1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 2Effects of AT1 receptor antagonist,Iosartan,on rat hepatic fibrosis induced by CCl_4显示文摘AIM To investigate effect of losartan,an AT1receptor antagonist,on hepatic fibrosis induced byCCl;and to determine whether or not AT1receptors are expressed on hepatic stellate cells,METHODS AND RESULTS Fifty male Sprague-Dawley rats,weighing(180±20)g,wererandomized into five groups(control group,modelgroup,and three losartan treated groups),inwhich all rats were given the subcutaneousinjection of 40% CCl4(every 3 days for 6 weeks)except for rats of control group.Rats of losartan-treated groups were treated with losartan(20 mg/kg,10 mg/kg,5 mg/kg,daily gavage),After 6weeks liver tissue and serum samples of all ratswere examined.Serum hyaluronic acid(HA),procollagen typeⅢ(PCⅢ)were detected byradioimmunoassays,van Giesion collagen stainingwas used to evaluate the extracellular matrix of ratswith liver fibrosis.The expression of AT1receptors,transforming growth factor-beta(TGF-β),and alpha-smooth muscle actin(a-SMA)inliver tissue were determined byimmunohistochemical techniques.Compared withmodel group,serum ALT and AST of losartan-treated groups were significantly reduced(t=4.20,P<0.01 and t=4.57,P<0.01).Serum HAand PCⅢalso had significant differences(t=3.53,P<0.01 and t=2.20,P<0.05).Thedegree of fibrosis was improved by losartan and correlated with the expressions of AT1 receptors,TGF-β,and α-SMA in liver tissue.CONCLUSION AT1 receptor antagonist,losartan,could limit the progression of the hepatic fibrosisinduced by CCl4.The mechanism may be related tothe decrease in the expression of AT1 receptorsand TGF-β,ameliorating the injury of hepatocytes;activation of local renin-angiotensin system mightrelate to hepatic fibrosis;and during progressionof fibrosis,activated hepatic stellate cells mightexpress AT1 receptors.Hong Shan Wei Ding Guo Li Han Ming Lu Yu Tao Zhan Zhi Rong Wang Xin Huang Jing Zhang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,4:42
3Expansion of the Cathaysian Oldland through the Ordovician-Silurian transition:Emerging evidence and possible dynamics显示文摘Located northwest of the Jiangshan-Shaoxing Fault Zone,western and northwestern Zhejiang and northeastern Jiangxi provinces experienced major changes in bio-and litho-facies and paleogeography through the Ordovician-Silurian transition (late Katian,Hirnantian,and early Rhuddanian),as manifested by stratigraphic,paleontologic and synecologic records.Three geographic units under consideration are South,Central,and North areas.The western margin of the South area was occupied by the Huaiyu Mountains,whereas the other two parts were covered by the Zhe-Gan Sea during late Katian (Late Ordovician) time.In the early stage of the Changwu Formation deposition (late Katian),the sea was deepening northeastward,but with shallower conditions in the southwest and deeper conditions in the northeast.During mid to late stages of Changwu Formation deposition (latest Katian),the sea became much shallower and the sea bottom was uplifted substantially,which occurred somewhat prior to the global trend.During the Hirnantian (latest Ordovician) and early Rhuddanian (earliest Silurian),the study region became a shallow bay under expansion of the Cathaysian Oldland.There occurred a major drop of sea level and great changes in benthic biota with the occurrence of many new immigrants through the Ordovician-Silurian transition,which are closely related to a unique regional orogeny.The Yangtze and Cathaysian blocks may have amalgamated within the South China Paleoplate during this interval to cause the continuous uplifting and northwestward expansion of the Cathaysian Oldland.RONG JiaYu ZHAN RenBin XU HongGen HUANG Bing YU GuoHua 2010Science China Earth Sciences2010,53,1:37

