| 1 | Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. | 2 | Effects of electroacupuncture and Chinese kidney-nourishing medicine on polycystic ovary syndrome in obese patients显示文摘OBJECTIVE: To explore the effect of electroacupuncture and Chinese kidney-nourishing medicine on insulin (INS), adiponectin (APN), leptin (LEP), and glucolipid metabolism of obese patients with polycystic ovary syndrome (PCOS). METHODS: Sixty-seven obese PCOS patients were randomly divided into two groups. Thirty-three patients in the acupuncture-medicine group were treated three times a week with electroacupuncture at the Tianshu (ST 25), Zhongwan (CV 12), Qihai (CV 6), Sanyinjiao (SP 6), Geshu (BL 17), and Cil-iao (BL 32) acupoints. They also took the Chinese drug, Tiankui capsule, for 3 months as a course of treatment. Point-taking and treatment in the electroacupuncture group of 34 patients was the same as those in the acupuncture-medicine group. We observed and compared the changes in the obesity-related indexes of body weight (BW), body mass index (BMI), and waist-hip ratio (WHR), as well as fasting plasma glucose (FPG), fasting insulin (FINS), APN, and LEP. RESULTS: BW, BMI, WHR, and FINS decreased and insulin sensitivity index (ISI) and APN were higher in the acupuncture-medicine group than in the electroacupuncture group (P<0.01). There was no obvious difference in LEP between the two groups (P>0.05). CONCLUSION: Acupuncture combined with medicine is better than just electroacupuncture for obese PCOS patients by improving obesity-related indexes, insulin sensitivity, and APN level. | 5 | Characteristics of Boundary Layer Structure during a Persistent Haze Event in the Central Liaoning City Cluster, Northeast China显示文摘The characteristics of boundary layer structure during a persistent regional haze event over the central Liaoning city cluster of Northeast China from 16 to 21 December 2016 were investigated based on the measurements of particulate matter(PM) concentration and the meteorological data within the atmospheric boundary layer(ABL). During the observational period, the maximum hourly mean PM_(2.5) and PM10 concentrations in Shenyang, Anshan, Fushun, and Benxi ranged from 276 to 355 μg m–3 and from 378 to 442 μg m–3, respectively, and the lowest hourly mean atmospheric visibility(VIS) in different cities ranged from 0.14 to 0.64 km. The central Liaoning city cluster was located in the front of a slowly moving high pressure and was mainly controlled by southerly winds. Wind speed(WS) within the ABL(< 2 km) decreased significantly and WS at 10-m height mostly remained below 2 m s–1 during the hazy episodes, which was favorable for the accumulation of air pollutants. A potential temperature inversion layer existed throughout the entire ABL during the earlier hazy episode [from 0500 Local Time(LT) 18 December to 1100 LT 19 December], and then a potential temperature inversion layer developed with the bottom gradually decreased from 900 m to 300 m. Such a stable atmospheric stratification further weakened pollutant dispersion. The atmospheric boundary layer height(ABLH) estimated based on potential temperature profiles was mostly lower than 400 m and varied oppositely with PM_(2.5) in Shenyang. | 8 | Orbital-scale Asian summer monsoon variations:Paradox and exploration显示文摘The Asian summer monsoon(ASM) is a vast climate system, whose variability is critical to the livelihoods of billions of people across the Asian continent. During the past half-century, much progress has been made in understanding variations on a wide range of timescales, yet several significant issues remain unresolved. Of note are two long-standing problems concerning orbital-scale variations of the ASM.(1) Chinese loess magnetic susceptibility records show a persistent glacial-interglacial dominated ~100 kyr(thousand years) periodicity, while the cave oxygen-isotope(δ18 O) records reveal periodicity in an almost pure precession band(~20 kyr periodicity)—the 'Chinese 100 kyr problem'.