sensitive to mechanical stimulation, has attracted our attention. Here,we review the critical significance of Piezo1 in mechanical stimulation of endothelial cells,NO production, lipid metabolism,DNA damage protection, the development of new blood vessels and maturation, narrowing of blood vessels, blood pressure regulation, vascular permeability, insulin sensitivity, and maintenance of red blood cell function.Besides,Piezo1 may participate in the occurrence and development of atherosclerosis, diabetes, hypertension, and other cardiovascular diseases. It is worth noting that Piezo1 has dual effects on maintaining cardiovascular health. On the one hand, the function of Piezo1 is necessary to maintain cardiovascular health;on the other hand, under some extreme mechanical stimulation, the overexpression of Piezo1 may bring adverse factors such as inflammation. Therefore, this review discusses the Janus-faced role of Piezo1 in maintaining cardiovascular health and puts forward new ideas to provide references for gene therapy or nanoagents targeting Piezo1. | Junyang Huang Kun Zhang Ruolin Du Wanling Liu Haijun Zhang Tian Tian Yazhou Wang Guixue Wang Tieying Yin | 2023 | Genes & Diseases2023,10,5: | 1
G3BP2 regulates oscillatory shear stress-induced endothelial dysfunction显示文摘GTPase-activating SH3 domain-binding protein 2(G3BP2)is a mediator that responds to environmental stresses through stress granule formation and is involved in the progression of chronic diseases. However,no studies have examined the contribution of G3BP2 in the oscillatory shear stress(OSS)-induced endothelial dysfunction. Here we assessed the effects of G3BP2 in endothelial cells(ECs)function and investigated the underlying mechanism.Using shear stress apparatus and partial ligation model,we identified that stress granulerelated genes in ECs could be induced by OSS with RNA-seq,and then confirmed that G3BP2 was highly and specifically expressed in athero-susceptible endothelia in the OSS regions.G3bp2e/eApoee/e mice had significantly decreased atherosclerotic lesions associated with deficiency of G3BP2 in protecting endothelial barrier function, decreasing monocyte adhesion to ECs and inhibiting the proinflammatory cytokine levels.Furthermore, loss of G3BP2 diminished OSS-induced inflammation in ECs by increasing YAP nucleocytoplasmic shuttling and phosphorylation. These data demonstrate that G3BP2 is a critical OSS regulated gene in regulating ECs function and that G3BP2 inhibition in ECs is a promising atheroprotective therapeutic strategy. | Tianhan Li Juhui Qiu Tingting Jia Yinming Liang Kun Zhang Wenhua Yan Zhengjun Hou Shiwei Yang Lushan Liu Wenhao Xiong Yaokai Chen Guixue Wang | 2022 | Genes & Diseases2022,9,6: | 1

Host defense peptide-mimicking peptide polymer-based antibacterial hydrogel enables efficient healing of MRSA-infected wounds显示文摘Wound infections are a compelling health issue caused by the invasion and proliferation of pathogens in wound sites.Antibioticloaded hydrogels are widely used to achieve anti-infectious wound healing. However, due to the quick emergence of drugresistant bacteria, such as methicillin-resistant Staphylococcus aureus(MRSA),wound infection has been a formidable challenge to human health. To address MRSA-infected wounds,an antibacterial peptide polymer-loaded hyaluronic acid(HA)hydrogel(Gel-HA@P)is prepared. The peptide polymer is designed to mimic host defense peptides as the antibiotic alternative showing potent antibacterial activity, low susceptibility to drug resistance and good stability against proteolysis.HA is biocompatible and biodegradable hydrogel substrate as a primary constituent of the extracellular matrix and suitable for cell migration and wound healing.Gel-HA@P shows potent activity against MRSA in vitro and in vivo, low toxicity during the treatment and promotes the wound healing in vivo. This design has proven to be an effective and antibiotic-free strategy to enable the healing of MRSA-infected wounds. | Zhongqian Qiao Wenjing Zhang Yueming Wu Weinan Jiang Ning Shao Jiayang Xie Guixue Xia Qimeng Chen Zhefeng Liu Jingcheng Zou Jiawei Gu Shifang Luan Haodong Lin Runhui Liu | 2023 | Science China Chemistry2023,66,6: | 1