6The regulatory role of AT 1 receptor on activated HSCs in hepat,c fibrogenesis,effects of RAS inhibitors on hepatic fibrosis induced by CCl_4显示文摘AIM To assess the effect of ACE inhibitor andAng Ⅱ type Ⅰ(AT1)receptor antagonist inpreventing hepatic fibrosis caused by CCl4administration in rats;to investigate whether ornot there are expression of AT 1 receptors onhepatic stellate cells;and to observe the effectof Ang Ⅱ on proliferation and ECM synthesis ofcultured HSCs.METHODS Studies were conducted in maleSprague-Dawley rats.Except for thehepatofibrotic model group and the controlgroup,in three treated groups,either enalapril(5 mg/kg),or Iosartan(10 mg/kg),or enalapril+Iosartan were given to the fibrotic rats bydaily gavage,and saline vehicle was given tomodel and normal control rats.After 6 weeks,liver fibrosis was assessed directly by hepaticmorphometric analysis,which has beenconsidered the gold standard for thequantification of fibrosis.The expressions of AT1 receptors and(α-mooth muscle actin,α-SMA)in liver tissue or isolated hepatic stellate cells(HSCs)were detected by immunohistochemicaltechniques.The effect of Ang Ⅱ on HSCproliferation was determined by MTT method.Effect of Ang Ⅱ on collagen synthesis of HSCswas determined by 3H-proline incorporation.RESULTS Contrasted to the fibrosis in rats ofthe model group,groups of rats treated with either enalapril or Iosartan,or a combination oftwo drugs showed a limited expansion of theinterstitium(4.23±3.70 vs 11.22±4.79,P<0.05),but no difference was observedamong three treated groups(5.38±3.43,4.96±2.96,4.23±2.70,P>0.05).Expression of AT 1receptors was found in fibrotic interstitium offibrotic rats,whereas in normal control rats theywere limited to vasculature only to a very slightdegree.AT 1 receptors were also expressed onactivated HSCs in the culture.At concentrationsfrom 10-9to 10-5mol/L,Ang Ⅱ stimulated HSCproliferation in culture in a dose-dependentmanner.Increasing Ang Ⅱ concentrationsproduced corresponding increases in 3H-prolineincorporation.Differences among groups were significant.CONCLUSION Angiotensin-converting enzyme inhibitors and AT I blocker may slow the progression of hepatic fibrosis; activated HSCs express AT 1 receptors, and Ang Ⅱ can stimulate the proliferation and collagen synthesis of HSCs in a dose-dependent manner; and activation of RAS may be related to hepatic fibrogenesis induced by CCI4.Hong Shan Wei Han Ming Lu Ding Guo Li Yu Tao Zhan Zhi Rong Wang Xin Huang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,6:27

9NEW METHOD OF SIMULATING CONCENTRATED DRAIN HOLES IN SEEPAGE CONTROL ANALYSIS显示文摘In this paper, based upon the basic solution of sink, the approximate solution of single drain hole in finite elements is derived by use of the superposition principle. Then, the theoretical solution is extended to the case of some drain holes in one finite element, and the method is used in seepage control analysis with quick convergence and high accuracy. On the other hand, if the positions of the drain holes are changed, only some control factors of drain holes are changed, but the finite element grid need not to be reformed. 16Biomechanical and biological responses of periodontium in orthodontic tooth movement:up-date in a new decade显示文摘Nowadays,orthodontic treatment has become increasingly popular.However,the biological mechanisms of orthodontic tooth movement(OTM)have not been fully elucidated.We were aiming to summarize the evidences regarding the mechanisms of OTM.Firstly,we introduced the research models as a basis for further discussion of mechanisms.Secondly,we proposed a new hypothesis regarding the primary roles of periodontal ligament cells(PDLCs)and osteocytes involved in OTM mechanisms and summarized the biomechanical and biological responses of the periodontium in OTM through four steps,basically in OTM temporal sequences,as follows:(1)Extracellular mechanobiology of periodontium:biological,mechanical,and material changes of acellular components in periodontium under orthodontic forces were introduced.(2)Cell strain:the sensing,transduction,and regulation of mechanical stimuli in PDLCs and osteocytes.(3)Cell activation and differentiation:the activation and differentiation mechanisms of osteoblast and osteoclast,the force-induced sterile inflammation,and the communication networks consisting of sensors and effectors.(4)Tissue remodeling:the remodeling of bone and periodontal ligament(PDL)in the compression side and tension side responding to mechanical stimuli and root resorption.Lastly,we talked about the clinical implications of the updated OTM mechanisms,regarding optimal orthodontic force(OOF),acceleration of OTM,and prevention of root resorption.Yuan Li Qi Zhan Minyue Bao Jianru Yi Yu Li 2021International Journal of Oral Science2021,13,2:15
17Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. 18Effects of cotransplantated Schwann cells and neural stem cells in a rat model of Alzheimer's disease显示文摘Schwann cells (SCs) are significantly better at promoting neural stem cell (NSCs) proliferation, differentiation and synaptic formation when cocultured with NSCs in vitro, compared with cultured in a single nerve growth factor. The present study transplanted NSCs and SCs into the brain of a rat model of Alzheimer's disease to investigate the effect of cotransplantation. Results show transplantation of both NSCs alone and NSCs + SCs significantly promoted learning and memory functions in Alzheimer's disease rats, decreased glial fibrillary acidic protein and calcium binding protein S100β expression, but increased expression of the cholinergic neuron marker choline acetyl transferase mRNA. The effect of NSCs + SCs cotransplantation was, however, more significant. 19Study of parachute inflation process using fluid–structure interaction method显示文摘A direct numerical modeling method for parachute is proposed firstly, and a model for the star-shaped folded parachute with detailed structures is established. The simplified arbitrary Lagrangian–Eulerian fluid–structure interaction(SALE/FSI) method is used to simulate the inflation process of a folded parachute, and the flow field calculation is mainly based on operator splitting technique. By using this method, the dynamic variations of related parameters such as flow field and structure are obtained, and the load jump appearing at the end of initial inflation stage is captured. Numerical results including opening load, drag characteristics, swinging angle, etc. are well consistent with wind tunnel tests. In addition, this coupled method can get more space–time detailed information such as geometry shape, structure, motion, and flow field. 20The electromagnetic wave experiment for CSES mission: Search coil magnetometer显示文摘The seismic activities on the Earth can produce a disturbance of the electromagnetic field and particles in the ionosphere. The search coil magnetometer(SCM) mounted on China Seismo-Electromagnetic satellite(CSES) is designed to measure the magnetic field fluctuation of low frequency electromagnetic waves in the frequency range of 10 Hz–20 k Hz. The SCM comprises a three-axis search coil sensor mounted on a 4.5 m boom and an electronic box inside satellite module. The sampling rate of the SCM is 51.2 k Hz and the time resolution of the power spectrum density(PSD) is 2 s. The frequency resolution is 12.5 Hz.There are three operation modes: survey, detailed survey and calibration. In the survey mode, the SCM can provide a PSD in the whole frequency range of 10 Hz–20 k Hz and wave forms in the low frequency range below 2 k Hz while in the detailed survey mode the SCM can provide both PSD and wave forms in the whole frequency range of 10 Hz–20 k Hz. The sensitivity of the SCM instrument is 5.0×10^(-4) n T Hz^(-1/2) at 10 Hz, 5.0×10^(–5) n T Hz^(-1/2) at 200 Hz, 3.4×10^(-5) n T Hz^(-1/2) at 2 k Hz and 1.1×10^(-4) n T Hz^(-1/2) at 20 k Hz. The telemetry rate is ~0.85 Mbps in the survey mode and ~3.0 Mbps in the detailed survey mode. The phase difference between three axes can be made generally with a precision of less than 1.0°. The dynamic range of the SCM instrument is over 100 d B. The orthogonality of three mechanical axes of search coil senor is better than 0.13°.