(2) ASM records from the Arabian Sea and other oceans surrounding the Asian continent show a significant lag of 8–10 kyr to Northern Hemisphere summer insolation(NHSI), whereas the Asian cave δ18 O records follow NHSI without a significant lag—a discrepancy termed the 'sea-land precession-phase paradox'. How can we reconcile these differences? Recent and more refined model simulations now provide spatial patterns of rainfall and wind across the precession cycle, revealing distinct regional divergences in the ASM domain, which can well explain a large portion of the disparities between the loess, marine, and cave proxy records. Overall, we also find that the loess, marine, and cave records are indeed complementary rather than incompatible, with each record preferentially describing a certain aspect of ASM dynamics. | 10 | Comparison of Two Air Pollution Episodes over Northeast China in Winter 2016/17 Using Ground-Based Lidar显示文摘This study analyzes and compares aerosol properties and meteorological conditions during two air pollution episodes in 19–22(E1) and 25–26(E2) December 2016 in Northeast China. The visibility, particulate matter(PM) mass concentration, and surface meteorological observations were examined, together with the planetary boundary layer(PBL) properties and vertical profiles of aerosol extinction coefficient and volume depolarization ratio that were measured by a ground-based lidar in Shenyang of Liaoning Province, China during December 2016–January 2017.Results suggest that the low PBL height led to poor pollution dilution in E1, while the high PBL accompanied by low visibility in E2 might have been due to cross-regional and vertical air transmission. The PM mass concentration decreased as the PBL height increased in E1 while these two variables were positively correlated in E2. The enhanced winds in E2 diffused the pollutants and contributed largely to the aerosol transport. Strong temperature inversion in E1 resulted in increased PM2.5 and PM10 concentrations, and the winds in E2 favoured the southwesterly transport of aerosols from the North China Plain into the region surrounding Shenyang. The large extinction coefficient was partially attributed to the local pollution under the low PBL with high ground-surface PM mass concentrations in E1,whereas the cross-regional transport of aerosols within a high PBL and the low PM mass concentration near the ground in E2 were associated with severe aerosol extinction at high altitudes. In the present work, an electroconductive nanocrystalline cellulose(NCC) film filled with TiO2-reducedgraphene oxide(TiO2-RGO) was developed. Initially, graphene oxide(GO) was prepared using the modified Hummers method and thereafter photocatalytically reduced using TiO2 as a catalyst. Subsequently, an electro-conductive NCC film was prepared via vacuum filtration with the as-prepared TiO2-RGO nanocomposite as a functional filler. The TiO2-RGO nanocomposite and the NCC/TiO2-RGO film were systematically characterized. The results showed that the obtained TiO2-RGO nanocomposite exhibited reduced oxygen-containing group content and enhanced electroconductivity as compared with those of GO. Moreover, the NCC film filled with TiO2-RGO nanocomposite displayed an electro-conductivity of up to 9.3 S/m and improved mechanical properties compared with that of the control. | 15 | Propofol attenuates high glucose-induced P66shc expression in human umbilical vein endothelial cells through Sirt1显示文摘Perioperative hyperglycemia is a common metabolic disorder in clinic settings.Hyperglycemia leads to endothelial inflammation,endothelial cell apoptosis,and dysfunction,thus resulting in endothelial injury.Propofol(2,6-diisopropylphenol)is a widely used intravenous anesthetic in clinic settings.Our previous study indicated that propofol inhibits mitochondrial reactive oxygen species(ROS)production via down-regulation of phosphatase A2(PP2A)expression,inhibition of Ser36-p66shc dephosphorylation and mitochondrial translocation,thus improving high glucose-induced endothelial injury.The expression of p66shc was inhibited by propofol in hyperglycemic human umbilical vein endothelial cells(HUVECs).However,the mechanism by which propofol inhibits p66shc expression in hyperglycemic HUVECs is still obscure.In the present study,we mainly examined how propofol inhibited high glucose-induced p66shc expression in HUVECs.Compared with 5 mM glucose treatment,high glucose increased p66shc expression and decreased sirt1 expression,which was inhibited by propofol treatment.Moreover,EX527(a sirt1 inhibitor)reversed the effect of propofol against high glucose-induced p66shc expression.However,EX527 did not reverse the effects of propofol against high glucose-induced ROS accumulation,endothelial inflammation,and apoptosis.Furthermore,when cells were incubated with propofol,EX527,and FTY720(a PP2A activator)simultaneously,the effects of propofol against high glucose-induced ROS accumulation,inflammation,and apoptosis were reversed.Our results suggested that propofol inhibited high glucose-induced p66shc expression via upregulation of sirt1 expression in hyperglycemic HUVECs.Moreover,propofol protects against high glucose-mediated ROS accumulation and endothelial injury via both inhibition of p66shc expression and dephosphorylation of Ser36-p66shc. | Jing Wang Jie Qi Qichao Wu Hui Jiang Yuehao Yin Yan Huanx Yanjun Zhao Minmin Zhu | 2019 | Acta Biochimica et Biophysica Sinica2019,51,2: | 5 |
