| 2 | A novel mutation in the MITF may be digenic with GJB2 mutations in a large Chinese family of Waardenburg syndrome type Ⅱ显示文摘Waardenburg syndrome typeⅡ(WS2) is associated with syndromic deafness.A subset of WS2,WS2A,accounting for approximately 15%of patients,is attributed to mutations in the microphthalmia-associated transcription factor(MITF) gene.We examined the genetic basis of WS2 in a large Chinese family.All 9 exons of the MITF gene,the single coding exon(exon 2) of the most common hereditary deafness gene GJB2 and the mitochondrial DNA(mtDNA) 12S rRNA were sequenced.A novel heterozygous mutation c.[742_743delAAinsT;746_747delCA]in exon 8 of the MITF gene co-segregates with WS2 in the family.The MITF mutation results in a premature termination codon and a truncated MITF protein with only 247 of the 419 wild type amino acids.The deaf proband had this MITF gene heterozygous mutation as well as a c.[109G>A]+ [235delC]compound heterozygous pathogenic mutation in the GJB2 gene.No pathogenic mutation was found in mtDNA 12S rRNA in this family.Thus,a novel compound heterozygous mutation,c.[742_743delAAinsT;746_747delCA]in MITF exon 8 was the key genetic reason for WS2 in this family,and a digenic effect of MITF and GJB2 genes may contribute to deafness of the proband. | Xukun Yan Tianyu Zhang Zhengmin Wang Yi Jiang Yan Chen Hongyan Wang Duan Ma Lei Wang Huawei Li | 2011 | Journal of Genetics and Genomics2011,38,12: | 8 |
| 3 | Immunogenicity of recombinant fowl-pox virus co-expressing structural protein precursor P1-2A and proteinase 3C of FMDV显示文摘Two recombinant plasmids, pUTA2P1 and pUTAL3CP1, were constructed by inserting structural pro-tein precursor P1-2A and proteinase 3C of foot-and-mouth disease virus (FMDV) into fowl-pox virus (FPV) recombi-nant vectors pUTA-2 and pUTA-16-LacZ respectively, and two recombinant FPVs (vUTA2P1 and vUTAL3CP1) screened by the RT-PCR, IFA assay and Western blotting assay were obtained successfully. Mice injected respectively with rFPVs were induced high level specific anti-FMDV an-tibodies, increasing of T subtypes, and higher cytotoxicities of splenocytes than those of control groups. | 4 | Pharmacokinetics and Pharmacodynamics of Subcutaneous Single Doses of Pegylated Human G-CSF Mutant(PEG30-rhG-CSF) in Beagle Dogs显示文摘OBJECTIVE To compare the pharmacokinetics and pharmacodynamics of PEG30-rhG-CSF administered to beagle dogs at three different dosages with PEG20-rhG-CSF administered at one dosage,and to provide an experimental basis for clinical trials.METHODS Beagle dogs received single,subcutaneous doses of PEG30-rhG-CSF at 100,200 and 400 μg/kg or PEG20-rhG-CSF at 200 μg/kg.PEG30-rhG-CSF and PEG20-rhG-CSF concentrations in serum were analyzed using an enzyme-linked immunosorbent assay(ELISA).WBC,ANC and PLT counts of whole blood samples were measured using fully automated analytic instrumentation.Pharmacokinetic and pharmacodynamic parameters were calculated using DAS 2.0 statistical analysis so ware.RESULTS The pharmacokinetic parameters of PEG30-rhG-CSF calculated from the serum concentration data determined by ELISA were as follows:the mean elimination half-life(t1/2ke) was 40.6 h(33.5~45.4 h);the mean time to reach peak concentration(Tmax) was 19.2 h(11.7~24.0 h);the drug clearance from the serum(CL) was decreased with increasing doses;the peak concentration(Cmax) and the area under the serum concentration-time curve(AUC) were increased with increasing doses.For PEG20-rhG-CSF,the half-life was shorter(12 h) and Tmax was achieved much earlier(10 h) relative to PEG30-rhG-CSF.The AUC of PEG30-rhG-CSF was much greater than that of PEG20-rhG-CSF,and the relative bioavailability with a subcutaneous injection was 158.7%.Administration of single doses of PEG30-rhG-CSF resulted in substantial increases in the absolute neutrophil count(ANC).The time to reach ANCmax(ANCTmax) was 72 h.The maximum observed absolute neutrophil counts(ANCmax) and the area over the baseline effect curve(AOBEC) was increased with increasing doses.The effect-elimination half-life(t1/2E) ranged from 60 h to 80 h a er subcutaneous administration.The PLT count was slightly elevated 8~12 h a er s.c.injection,and declined a er 24 h.CONCLUSION The mean elimination half-life of PEG30-rhG-CSF was longer than that of PEG20-rhG-CSF at the same dose,and the other main pharmacokinetic and pharmacodynamic parameters of PEG30-rhG-CSF,including Cmax,ANCmax,AUC and AOBEC were much greater than those following PEG20-rhG-CSF injection. | Yongming Cai Zhengmin Chen Ling Jiang Ming Li Changxiao Liu | 2008 | Chinese Journal of Clinical Oncology2008,5,5: | 2 |
| 5 | Ambient air pollution and low temperature associated with case fatality of COVID-19: A nationwide retrospective cohort study in China显示文摘The evidence for the effects of environmental factors on COVID-19 case fatality remains controversial,and it is crucial to understand the role of preventable environmental factors in driving COVID-19 fatality.We thus conducted a nationwide cohort study to estimate the effects of environmental factors(temperature,particulate matter[PM2.5,PM10],sulfur dioxide[SO2],nitrogen dioxide[NO2],and ozone[O3])on COVID-19 case fatality.A total of 71,808 confirmed COVID-19 cases were identified and followed up for their vital status through April 25,2020.Exposures to ambient air pollution and temperature were estimated by linking the city-and county-level monitoring data to the residential community of each participant.For each participant,two windows were defined:the period from symptom onset to diagnosis(exposure window I)and the period from diagnosis date to date of death/recovery or end of the study period(exposure window II).Cox proportional hazards models were used to estimate the associations between these environmental factors and COVID-19 case fatality.COVID-19 case fatality increased in association with environmental factors for the two exposure windows.For example,each 10 mg/m^(3) increase in PM2.5,PM10,O3,and NO2 in window I was associated with a hazard ratio of 1.11(95%CI 1.09,1.13),1.10(95%CI 1.08,1.13),1.09(95 CI 1.03,1.14),and 1.27(95%CI 1.19,1.35)for COVID-19 fatality,respectively.A significant effect was also observed for low temperature,with a hazard ratio of 1.03(95%CI 1.01,1.04)for COVID-19 case fatality per 1C decrease.Subgroup analysis indicated that these effects were stronger in the elderly,as well as in those with mild symptoms and living in Wuhan or Hubei.Overall,the sensitivity analyses also yielded consistent estimates.Short-term exposure to ambient air pollution and low temperature during the illness would play a nonnegligible part in causing case fatality due to COVID-19.Reduced exposures to high concentrations of PM2.5,PM10,O3,SO2,and NO2 and low temperature would help improve the prognosis and reduce public health burden. | Fei Tian Xiaobo Liu Qingchen Chao Zhengmin(Min)Qian Siqi Zhang Li Qi Yanlin Niu Lauren DArnold Shiyu Zhang Huan Li Hualiang Lin Qiyong Liu | 2021 | The Innovation2021,2,3: | 2 |
