4Oxidized galectin-1 in SLE fails to bind the inhibitory receptor VSTM1 and increases reactive oxygen species levels in neutrophils显示文摘Inhibitory immune receptors set thresholds for immune cell activation,and their deficiency predisposes a person to autoimmune responses.However,the agonists of inhibitory immune receptors remain largely unknown,representing untapped sources of treatments for autoimmune diseases.Here,we show that V-set and transmembrane domain-containing 1(VSTM1)is an inhibitory receptor and that its binding by the competent ligand soluble galectin-1(Gal1)is essential for maintaining neutrophil viability mediated by downregulated reactive oxygen species production.However,in patients with systemic lupus erythematosus(SLE),circulating Gal1 is oxidized and cannot be recognized by VSTM1,leading to increased intracellular reactive oxygen species levels and reduced neutrophil viability.Dysregulated neutrophil function or death contributes significantly to the pathogenesis of SLE by providing danger molecules and autoantigens that drive the production of inflammatory cytokines and the activation of autoreactive lymphocytes.Interestingly,serum levels of glutathione,an antioxidant able to convert oxidized Gal1 to its reduced form,were negatively correlated with SLE disease activity.Taken together,our findings reveal failed inhibitory Gal1/VSTM1 pathway activation in patients with SLE and provide important insights for the development of effective targeted therapies.Xu Jiang Xinyue Xiao Hao Li Yiyi Gong Min Wang Huaxia Yang Lidan Zhao Ying Jiang Yanping Wei Chongchong Zhao Jin Li Yuling Chen Shan Feng Haiteng Deng Shiliang Ma Yue Xu Yudong Liu George C.Tsokos Minghong Jiang Xuan Zhan 2023Cellular & Molecular Immunology2023,20,11:1
5槲皮素对人肺癌细胞株A549化疗药物增敏作用的实验研究(英文)显示文摘Objective: The present study was designed to investigate whether quercetin exerts increasing chemosensitivity on human lung adenocarcinoma cells when quercetin combined with cisplatin (DDP) and vincristine (VCR) in vitro respectively and its possible antitumor mechanism. To provide experimental proof for clinical combination application. Methods: Using intermittent administration of high dose VCR, human lung adenocarcinoma sensitive cell line (A549/S) was induced to VCR-resistant human lung adenocarcinoma cell line (A549/VCR). MTT assay was adapted for examing the 50% inhibition (IC50) value of DDP and VCR on A549/S and A549/VCR when quercetin combined with DDP and VCR respectively. Results: IC50 of DDP on A549/S and A549/VCR was 10.18 and 12.35 mg/L, and the IC50 of VCR on the two cell lines was 1.21 and 12.77 mg/L, respectively. The resistance fold of A549/VCR on VCR and DDP was 10.55 and 1.21, respectively. When quercetin at concentration of 50, 100 and 200 μmol/L in combination with DDP and VCR respectively, the IC50 of DDP and VCR on A549/S and A549/VCR were obvious decreased (P < 0.05 - P < 0.01). 6Photokinesis and Djopsin gene expression analysis during the regeneration of planarian eyes显示文摘Planarians provide the ideal model for studying eye development,with their simple eye structure and exceptionally rapid regeneration.Here,we observed the eye morphogenesis,photophobic behavior,spectral sensitivity and expression pattern of Djopsin in the freshwater planarian Dugesia japonica.The results showed that:(i)Djopsin encoding the putative protein belonged to the rhabdomeric opsins group and displayed high conservation during animal evolution;(ii)planarians displayed diverse photophobic response to different visible wavelengths and were more sensitive to light blue(495 nm)and yellow(635 nm);(iii)the morphogenesis and functional recovery of eyes were related to the expression pattern of Djopsin during head regeneration;and(iv)Djopsin gene plays a major role in functional recovery during eye regeneration and visual system maintenance in adult planarians.Zimei DONG Yanqing YUWEN Yingxu SIMA Yanping DONG Huina ZHAN Guangwen CHEN Dezeng LIU 2017Integrative Zoology2017,12,2:0
7M-X Ray of Ta Excited by Highly Charged Ar and Xe Ions显示文摘The X-ray spectra induced by the highly charged xenon and argon ions impinging on a tantalum surface are reported. The experiment was done at the ECR ion source of H1RFL. It is found that the yield of the tantalum M-X rays increases intensively as the projectile kinetic energy increasing.