| 16 | A Virulent PEDV Strain FJzz1 with Genomic Mutations and Deletions at the High Passage Level Was Attenuated in Piglets via Serial Passage In Vitro显示文摘Highly virulent porcine epidemic diarrhea virus(PEDV)strains re-emerged and circulated in China at the end of 2010,causing significant economic losses in the pork industry worldwide.To understand the genetic dynamics of PEDV during its passage in vitro,the PEDV G2 strain FJzzl was serially propagated in Vero cells for up to 200 passages.The susceptibility and adaptability of the FJzzl strain increased gradually as it was serially passaged in vitro.Sequence analysis revealed that amino acid(aa)changes were mainly concentrated in the S glycoprotein,which accounted for 72.22%-85.71%of all aa changes.A continuous aa deletion(^(55)I^(56)G^(57)E→^(55)K^(56)Δ^(57)Δ)occurred in the N-terminal domain of S1(Sl-NTD).To examine how the aa changes affected its virulence,FJzzl-F20 and FJzzl-F200 were selected to simultaneously evaluate their pathogenicity in suckling piglets.All the piglets in the FJzzl-F20-infected group showed typical diarrhea at 24 h postinfection,and the piglets died successively by 48 h postinfection.However,the clinical signs of the piglets in the FJzzl-F200-infected group were significantly weaker,and no deaths occurred.The FJzzl-F200-infected group also showed a lower level of fecal viral shedding and lower viral loads in the intestinal tissues,and no obvious histopathological lesions.TypeⅠandⅢinterferon were induced in the FJzzl-F200 infection group,together with pro-inflammatory cytokines,such as TNF-α,IL-1βand IL-8.These results indicate that the identified genetic changes may contribute to the attenuation of FJzzl strain,and the attenuated FJzzl-F200 may have the potential for developing PEDV live-attenuated vaccines. | Pengfei Chen Xiongwei Zhao Shuting Zhou Tianxing Zhou Xiangmei Tan Xia Wu Wu Tong Fei Gao Lingxue Yu Yifeng Jiang Hai Yu Zhibiao Yang Guangzhi Tong Yanjun Zhou | 2021 | Virologica Sinica2021,36,5: | 4 |

| 19 | Identification of genes for drought resistance and prediction of gene candidates in soybean seedlings based on linkage and association mapping显示文摘Drought is one of the primary abiotic stress factors affecting the yield,growth,and development of soybeans.In extreme cases,drought can reduce yield by more than 50%.The seedling stage is an important determinant of soybean growth:the number and vigor of seedlings will affect growth and yield at harvest.Therefore,it is important to study the drought resistance of soybean seedlings.In this study,a recombinant inbred line(RIL)population comprising 234 F_(6:10)lines(derived from Zhonghuang35×Jindou 21)and a panel of 259 soybean accessions was subjected to drought conditions to identify the effects on phenotypic traits under these conditions.Using a genetic map constructed by single nucleotide polymorphism(SNPs)markers,18 quantitative trait loci(QTL)on 7 soybean chromosomes were identified in two environments.This included 9 QTL clusters identified in the RIL population.Fifty-three QTL were identified in 19 soybean chromosomes by genome-wide association analysis(GWAS)in the panel of accessions,including 69 significant SNPs(-log_(10)(P)≥3.97).A combination of the two populations revealed that two SNPs(-log_(10)(P)≥3.0)fell within two of the QTL(qPH7-4 and qPH7-6)confidence intervals.We not only re-located several previously reported drought-resistance genes in soybean and other crops but also identified several non-synonymous stress-related mutation site differences between the two parents,involving Glyma.07 g093000,Glyma.07 g093200,Glyma.07 g094100 and Glyma.07 g094200.One previously unreported new gene related to drought stress,Glyma.07 g094200,was found by regional association analysis.The significant SNP CHR7-17619(G/T)was within an exon of the Glyma.07 g094200 gene.In the RIL population,the DSP value of the'T'allele of CHR7-17619 was significantly(P<0.05)larger than the'G'allele in different environments.The results of our study lay the groundwork for cloning and molecular marker-assisted selection of droughtresistance genes in soybeans at the seedling stage. | Yanjun Zhang Zhangxiong Liu Xingrong Wang Yue Li Yongsheng Li Zuowang Gou Xingzhen Zhao Huilong Hong Honglei Ren Xusheng Qi Lijuan Qiu | 2022 | The Crop Journal2022,10,3: | 3 |
