1Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis显示文摘Objective PERK/eI F2α/CHOP is a major signaling pathway mediating endoplasmic reticulum(ER) stress related with atherosclerosis.Oxidized LDL(ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis.The present study was conducted to explore the regulatory effect of ox-LDL on PERK/e IF2α/CHOP signaling pathway in vascular endothelial cells.Methods The effects of ox-LDL on PERK and p-e IF2α protein expression of primary human umbilical vein endothelial cells(HUVECs) were investigated by Western blot analysis.PERK gene silencing and selective eI F2α phosphatase inhibitor,salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis,caspase-3 activity,and CHOP mR NA level.Results Ox-LDL treatment significantly increased the expression of PERK,PERK-mediated inactivation of e IF2α phosphorylation,and the expression of CHOP,as well as the caspase-3 activity and apoptosis.The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral sh RNA or by selective eI F2α phosphatase inhibitor,salubrinal.Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eI F2α/CHOP ER-stress pathway.It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis.TAO Yong Kang YU Pu Lin BAI Yong Ping YAN Sheng Tao ZHAO Shui Ping ZHANG Guo Qiang 2016Biomedical and Environmental Sciences2016,29,12:21
2A structural view of the antibiotic degradation enzyme NDM-1 from a superbug显示文摘Gram-negative Enterobacteriaceae with resistance to carbapenem conferred by New Delhi metallo-β-lactamase 1(NDM-1)are a type of newly discovered antibioticresistant bacteria.The rapid pandemic spread of NDM-1 bacteria worldwide(spreading to India,Pakistan,Europe,America,and Chinese Taiwan)in less than 2 months characterizes these microbes as a potentially major global health problem.The drug resistance of NDM-1 bacteria is largely due to plasmids containing the blaNDM-1 gene shuttling through bacterial populations.The NDM-1 enzyme encoded by the blaNDM-1 gene hydrolyzes β-lactam antibiotics,allowing the bacteria to escape the action of antibiotics.Although the biological functions and structural features of NDM-1 have been proposed according to results from functional and structural investigation of its homologues,the precise molecular characteristics and mechanism of action of NDM-1 have not been clarified.Here,we report the threedimensional structure of NDM-1 with two catalytic zinc ions in its active site.Biological and mass spectroscopy results revealed that D-captopril can effectively inhibit the enzymatic activity of NDM-1 by binding to its active site with high binding affinity.The unique features concerning the primary sequence and structural conformation of the active site distinguish NDM-1 from other reported metallo-β-lactamases(MBLs)and implicate its role in wide spectrum drug resistance.We also discuss the molecular mechanism of NDM-1 action and its essential role in the pandemic of drug-resistant NDM-1 bacteria.Our results will provide helpful information for future drug discovery targeting drug resistance caused by NDM-1 and related metallo-β-lactamases.Yu Guo Jing Wang Guojun Niu Wenqing Shui Yuna Sun Honggang Zhou Yaozhou Zhang Cheng Yang Zhiyong Lou Zihe Rao 2011Protein & Cell2011,2,5:16

6EFFECT: an efficient flexible privacy-preserving data aggregation scheme with authentication in smart grid显示文摘Smart grid is considered as a promising approach to solve the problems of carbon emission and energy crisis. In smart grid, the power consumption data are collected to optimize the energy utilization.However, security issues in communications still present practical concerns. To cope with these challenges, we propose EFFECT, an efficient flexible privacy-preserving aggregation scheme with authentication in smart grid. Specifically, in the proposed scheme, we achieve both data source authentication and data aggregation in high efficiency. 7Boosting electrocatalytic water splitting via metal-metalloid combined modulation in quaternary Ni-Fe-P-B amorphous compound显示文摘Design and synthesis of highly efficient and cost-effective bifunctional catalysts for hydrogen evolution reaction(HER)and oxygen evolution reaction(OER)remain a big challenge.Herein,a quaternary amorphous nanocompound Ni-Fe-P-B has been synthesized by a facile,scalable co-reduction method.The Ni-Fe-P-B exhibits high electrocatalytic activity and outstanding durability for both HER and OER,delivering a current density of 10 mA·cm^-2 at overpotentials of 220 and 269 mV,respectively.When loaded on carbon fiber paper(CFP)as a bifunctional catalyst,the Ni-Fe-P-B@CFP electrode requires a low cell voltage of 1.58 V to obtain 10 mA·cm^-2 for overall water splitting with negligible recession over 60 h.The excellent catalytic performances of Ni-Fe-P-B mainly benefit from the metal-metalloid combined composition modulation and the unique amorphous structure.This work provides new insights into the design of robust bifunctional catalysts for water splitting,and may promote the development of multicomponent amorphous catalysts.Wukui Tang Xiaofang Liu Ya Li Yanhui Pu Yao Lu Zhiming Song Qiang Wang Ronghai Yu Jianglan Shui 2020Nano Research2020,13,2:8

