1Chromosome-scale genome assembly provides insights into the evolution and flavor synthesis of passion fruit (Passiflora edulis Sims)显示文摘Passion fruit(Passiflora edulis Sims)is an economically valuable fruit that is cultivated in tropical and subtropical regions of the world.Here,we report an~1341.7Mb chromosome-scale genome assembly of passion fruit,with 98.91%(~1327.18Mb)of the assembly assigned to nine pseudochromosomes.The genome includes 23,171 protein-coding genes,and most of the assembled sequences are repetitive sequences,with long-terminal repeats(LTRs)being the most abundant.Phylogenetic analysis revealed that passion fruit diverged after Brassicaceae and before Euphorbiaceae.Ks analysis showed that two whole-genome duplication events occurred in passion fruit at 65 MYA and 12 MYA,which may have contributed to its large genome size.An integrated analysis of genomic,transcriptomic,and metabolomic data showed that‘alpha-linolenic acid metabolism’,‘metabolic pathways’,and‘secondary metabolic pathways’were the main pathways involved in the synthesis of important volatile organic compounds(VOCs)in passion fruit,and this analysis identified some candidate genes,including GDP-fucose Transporter 1-like,Tetratricopeptide repeat protein 33,protein NETWORKED 4B isoform X1,and Golgin Subfamily A member 6-like protein 22.In addition,we identified 13 important gene families in fatty acid pathways and eight important gene families in terpene pathways.Gene family analysis showed that the ACX,ADH,ALDH,and HPL gene families,especially ACX13/14/15/20,ADH13/26/33,ALDH1/4/21,and HPL4/6,were the key genes for ester synthesis,while the TPS gene family,especially PeTPS2/3/4/24,was the key gene family for terpene synthesis.This work provides insights into genome evolution and flavor trait biology and offers valuable resources for the improved cultivation of passion fruit.Zhiqiang Xia Dongmei Huang Shengkui Zhang Wenquan Wang Funing Ma Bin Wu Yi Xu Bingqiang Xu Di Chen Meiling Zou Huanyu Xu Xincheng Zhou Rulin Zhan Shun Song 2021Horticulture Research2021,8,1:15
2Minor alloying behavior in bulk metallic glasses and high-entropy alloys显示文摘The effect of minor alloying on several bulk metallic glasses and high-entropy al-loys was studied. It was found that minor Nb addition can optimize the interface structure between the W fiber and the Zr-based bulk metallic glass in the compos-ites,and improve the mechanical properties. Minor Y addition can destabilize the crystalline phases by inducing lattice distortion as a result to improve the glass-forming ability,and the lattice distortion energy is closely related to the effi-ciency of space filling of the competing crystalline phases. A long-period ordered structure can precipitate in the Mg-based bulk metallic glass by yttrium alloying. 5Spatial-resolved metabolomics reveals tissue-specific metabolic reprogramming in diabetic nephropathy by using mass spectrometry imaging显示文摘Detailed knowledge on tissue-specific metabolic reprogramming in diabetic nephropathy(DN)is vital for more accurate understanding the molecular pathological signature and developing novel therapeutic strategies.In the present study,a spatial-resolved metabolomics approach based on air flowassisted desorption electrospray ionization(AFADESI)and matrix-assisted laser desorption ionization(MALDI)integrated mass spectrometry imaging(MSI)was proposed to investigate tissue-specific metabolic alterations in the kidneys of high-fat diet-fed and streptozotocin(STZ)-treated DN rats and the therapeutic effect of astragalosideⅣ,a potential anti-diabetic drug,against DN.As a result,a wide range of functional metabolites including sugars,amino acids,nucleotides and their derivatives,fatty acids,phospholipids,sphingolipids,glycerides,carnitine and its derivatives,vitamins,peptides,and metal ions associated with DN were identified and their unique distribution patterns in the rat kidney were visualized with high chemical specificity and high spatial resolution.These region-specific metabolic disturbances were ameliorated by repeated oral administration of astragaloside Ⅳ(100 mg/kg)for 12 weeks.This study provided more comprehensive and detailed information about the tissue-specific metabolic reprogramming and molecular pathological signature in the kidney of diabetic rats.These findings highlighted the promising potential of AFADESI and MALDI integrated MSI based metabolomics approach for application in metabolic kidney diseases.Zhonghua Wang Wenqing Fu Meiling Huo Bingshu He Yaqi Liu Lu Tian Wanfang Li Zhi Zhou Baili Wang Jianzhen Xia Yanhua Chen Jinfeng Wei Zeper Abliz 2021Acta Pharmaceutica Sinica B2021,11,11:7

