2The lifestyle transition of Arthrobotrys oligospora is mediated by microRNA-like RNAs显示文摘The lifestyle transition of fungi,defined as switching from taking organic material as nutrients to pathogens,is a fundamental phenomenon in nature.However,the mechanisms of such transition remain largely unknown.Here we show microRNA-like RNAs(milRNAs)play a key role in fungal lifestyle transition for the first time.We identified milRNAs by small RNA sequencing in Arthrobotrys oligospora,a known nematode-trapping fungus.Among them,7 highly expressed milRNAs were confirmed by northern-blot analysis.Knocking out two milRNAs significantly decreased A.oligospora’s ability to switch lifestyles.We further identified that two of these milRNAs were associated with argonaute protein QDE-2 by RNA-immunoprecipitation(RIP)analysis.Three of the predicted target genes of milRNAs were found in immunoprecipitation(IP)products of QDE-2.Disruption of argonaute gene qde-2 also led to serious defects in lifestyle transition.Interestingly,knocking out individual milRNAs or qde-2 lead to diverse responses under different conditions,and qde-2 itself may be targeted by the milRNAs.Collectively,it indicates the lifestyle transition of fungi is mediated by milRNAs through RNA interference(RNAi)machinery,revealing the wide existence of miRNAs in fungi kingdom and providing new insights into understanding the adaptation of fungi from scavengers to predators and the mechanisms underlying fungal infections.Xinglai Ji Heng Li Weihua Zhang Jiai Wang Lianming Liang Chenggang Zou Zefen Yu Shuqun Liu Ke-Qin Zhang 2020Science China(Life Sciences)2020,63,4:4
3Further understanding of an uncommon disease of combined small cell lung cancer: clinical features and prognostic factors of 114 cases显示文摘Objective: Combined small cell lung cancer(C-SCLC) is an uncommon subgroup of small cell lung cancer(SCLC) and few clinical data can be referred.Our study is to investigate the clinical features and prognostic factors of C-SCLC, as well as the role of multimodality treatment.Methods: Between January 2004 and December 2012, patients with histologically diagnosed C-SCLC were retrospectively analyzed.The survivals were evaluated with the Kaplan-Meier method.Univariate and multivariate analyses were used to evaluate potential prognostic factors.Results: One hundred and fourteen patients were enrolled, with a median age of 59(range: 20-79) years old.The most common combined component was squamous cell carcinoma(52.6%).Among these patients, the disease was stage Ⅰ, Ⅱ, Ⅲ and Ⅳ in 9.6%, 19.3%, 46.5% and 24.6% of the patients, respectively.Eighty patients(70.2%)received at least two of the three modalities containing chemotherapy, radiotherapy and surgery.The median follow-up was 32.5 months.The median time of overall survival(OS) was 26.2 months.On univariate analysis,smoking(P=0.029), Karnofsky performance score(KPS) <80(P=0.000), advanced TNM stage(P=0.000), no surgery(P=0.010), positive resection margin(P=0.000), positive lymph nodes ≥4(P=0.000), positive lymph node ratio >10%(P=0.000) and non-multimodality treatment(P=0.004) were associated with poor OS.Multivariate analysis confirmed that smoking, advanced TNM stage, positive resection margin and positive lymph nodes ratio>10% were poor prognostic features.Conclusions: C-SCLC has a relatively early stage and good prognosis, which may due to the underestimated diagnosis in non-surgical patients.Multimodality therapy is recommended, especially for limited disease.Smoking,advanced TNM stage, positive resection margin and positive lymph nodes ratio >10% are poor prognostic factors.Yu Men Zhouguang Hui Jun Liang Qinfu Feng Dongfu Chen Hongxing Zhang Zefen Xiao Zongmei Zhou Weibo Yin Luhua Wang 2016Chinese Journal of Cancer Research2016,28,5:2