9Predictive value of K-ras and PIK3CA in non-small cell lung cancer patients treated with EGFR-TKIs: a systemic review and meta-analysis显示文摘Objective: A meta-analysis was performed to augment the insufficient data on the impact of mutative EGFR downstream phosphatidylinositol-3-kinase(PI3K) and mitogen-activated protein kinase(MAPK) pathways on the clinical efficiency of epidermal growth factor receptor tyrosine kinase inhibitor(EGFR-TKI) treatment of non-small cell lung cancer(NSCLC) patients.Methods: Network databases were explored in April, 2015. Papers that investigated the clinical outcomes of NSCLC patients treated with EGFR-TKIs according to the status of K-ras and/or PIK3 CA gene mutation were included. A quantitative meta-analysis was conducted using standard statistical methods. Odds ratios(ORs) for objective response rate(ORR) and hazard ratios(HRs) for progression-free survival(PFS) and overall survival(OS) were calculated.Results: Mutation in K-ras significantly predicted poor ORR [OR =0.22; 95% confidence interval(CI), 0.13-0.35], shorter PFS(HR =1.56; 95% CI, 1.27-1.92), and shorter OS(HR =1.59; 95% CI, 1.33-1.91) in NSCLC patients treated with EGFR-TKIs. Mutant PIK3 CA significantly predicted shorter OS(HR =1.83; 95% CI, 1.05-3.20), showed poor ORR(OR =0.70; 95% CI, 0.22-2.18), and shorter PFS(HR =1.79; 95% CI, 0.91-3.53) in NSCLC patients treated with EGFR-TKIs.Conclusion: K-ras mutation adversely affected the clinical response and survival of NSCLC patients treated with EGFRTKIs. PIK3 CA mutation showed similar trends. 10Estrogen receptor-low breast cancer:Biology chaos and treatment paradox显示文摘Hormone receptor testing mainly serves the purpose of guiding treatment choices for breast cancer patients.Patients with estrogen receptor(ER)-positive breast cancers show significant response to endocrine therapy.However,the methods to define ER status and eligibility for treatment remain controversial.Despite recent guidelines considering staining≥1% of tumor nuclei by immunohistology as ER-positive,it has raised concerns on the benefit of endocrine therapy for tumors with ER 1%-10% expression,termed“ER-low positive”.This subgroup accounts for 3% to 9% of all patients and is likely to have unique molecular features,and therefore distinct therapeutic response to endocrine therapy compared with ER-high positive tumors.The latest guidelines did not provide detailed descriptions for those patients,resulting in inconsistent treatment strategies.Consequently,we aimed to resolve this dilemma comprehensively.This review discusses molecular traits and recent ER-low positive breast cancer innovations,highlighting molecular-targeted treatment rather than traditional unified endocrine therapy for future basic and clinical research.Ke-Da Yu Yu-Wen Cai Song-Yang Wu Ruo-Hong Shui Zhi-Ming Shao 2021Cancer Communications2021,41,10:6
11Environmentally Tough and Stretchable MXene Organohydrogel with Exceptionally Enhanced Electromagnetic Interference Shielding Performances显示文摘Conductive hydrogels have potential applications in shielding electromagnetic(EM)radiation interference in deformable and wearable electronic devices,but usually suffer from poor environmental stability and stretching-induced shielding performance degradation.Although organohydrogels can improve the environmental stability of materials,their development is at the expense of reducing electrical conductivity and thus weakening EM interference shielding ability.Here,a MXene organohydrogel is prepared which is composed of MXene network for electron conduction,binary solvent channels for ion conduction,and abundant solvent-polymer-MXene interfaces for EM wave scattering.This organohydrogel possesses excellent anti-drying ability,low-temperature tolerance,stretchability,shape adaptability,adhesion and rapid self-healing