1A shear-thinning,ROS-scavenging hydrogel combined with dental pulp stem cells promotes spinal cord repair by inhibiting ferroptosis显示文摘Spinal cord injury(SCI)is a serious clinical disease.Due to the deformability and fragility of the spinal cord,overly rigid hydrogels cannot be used to treat SCI.Hence,we used TPA and Laponite to develop a hydrogel with shear-thinning ability.This hydrogel exhibits good deformation,allowing it to match the physical properties of the spinal cord;additionally,this hydrogel scavenges ROS well,allowing it to inhibit the lipid peroxidation caused by ferroptosis.According to the in vivo studies,the TPA@Laponite hydrogel could synergistically inhibit ferroptosis by improving vascular function and regulating iron metabolism.In addition,dental pulp stem cells(DPSCs)were introduced into the TPA@Laponite hydrogel to regulate the ratios of excitatory and inhibitory synapses.It was shown that this combination biomaterial effectively reduced muscle spasms and promoted recovery from SCI.Yibo Ying Zhiyang Huang Yurong Tu Qiuji Wu Zhaoyu Li Yifan Zhang Huilei Yu Annian Zeng Hanzhi Huang Jiahui Ye Weiyang Ying Min Chen Zhiyi Feng Ziyue Xiang Qingsong Ye Sipin Zhu Zhouguang Wang 2023Bioactive Materials2023,,4:2
2Stannous oxalate: An efficient catalyst for poly(trimethylene terephthalate) synthesis显示文摘A complete study on the catalytic activity of stannous oxalate for poly(trimethylene terephthalate) (PTT) synthesis via esterification method is carried out by comparison to the well known catalysts (tetrabutyl titanate (TBT), dibutyltin oxide (Bu2SnO), and stannous octoate (SOC)). Their catalytic activity in the esterification process is monitored by measuring the amount of water generated, while intrinsic viscosity (IV) and content of terminal carboxyl groups (CTCG) are used as the index in the polycondensation process. Stannous oxalate shows higher activity than the other catalysts. Decrease in reaction time and improvements in PTT property are observed. 10Detection of Tumor Suppressor Gene and Oncogene in SO-Rb_(50) Human Retinoblastoma Cell Line显示文摘Retinoblastoma (Rb) is the most common malignant'cancer of eye. So-Rb_(50) is the first Rb cell line established in China in 1988. It has passed to the 387th passage now. We collected cells of the 327th passage of SO-Rb_(50), purified its genomic DNA and detected it with Rb and c-myc cDNA probes respectively (normal human white blood cells DNA was the control). 12Construction method of quantitative evaluation model for the maturity of Korla fragrant pear显示文摘The maturity variation laws of Korla fragrant pears were explored for a quantitative evaluation of harvest maturity to solve the reasonable matching between harvest maturity of Korla fragrant pears and market quality demands.Korla fragrant pears from different harvesting periods were chosen as the research objects.Some quality indexes were chosen as the evaluation indexes per industry standards,including hardness,soluble solid content(SSC),single-fruit weight,fruit longitudinal diameter,fruit equatorial diameter,pericarp color parameters(L*,a*,and b*),and titratable acid.Variation data of these quality indexes with accumulated temperature were collected.Scores of several quality indexes were gained through principal component analysis.A mathematical model of scores and accumulated temperatures was constructed.On this basis,a quantitative maturity model of Korla fragrant pears was constructed.Results demonstrate that SSC,single-fruit weight,fruit longitudinal diameter,fruit equatorial diameter,L*,a*,and b*are significantly and positively correlated with the accumulated temperature.Meanwhile,hardness and titratable acid showed significant negative correlations with the accumulated temperature.Relations between scores of principal components and accumulated temperature conform to the Sigmoidal model.The constructed quantitative maturity model of Korla fragrant pears can quantify the maturity of pears.Research conclusions can provide insight into the harvest periods,evaluate Korla fragrant pears’maturity,and lay a theoretical foundation for quantitative research on fruit maturity.Shihui Yu Yurong Tang Haipeng Lan Xiaolong Li Hong Zhang Yong Zeng Hao Niu Xiaozhen Jin Yang Liu 2022International Journal of Agricultural and Biological Engineering2022,15,4:0
13RNA sequencing-based optimization of biological lipid droplets for sonodynamic therapy to reverse tumor hypoxia and elicit robust immune response显示文摘Mitochondria-targeted sonodynamic therapy(SDT)is a promising strategy to inhibit tumor growth and activate the anti-tumor immune responses.Identifying the mechanisms underlying mitochondria-targeted SDT,further optimizing its efficacy,developing novel sonosensitizer carriers with good biocompatibility pose major challenges to the clinical practice of SDT.In this study,we investigated the mechanisms of mitochondria-targeted SDT and demonstrated that it suppressed the mitochondrial electron transport chain(ETC)in pancreatic cancer cells through RNA-sequencing analysis.Based on these findings,we constructed the functional lipid droplets(LDs)(CPI-613/IR780@LDs),which combined mitochondria-targeted SDT with the tricarboxylic acid(TCA)cycle inhibitor CPI-613.CPI-613/IR780@LDs synergistically inhibited the TCA cycle and the ETC of mitochondrial aerobic respiration to reduce oxygen consumption and increase reactive oxygen species(ROS)generation at the tumor site,thus enhancing the efficacy of SDT in hypoxic pancreatic cancer.Moreover,the combination of mitochondria-targeted SDT and anti-PD-1 antibody exhibited excellent tumor inhibition and activated anti-tumor immune responses by increasing tumorinfiltrating CD8+T cells and reducing regulatory T cells,synergistically arresting the growth of both primary and metastatic pancreatic tumors.Meanwhile,lipid droplets are cell-derived biological carriers with natural mitochondrial targeting ability and can achieve efficient hydrophobic drug loading through active phagocytosis.Therefore,the functional lipid droplet-based SDT combined with anti-PD-1 antibody holds great potential in the clinical treatment of hypoxic pancreatic cancer.Zhan Shi Xue Wang Jiali Luo Yiqing Zeng Qing Wen Yurong Hong Tao Zhang Pintong Huang 2023Nano Research2023,16,5:0
