| 2 | Colonization of endophyte Acremonium sp. D212 in Panax notoginseng and rice mediated by auxin and jasmonic acid显示文摘Endophytic fungi can be beneficial to plant growth. However, the molecular mechanisms under-lying colonization of Acremonium spp. remain unclear. In this study, a novel endophytic Acremonium strain was isolated from the buds of Panax notoginseng and named Acremonium sp. D212. The Acremonium sp. D212 could colonize the roots of P. notoginseng, enhance the resistance of P. notoginseng to root rot disease, and promote root growth and saponin bio-synthesis in P. notoginseng. Acremonium sp. D212 could secrete indole-3-acetic acid (IAA) and jasmonic acid (JA), and inoculation with the fungus increased the endogenous levels of IAA and JA in P. noto-ginseng. Colonization of the Acremonium sp. D212 in the roots of the rice line Nipponbare was dependent on the concentration of methyl jasmonate (MeJA) (2–15μmol/L) and 1-naphthalenacetic acid (NAA) (10–20μmol/L). Moreover, the roots of the JA signaling-defective coi1-18 mutant were colonized by Acremonium sp. D212 to a lesser degree than those of the wild-type Nipponbare and miR393b-overexpressing lines, and the colonization was res-cued by MeJA but not by NAA. It suggests that the cross-talk between JA signaling and the auxin biosynthetic pathway plays a crucial role in the colonization of Acremonium sp. | 4 | Determination of paraquat in water samples using a sensitive fluorescent probe titration method显示文摘Paraquat(PQ),a nonselective herbicide,is non-fluorescent in aqueous solutions.Thus,its determination through direct fluorescent methods is not feasible.The supramolecular inclusion interaction of PQ with cucurbit[7]uril was studied by a fluorescent probe titration method.Significant quenching of the fluorescence intensity of the cucurbit[7]uril-coptisine fluorescent probe was observed with the addition of PQ.A new fluorescent probe titration method with high selectivity and sensitivity at the ng/mL level was developed to determine PQ in aqueous solutions with good precision and accuracy based on the significant quenching of the supramolecular complex fluorescence intensity.The proposed method was successfully used in the determination of PQ in lake water,tap water,well water,and ditch water in an agricultural area,with recoveries of 96.73% to 105.77%.The fluorescence quenching values(F) showed a good linear relationship with PQ concentrations from 1.0 × 108 to 1.2× 105 mol/L with a detection limit of 3.35 × 109 mol/L.In addition,the interaction models of the supramolecular complexes formed between the host and the guest were established using theoretical calculations.The interaction mechanism between the cucurbit[7]uril and PQ was confirmed by 1 H NMR spectroscopy. | Feihu Yao Hailong Liu Guangquan Wang Liming Du Xiaofen Yin Yunlong Fu | 2013 | Journal of Environmental Sciences2013,25,6: | 2 |

| 7 | SAMD4 family members suppress human hepatitis B virus by directly binding to the Smaug recognition region of viral RNA显示文摘HBV infection initiates hepatitis B and promotes liver cirrhosis and hepatocellular carcinoma.IFN-α is commonly used in hepatitis B therapy,but how it inhibits HBV is not fully understood.We screened 285 human interferon-stimulated genes(ISGs)for anti-HBV activity using a cell-based assay,which revealed several anti-HBV ISGs.Among these ISGs,SAMD4A was the strongest suppressor of HBV replication.We found the binding site of SAMD4A in HBV RNA,which was a previously unidentified Smaug recognition region(SRE)sequence conserved in HBV variants.SAMD4A binds to the SRE site in viral RNA to trigger its degradation.The SAM domain in SAMD4A is critical for RNA binding and the C-terminal domain of SAMD4A is required for SAMD4A anti-HBV function.Human SAMD4B is a homolog of human SAMD4A but is not an ISG,and the murine genome encodes SAMD4.All these SAMD4 proteins suppressed HBV replication when overexpressed in vitro and in vivo.We also showed that knocking out the Samd4 gene in hepatocytes led to a higher level of HBV replication in mice and AAV-delivered SAMD4A expression reduced the virus titer in HBVproducing transgenic mice.In addition,a database analysis revealed a negative correlation between the levels of SAMD4A/B and HBV in patients.Our data suggest that SAMD4A is an important anti-HBV ISG for use in IFN therapy of hepatitis B and that the levels of SAMD4A/B expression are related to HBV sensitivity in humans. | Yuze Wang Xinrui Fan Yunlong Song Yifei Liu Ruixin Liu Jianfeng Wu Xiaoling Li Quan Yuan Guo Fu Ningshao Xia Jiahuai Han | 2021 | Cellular & Molecular Immunology2021,18,4: | 1 |

For both the underwater and in-air deposition layers, both were well-formed and incomplete fusion, cracks, or other defects did not exist.Compared with the single-track deposition layer in air, the oxidation degree of the underwater single-track deposition layer was slightly higher.In both the underwater and in-air deposition layers, columnar dendrites nucleated close to the fusion line and grew along the direction of the maximum cooling rate in the fusion region(FR), while equiaxed grains formed in the deposited region(DR). As the environment changed from air to water, the width of DR and height of FR decreased, but the deposition angle and height of DR increased. The grain size and ratio of the high-angle boundaries also decreased due to the large cooling rate and low peak temperature in the water environment.Besides, the existence of a water environment benefitted the reduction of magnesium element burning loss in the DR.