ability.Two effective strategies have been proposed to solve the problems of current organohydrogel shielding materials.By reasonably controlling the MXene content and the glycerol-water ratio in the gel,MXene organohydrogel can exhibit exceptionally enhanced EM interference shielding performances compared to MXene hydrogel due to the increased physical cross-linking density of the gel.Moreover,MXene organohydrogel shows attractive stretching-enhanced interference effectiveness,caused by the connection and parallel arrangement of MXene nanosheets.This well-designed MXene organohydrogel has potential applications in shielding EM interference in deformable and wearable electronic devices.Yuanhang Yu Peng Yi Wenbin Xu Xin Sun Gao Deng Xiaofang Liu Jianglan Shui Ronghai Yu 2022Nano-Micro Letters2022,14,5:6

13Microwave ablation of primary breast cancer inhibits metastatic progression in model mice via activation of natural killer cells显示文摘Surgery is essential for controlling the symptoms and complications of stage IV breast cancer.However,locoregional treatment of primary tumors often results in distant progression,including lung metastasis,the most common type of visceral metastasis.As a minimally invasive thermal therapy,microwave ablation(MWA)has been attempted in the treatment of breast cancer,but the innate immune response after MWA has not yet been reported.Using two murine models of stage IV breast cancer,we found that MWA of primary breast cancer inhibited the progression of lung metastasis and improved survival.NK cells were activated after MWA of the primary tumor and exhibited enhanced cytotoxic functions,and the cytotoxic pathways of NK cells were activated.Depletion experiments showed that NK cells but not CD4+or CD8+T cells played a pivotal role in prolonging survival.Then,we found that compared with surgery or control treatment,MWA of the primary tumor induced completely different NK-cell-related cytokine profiles.Macrophages were activated after MWA of the primary tumor and produced IL-15 that activated NK cells to inhibit the progression of metastasis.In addition,MWA of human breast cancer stimulated an autologous NK-cell response.These results demonstrate that MWA of the primary tumor in metastatic breast cancer inhibits metastatic progression via the macrophage/IL-15/NK-cell axis.MWA of the primary tumor may be a promising treatment strategy for de novo stage IV breast cancer,although further substantiation is essential for clinical testing.Muxin Yu Hong Pan Nan Che Li Li Cong Wang Yue Wang Ge Ma Mengjia Qian Jiawei Liu Mingjie Zheng Hui Xie Lijun Ling Yi Zhao Xiaoxiang Guan Qiang Ding Wenbin Zhou Shui Wang 2021Cellular & Molecular Immunology2021,18,9:5
14Elevated Platelet Activating Factor Level in Ischemia-Related Arrhythmia and Its Electrophysiological Effect on Myocardium显示文摘Objective The mechanism through which platelet activating factor (PAF) induces cardiac electrical activity and arrhythmia is not well understood and previous studies have suggested a potential involvement of ion channels in its action. The present study was aimed to clarify the role of PAF in fatal arrhythmias following acute myocardia infarction (AMI) and the underlying mechanism. Methods (1) Blood PAF levels were measured among 72 AMI patients at the time of diagnosis with AMI and 48 h later, and their electrocardiogram (ECG) was recorded continuously. (2) Ischemia simulation and surface electrocardiogram were conducted in 20 pigs and their PAF levels were measured. (3) PAF perfusion and standard microelectrode recording were performed on guinea pig papillarymuscles. Results In both humans and pigs, elevated PAF levels were detected in AMI and simulated ischemia, respectively, and even higher PAF levels were found when fatal arrhythmias occurred. In guinea pig myocardium, PAF induced a shortening of action potential duration at 90% level of repolarization (APD 90 )under non-ischemic conditions and a more pronounced shortening under early simulated ischemic conditions. Conclusion AMI and ischemia are associated with increased PAF levels in humans and pigs, which are further raised when fatal arrhythmia follows. 