8Ophiostomatales (Ascomycota) associated with Tomicus species in southwestern China with an emphasis on Ophiostoma canum显示文摘Ophiostomatalean fungi may facilitate bark beetle colonization and reproduction.In the present study of the fungal community associated with bark beetle species belonging to Tomicus in Yunnan,China,six ophiostomatalean fungi(Ophiostoma canum,O.ips,O.tingens,Leptographium yunnanense,Leptographium sp.1 and Leptographium sp.2)were isolated from the beetles or their galleries;O.canum was the most common fungal species.The distribution of O.canum was associated with stands heavily damaged by Tomicus species and a higher percentage of valid galleries of Tomicus yunnanensis and T.minor in Yunnan pine(Pinus yunnanensis).After inoculation of Yunnan pine with the fungus,a phloem reaction zone formed and monoterpenes accumulated in the phloem.These results suggested that O.canum was pathogenic to Yunnan pine and that the wide distribution of the fungus might be benefi cial to reproduction of pine shoot beetles in Yunnan pine.However,because the reaction zone and monoterpene accumulation were mild,fungal damage of Yunnan pine might be limited.A more integrated study considering all the fungal species should be done to better understand the interactions among bark beetles,blue-stain fungi,and the tree hosts in the region.Yue Pan Jun Lu Peng Chen Zefen Yu Huihong Zhang Hui Ye Tao Zhao 2020Journal of Forestry Research2020,31,6:1
9Molecular Weight and Distribution of Cellulose and Cellulose Microspheres显示文摘[Objective] This study aimed to provide basic data for studying the relationship between structure and property of cellulose microspheres by measuring molecular weight of cellulose and cellulose microspheres with viscosity method and gel permeation chromatography(GPC) method.[Method] In viscosity method,cadmium ethylenediamine was used as the solvent,intrinsic viscosity η of the solution was determined at 25 ℃ by using a Ubbelohde viscometer,to calculate the molecular weight of cellulose;in GPC method,8% LiCl /N,N-dimethylacetamide(LiCl/DMAc) was used as the solvent and 0.5% LiCl/DMAc was used as the mobile phase to determine the relative molecular weight and distribution of cellulose and cellulose microspheres.In addition,the determination results were analyzed to compare these two methods.[Result] Viscosity-average molecular weight Mη of cellulose and cellulose microspheres determined with viscosity method were 224,532 and 16,686,respectively;weight-average molecular weight Mw of cellulose and cellulose microspheres determined with GPC method were 284,196 and 22,345,respectively.[Conclusion] The determination results of GPC method are relatively close to the actual value and could truly reflect the characteristics of molecular weight distribution of cellulose and cellulose microspheres.Zefen WANG Kelin HUANG Ben WANG Rui WU Jing TAO Xiaoyu PENG Dankui LIAO 2012Agricultural Biotechnology2012,1,1:1

11Molecular Dynamics Simulation on Friction and Thermal Properties of FCC Copper in Nanoscale Sliding Contacts显示文摘In nanoscale sliding contact,adhesion effects and adhesive force are predominant,and high friction force will be produced.Friction energy is mainly converted into heat,and the heat will make nanomaterials become soft to affect friction behaviors,so it is important to investigate the friction and thermal properties of the nanoscale sliding contacts.A model of a nanoscale sliding contact between a rigid cylindrical tip and an FCC copper substrate is developed by molecular dynamics simulation.The thermal properties of the substrate and the friction behaviors are studied at different sliding velocities and different tip radii.The results show that at a low sliding velocity,the friction force fluctuation is mainly caused by material melting⁃solidification,while at a high sliding velocity the material melting is a main factor for the friction reduction.The average friction forces increase at initial phase and then decrease with increasing sliding velocity,and the average temperature of the substrate increases as sliding velocity increases.Increasing tip radius significantly increases the temperature,while the coupled effects of tip radius and temperature rise make friction force increase slightly.Ruiting Tong Bin Han Tao Zhang Zefen Quan Geng Liu 2022Journal of Harbin Institute of Technology(New Series)2022,29,2:0

13Advances in the Immunotherapeutic Potential of Isocitrate Dehydrogenase Mutations in Glioma显示文摘Isocitrate dehydrogenase(IDH)is an essential metabolic enzyme in the tricarboxylic acid cycle(TAC).The high mutation frequency of the IDH gene plays a complicated role in gliomas.In addition to affecting gliomas directly,mutations in IDH can also alter their immune microenvironment and can change immune-cell function in direct and indirect ways.IDH mutations mediate immune-cell infiltration and function by modulating immune-checkpoint gene expression and chemokine secretion.In addition,IDH mutation-derived D2-hydroxyglutarate can be absorbed by surrounding immune cells,also affecting their functioning.In this review,we summarize current knowledge about the effects of IDH mutations as well as other gene mutations on the immune microenvironment of gliomas.We also describe recent preclinical and clinical data related to IDH-mutant inhibitors for the treatment of gliomas.Finally,we discuss different types of immunotherapy and the immunotherapeutic potential of IDH mutations in gliomas.Feng Tang Zhiyong Pan Yi Wang Tian Lan Mengyue Wang Fengping Li Wei Quan Zhenyuan Liu Zefen Wang Zhiqiang Li 2022Neuroscience Bulletin2022,38,9:0