10Advances in Thermionic Cathode of Tungsten and Molybdenum显示文摘Several kinds of tungsten thermonic cathodes have been introduced. As a promising alternative for thoriated tungsten, rare earth doped molybdenum cathode has been studied. Compared with the traditional thoriated tungsten, La- Mo cathode has higher emission current density at lower temperature, but it has poor emission stability. In order to im- prove the emission stability, systematical Study on the emission mechanism of La-Mo cathode has been carried out. The life of La-Mo cathode has been improved and has achieved 1400 h, which exceeds the minimum life for practical uses (1000 h). As another alternative for thoriated tungsten cathode, Y-Mo cathode has shown better performance. 11Anisotropic corrosion behavior of sintered(Ce_(0.15)Nd_(0.85))_(30)Fe_(bal)B permanent magnets显示文摘In this work, the corrosion behavior of the surface parallel to the oriented c-axis(c‖) and perpendicular to c-axis(c⊥)in the(Ce_(0.15)Nd_(0.85))_(30)Fe_(bal)B dual main phase magnet was studied. With the addition of Ce,the volume fraction of RE-rich phase shown in the backscattered electron(BSE) images is basically approximate on the two surfaces. The free corrosion potential(E_(corr)) of the c‖ surface is more negative,which shows the worse corrosion resistance from the perspective of thermodynamics. While the reaction kinetics parameters with the smaller free corrosion current(i_(corr)) and the larger transfer resistance(R_(ct)) react the opposite conclusion in 3.5 wt% NaCl solution. Moreover, the c‖ surface performs smaller ions concentration of corrosion product and less damaged corrosion morphology compared to c⊥ surface after the free corrosion. 12Wheat breeding history reveals synergistic selection of pleiotropic genomic sites for plant architecture and grain yield显示文摘Diversity surveys of crop germplasm are important for gaining insights into the genomic basis for plant architecture and grain yield improvement,which is still poorly understood in wheat.In this study,we exome sequenced 287 wheat accessions that were collected in the past 100 years.Population genetics analysis identified that 6.7%of the wheat genome falls within the selective sweeps between landraces and cultivars,which harbors the genes known for yield improvement.These regions were asymmetrically distributed on the A and B subgenomes with regulatory genes being favorably selected.Genome-wide association study(GWAS)identified genomic loci associated with traits for yield potential,and two underlying genes,TaARF12 encoding an auxin response factor and TaDEP1 encoding the G-proteinγ-subunit,were located and characterized to pleiotropically regulate both plant height and grain weight.Elite single-nucleotide haplotypes with increased allele frequency in cultivars relative to the landraces were identified and found to have accumulated over the course of breeding.Interestingly,we found that TaARF12 and TaDEP1 function in epistasis with the classical plant height Rht-1 locus,leading to propose a“Green Revolution”-based working model for historical wheat breeding.Collectively,our study identifies selection signatures that fine-tune the gibberellin pathway during modern wheat breeding and provides a wealth of genomic diversity resources for the wheat research community.Aili Li Chenyang Hao Zhenyu Wang Shuaifeng Geng Meiling Jia Fang Wang Xiang Han Xingchen Kong Lingjie Yin Shu Tao Zhongyin Deng Ruyi Liao Guoliang Sun Ke Wang Xingguo Ye Chengzhi Jiao Hongfeng Lu Yun Zhou Dengcai Liu Xiangdong Fu Xueyong Zhang Long Mao 2022Molecular Plant2022,15,3:4

14Ground-based GPS Used in the Snow Depth Survey of Greenland显示文摘Snow cover is one of the important components of land cover,and it is necessary to accurately monitor the depth and coverage of snow cover.Using the GPS signal receiver data and the basic principle of snow depth detection based on GPS-MR technology,the snow depth of the three sites on the Greenland PBO network GLS1,GLS2,and GLS3 from 2012 to 2018 was obtained.The inversion snow depth is affected by site drift,which is a quite difference from the measured snow depth.Combined with the stable reference point,the velocity field distribution of Greenland Island and the U-direction component change value of the station can be obtained through GAMIT calculation.By analyzing the glacial flow and U-direction component,the influence of the site drift on the snow depth was deducted,and finally compared the corrected inversion snow depth and measured snow depth found that the two were better than before the correction,the results were significantly improved,and the consistency was good.The analysis of the experimental results showed that in extremely cold areas such as Greenland Island,affected by glaciers,the continuous,real-time,high-time resolution snow depth around the measured station obtained by ground-based GPS tracking stations has a large gap with the measured snow depth value,and the gap will gradually increase with time.By deducting the impact of glacier drift,the trend of the two is the same and the consistency is good.The correctness and feasibility of the application of ground-based GPS snow cover theory in the polar area further expand the application scope and practical value of ground-based GPS in snow monitoring.Shuangcheng ZHANG Meiling ZHOU Yajie WANG Ning LIU Qi LIU Jilun PENG 2021Journal of Geodesy and Geoinformation Science2021,4,2:3