17Synergy between metallic components of MoNi alloy for catalyzing highly efficient hydrogen storage of MgH2显示文摘Catalysts play a critical role in improving the hydrogen storage kinetics in Mg/MgH2 system.Exploring highly efficient catalysts and catalyst design principles are hot topics but challenging.The catalytic activity of metallic elements on dehydrogenation kinetics generally follows a sequence of Ti>Nb>Ni>V>Co>Mo.Herein,we report a highly efficient alloy catalyst composed of low-active elements of Mo and Ni(i.e.MoNi alloy)for MgH2 particles.MoNi alloy nanoparticles show excellent catalytic effect,even outperforming most advanced Ti-based catalysts.The synergy between Mo and Ni elements can promote the break of Mg-H bonds and the dissociation of hydrogen molecules,thus significantly improves the kinetics of Mg/MgH2 system.The MoNi-catalyzed Mg/MgH2 system can absorb and release 6.7 wt.%hydrogen within 60 s and 10 min at 300℃,respectively,and exhibits excellent cycling stability and low-temperature hydrogen storage performance.This study provides a strategy for designing efficient catalysts for hydrogen storage materials using the synergy of metal elements.Meng Chen Yanhui Pu Zhenyang Li Gang Huang Xiaofang Liu Yao Lu Wukui Tang Li Xu Shuangyu Liu Ronghai Yu Jianglan Shui 2020Nano Research2020,13,8:5

19Fast-Suppressor Screening for New Components in Protein Trafficking,Organelle Biogenesis and Silencing Pathway in Arabidopsis thaliana Using DEX-Inducible FREE1-RNAi Plants显示文摘Membrane trafficking is essential for plant growth and responses to external signals.The plant unique FYVE domain-containing protein FREE1 is a component of the ESCRT complex(endosomal sorting complex required for transport).FREE1 plays multiple roles in regulating protein trafficking and organelle biogenesis including the formation of intraluminal vesicles of multivesicular body(MVB),vacuolar protein transport and vacuole biogenesis,and autophagic degradation.FREE1 knockout plants show defective MVB formation,abnormal vacuolar transport,fragmented vacuoles,accumulated autophagosomes,and seedling lethality.To further uncover the underlying mechanisms of FREE1 function in plants,we performed a forward genetic screen for mutants that suppressed the seedling lethal phenotype of FREE1-RNAi transgenic plants.The obtained mutants are termed as suppressors of free1(sof).To date,229 putative sof mutants have been identified.Barely detecting of FREE1 protein with M3plants further identified 84 FREE1-related suppressors.Also145 mutants showing no reduction of FREE1 protein were termed as RNAi-related mutants.Through next-generation sequencing(NGS)of bulked DNA from F2mapping population of two RNAi-related sof mutants,FREE1-RNAi T-DNA inserted on chromosome 1 was identified and the causal mutation of putative sof mutant is being identified similarly.These FREE1-and RNAi-related sof mutants will be useful tools and resources for illustrating the underlying mechanisms of FREE1 function in intracellular trafficking and organelle biogenesis,as well as for uncovering the new components involved in the regulation of silencing pathways in plants.Qiong Zhao Caiji Gao PoShing Lee Lin Liu Shaofang Li Tangjin Hu Jinbo Shen Shuying Pan Hao Ye Yunru Chen Wenhan Cao Yong Cui Peng Zeng Sheng Yu Yangbin Gao Liang Chen Beixin Mo Xin Liu Shi Xiao Yunde Zhao Silin Zhong Xuemei Chen Liwen Jiang 2015Journal of Genetics and Genomics2015,42,6:4
20Synthesis and characterization of magnesium hydroxide by batch reaction crystallization显示文摘Magnesium hydroxide with high purity and uniform particle size distribution was synthesized by the direct precipitation method using MgCl_(2) and NaOH as reactive materials and NaCl as additive to improve the crystallization behavior of the product.The particle size distribution,crystal phase,morphology,and surface area of magnesium hydroxide were characterized by Malvern laser particle size analyzer,X-ray diffraction(XRD),scanning electron microscope(SEM)and Branauer-Emmett-Teller(BET)method,respectively.The purity of products was analyzed by the chemical method.The effects of synthesis conditions on the particle size distribution and water content(filtration cake)of magnesium hydroxide were investigated.The results indicated that feeding mode and rate,and reaction temperature had important effects on water content and the particle size distribution of the product,and sodium chloride improved the crystallization behavior of magnesium hydroxide.The ball-like magne-sium hydroxides with the particle size distribution of 6.0–30.0μm and purity higher than 99.0%were obtained.This simple and mild synthesis method was promising to be scaled up for the industrial production of magnesium hydroxide.Xingfu SONG Shuying SUN Dengke ZHANG Jin WANG Jianguo YU 2011Frontiers of Chemical Science and Engineering2011,5,4:4
