1Development of a Haploid-Inducer Mediated Genome Editing System for Accelerating Maize Breeding显示文摘Crop breeding aims to generate pure in bred lines with multiple desired traits. Doubled haploid (DH) and genome editing using CRISPR/Cas9 are two powerful game-changing technologies in crop breeding. However, both of them still fall short for rapid generation of pure elite lines with integrated favorable traits. Here, we report the development of a Haploid-Inducer Mediated Genome Editing (IMGE) approach, which utilizes a maize haploid inducer line carrying a CRISPR/Cas9 cassette targeting for a desired agronomic trait to pollinate an elite maize in bred line and to generate genome-edited haploids in the elite maize background. 35-Hydroxymethylome in Circulating Cell-free DNA as A Potential Biomarker for Non-small-cell Lung Cancer显示文摘Non-small-cell lung cancer(NSCLC), the most common type of lung cancer accounting for 85% of the cases, is often diagnosed at advanced stages owing to the lack of efficient early diagnostic tools. 5-Hydroxymethylcytosine(5 hmC) signatures in circulating cell-free DNA(cfDNA) that carries the cancer-specific epigenetic patterns may represent the valuable biomarkers for discriminating tumor and healthy individuals, and thus could be potentially useful for NSCLC diagnosis. Here,we employed a sensitive and reliable method to map genome-wide 5 hmC in the cfDNA of Chinese NSCLC patients and detected a significant 5 hmC gain in both the gene bodies and promoter regions in the blood samples from tumor patients compared with healthy controls. Specifically, we identified six potential biomarkers from 66 patients and 67 healthy controls(mean decrease accuracy>3.2, P < 3.68E-19) using machine-learning-based tumor classifiers with high accuracy. 5Fabrication and characterization of novel meso-porous carbon/n-octadecane as form-stable phase change materials for enhancement of phase-change behavior显示文摘In this study, series of novel composite phase change materials(PCMs) were prepared through vacuum impregnation by using meso-porous carbon as a supporting matrix and n-octadcane as PCMs.The meso-porous carbon material was prepared through one-pot co-assembly method, using resorcinol and formaldehyde as carbon precursor, tetraethoxysilane as silica sources and triblock copolymer F127 as a template. And the phase behaviors of n-octadcane confined in the nano-porous structure of the meso-porous carbon were further investigated. Fourier transform-infrared spectroscopy spectra show that n-octadecane was effectively encapsulated in the porous structure of mesoporous carbon and the composite PCMs were successfully prepared. Differential scanning calorimetry results confirm that the composite PCMs possess a good phase change behavior, fast thermal-response rate and excellent thermal cycling stability. In addition, the composite PCMs possess expected heat storage and heat release properties. 6All-climate aqueous supercapacitor enabled by a deep eutectic solvent electrolyte based on salt hydrate显示文摘Aqueous supercapacitors(SCs)have received considerable attention owing to the utilization of low-cost,non-flammable,and low-toxicity aqueous electrolytes thus could eliminate the safety and cost concerns,but their wide temperature range applications have generally suffered from frozen of electrolyte and insufficient ionic conductivity at low temperatures.Herein,we demonstrate the feasibility of using an unconventional Deep Eutectic Solvent(DES)based on H2O-Mg(ClO4)2·6 H2O binary system as electrolyte to construct all-climate aqueous carbon-based SC.This unconventional class DES completely base on inorganic substances and achieving simply mix inexpensive salts and water together at the right proportions.Attributed to the attractive feature of extremely low freeze temperature of-69℃,this electrolyte can enable the 1.8 V carbon-based SC to fully work at-40℃with outstanding cycling stability.This DES electrolyte comprising of a single salt and a single solvent without any additive will open up an avenue for developing simple and green electrolytes to construct all-climate SC.Xudong Bu Yurong Zhang Yinglun Sun Lijun Su Jianing Meng Xionggang Lu Xingbin Yan 2020Journal of Energy Chemistry2020,29,10:2

17China’s Recent Progresses in Polar Climate Change and Its Interactions with the Global Climate System显示文摘During the recent four decades since 1980,a series of modern climate satellites were launched,allowing for the measurement and record-keeping of multiple climate parameters,especially over the polar regions where traditional observations are difficult to obtain.China has been actively engaging in polar expeditions.Many observations were conducted during this period,accompanied by improved Earth climate models,leading to a series of insightful understandings concerning Arctic and Antarctic climate changes.Here,we review the recent progress China has made concerning Arctic and Antarctic climate change research over the past decade.The Arctic temperature increase is much higher than the global-mean warming rate,associated with a rapid decline in sea ice,a phenomenon called the Arctic Amplification.The Antarctic climate changes showed a zonally asymmetric pattern over the past four decades,with most of the fastest changes occurring over West Antarctica and the Antarctic Peninsula.The Arctic and Antarctic climate changes were driven by anthropogenic greenhouse gas emissions and ozone loss,while tropical-polar teleconnections play important roles in driving the regional climate changes and extreme events over the polar regions.Polar climate changes may also feedback to the entire Earth climate system.The adjustment of the circulation in both the troposphere and the stratosphere contributed to the interactions between the polar climate changes and lower latitudes.Climate change has also driven rapid Arctic and Southern ocean acidification.Chinese researchers have made a series of advances in understanding these processes,as reviewed in this paper.Xichen LI Xianyao CHEN Bingyi WU Xiao CHENG Minghu DING Ruibo LEI Di QI Qizhen SUN Xiaoyu WANG Wenli ZHONG Lei ZHENG Meijiao XIN Xiaocen SHEN Chentao SONG Yurong HOU 2023Advances in Atmospheric Sciences2023,40,8:1

19Role of Elevated Thrombospondin-1 in Kainic Acid-Induced Status Epilepticus显示文摘Previous studies have suggested that thrombospondin-1(TSP-1) regulates the transforming growth factor beta 1(TGF-b1)/phosphorylated Smad2/3(p Smad2/3) pathway. Moreover, TSP-1 is closely associated with epilepsy. However, the role of the TSP-1-regulated TGFb1/p Smad2/3 pathway in seizures remains unclear. In this study, changes in this pathway were assessed following kainic acid(KA)-induced status epilepticus(SE) in rats.The results showed that increases in the TSP-1/TGF-b1/p Smad2/3 levels spatially and temporally matched the increases in glial fibrillary acidic protein(GFAP)/chondroitin sulfate(CS56) levels following KA administration.Inhibition of TSP-1 expression by small interfering RNA or inhibition of TGF-b1 activation with a Leu-Ser-Lys-Leu peptide significantly reduced the severity of KA-induced acute seizures. These anti-seizure effects were accompanied by decreased GFAP/CS56 expression and Smad2/3 phosphorylation. Moreover, inhibiting Smad2/3 phosphorylation with ponatinib or SIS3 also significantly reduced seizure severity, alongside reducing GFAP/CS56 immunoreactivity.