15Comparative transcriptomics revealed enhanced light responses, energy transport and storage in domestication of cassava(Manihot esculenta)显示文摘Cassava is a staple food, feed and bioenergy crop important to the world especially in the tropics.Domesticated cassava is characterized by powerful carbohydrate accumulation but its wild progenitor is not.Here, we investigated the transcriptional differences of eight c DNA libraries derived from developing leaf, stem and storage root of cassava cv. Arg7 and an ancestor line,W14, using next generation sequencing system. A total of41302 assembled transcripts were obtained and from these,25961 transcripts with FPKM≥3 in at least one library were named the expressed genes. A total of 2117, 1963 and3584 transcripts were found to be differentially expressed in leaf, stem and storage root(150 d after planting),respectively, between Arg7 and W14 and ascribed to 103,93 and 119 important pathways in leaf, stem and storage root, respectively. The highlight of this work is that the genes involved in light response, such as those for photosystem I(PSA) and photosystem II(PSB), other genes involved in light harvesting, and some of the genes in the Calvin cycle of carbon fixation were specially upregulated in leaf. Genes for transport and also for key rate-limiting enzymes(PFK, PGK and PK, GAPDH)coupling ATP consumption in glycolysis pathway were predominantly expressed in stem, and genes for sucrose degradation(INVs), amylose synthesis(GBSS) and hydrolysis(RCP1, AMYs), the three key steps of starch metabolism, and transport associated with energy translocation(ABC, AVPs and ATPase) and their upstream transcription factors had enhanced expression in storage root in domesticated cassava. 16Abnormal corrosion behavior of dual-main phase sintered(Ce,Nd)-Fe-B magnets in different sodium solutions显示文摘Sintered(Ce,Nd)-Fe-B magnets with high cost performance were prepared by substituting of Ce for Nd with the dual-main phase method.The corrosion behaviors of dual-main phase sintered(CexNd1-x)30Fe69B1(x=0.15 wt%,0.3 wt%) magnets,which were named as Ce15 and Ce30,were studied in 3.5 wt% NaCl,6.6 wt% Na2 SO4 and 3.0 wt% NaOH aqueous solutions,respectively.The galvanic corrosion of Ce30 is stronger than that of Ce15.However,the mass loss value of Ce30 is smaller than Ce15 after free corrosion for 33 d in NaCl solution.In the other two solutions,all the results show that the corrosion resistance of Ce30 is better than that of Ce15.The scanning electron microscope results show that the crack and shedding of the magnetic phase of Ce30 is less than that of Ce15.Combined above,the abnormal corrosion behavior shows that Ce30 magnets perform better corrosion resistance than Ce15,and the degradation degree of magnetic phase mainly affects the corrosion resistance of magnets.Xiaoning Shi Minggang Zhu Xiangdong Wang Shaoxiong Zhou Xin Wang Meiling Zhang Wei Li 2020Journal of Rare Earths2020,38,7:2

18Termination of Transcription of LAT Increases the Amounts of ICP0 mRNA but Does Not Alter the Course of HSV-1 Infection in Latently Infected Murine Ganglia显示文摘On entering sensory ganglia,herpes simplex viruses 1(HSV-1)establishes a latent infection with the synthesis of a latency associated transcript(LAT)or initiates productive infection with expression of a set of immediate early viral proteins.The precise mechanisms how expression of a genes is suppressed during the latency are unknown.One mechanism that has been proposed is illustrated in the case of ICP0,a key immediate early viral regulatory protein.Specifically,the 2 kb LAT intron is complementary to the 30 terminal portion of ICP0 m RNA.To test the hypothesis that accumulation of LAT negatively affects the accumulation of ICP0 m RNA,we inserted a DNA fragment encoding two poly(A)sequences into LAT to early terminate LAT transcript without interrupting the complementary sequence of ICP0 transcript(named as SR1603).Comparisons of the parent(SR1601)and mutant(SR1603)HSV-1 viruses showed the following:Neurons harboring latent SR1603 virus accumulated equivalent amounts of viral DNA but higher amounts of ICP0 m RNA and lower amounts of LAT,when compared to neurons harboring the SR1601 virus.One notable difference between the two viruses is that viral RNA accumulation in explanted ganglia harboring SR1603 virus initiated significantly sooner than that in neurons harboring SR1601 virus,suggesting that ICP0 may act as an activator of viral gene expression in permissive cells.Collectively,these data suggest that increased ICP0 m RNA by suppressed LAT did not affect the establishment of latency in latently infected murine ganglia.Haifang Jiang Jiaming Wu Xianjie Liu Ruitao Lu Manling Zhou Meiling Chen Yonghong Liu Grace Guoying Zhou Wenmin Fu 2021Virologica Sinica2021,36,2:2
